The objective of the study was to verify the relationships between sport skill levels and to identify the tests that accurately diagnose flexibility of sport climbers. Project description:Masticatory muscle activity during teeth clenching is associated with changes in many physiological parameters throughout the body. Clenching can improve muscle activity, force production, rate of force development, and joint fixation. Hence, teeth clenching and masticatory muscle activity can be important in competitive sports activities. Sport climbing is becoming increasingly popular and will be included for the first time in the Summer Olympic Games, Tokyo, 2020. However, masticatory muscle activity in sport climbers has not yet been studied. The aim of the presented study is to compare the bioelectrical activity of the masticatory muscles in sport climbers and non-climbers in order to determine the relationship between these muscles and climbing activity. 44 subjects without masticatory system disorders (16 women and 28 men, average age 26.3) were divided into two groups of 22 sport climbers (8 women, 14 men, climbing experience >4 years), while 22 people (8 women, 14 men, with no regular sports activity) were assigned to the control group. Electromyographic examination of temporalis anterior (TA) and masseter muscle (MM) was evaluated in three conditions: during resting mandibular position, during maximum intercuspation clenching, and during maximum voluntary clenching with cotton rolls between teeth. For statistical analysis, the W Shapiro-Wilk test and the Mann-Whitney U test were used. Sport climbers showed significantly higher bioelectrical activities of MM during maximum intercuspation clenching (238.45 ?V vs. 83.87 ?V, p = 0.002), and during maximum voluntary clenching with cotton rolls between teeth (300.01 ?V vs. 101.38 ?V, p = 0.001) compared to controls. The differences between groups in relation to the resting bioelectrical activity of the MM muscles, and TA muscles in all conditions were not statistically significant (p > 0.05). Higher bioelectrical activity of masseter muscles during clenching in climbers can be associated with this sports activity. However, the mechanism remains unknown and requires future research.

Project description:Urine PCR has been used for the diagnosis of Borrelia burgdorferi infection in recent years but has been abandoned because of its low sensitivity and the irreproducibility of the results. Our study aimed to analyze technical details related to sample preparation and detection methods. Crucial for a successful urine PCR were (i) avoidance of the first morning urine sample; (ii) centrifugation at 36,000 x g; and (iii) the extraction method, with only DNAzol of the seven different extraction methods used yielding positive results with patient urine specimens. Furthermore, storage of frozen urine samples at -80 degrees C reduced the sensitivity of a positive urine PCR result obtained with samples from 72 untreated erythema migrans (EM) patients from 85% in the first 3 months to <30% after more than 3 months. Bands were detected at 276 bp on ethidium bromide-stained agarose gels after amplification by a nested PCR. The specificity of bands for 32 of 33 samples was proven by hybridization with a GEN-ETI-K-DEIA kit and for a 10 further positive amplicons by sequencing. By using all of these steps to optimize the urine PCR technique, B. burgdorferi infection could be diagnosed by using urine samples from EM patients with a sensitivity (85%) substantially better than that of serological methods (50%). This improved method could be of future importance as an additional laboratory technique for the diagnosis of unclear, unrecognized borrelia infections and diseases possibly related to Lyme borreliosis.

