Chemical trigger toward phase separation in the aqueous Al(III) system revealed.

ABSTRACT: Although Al(III) hydrolysis, condensation, and nucleation play pivotal roles in the synthesis of Al-based compounds and determine their chemical behavior, we still lack experimental evidence regarding the chemistry of nucleation from solution. Here, by combining advanced titration assays, high-resolution transmission electron microscopy (HR-TEM), and 27Al-nuclear magnetic resonance spectroscopy, we show that highly dynamic solute prenucleation clusters (PNCs) are fundamental precursors of nanosolid formation. Chemical changes from olation to oxolation bridging within PNCs rely on the formation of tetrahedral AlO4 in solution and trigger phase separation at low driving force (supersaturation). This does not include the formation of Keggin-Al13 ions, at least during the earliest stages. The PNC pathway of the formation of Al(III) (oxy)(hydr)oxides offers new possibilities toward the development of strategies for controlling the entire crystallization process.

Project description:(1) We have measured the incompatible phase separation that occurs in a polyethylene glycol-sodium dextran sulphate-sodium chloride-water system and have determined a critical point. (2) We have measured the activity coefficients of sodium chloride in critical-point concentrations of polyethylene glycol and sodium dextran sulphate respectively, and the osmotic coefficient of sodium dextran sulphate at the critical-point concentration. (3) With use of the relevant thermodynamic equations for a quaternary ionic system, we have determined the interaction coefficients between polyethylene glycol and dextran sulphate and between polyethylene glycol and sodium chloride. The former could be due mainly to volume exclusion, but the latter is too large to be explained on that basis.

G-quadruplex structures trigger RNA phase separation.

Project description:Liquid-liquid phase separation plays an important role in a variety of cellular processes, including the formation of membrane-less organelles, the cytoskeleton, signalling complexes, and many other biological supramolecular assemblies. Studies on the molecular basis of phase separation in cells have focused on protein-driven phase separation. In contrast, there is limited understanding on how RNA specifically contributes to phase separation. Here, we described a phase-separation-like phenomenon that SHORT ROOT (SHR) RNA undergoes in cells. We found that an RNA G-quadruplex (GQ) forms in SHR mRNA and is capable of triggering RNA phase separation under physiological conditions, suggesting that GQs might be responsible for the formation of the SHR phase-separation-like phenomenon in vivo. We also found the extent of GQ-triggered-phase-separation increases on exposure to conditions which promote GQ. Furthermore, GQs with more G-quartets and longer loops are more likely to form phase separation. Our studies provide the first evidence that RNA can adopt structural motifs to trigger and/or maintain the specificity of RNA-driven phase separation.

Aqueous/Aqueous Micro Phase Separation: Construction of an Artificial Model of Cellular Assembly.

Project description:To artificially construct a three-dimensional cell assembly, we investigated the availability of long-duration microdroplets that emerged near a critical point in an aqueous two-phase system (ATPS) with the hydrophilic binary polymers, polyethylene glycol (PEG), and dextran (DEX), as host containers. We found that erythrocytes (horse red blood cells; RBCs) and NAMRU mouse mammary gland epithelial cells (NMuMG cells) were completely and spontaneously entrapped inside DEX-rich microdroplets. RBCs and NMuMG cells were located in the interior and at the periphery of the droplets at PEG/DEX = 5%:5%. In contrast, the cells exhibited opposite localizations at PEG/DEX = 10%:5%, where, interestingly, NMuMG cells apparently assembled to achieve cell adhesion. We simply interpreted such specific localizations by considering the alternative responses of these cells to the properties of the PEG/DEX interfaces with different gradients in polymer concentrations.

Phase Separation of Aqueous Poly(diisopropylaminoethyl methacrylate) upon Heating.

Project description:Poly(diisopropylaminoethyl methacrylate) (PDPA) is a pH- and thermally responsive water-soluble polymer. This study deepens the understanding of its phase separation behavior upon heating. Phase separation upon heating was investigated in salt solutions of varying pH and ionic strength. The effect of the counterion on the phase transition upon heating is clearly demonstrated for chloride-, phosphate-, and citrate-anions. Phase separation did not occur in pure water. The buffer solutions exhibited similar cloud points, but phase separation occurred in different pH ranges and with different mechanisms. The solution behavior of a block copolymer comprising poly(dimethylaminoethyl methacrylate) (PDMAEMA) and PDPA was investigated. Since the PDMAEMA and PDPA blocks phase separate within different pH- and temperature ranges, the block copolymer forms micelle-like structures at high temperature or pH.

