Parasite species co-occurrence patterns on Peromyscus: Joint species distribution modelling.

ABSTRACT: Hosts are often infested by multiple parasite species, but it is often unclear whether patterns of parasite co-occurrence are driven by parasite habitat requirements or parasite species interactions. Using data on infestation patterns of ectoparasitic arthropods (fleas, trombiculid mites, cuterebrid botflies) from deer mice (Peromyscus maniculatus), we analyzed species associations using joint species distribution modelling. We also experimentally removed a flea (Orchopeas leucopus) from a subset of deer mice to examine the effect on other common ectoparasite species. We found that the mite (Neotrombicula microti) and botfly (Cuterebra sp.) had a negative relationship that is likely a true biotic species interaction. The flea had a negative association with the mite and a positive association with the botfly species, both of which appeared to be influenced by host traits or parasite life-history traits. Furthermore, experimental removal of the flea did not have a significant effect on ectoparasite prevalence of another species. Overall, these findings suggest that complex parasite species associations can be present among multiple parasite taxa, and that aggregation is not always the rule for ectoparasite communities of small mammals.

Project description:Interactions among parasite species coinfecting the same host individual can have far reaching consequences for parasite ecology and evolution. How these within-host interactions affect epidemics may depend on two non-exclusive mechanisms: parasite growth and reproduction within hosts, and parasite transmission between hosts. Yet, how these two mechanisms operate under coinfection, and how sensitive they are to the composition of the coinfecting parasite community, remains poorly understood. Here, we test the hypothesis that the relationship between within- and between-host transmission of the fungal pathogen, Phomopsis subordinaria, is affected by co-occurring parasites infecting the host plant, Plantago lanceolata. We conducted a field experiment manipulating the parasite community of transmission source plants, then tracked P. subordinaria within-host transmission, as well as between-host transmission to naïve recipient plants. We find that coinfection with the powdery mildew pathogen, Podosphaera plantaginis, causes increased between-host transmission of P. subordinaria by affecting the number of infected flower stalks in the source plants, resulting from altered auto-infection. In contrast, coinfection with viruses did not have an effect on either within- or between-host transmission. We then analyzed data on the occurrence of P. subordinaria in 2018 and the powdery mildew in a multi-year survey data set from natural host populations to test whether the positive association predicted by our experimental results is evident in field epidemiological data. Consistent with our experimental findings, we observed a positive association in the occurrence of P. subordinaria and historical powdery mildew persistence. Jointly, our experimental and epidemiological results suggest that within- and between-host transmission of P. subordinaria depends on the identity of coinfecting parasites, with potentially far-reaching effects on disease dynamics and parasite co-occurrence patterns in wild populations.Supplementary informationThe online version contains supplementary material available at 10.1007/s10682-022-10182-9.

Project description:Disentangling the role of competition in regulating the distribution of sympatric species can be difficult because species can have different habitat preferences or time use that introduce non-random patterns that are not related to interspecific interactions. We adopted a multi-step approach to systematically incorporate habitat preferences while investigating the co-occurrence of two presumed competitors, morphologically similar, and closely related ground-dwelling birds: the brown tinamou (Crypturellus obsoletus) and the tataupa tinamou (C. tataupa). First, we used single-species occupancy models to identify the main landscape characteristics affecting site occupancy, while accounting for detection probability. We then used these factors to control for the effect of habitat while investigating species co-occurrence. In addition, we investigated species present-time partitioning by measuring the degree of overlap in their activity time. Both species were strictly diurnal and their activity time highly overlapped (i.e., the species are not present-time partitioning). The distribution of the two species varied across the landscape, and they seemed to occupy opposite portions of the study area, but co-occurrence models and species interaction factors suggested that the tinamous have independent occupancy and detection. In addition, co-occurrence models that accounted for habitat performed better than models without habitat covariates. The observed co-occurrence pattern is more likely related to habitat preferences, wherein species segregated by elevation. These results provide evidence that habitat characteristics can play a bigger role than interspecific interactions in regulating co-existence of some species. Therefore, exploring habitat preferences while analyzing co-occurrence patterns is essential, in addition to being a feasible approach to achieve more accurate estimation of parameters reflecting species interactions. Occupancy models can be a valuable tool in such modeling.

Project description:Parasites, especially brain-encysting trematodes, can have an impact on host behaviour, facilitating the transmission to next host and completion of the life cycle, but insufficient research has been done on whether specific brain regions are targeted. Using Cardiocephaloides longicollis as a laboratory model, the precise distribution of metacercariae in experimentally-infected, wild and farmed fish was mapped. The brain regions targeted by this parasite were explored, also from a histologic perspective, and potential pathogenic effects were evaluated. Experimental infections allowed to reproduce the natural infection intensity of C. longicollis, with four times higher infection intensity at the higher dose (150 vs 50 cercariae). The observed metacercarial distribution, similar among all fish groups, may reflect a trematode species-specific pattern: metacercariae occur with highest density in the optic lobe area (primarily infecting the periventricular gray zone of optic tectum) and the medulla oblongata, whereas other areas such as the olfactory lobes and cerebellar lobes may be occupied when the more frequently invaded parts of the brain were crowded. Mono- and multicysts (i.e. formed either with a single metacercaria, or with 2-25 metacercariae encapsulated together) may be formed depending on the aggregation and timing of metacercariae arrival, with minor host inflammatory response. Larvae of C. longicollis colonizing specific brain areas may have an effect on the functions associated with these areas, which are generally related to sensory and motor functions, but are also related to other host fitness traits such as school maintenance or recognition of predators. The detailed information on the extent and distribution of C. longicollis in fish encephalon sets the ground to understand the effects of brain parasites on fish, but further investigation to establish if C. longicollis, through purely mechanical damage (e.g., occupation, pressure and displacement), has an actual impact on host behaviour remains to be tested under controlled experimental conditions.