Project description:Background: Sport climbing places high mechanical loads on fingers. In 2012, our research group demonstrated adaptations of climbers’ cortical bones with the presence of osteophytes compared to non-climbing controls. Objectives: 1) To investigate 10-year changes in cortical bone thickness, base osteophyte occurrence and radiological signs of osteoarthritis in the fingers of elite male sport climbers with more than 25 years of climbing history and 2) to compare cortical bone thickness, base osteophyte occurrence and radiological signs of osteoarthritis between male sport climbers and age-matched controls at the 10-year follow-up. Methods: All 31 elite sport climbers who participated in both the baseline and 10-year follow-up assessments (follow-up rate 100%) were examined by means of X-rays. Cortical bone thickness, presence of osteophytes and signs of osteoarthritis according to Kellgren-Lawrence were obtained and compared to the baseline values 10 years earlier and to age-matched controls at the follow-up (n = 15). Results: Significantly increased cortical bone thickness over the past 10 years was observed in climbers (mean absolute difference with 95% CI:0.98 mm (0.77 mm, 1.19 mm); p <0.001). Moreover, compared to age-matched controls, climbers had significantly thicker cortical bone at the 10-year follow-up (mean absolute difference with 95% CI:0.86 mm (0.61 mm, 1.12 mm); p <0.001). In climbers, osteophytes and clear signs of osteoarthritis were mainly seen in DIP joints. Signs of osteoarthritis according to Kellgren-Lawrence were more prevalent than 10 years before in most joints. In lateral radiographs, base osteophytes were not significantly more prevalent than 10 years before in most of the joints. The percentage of climbers who had osteophytes in any DIP (PIP) joint increased from 93.5% (67.7%) at baseline to 100% (74.2%) at the 10-year follow-up. The percentage of climbers who had clear signs of osteoarthritis according to Kellgren-Lawrence in any DIP (PIP) joint increased from 12.9% (9.7%) at baseline to 74.2% (64.5%) at 10-year follow-up. Only a few such degenerative changes were found in age-matched controls. Conclusion: An accumulation of repetitive climbing-related stress to the fingers of elite sport climbers over the career may induce lifelong mechano-adaptation of the cortical bone thickness of all phalanges. At the 10-year follow-up, a further significant increase in radiographic signs of osteoarthritic changes was observed.

Project description:Nicotinamide adenine dinucleotide (NAD+) is an essential coenzyme regulating cellular energy metabolism across all species. Recent studies have shown the pleiotropic roles of adipose tissue NAD+ biology in adipose tissue and whole-body metabolism. The major purpose of the present study was to test the hypothesis that adipose tissue NAD+ biosynthesis, mediated by nicotinamide phosphoribosyltransferase (NAMPT), is a physiological regulator of whole-body metabolic flexibility. To this end, we analyzed adipocyte-specific Nampt knockout (ANKO) mice and subcutaneous white adipose tissue (WAT) biopsy samples obtained from human subjects. We found ANKO mice preferably utilized glucose substrate during the light period or after prolonged fasting. In contrast, ANKO mice had marked decreases in stimulation of glucose oxidation and suppression of fat oxidation during the postprandial status despite hyperinsulinemia. Data obtained from RNA-sequencing of WAT suggest that loss of NAMPT causes inflammation, oxidative stress, defective fatty acid oxidation, and mitochondrial dysfunction in WAT, key features of obesity and metabolic inflexibility. We also found WAT NAD+ concentration was decreased in people with obesity and insulin resistance compared with those who were non-obese and insulin-sensitive. In addition, bariatric surgery-induced 20% weight loss increased WAT NAD+ concentration in people with obesity. Taken together, our results reveal the importance of adipose tissue NAMPT-mediated NAD+ biosynthesis in regulating whole-body metabolic flexibility. Our results also provide translational and clinical insight into adipose tissue NAD+ biology in obesity and whole-body metabolic health.

Project description:PurposeThe critical force (CF) concept, differentiating steady and non-steady state conditions, extends the critical power paradigm for sport climbing. This study aimed to validate CF for finger flexors derived from the 4 min all-out test as a boundary for the highest sustainable work intensity in sport climbers.MethodsTwelve participants underwent multiple laboratory visits. Initially, they performed the 4 min intermittent contraction all-out test for CF determination. Subsequent verification visits involved finger-flexor contractions at various intensities, including CF, CF -2 kg, CF -4 kg, and CF -6 kg, lasting for 720 s or until failure, while monitoring muscle-oxygen dynamics of forearm muscles.ResultsCF, determined from the mean force of last three contractions, was measured at 20.1 ± 5.7 kg, while the end-force at 16.8 ± 5.2 kg. In the verification trials, the mean time to failure at CF was 440 ± 140 s, with only one participant completing the 720 s task. When the load was continuously lowered (-2 kg, -4 kg, and -6 kg), a greater number of participants (38%, 69%, and 92%, respectively) successfully completed the 720 s task. Changes of muscle-oxygen dynamics showed a high variability and could not clearly distinguish between exhaustive and non-exhaustive trials.ConclusionsCF, based on the mean force of the last three contractions, failed to reliably predict the highest sustainable work rate. In contrast, determining CF as the end-force of the last three contractions exhibited a stronger link to sustainable work. Caution is advised in interpreting forearm muscle-oxygen dynamics, lacking sensitivity for nuanced metabolic responses during climbing-related tasks.

