A New Model for Predicting Rate of Penetration Using an Artificial Neural Network.

ABSTRACT: The drilling rate of penetration (ROP) is defined as the speed of drilling through rock under the bit. ROP is affected by different interconnected factors, which makes it very difficult to infer the mutual effect of each individual parameter. A robust ROP is required to understand the complexity of the drilling process. Therefore, an artificial neural network (ANN) is used to predict ROP and capture the effect of the changes in the drilling parameters. Field data (4525 points) from three vertical onshore wells drilled in the same formation using the same conventional bottom hole assembly were used to train, test, and validate the ANN model. Data from Well A (1528 points) were utilized to train and test the model with a 70/30 data ratio. Data from Well B and Well C were used to test the model. An empirical equation was derived based on the weights and biases of the optimized ANN model and compared with four ROP models using the data set of Well C. The developed ANN model accurately predicted the ROP with a correlation coefficient (R) of 0.94 and an average absolute percentage error (AAPE) of 8.6%. The developed ANN model outperformed four existing models with the lowest AAPE and highest R value.

Project description:BackgroundThe performance of previously published glomerular filtration rate (GFR) estimation equations degrades when directly used in Chinese population. We incorporated more independent variables and using complicated non-linear modeling technology (artificial neural network, ANN) to develop a more accurate GFR estimation model for Chinese population.MethodsThe enrolled participants came from the Third Affiliated Hospital of Sun Yat-sen University, China from Jan 2012 to Jun 2016. Participants with age < 18, unstable kidney function, taking trimethoprim or cimetidine, or receiving dialysis were excluded. Among the finally enrolled 1952 participants, 1075 participants (55.07%) from Jan 2012 to Dec 2014 were assigned as the development data whereas 877 participants (44.93%) from Jan 2015 to Jun 2016 as the internal validation data. We in total developed 3 GFR estimation models: a 4-variable revised CKD-EPI (chronic kidney disease epidemiology collaboration) equation (standardized serum creatinine and cystatin C, age and gender), a 9-variable revised CKD-EPI equation (additional auxiliary variables: body mass index, blood urea nitrogen, albumin, uric acid and hemoglobin), and a 9-variable ANN model.ResultsCompared with the 4-variable equation, the 9-variable equation could not achieve superior performance in the internal validation data (mean of difference: 5.00 [3.82, 6.54] vs 4.67 [3.55, 5.90], P = 0.5; interquartile range (IQR) of difference: 18.91 [17.43, 20.48] vs 20.11 [18.46, 21.80], P = 0.05; P30: 76.6% [73.7%, 79.5%] vs 75.8% [72.9%, 78.6%], P = 0.4), but the 9-variable ANN model significantly improve bias and P30 accuracy (mean of difference: 2.77 [1.82, 4.10], P = 0.007; IQR: 19.33 [17.77, 21.17], P = 0.3; P30: 80.0% [77.4%, 82.7%], P < 0.001).ConclusionsIt is suggested that using complicated non-linear models like ANN could fully utilize the predictive ability of the independent variables, and then finally achieve a superior GFR estimation model.

Project description:BackgroundAccurate evaluation of glomerular filtration rates (GFRs) is of critical importance in clinical practice. A previous study showed that models based on artificial neural networks (ANNs) could achieve a better performance than traditional equations. However, large-sample cross-sectional surveys have not resolved questions about ANN performance.MethodsA total of 1,180 patients that had chronic kidney disease (CKD) were enrolled in the development data set, the internal validation data set and the external validation data set. Additional 222 patients that were admitted to two independent institutions were externally validated. Several ANNs were constructed and finally a Back Propagation network optimized by a genetic algorithm (GABP network) was chosen as a superior model, which included six input variables; i.e., serum creatinine, serum urea nitrogen, age, height, weight and gender, and estimated GFR as the one output variable. Performance was then compared with the Cockcroft-Gault equation, the MDRD equations and the CKD-EPI equation.ResultsIn the external validation data set, Bland-Altman analysis demonstrated that the precision of the six-variable GABP network was the highest among all of the estimation models; i.e., 46.7 ml/min/1.73 m(2) vs. a range from 71.3 to 101.7 ml/min/1.73 m(2), allowing improvement in accuracy (15% accuracy, 49.0%; 30% accuracy, 75.1%; 50% accuracy, 90.5% [P<0.001 for all]) and CKD stage classification (misclassification rate of CKD stage, 32.4% vs. a range from 47.3% to 53.3% [P<0.001 for all]). Furthermore, in the additional external validation data set, precision and accuracy were improved by the six-variable GABP network.ConclusionsA new ANN model (the six-variable GABP network) for CKD patients was developed that could provide a simple, more accurate and reliable means for the estimation of GFR and stage of CKD than traditional equations. Further validations are needed to assess the ability of the ANN model in diverse populations.

Project description:We developed a method to apply artificial neural networks (ANNs) for predicting time-series pharmacokinetics (PKs), and an interpretable the ANN-PK model, which can explain the evidence of prediction by applying Shapley additive explanations (SHAP). A previous population PK (PopPK) model of cyclosporin A was used as the comparison model. The patients' data were used for the ANN-PK model input, and the output by ANN was the clearance (CL). The estimated CL value from the ANN were substituted into the one-compartment with one-order absorption model, the concentrations were calculated, and the parameters of ANN were updated by the back-propagation method. Kernel SHAP was applied to the trained model and the SHAP value of each input was calculated. The root mean squared error for the PopPK model and the ANN-PK model were 41.1 and 31.0 ng/ml, respectively. The goodness of fit plots for the ANN-PK model represented more convergence to y = x compared with that for the PopPK model, with good model performance for the ANN-PK model. The most influential factors on CL output were age and body weight from the evaluation using Kernel SHAP, and these factors were incorporated into the PopPK model as the significant covariates of CL. The ANN-PK model could handle time-series data and showed higher prediction accuracy then the conventional PopPK model, and the scientific validity for the model could be evaluated by applying SHAP. A black-box property of an artificial neural network (ANN) decreases the scientific confidence of the model, and making it difficult to utilize the ANN in the medical field. Moreover, difficulty in handling the time-series data is a significant problem for applying the ANN for pharmacometrics study. WHAT QUESTION DID THIS STUDY ADDRESS? How can we apply the ANN for predicting the time-series pharmacokinetics (PKs) , and confirm the scientific validity of the ANN model? WHAT DOES THIS STUDY ADD TO OUR KNOWLEDGE? Using the ANN in combination with a conventional compartment (ANN-PK) model enabled to handle the time-series PK data, and the predicting performance of the model was higher than that of the population PK model. Furthermore, we could evaluate the scientific validity of the ANN model by applying the Shapley additive explanations. HOW MIGHT THIS CHANGE DRUG DISCOVERY, DEVELOPMENT, AND/OR THERAPEUTICS? We expect that our study will contribute to develop the interpretable ANN model, which can predict the time-series PKs, drug efficacies, and side effects with high prediction performance.
