Project description:In linear regression models with high dimensional data, the classical z-test (or t-test) for testing the significance of each single regression coefficient is no longer applicable. This is mainly because the number of covariates exceeds the sample size. In this paper, we propose a simple and novel alternative by introducing the Correlated Predictors Screening (CPS) method to control for predictors that are highly correlated with the target covariate. Accordingly, the classical ordinary least squares approach can be employed to estimate the regression coefficient associated with the target covariate. In addition, we demonstrate that the resulting estimator is consistent and asymptotically normal even if the random errors are heteroscedastic. This enables us to apply the z-test to assess the significance of each covariate. Based on the p-value obtained from testing the significance of each covariate, we further conduct multiple hypothesis testing by controlling the false discovery rate at the nominal level. Then, we show that the multiple hypothesis testing achieves consistent model selection. Simulation studies and empirical examples are presented to illustrate the finite sample performance and the usefulness of the proposed method, respectively.

Project description:The aim of this paper is to develop a low-rank linear regression model (L2RM) to correlate a high-dimensional response matrix with a high dimensional vector of covariates when coefficient matrices have low-rank structures. We propose a fast and efficient screening procedure based on the spectral norm of each coefficient matrix in order to deal with the case when the number of covariates is extremely large. We develop an efficient estimation procedure based on the trace norm regularization, which explicitly imposes the low rank structure of coefficient matrices. When both the dimension of response matrix and that of covariate vector diverge at the exponential order of the sample size, we investigate the sure independence screening property under some mild conditions. We also systematically investigate some theoretical properties of our estimation procedure including estimation consistency, rank consistency and non-asymptotic error bound under some mild conditions. We further establish a theoretical guarantee for the overall solution of our two-step screening and estimation procedure. We examine the finite-sample performance of our screening and estimation methods using simulations and a large-scale imaging genetic dataset collected by the Philadelphia Neurodevelopmental Cohort (PNC) study.

Project description:Motivated by applications in genomics, we consider in this paper global and multiple testing for the comparisons of two high-dimensional linear regression models. A procedure for testing the equality of the two regression vectors globally is proposed and shown to be particularly powerful against sparse alternatives. We then introduce a multiple testing procedure for identifying unequal coordinates while controlling the false discovery rate and false discovery proportion. Theoretical justifications are provided to guarantee the validity of the proposed tests and optimality results are established under sparsity assumptions on the regression coefficients. The proposed testing procedures are easy to implement. Numerical properties of the procedures are investigated through simulation and data analysis. The results show that the proposed tests maintain the desired error rates under the null and have good power under the alternative at moderate sample sizes. The procedures are applied to the Framingham Offspring study to investigate the interactions between smoking and cardiovascular related genetic mutations important for an inflammation marker.

Project description:High-dimensional inference is one of fundamental problems in modern biomedical studies. However, the existing methods do not perform satisfactorily. Based on the Markov property of graphical models and the likelihood ratio test, this article provides a simple justification for the Markov neighborhood regression method such that it can be applied to statistical inference for high-dimensional generalized linear models with mixed features. The Markov neighborhood regression method is highly attractive in that it breaks the high-dimensional inference problems into a series of low-dimensional inference problems. The proposed method is applied to the cancer cell line encyclopedia data for identification of the genes and mutations that are sensitive to the response of anti-cancer drugs. The numerical results favor the Markov neighborhood regression method to the existing ones.

Project description:The aim of this paper is to develop a sparse projection regression modeling (SPReM) framework to perform multivariate regression modeling with a large number of responses and a multivariate covariate of interest. We propose two novel heritability ratios to simultaneously perform dimension reduction, response selection, estimation, and testing, while explicitly accounting for correlations among multivariate responses. Our SPReM is devised to specifically address the low statistical power issue of many standard statistical approaches, such as the Hotelling's T2 test statistic or a mass univariate analysis, for high-dimensional data. We formulate the estimation problem of SPREM as a novel sparse unit rank projection (SURP) problem and propose a fast optimization algorithm for SURP. Furthermore, we extend SURP to the sparse multi-rank projection (SMURP) by adopting a sequential SURP approximation. Theoretically, we have systematically investigated the convergence properties of SURP and the convergence rate of SURP estimates. Our simulation results and real data analysis have shown that SPReM out-performs other state-of-the-art methods.