Aqueous Liquid-Liquid Phase Separation of Natural and Synthetic Polyguanidiniums.

Project description:Protamines are natural polyguanidiniums, arginine(R)-rich proteins involved in the compaction of chromatin during vertebrate spermatogenesis. Salmine, a protamine isolated from salmon sperm, contains 65 mol% R residues, with positively charged guanidino (Gdm⁺) sidechains, and no other amino acids with ionizable or aromatic sidechains. Salmine sulfate solutions undergo liquid-liquid phase separation (LLPS) with a concentration-dependent upper critical solution temperature (UCST). The condensed liquid phase comprises 50 wt % water and >600 mg·ml-¹ salmine with a constant 1:2 ratio of sulfate (SO₄²-) to Gdm⁺. Isothermal titration calorimetry, titrating Na₂SO₄ into salmine chloride above and below the UCST, allowed isolation of exothermic sulfate binding to salmine chloride from subsequent endothermic condensation and exothermic phase separation events. Synthetic random polyacrylate analogs of salmine, with 3-guanidinopropyl sidechains, displayed similar counterion dependent phase behavior, demonstrating that the LLPS of polyguanidiniums does not depend upon subunit sequence or polymer backbone chirality, and was due entirely to Gdm⁺ sidechain interactions. The results provide experimental evidence for like-charge pairing of Gdm⁺ sidechains, and an experimental approach for further characterizing these interactions.

Polyelectrolyte Complex Hollow Fiber Membranes Prepared via Aqueous Phase Separation.

Project description:Hollow fiber (HF) membrane geometry is the preferred choice for most commercial membrane operations. Such fibers are conventionally prepared via the non-solvent-induced phase separation technique, which heavily relies on hazardous and reprotoxic organic solvents such as N-methyl pyrrolidone. A more sustainable alternative, i.e., aqueous phase separation (APS), was introduced recently that utilizes water as a solvent and non-solvent for the production of polymeric membranes. Herein, for the first time, we demonstrate the preparation of sustainable and functional HF membranes via the APS technique in a dry-jet wet spinning process. The dope solution comprising poly(sodium 4-styrenesulfonate) (PSS) and polyethyleneimine (PEI) at high pH along with an aqueous bore liquid is pushed through a single orifice spinneret into a low pH acetate buffer coagulation bath. Here, PEI becomes charged resulting in the formation of a polyelectrolyte complex with PSS. The compositions of the bore liquid and coagulation bath were systematically varied to study their effect on the structure and performance of the HF membranes. The microfiltration-type membranes (permeability ∼500 to 800 L·m-2·h-1·bar-1) with complete retention of emulsion droplets were obtained when the precipitation rate was slow. Increasing the concentration of the acetate buffer in the bath led to the increase in precipitation rate resulting in ultrafiltration-type membranes (permeability ∼12 to 15 L·m-2·h-1·bar-1) having molecular weight cut-offs in the range of ∼7.8-11.6 kDa. The research presented in this work confirms the versatility of APS and moves it another step closer to large-scale use.

Polyelectrolyte Complex Membranes via Salinity Change Induced Aqueous Phase Separation.

Project description:Polymeric membranes are used on very large scales for drinking water production and kidney dialysis, but they are nearly always prepared by using large quantities of unsustainable and toxic aprotic solvents. In this study, a water-based, sustainable, and simple way of making polymeric membranes is presented without the need for harmful solvents or extreme pH conditions. Membranes were prepared from water-insoluble polyelectrolyte complexes (PECs) via aqueous phase separation (APS). Strong polyelectrolytes (PEs), poly(sodium 4-styrenesulfonate) (PSS), and poly(diallyldimethylammonium chloride) (PDADMAC) were mixed in the presence of excess of salt, thereby preventing complexation. Immersing a thin film of this mixture into a low-salinity bath induces complexation and consequently the precipitation of a solid PEC-based membrane. This approach leads to asymmetric nanofiltration membranes, with thin dense top layers and porous, macrovoid-free support layers. While the PSS molecular weight and the total polymer concentrations of the casting mixture did not significantly affect the membrane structure, they did affect the film formation process, the resulting mechanical stability of the films, and the membrane separation properties. The salt concentration of the coagulation bath has a large effect on membrane structure and allows for control over the thickness of the separation layer. The nanofiltration membranes prepared by APS have a low molecular weight cutoff (<300 Da), a high MgSO4 retention (∼80%), and good stability even at high pressures (10 bar). PE complexation induced APS is a simple and sustainable way to prepare membranes where membrane structure and performance can be tuned with molecular weight, polymer concentration, and ionic strength.