Project description:BackgroundIndoor climbing involves overloading the shoulder girdle, including the rotator cuff and upper trapezius muscles. This on the field study aimed to investigate the effects of repeated climbing bouts on morphological and mechanical measures of the upper trapezius muscle.Materials and methodsFifteen experienced male climbers participated in the study. Rate of perceived exertion (RPE), blood lactate concentration ([La-]b), and stiffness and thickness over four points of the upper trapezius were assessed before and after a repeated climbing exercise. The procedure for the climbing exercise consisted of five climbs for a total time of 5-minutes per climb, followed by a 5-minute rest.ResultsThe analysis showed an increase from baseline to after the 3rd climb (p ≤ 0.01) for RPE and after the 5th climb for [La-]b (p ≤ 0.001). Muscle stiffness and thickness increased at all points (1-2-3-4) after the 5th climb (p ≤ 0.01). We found spatial heterogeneity in muscle stiffness and thickness; muscle stiffness was the highest at Point 4 (p ≤ 0.01), while muscle thickness reached the highest values at points 1-2 (both p ≤ 0.01). Moreover, the analysis between the dominant and non-dominant shoulder showed greater stiffness after the 1st climb at Point 1 (p = 0.004) and after the 5th climb at Point 4 (p ≤ 0.001).ConclusionsFor muscle thickness, the analysis showed significant changes in time and location between the dominant and the non-dominant shoulder. Bilateral increases in upper trapezius muscle stiffness and thickness, with simultaneous increases in RPE and blood lactate in response to consecutive climbs eliciting fatigue.

Project description:Background and objective: The use of suboptimal weight loss strategies in order to reach specific weight ranges as observed in combat sport disciplines can give rise to severe health problems. However, particular aspects regarding management of weight category comparing three sport disciplines remain to be investigated. Therefore, the aim of the present study was to obtain information regarding the weight loss strategies that competitors performed before a tournament. Materials and Methods: This article describes the most common dietary-nutritional strategies used by 140 national university male competitors of judo (n = 52), karate (n = 40) and taekwondo (n = 48) in order to achieve a specific weight, according to the rapid weight loss questionnaire (RWLQ) and the EAT-27 questionnaire. Results: Around 50% of participants were not involved in a weight loss process. Among the remaining participants, we considered three periods for weight reduction: less than 1 week (35% in judo, 8% in karate and 19% in taekwondo), less than 1 month (17% in judo, 15% in karate and 26% in taekwondo) and more than 1 month (0% in judo, 5% in karate and 21% in taekwondo). Severe fasting, focused on food/water restriction, was the most commonly used strategy, being more frequent in judo players. Light weight judo practitioners generally lost 2-5 kg before the contest. One third of participants avoided carbohydrate consumption when performing food restriction. Finally, individuals that reduced weight in the last week seemed to develop an unhealthy psychological relationship with food. Conclusion: All these aspects could be particularly relevant, providing information regarding how competitors manage basic nutritional concepts that guide dieting strategies. This information is relevant to prepare future educational interventions in the area of nutrition for competitors, coaches and technical staff.

