Project description:A network of predominantly left-lateralized brain regions has been linked to verbal working memory (VWM) performance. However, the impact of memory load on the oscillatory dynamics serving VWM is far less understood. To further investigate this, we had 26 healthy adults perform a high-load (6 letter) and low-load (4 letter) variant of a VWM task while undergoing magnetoencephalography (MEG). MEG data were evaluated in the time-frequency domain and significant oscillatory responses spanning the encoding and maintenance phases were reconstructed using a beamformer. To determine the impact of load on the neural dynamics, the resulting images were examined using paired-samples t-tests and virtual sensor analyses. Our results indicated stronger increases in frontal theta activity in the high- relative to low-load condition during early encoding. Stronger decreases in alpha/beta activity were also observed during encoding in bilateral posterior cortices during the high-load condition, and the strength of these load effects increased as encoding progressed. During maintenance, stronger decreases in alpha activity in the left inferior frontal gyrus, middle temporal gyrus, supramarginal gyrus, and inferior parietal cortices were detected during high- relative to low-load performance, with the strength of these load effects remaining largely static throughout maintenance. Finally, stronger increases in occipital alpha activity were observed during maintenance in the high-load condition, and the strength of these effects grew stronger with time during the first half of maintenance, before dissipating during the latter half of maintenance. Notably, this was the first study to utilize a whole-brain approach to statistically evaluate the temporal dynamics of load-related oscillatory differences during encoding and maintenance processes, and our results highlight the importance of spatial, temporal, and spectral specificity in this regard.

Project description:The maintenance of items in working memory relies on persistent neural activity in a widespread network of brain areas. To investigate the influence of load on working memory, we asked human subjects to maintain sets of letters in memory while we recorded single neurons and intracranial encephalography (EEG) in the medial temporal lobe and scalp EEG. Along the periods of a trial, hippocampal neural firing differentiated between success and error trials during stimulus encoding, predicted workload during memory maintenance, and predicted the subjects' behavior during retrieval. During maintenance, neuronal firing was synchronized with intracranial hippocampal EEG. On the network level, synchronization between hippocampal and scalp EEG in the theta-alpha frequency range showed workload dependent oscillatory coupling between hippocampus and cortex. Thus, we found that persistent neural activity in the hippocampus participated in working memory processing that is specific to memory maintenance, load sensitive and synchronized to the cortex.

Project description:Can cognitive load enhance concentration on task-relevant information and help filter out distractors? Most of the prior research in the area of selective attention has focused on visual attention or cross-modal distraction and has yielded controversial results. Here, we studied whether working memory load can facilitate selective attention when both target and distractor stimuli are auditory. We used a letter n-back task with four levels of working memory load and two levels of distraction: congruent and incongruent distractors. This combination of updating and inhibition tasks allowed us to manipulate working memory load within the selective attention task. Participants sat in front of three loudspeakers and were asked to attend to the letter presented from the central loudspeaker while ignoring that presented from the flanking ones (spoken by a different person), which could be the same letter as the central one (congruent) or a different (incongruent) letter. Their task was to respond whether or not the central letter matched the letter presented n (0, 1, 2, or 3) trials back. Distraction was measured in terms of the difference in reaction time and accuracy on trials with incongruent versus congruent flankers. We found reduced interference from incongruent flankers in 2- and 3-back conditions compared to 0- and 1-back conditions, whereby higher working memory load almost negated the effect of incongruent flankers. These results suggest that high load on verbal working memory can facilitate inhibition of distractors in the auditory domain rather than make it more difficult as sometimes claimed.

Project description:There is accumulating evidence that training working memory (WM) leads to beneficial effects in tasks that were not trained, but the mechanisms underlying this transfer remain elusive. Brain imaging can be a valuable method to gain insights into such mechanisms. Here, we discuss the impact of cognitive training on neural correlates with an emphasis on studies that implemented a WM intervention. We focus on changes in activation patterns, changes in resting state connectivity, changes in brain structure, and changes in the dopaminergic system. Our analysis of the existing literature reveals that there is currently no clear pattern of results that would single out a specific neural mechanism underlying training and transfer. We conclude that although brain imaging has provided us with information about the mechanisms of WM training, more research is needed to understand its neural impact.

