All-optical, an ultra-thin endoscopic photoacoustic sensor using multi-mode fiber.

ABSTRACT: Photoacoustic endoscopy (PAE) is a method of in-vivo imaging that uses tissue absorption properties. In PAE, the main tools used to detect the acoustic signal are mechanical ultrasound transducers, which require direct contact and which are difficult to miniaturize. All-optic photoacoustic sensors can challenge this issue as they can provide contact-free sensing. Here, we demonstrate sensing of photo-acoustic signals through a multimode fiber (MMF) which can provide an ultra-thin endoscopic photoacoustic sensor. Furthermore, we show the advantage of using the optical-flow method for speckle sensing and extract the photoacoustic signal despite the mode-mixing along the MMF. Moreover, it is demonstrated for the first time that the speckle reconstruction method can be used without the need for imaging of the speckles as this enables the use of multimode fibers for the speckle method.

Automated 3D segmentation of methyl isocyanate-exposed rat trachea using an ultra-thin, fully fiber optic optical coherence endoscopic probe.

Project description:Development of effective rescue countermeasures for toxic inhalational industrial chemicals, such as methyl isocyanate (MIC), has been an emerging interest. Nonetheless, current methods for studying toxin-induced airway injuries are limited by cost, labor time, or accuracy, and only provide indirect or localized information. Optical Coherence Tomography (OCT) endoscopic probes have previously been used to visualize the 3-D airway structure. However, gathering such information in small animal models, such as rat airways after toxic gas exposure, remains a challenge due to the required probe size necessary for accessing the small, narrow, and partially obstructed tracheas. In this study, we have designed a 0.4 mm miniature endoscopic probe and investigated the structural changes in rat trachea after MIC inhalation. An automated 3D segmentation algorithm was implemented so that anatomical changes, such as tracheal lumen volume and cross-sectional areas, could be quantified. The tracheal region of rats exposed to MIC by inhalation showed significant airway narrowing, especially within the upper trachea, as a result of epithelial detachment and extravascular coagulation within the airway. This imaging and automated reconstruction technique is capable of rapid and minimally-invasive identification of airway obstruction. This method can be applied to large-scale quantitative analysis of in vivo animal models.

Reflection-mode multifocal optical-resolution photoacoustic microscopy.

Project description:Compared with single-focus optical-resolution photoacoustic microscopy (OR-PAM), multifocal OR-PAM utilizes both multifocal optical illumination and an ultrasonic array transducer, significantly increasing the imaging speed. A reflection-mode multifocal OR-PAM system based on a microlens array that provides multiple foci as well as an ultrasonic array transducer that receives the excited photoacoustic waves from all foci simultaneously is presented. Using a customized microprism to reflect the incident laser beam to the microlens array, the multiple optical foci are aligned confocally with the focal zone of the ultrasonic array transducer. Experiments show the reflection-mode multifocal OR-PAM is capable of imaging microvessels in vivo, and it can image a 6×5×2.5??mm³ volume at 16 ?m lateral resolution in ?2.5??min, which was limited by the signal multiplexing ratio and laser pulse repetition rate.

Fiber-Laser-Based Ultrasound Sensor for Photoacoustic Imaging.

Project description:Photoacoustic imaging, especially for intravascular and endoscopic applications, requires ultrasound probes with miniature size and high sensitivity. In this paper, we present a new photoacoustic sensor based on a small-sized fiber laser. Incident ultrasound waves exert pressures on the optical fiber laser and induce harmonic vibrations of the fiber, which is detected by the frequency shift of the beating signal between the two orthogonal polarization modes in the fiber laser. This ultrasound sensor presents a noise-equivalent pressure of 40 Pa over a 50-MHz bandwidth. We demonstrate this new ultrasound sensor on an optical-resolution photoacoustic microscope. The axial and lateral resolutions are 48 μm and 3.3 μm. The field of view is up to 1.57 mm2. The sensor exhibits strong resistance to environmental perturbations, such as temperature changes, due to common-mode cancellation between the two orthogonal modes. The present fiber laser ultrasound sensor offers a new tool for all-optical photoacoustic imaging.

Photoacoustic Mouse Brain Imaging Using an Optical Fabry-Perot Interferometric Ultrasound Sensor.

Project description:Photoacoustic (PA, or optoacoustic, OA) mesoscopy is a powerful tool for mouse cerebral imaging, which offers high resolution three-dimensional (3D) images with optical absorption contrast inside the optically turbid brain. The image quality of a PA mesoscope relies on the ultrasonic transducer which detects the PA signals. An all-optical ultrasound sensor based on a Fabry-Pérot (FP) polymer cavity has the following advantages: broadband frequency response, wide angular coverage and small footprint. Here, we present 3D PA mesoscope for mouse brain imaging using such an optical sensor. A heating laser was used to stabilize the sensor's cavity length during the imaging process. To acquire data for a 3D angiogram of the mouse brain, the sensor was mounted on a translation stage and raster scanned. 3D images of the mouse brain vasculature were reconstructed which showed cerebrovascular structure up to a depth of 8 mm with high quality. Imaging segmentation and dual wavelength imaging were performed to demonstrate the potential of the system in preclinical brain research.

Ultra-fast optical ranging using quantum-dash mode-locked laser diodes.