Separation and Purification of <i>Astragalus membranaceus</i> Polysaccharides by Deep Eutectic Solvents-Based Aqueous Two-Phase System.

Project description:(1) Background: Aqueous two-phase systems (ATPSs) have been widely used in the separation and purification of bioactive substances in recent years. (2) Methods: In this study, deep eutectic solvents (DESs)-based ATPSs were employed for the extraction and separation of Astragalus membranaceus polysaccharides (AMP). The optimal DES (choline chloride:urea = 1:1) was first screened to extract AMP, and the effect of DES concentration, solid-liquid ratio, pH, extraction temperature, and extraction time on the extraction yield of AMP were investigated. (3) Results: The maximum extraction yield was 141.11 mg/g under the optimum conditions. AMP was then preliminarily purified by ATPS, to further realize the recycling and reuse of DES. The effect of type of salts, salt concentration, and extraction temperature on extraction efficiency was investigated. The extraction efficiency was 97.85% for AMP under the optimum ATPS conditions. Finally, the obtained AMP was studied by molecular weight determination, infrared spectroscopy analysis, and monosaccharide composition analysis. (4) Conclusions: This ATPS extraction based on DESs is simple, environmentally friendly, low-cost, and has high extraction efficiency, which provides new ideas for the extraction of plant polysaccharides and other bioactive compounds.

Theory for the Liquid-Liquid Phase Separation in Aqueous Antibody Solutions.

Project description:This study presents the theory for liquid-liquid phase separation for systems of molecules modeling monoclonal antibodies. Individual molecule is depicted as an assembly of seven hard spheres, organized to mimic the Y-shaped antibody. We consider the antibody-antibody interactions either through Fab, Fab' (two Fab fragments may be different), or Fc domain. Interaction between these three domains of the molecule (hereafter denoted as A, B, and C, respectively) is modeled by a short-range square-well attraction. To obtain numerical results for the model under study, we adapt Wertheim's thermodynamic perturbation theory. We use this model to calculate the liquid-liquid phase separation curve and the second virial coefficient B2. Various interaction scenarios are examined to see how the strength of the site-site interactions and their range shape the coexistence curve. In the asymmetric case, where an attraction between two sites is favored and the interaction energies for the other sites kept constant, critical temperature first increases and than strongly decreases. Some more microscopic information, for example, the probability for the particular two sites to be connected, has been calculated. Analysis of the experimental liquid-liquid phase diagrams, obtained from literature, is presented. In addition, we calculate the second virial coefficient under conditions leading to the liquid-liquid phase separation and present this quantity on the graph B2 versus protein concentration.

Selective solvent filters for non-aqueous phase liquid separation from water.

Project description:Injectable filters permeable to water but impermeable to non-polar solvents were developed to contain non-aqueous phase liquids (NAPL) in contaminated aquifers, hence protecting downstream receptors during NAPL remediation. Filters were produced by injecting aqueous solutions of 0.01% chitosan, hydroxyethylcellulose and quaternized hydroxyethylcellulose into sand columns, followed by rinsing with water. Polymer sorption onto silica was verified using a quartz-crystal microbalance with dissipation monitoring. Fluorescence and gas chromatography mass spectroscopy showed low ppm range concentrations of non-polar solvents (e.g., hexane and toluene) in water eluted from the filters (in the absence of emulsifiers). The contact angles between polymer-coated surfaces and hexane or toluene were > 90°, indicating surface oleophobicity. Organic, polar solvents (e.g. tetrahydrofuran and tetrachloroethylene, TCE) were not separated from water. The contact angles between polymer-coated surfaces and TCE was also > 90°. However, the contact area with polymer coated surfaces was greater for TCE than non-polar solvents, suggesting higher affinity between TCE and the surfaces. Emulsifiers can be used to facilitate NAPL extraction from aquifers. Emulsion separation efficiency depended on the emulsifier used. Emulsions were not separated with classical surfactants (e.g. Tween 20 and oleic acid) or alkaline zein solutions. Partial emulsion separation was achieved with humic acids and zein particles.