Project description:We study the heteroscedastic partially linear single-index model with an unspecified error variance function, which allows for high dimensional covariates in both the linear and the single-index components of the mean function. We propose a class of consistent estimators of the parameters by using a proper weighting strategy. An interesting finding is that the linearity condition which is widely assumed in the dimension reduction literature is not necessary for methodological or theoretical development: it contributes only to the simplification of non-optimal consistent estimation. We also find that the performance of the usual weighted least square type of estimators deteriorates when the non-parametric component is badly estimated. However, estimators in our family automatically provide protection against such deterioration, in that the consistency can be achieved even if the baseline non-parametric function is completely misspecified. We further show that the most efficient estimator is a member of this family and can be easily obtained by using non-parametric estimation. Properties of the estimators proposed are presented through theoretical illustration and numerical simulations. An example on gender discrimination is used to demonstrate and to compare the practical performance of the estimators.

Project description:Drawing inferences for high-dimensional models is challenging as regular asymptotic theories are not applicable. This article proposes a new framework of simultaneous estimation and inferences for high-dimensional linear models. By smoothing over partial regression estimates based on a given variable selection scheme, we reduce the problem to low-dimensional least squares estimations. The procedure, termed as Selection-assisted Partial Regression and Smoothing (SPARES), utilizes data splitting along with variable selection and partial regression. We show that the SPARES estimator is asymptotically unbiased and normal, and derive its variance via a nonparametric delta method. The utility of the procedure is evaluated under various simulation scenarios and via comparisons with the de-biased LASSO estimators, a major competitor. We apply the method to analyze two genomic datasets and obtain biologically meaningful results.

Project description:This paper is concerned with testing linear hypotheses in high-dimensional generalized linear models. To deal with linear hypotheses, we first propose constrained partial regularization method and study its statistical properties. We further introduce an algorithm for solving regularization problems with folded-concave penalty functions and linear constraints. To test linear hypotheses, we propose a partial penalized likelihood ratio test, a partial penalized score test and a partial penalized Wald test. We show that the limiting null distributions of these three test statistics are ?2 distribution with the same degrees of freedom, and under local alternatives, they asymptotically follow non-central ?2 distributions with the same degrees of freedom and noncentral parameter, provided the number of parameters involved in the test hypothesis grows to ? at a certain rate. Simulation studies are conducted to examine the finite sample performance of the proposed tests. Empirical analysis of a real data example is used to illustrate the proposed testing procedures.

Project description:We consider a high-dimensional linear regression problem. Unlike many papers on the topic, we do not require sparsity of the regression coefficients; instead, our main structural assumption is a decay of eigenvalues of the covariance matrix of the data. We propose a new family of estimators, called the canonical thresholding estimators, which pick largest regression coefficients in the canonical form. The estimators admit an explicit form and can be linked to LASSO and Principal Component Regression (PCR). A theoretical analysis for both fixed design and random design settings is provided. Obtained bounds on the mean squared error and the prediction error of a specific estimator from the family allow to clearly state sufficient conditions on the decay of eigenvalues to ensure convergence. In addition, we promote the use of the relative errors, strongly linked with the out-of-sample R 2. The study of these relative errors leads to a new concept of joint effective dimension, which incorporates the covariance of the data and the regression coefficients simultaneously, and describes the complexity of a linear regression problem. Some minimax lower bounds are established to showcase the optimality of our procedure. Numerical simulations confirm good performance of the proposed estimators compared to the previously developed methods.

Project description:In this paper, we propose a new projection test for linear hypotheses on regression coefficient matrices in linear models with high dimensional responses. We systematically study the theoretical properties of the proposed test. We first derive the optimal projection matrix for any given projection dimension to achieve the best power and provide an upper bound for the optimal dimension of projection matrix. We further provide insights into how to construct the optimal projection matrix. One- and two-sample mean problems can be formulated as special cases of linear hypotheses studied in this paper. We both theoretically and empirically demonstrate that the proposed test can outperform the existing ones for one- and two-sample mean problems. We conduct Monte Carlo simulation to examine the finite sample performance and illustrate the proposed test by a real data example.