Project description:Laser-based light detection and ranging (LiDAR) is key to many applications in science and industry. For many use cases, compactness and power efficiency are key, especially in high-volume applications such as industrial sensing, navigation of autonomous objects, or digitization of 3D scenes using hand-held devices. In this context, comb-based ranging systems are of particular interest, combining high accuracy with high measurement speed. However, the technical complexity of miniaturized comb sources is still prohibitive for many applications, in particular when high optical output powers and high efficiency are required. Here we show that quantum-dash mode-locked laser diodes (QD-MLLD) offer a particularly attractive route towards high-performance chip-scale ranging systems. QD-MLLDs are compact, can be easily operated by a simple DC drive current, and provide spectrally flat frequency combs with bandwidths in excess of 2 THz, thus lending themselves to coherent dual-comb ranging. In our experiments, we show measurement rates of up to 500 MHz-the highest rate demonstrated with any ranging system so far. We attain reliable measurement results with optical return powers of only - 40 dBm, corresponding to a total loss of 49 dB in the ranging path, which corresponds to the highest loss tolerance demonstrated so far for dual-comb ranging with chip-scale comb sources. Combing QD-MLLDs with advanced silicon photonic receivers offers an attractive route towards robust and technically simple chip-scale LiDAR systems.

Excitation of epsilon-near-zero resonance in ultra-thin indium tin oxide shell embedded nanostructured optical fiber.

Project description:We report a novel optical waveguide design of a hollow step index fiber modified with a thin layer of indium tin oxide (ITO). We show an excitation of highly confined waveguide mode in the proposed fiber near the wavelength where permittivity of ITO approaches zero. Due to the high field confinement within thin ITO shell inside the fiber, the epsilon-near-zero (ENZ) mode can be characterized by a peak in modal loss of the hybrid waveguide. Our results show that such in-fiber excitation of ENZ mode is due to the coupling of the guided core mode to the thin-film ENZ mode. We also show that the phase matching wavelength, where the coupling takes place, varies depending on the refractive index of the constituents inside the central bore of the fiber. These ENZ nanostructured optical fibers have many potential applications, for example, in ENZ nonlinear and magneto-optics, as in-fiber wavelength-dependent filters, and as subwavelength fluid channel for optical and bio-photonic sensing.

Automated Laser-Fiber Coupling Module for Optical-Resolution Photoacoustic Microscopy.

Project description:Photoacoustic imaging has emerged as a promising biomedical imaging technique that enables visualization of the optical absorption characteristics of biological tissues in vivo. Among the different photoacoustic imaging system configurations, optical-resolution photoacoustic microscopy stands out by providing high spatial resolution using a tightly focused laser beam, which is typically transmitted through optical fibers. Achieving high-quality images depends significantly on optical fluence, which is directly proportional to the signal-to-noise ratio. Hence, optimizing the laser-fiber coupling is critical. Conventional coupling systems require manual adjustment of the optical path to direct the laser beam into the fiber, which is a repetitive and time-consuming process. In this study, we propose an automated laser-fiber coupling module that optimizes laser delivery and minimizes the need for manual intervention. By incorporating a motor-mounted mirror holder and proportional derivative control, we successfully achieved efficient and robust laser delivery. The performance of the proposed system was evaluated using a leaf-skeleton phantom in vitro and a human finger in vivo, resulting in high-quality photoacoustic images. This innovation has the potential to significantly enhance the quality and efficiency of optical-resolution photoacoustic microscopy.

Biofilm Detection by a Fiber-Tip Ball Resonator Optical Fiber Sensor.

Project description:Bacterial biofilms are one of the most important challenges that modern medicine faces due to the difficulties of diagnosis, antibiotic resistance, and protective mechanisms against aggressive environments. For these reasons, methods that ensure the inexpensive and rapid or real-time detection of biofilm formation on medical devices are needed. This study examines the possibilities of using optical- and fiber-based biosensors to detect and analyze early bacterial biofilms. In this study, the biofilm-forming model organism Pseudomonas aeruginosa was inoculated on the surface of the optical sensor and allowed to attach for 2 h. The biosensors were made by a fiber-tip ball resonator, fabricated through a CO2 laser splicer on a single-mode fiber, forming a weak reflective spectrum. An optical backscatter reflectometer was used to measure the refractive index detected by the sensors during different growth periods. The early biofilm concentration was determined by crystal violet (CV) binding assay; however, such a concentration was lower than the detection limit of this assay. This work presents a new approach of biofilm sensing in the early attachment stage with a low limit of detection up to 10-4 RIU (refractive index units) or 35 ± 20 × 103 CFU/mL (colony formed units).

Quantitative phase imaging through an ultra-thin lensless fiber endoscope.

Project description:Quantitative phase imaging (QPI) is a label-free technique providing both morphology and quantitative biophysical information in biomedicine. However, applying such a powerful technique to in vivo pathological diagnosis remains challenging. Multi-core fiber bundles (MCFs) enable ultra-thin probes for in vivo imaging, but current MCF imaging techniques are limited to amplitude imaging modalities. We demonstrate a computational lensless microendoscope that uses an ultra-thin bare MCF to perform quantitative phase imaging with microscale lateral resolution and nanoscale axial sensitivity of the optical path length. The incident complex light field at the measurement side is precisely reconstructed from the far-field speckle pattern at the detection side, enabling digital refocusing in a multi-layer sample without any mechanical movement. The accuracy of the quantitative phase reconstruction is validated by imaging the phase target and hydrogel beads through the MCF. With the proposed imaging modality, three-dimensional imaging of human cancer cells is achieved through the ultra-thin fiber endoscope, promising widespread clinical applications.

Scanner-free and wide-field endoscopic imaging by using a single multimode optical fiber.

Project description:A single multimode fiber is considered an ideal optical element for endoscopic imaging due to the possibility of direct image transmission via multiple spatial modes. However, the wave distortion induced by the mode dispersion has been a fundamental limitation. In this Letter, we propose a method for eliminating the effect of mode dispersion and therefore realize wide-field endoscopic imaging by using only a single multimode fiber with no scanner attached to the fiber. Our method will potentially revolutionize endoscopy in various fields encompassing medicine and industry.