Project description:The purpose of this study was to compare the frequency of COL1A1 rs1107946 polymorphism between sport climbers and controls from three ethnic groups (Japanese, Polish, and Russian) and investigate the effect of the COL1A1 rs1107946 polymorphism on the age-related decrease in flexibility in the general population. Study I consisted of 1929 healthy people (controls) and 218 climbers, including Japanese, Polish, and Russian participants. The results of the meta-analysis showed that the frequency of the AC genotype was higher in climbers than in the controls (p = 0.03). Study II involved 1093 healthy Japanese individuals (435 men and 658 women). Flexibility was assessed using a sit-and-reach test. There was a tendency towards association between sit-and-reach and the COL1A1 rs1107946 polymorphism (genotype: p = 0.034; dominant: p = 0.435; recessive: p = 0.035; over-dominant: p = 0.026). In addition, there was a higher negative correlation between sit-and-reach and age in the AA + CC genotype than in the AC genotype (AA + CC: r = −0.216, p < 0.001; AC: r = −0.089, p = 0.04; interaction p = 0.037). However, none of these results survived correction for multiple testing. Further studies are warranted to investigate the association between the COL1A1 gene variation and exercise-related phenotypes.

Project description:Nicotinamide adenine dinucleotide (NAD+) is an essential coenzyme that regulates cellular energy metabolism in many cell types. The major purpose of the present study was to test the hypothesis that NAD+ in white adipose tissue (WAT) is a regulator of whole-body metabolic flexibility in response to changes in insulin sensitivity and with respect to substrate availability and use during feeding and fasting conditions. To this end, we first evaluated the relationship between WAT NAD+ concentration and metabolic flexibility in mice and humans. We found that WAT NAD+ concentration was increased in mice after calorie restriction and exercise, 2 enhancers of metabolic flexibility. Bariatric surgery-induced 20% weight loss increased plasma adiponectin concentration, skeletal muscle insulin sensitivity, and WAT NAD+ concentration in people with obesity. We next analyzed adipocyte-specific nicotinamide phosphoribosyltransferase (Nampt) knockout (ANKO) mice, which have markedly decreased NAD+ concentrations in WAT. ANKO mice oxidized more glucose during the light period and after fasting than control mice. In contrast, the normal postprandial stimulation of glucose oxidation and suppression of fat oxidation were impaired in ANKO mice. Data obtained from RNA-sequencing of WAT suggest that loss of NAMPT increases inflammation, and impairs insulin sensitivity, glucose oxidation, lipolysis, branched-chain amino acid catabolism, and mitochondrial function in WAT, which are features of metabolic inflexibility. These results demonstrate a novel function of WAT NAMPT-mediated NAD+ biosynthesis in regulating whole-body metabolic flexibility, and provide new insights into the role of adipose tissue NAD+ biology in metabolic health.

Project description:The work reported here aims toward the optimization of electrode preparation methodologies for superior performance of supercapacitors through a rigorous understanding of underlying physical parameters. Oxygen-functionalized few-layer graphene was employed as an active material while binders [Nafion, polyvinylidene fluoride (PVDF), and polytetrafluoroethylene], solvents for active material dispersion [ethylene glycol and N-methyl-2-pyrrolidone (NMP)], and electrode-drying temperatures (100, 170, and 190 °C) were varied. Maximum specific capacitances at different electrode preparation conditions ranged from 240 to 318 F g-1 at 1 mV s-1 scan rate of cyclic voltammetry for the same active material. The study revealed that the electrodes prepared using the PVDF binder, the NMP solvent for active material dispersion, 170 °C electrode-drying temperature (slightly below the boiling temperature of the solvent) provided the best electrochemical performance. Electrochemical impedance spectroscopy revealed that the resistance for electron transfer at the electrode/electrolyte interface can be minimized while mass transport and pseudocapacitive charging can be improved significantly by tuning electrode preparation methodologies which resulted in smaller time constants and hence better capacitor performances. Scanning electron microscopy images revealed that graphene layers were properly stacked much similar to the synthesized nanomaterial wherein better electrochemical performances were achieved, avoiding the agglomeration of nanomaterials on the electrode surface. Low viscosity of the solvent for active material dispersion and better solubility of the binder in the solvent helped to reduce the agglomeration of nanomaterials by minimizing the strong van der Waals interaction which causes agglomeration.