Project description:Recent studies have shown that past selection history affects the allocation of attention on target selection. However, it is unclear whether context-driven selection history can modulate the efficacy of attention allocation on working memory (WM) representations. This study tests the influences of selection history on WM capacity. A display of one item (low load) or three/four items (high load) was shown for the participants to hold in WM in a delayed response task. Participants then judged whether a probe item was in the memory display or not. Selection history was defined as the number of items attended across trials in the task context within a block, manipulated by the stimulus set-size in the contexts with fewer possible stimuli (4-item or 5-item context) or more possible stimuli (8-item or 9-item context) from which the memorized content was selected. The capacity measure (i.e., the K measure) was estimated to reflect the number of items that can be held in WM. Across four behavioral experiments, the results revealed that the capacity was significantly reduced in the context with more possible stimuli relative to the context with fewer possible stimuli. Moreover, the reduction in capacity was significant for high WM load and not observed when the focus was on only a single item. Together, these findings indicate that context-driven selection history and focused attention influence WM capacity.

Project description:Working memory (WM) enables the maintenance and manipulation of information relevant to behavioral goals. Variability in WM ability is strongly correlated with IQ [1], and WM function is impaired in many neurological and psychiatric disorders [2, 3], suggesting that this system is a core component of higher cognition. WM storage is thought to be mediated by patterns of activity in neural populations selective for specific properties (e.g., color, orientation, location, and motion direction) of memoranda [4-13]. Accordingly, many models propose that differences in the amplitude of these population responses should be related to differences in memory performance [14, 15]. Here, we used functional magnetic resonance imaging and an image reconstruction technique based on a spatial encoding model [16] to visualize and quantify population-level memory representations supported by multivoxel patterns of activation within regions of occipital, parietal and frontal cortex while participants precisely remembered the location(s) of zero, one, or two small stimuli. We successfully reconstructed images containing representations of the remembered-but not forgotten-locations within regions of occipital, parietal, and frontal cortex using delay-period activation patterns. Critically, the amplitude of representations of remembered locations and behavioral performance both decreased with increasing memory load. These results suggest that differences in visual WM performance between memory load conditions are mediated by changes in the fidelity of large-scale population response profiles distributed across multiple areas of human cortex.

Project description:Transfer of learning refers to successful application of previously acquired knowledge or skills to novel settings. Although working memory (WM) is thought to play a role in transfer learning, direct evidence of the effect of limitations in WM on transfer learning is lacking. To investigate, we used an acquired equivalence paradigm that included tests of association and transfer learning. The effects of imposing an acute WM limitation on young adults was tested (within-subjects design: N = 27 adults; Mage = 24 years) by conducting learning transfer tests concurrent with a secondary task that required carrying a spatial WM load when performing the learned/transfer trial (Load condition) to acutely limit WM resources or no WM load (No-Load condition; WM was unloaded prior to performing the learned/transfer trial). Analyses showed that although success on the transfer trials was high in the No-Load condition, performance dropped to chance in the Load condition. Performance on tests of learned associations remained high in both conditions. These results indicate that transfer of learning depends on access to WM resources and suggest that even healthy young individuals may be affected in their ability to cross-utilize knowledge when cognitive resources become scarce, such as when engaging in two tasks simultaneously (e.g., using satellite navigation while driving).

