First report of isolation of Aeromonas taiwanensis from India.

ABSTRACT: Genus Aeromonas consists of facultative anaerobic, Gram negative Bacilli which are primary environmental inhabitants worldwide. A recently reported strain of the genus, Aeromonas taiwanensis, was found while studying the presence of infectious marine microbes in a lacustrine wetland in India, making this the first isolation report from the country.

First report on isolation of Mycobacterium monacense from sputum specimen in India.

Project description:We report a case of Mycobacterium monacense (M. monacense) isolated from sputum of a female patient for the first time in India. The chest radiograph and symptoms were suggestive of pulmonary tuberculosis. M. monacense was isolated from the sputum specimen at the end of 11 days of incubation. The identification was confirmed by conventional biochemical tests and polymerase chain reaction (PCR) -based Restriction Fragment Length Polymorphism (RFLP) and DNA sequencing targeting Internal Transcribed Spacer (ITS) region and hsp65 gene. The patient was treated with conventional anti-tuberculous drugs.

Efficient degradation of ivermectin by newly isolated Aeromonas taiwanensis ZJB-18,044.

Project description:Ivermectin (IVM) is a widely used antiparasitic agent and acaricide. Despite its high efficiency against nematodes and arthropods, IVM may pose a threat to the environment due to its ecotoxcity. In this study, degradation of IVM by a newly isolated bacterium Aeromonas taiwanensis ZJB-18,044 was investigated. Strain ZJB-18,044 can completely degrade 50 mg/L IVM in 5 d with a biodegradation ability of 0.42 mg/L/h. Meanwhile, it exhibited high tolerance (50 mg/L) to doramectin, emamectin, rifampicin, and spiramycin. It can also efficiently degrade doramectin, emamectin, and spiramycin. The IVM degradation of strain ZJB-18,044 can be inhibited by erythromycin, azithromycin, spiramycin or rifampicin. However, supplement of carbonyl cyanide m-chlorophenylhydrazone, an uncoupler of oxidative phosphorylation, can partially recover the IVM degradation. Moreover, strain ZJB-18,044 cells can pump out excess IVM to maintain a low intracellular IVM concentration. Therefore, the IVM tolerance of strain ZJB-18,044 may be due to the regulation of the intracellular IVM concentration by the activated macrolide efflux pump(s). With the high IVM degradation efficiency, A. taiwanensis ZJB-18,044 may serve as a bioremediation agent for IVM and other macrolides in the environment.

First report of isolation and antibiotic susceptibility pattern of Raoultella electrica from table eggs in Jaipur, India.

Project description:Raoultella electrica, a Gram-negative, non-spore-forming, rod-shaped facultative anaerobe, was identified during a regular investigation of bacterial contamination in table eggs in the winter season. A total of 165 hen's eggs were collected in the winter season from 15 different areas of the city of Jaipur, India. Gram-negative Enterobacteriales were isolated on selective and differential media by the conventional plate method and were further identified by several biochemical tests and 16S rRNA gene sequencing. Commonly prescribed antibiotics for enteric infection were used for antibiotic susceptibility testing. For isolated microorganisms, different resistance patterns were found against the different antibiotics used (p < 0.01). The multiple antibiotic resistance index of bacterial isolates ranged from 0.10 to 0.60. R. electrica strain 1GB/NBRC 109676/KCTC 32430 was isolated for the first time from commercial chicken's eggs.

First report of canine bufavirus in India.

Project description:Canine bufavirus (CBuV), a novel protoparvovirus of dogs that is associated with enteric and respiratory symptoms, has been reported only in Italy and China. The enteric prevalence of CBuV in India was investigated, and the nearly complete genome sequence (4292 bp) was amplified and reconstructed for one strain. A nucleotide sequence alignment indicated 93.42-98.81% identity to the other available CBuV sequences and 70.88-73.39% and 54.4-54.8% identity to human bufavirus and canine parvovirus 2 (CPV-2), respectively. The current strain is most closely related to Chinese CBuV strains, which together form an Asian lineage. This first report of the prevalence of CBuV in India emphasizes the need for further epidemiological surveillance.

Vaccine-induced thrombotic thrombocytopenia (VITT): first report from India.

Project description:BackgroundVaccine-induced thrombotic thrombocytopenia (VITT) is a rare but devastating adverse event following adenoviral vector-based vaccinations for COVID-19, resulting in thrombosis, especially of the cerebral and splanchnic vasculature. Despite the progress in laboratory techniques for early diagnosis, VITT remains a clinical diagnosis supplemented by coagulation studies. We report on VITT for the first time from India.CaseWe describe cortical venous sinus thrombosis and intracerebral bleed associated with severe thrombocytopenia in two young men who had no other contributory cause besides a recent ChAdOx1 nCoV-19 vaccination. The diagnosis was supported with PF-4 antibodies in one patient. The second patient's test could not be processed to technical limitations. Both patients were treated with IVIG at 1 g/kg for 2 days and anticoagulation (Apixaban). One patient fully recovered with no residual deficits, and the other is under treatment and recovering.ConclusionVITT can cause devastating fatality and morbidity in otherwise healthy patients via potential immune-mediated effects. Clinicians should have a high suspicion index and treat VITT in the appropriate setting even if the PF-4 antibody testing by ELISA is unavailable or delayed. Though counterintuitive, clinicians must not delay the administration of non-heparin anticoagulation, IVIG and restrict platelet transfusion even in the presence of intracerebral haemorrhage.

First report of environmental bla PAC-1-carrying Aeromonas enteropelogenes.

Project description:The bla PAC-1 has been reported in Central Asia and Europe countries like Afghanistan and France in Aeromonas caviae and Pseudomonas aeruginosa strains from animals and patients, respectively. However, there is no record of bla PAC-1-carrying strain from the natural environment, and bla PAC-1-carrying Aeromonas has not been reported in the Asia Pacific. Here, we report the first known enviromental bla PAC-1-carrying Aeromonas enteropelogenes in the world from reservoir water in Singapore. We have performed a comprehensive genetic environment alignment and comparison of bla PAC-1 between our strain and other strains from different countries and sources and found the bla PAC-1 located on a highly conserved gene cluster. We suggest that environmental Aeromonas strains may act as a hidden reservoir involved in the circulating of bla PAC-1. The finding of conserved bla PAC-1 cluster also suggested the existence of multiple transmission pathways of bla PAC-1 in the Asia-Pacific region, involving multiple sources and different species.