Project description:Temporal attention refers to the ability to orient attention in time, which serves to enhance performance such as target detection and discrimination and is a fundamental component of cognitive function. Although some research indicates that temporal attention ability is affected by working memory updating, it is unclear whether temporal attention is also affected by the availability of working memory stores. To address this, participants were presented a dual-task paradigm requiring zero, three, or six digits to be held in working memory while engaged in a temporally cued visual discrimination task. Results show that working memory load did not differentially affect the ability to benefit from predictive temporal cues during the visual discrimination task. This indicates that temporal attention is not affected by available working memory stores. Interestingly, posterior beta band (12-30 Hz) activity was differentially modulated by temporal attention and working memory load, such that it decreased prior to expected targets and increased with load. Analysis across participants indicated that those individuals who exhibited greater temporal attention-based modulation of beta activity (i.e., predictive < neutrally cued) displayed improved discrimination performance, but also yielded lowered working memory accuracy. Thus, the ability to benefit from temporal attention processes while multitasking comes at the cost of lowered secondary task performance. Together, these results indicate that available working memory stores do not affect temporal attention ability. Rather, limitations in divided attention ability result in a performance cost that prioritizes one task over another, which may be indexed by beta band activity.

Project description:Several cognitive processes, including instrumental behavior and working memory, are controlled by endocannabinoids acting on cannabinoid receptor 1 (CB1R) in the brain through retrograde and presynaptic inhibition of GABA or glutamate release. However, the temporal mechanisms underlying the control of these cognitive processes by CB1Rs remain largely unknown. Here, we have developed a light-sensitive CB1R chimera (optoCB1R) by replacing the intracellular domains of bovine rhodopsin with those of human CB1R. We demonstrated that light stimulation of optoCB1R triggered canonical CB1R signaling by inhibiting cAMP (but not cGMP or IP1) signaling and activating the MAPK pathway in vitro or in vivo. Moreover, light stimulation of optoCB1R in corticostriatal glutamatergic neurons could temporally inhibit excitatory postsynaptic currents (EPSCs) at the level of seconds. Importantly, transient (3 s) and "time-locked", but not random, activation of optoCB1R signaling in corticostriatal neurons at the time of reward affected animal sensitivity to outcome devaluation and inhibited goal-directed behavior. However, prolonged (~30 min) but not transient (10 or 30 s) activation of astrocytic CB1R signaling in the hippocampus impaired working memory. Consequently, neuronal and astrocytic CB1R signaling differentially regulate working memory and goal-directed behavior through distinct temporal and cellular mechanisms. Ultimately, the pharmacological blockade of adenosine A2AR improved the neuronal and astrocytic CB1R-induced impairments in goal-directed behavior and working memory, possibly through modulation of EPSCs and c-Fos, respectively. Therefore, A2AR may represent a promising target for managing cognitive dysfunction resulting from the use of CB1R drugs.

Project description:Many of the cognitive deficits of normal ageing (forgetfulness, distractibility, inflexibility and impaired executive functions) involve prefrontal cortex (PFC) dysfunction. The PFC guides behaviour and thought using working memory, which are essential functions in the information age. Many PFC neurons hold information in working memory through excitatory networks that can maintain persistent neuronal firing in the absence of external stimulation. This fragile process is highly dependent on the neurochemical environment. For example, elevated cyclic-AMP signalling reduces persistent firing by opening HCN and KCNQ potassium channels. It is not known if molecular changes associated with normal ageing alter the physiological properties of PFC neurons during working memory, as there have been no in vivo recordings, to our knowledge, from PFC neurons of aged monkeys. Here we characterize the first recordings of this kind, revealing a marked loss of PFC persistent firing with advancing age that can be rescued by restoring an optimal neurochemical environment. Recordings showed an age-related decline in the firing rate of DELAY neurons, whereas the firing of CUE neurons remained unchanged with age. The memory-related firing of aged DELAY neurons was partially restored to more youthful levels by inhibiting cAMP signalling, or by blocking HCN or KCNQ channels. These findings reveal the cellular basis of age-related cognitive decline in dorsolateral PFC, and demonstrate that physiological integrity can be rescued by addressing the molecular needs of PFC circuits.
