Project description:Sensory differences between autism and neuro-typical populations are well-documented and have often been explained by either weak-central-coherence or excitation/inhibition-imbalance cortical theories. We tested these theories with perceptual multi-stability paradigms in which dissimilar images presented to each eye generate dynamic cyclopean percepts based on ongoing cortical grouping and suppression processes. We studied perceptual multi-stability with Interocular Grouping (IOG), which requires the simultaneous integration and suppression of image fragments from both eyes, and Conventional Binocular Rivalry (CBR), which only requires global suppression of either eye, in 17 autistic adults and 18 neurotypical participants. We used a Hidden-Markov-Model as tool to analyze the multistable dynamics of these processes. Overall, the dynamics of multi-stable perception were slower (i.e. there were longer durations and fewer transitions among perceptual states) in the autistic group compared to the neurotypical group for both IOG and CBR. The weighted Markovian transition distributions revealed key differences between both groups and paradigms. The results indicate overall lower levels of suppression and decreased levels of grouping in autistic than neurotypical participants, consistent with elements of excitation/inhibition imbalance and weak-central-coherence theories.

Project description:Prolonged exposure to a high contrast stimulus reduces the neural sensitivity to subsequent similar patterns. Recent work has disclosed that contrast adaptation is controlled by multiple mechanisms operating over differing timescales. Adaptation to high contrast for a relatively longer period can be rapidly eliminated by adaptation to a lower contrast (or meanfield in the present study). Such rapid deadaptation presumably causes a short-term mechanism to signal for a sensitivity increase, canceling ongoing signals from long-term mechanisms. Once deadaptation ends, the short-term mechanism rapidly returns to baseline, and the slowly decaying effects in the long-term mechanisms reemerge, allowing the perceptual aftereffects to recover during continued testing. Although this spontaneous recovery effect is considered strong evidence supporting the multiple mechanisms theory, it remains controversial whether the effect is mainly driven by visual memory established during the initial longer-term adaptation period. To resolve this debate, we used a modified Continuous Flash Suppression (CFS) and visual crowding paradigms to render the adapting stimuli invisible, but still observed the spontaneous recovery phenomenon. These results exclude the possibility that spontaneous recovery found in the previous work was merely the consequence of explicit visual memory. Our findings also demonstrate that contrast adaptation, even at the unconscious processing levels, is controlled by multiple mechanisms.

Project description:Expectations, derived from previous experience, can help in making perception faster, more reliable and informative. A key neural signature of perceptual expectations is expectation suppression, an attenuated neural response to expected compared with unexpected stimuli. While expectation suppression has been reported using a variety of paradigms and recording methods, it remains unclear what neural modulation underlies this response attenuation. Sharpening models propose that neural populations tuned away from an expected stimulus are particularly suppressed by expectations, thereby resulting in an attenuated, but sharper population response. In contrast, dampening models suggest that neural populations tuned toward the expected stimulus are most suppressed, thus resulting in a dampened, less redundant population response. Empirical support is divided, with some studies favoring sharpening, while others support dampening. A key limitation of previous neuroimaging studies is the ability to draw inferences about neural-level modulations based on population (e.g. voxel) level signals. Indeed, recent simulations of repetition suppression showed that opposite neural modulations can lead to comparable population-level modulations. Forward models provide one solution to this inference limitation. Here, we used forward models to implement sharpening and dampening models, mapping neural modulations to voxel-level data. We show that a feature-specific gain modulation, suppressing neurons tuned toward the expected stimulus, best explains the empirical fMRI data. Thus, our results support the dampening account of expectation suppression, suggesting that expectations reduce redundancy in sensory cortex, and thereby promote updating of internal models on the basis of surprising information.

Project description:Vision relies on both specific knowledge of visual attributes, such as object categories, and general brain states, such as those reflecting arousal. We hypothesized that these phenomena independently influence recognition of forthcoming stimuli through distinct processes reflected in spontaneous neural activity. Here, we recorded magnetoencephalographic (MEG) activity in participants (N = 24) who viewed images of objects presented at recognition threshold. Using multivariate analysis applied to sensor-level activity patterns recorded before stimulus presentation, we identified two neural processes influencing subsequent subjective recognition: a general process, which disregards stimulus category and correlates with pupil size, and a specific process, which facilitates category-specific recognition. The two processes are doubly-dissociable: the general process correlates with changes in criterion but not in sensitivity, whereas the specific process correlates with changes in sensitivity but not in criterion. Our findings reveal distinct mechanisms of how spontaneous neural activity influences perception and provide a framework to integrate previous findings.

Project description:Sensory deprivation, following a total loss of one sensory modality e.g. vision, has been demonstrated to result in compensatory plasticity. It is yet not known to which extent neural changes, e.g. higher resting-state activity in visual areas (cross-modal plasticity) as a consequence of blindness, reverse, when sight is restored. Here, we used functional MRI to acquire blood oxygen level-dependent resting-state activity during an eyes open and an eyes closed state in congenital cataract-reversal individuals, developmental cataract-reversal individuals, congenitally permanently blind individuals and sighted controls. The amplitude of low frequency fluctuation of the blood oxygen level-dependent signal-a neural marker of spontaneous brain activity during rest-was analyzed. In accordance with previous reports, in normally sighted controls we observed an increase in amplitude of low-frequency fluctuation during rest with the eyes open compared with rest with eyes closed in visual association areas and in parietal cortex but a decrease in auditory and sensorimotor regions. In congenital cataract-reversal individuals, we found an increase of the amplitude of slow blood oxygen level-dependent fluctuations in visual cortex during rest with eyes open compared with rest with eyes closed too but this increase was larger in amplitude than in normally sighted controls. In contrast, congenital cataract-reversal individuals lagged a similar increase in parietal regions and did not show the typical decrease of amplitude of low-frequency fluctuation in auditory cortex. Congenitally blind individuals displayed an overall higher amplitude in slow blood oxygen level-dependent fluctuations in visual cortex compared with sighted individuals and compared with congenital cataract-reversal individuals in the eyes closed condition. Higher amplitude of low-frequency fluctuation in visual cortex of congenital cataract-reversal individuals than in normally sighted controls during eyes open might indicate an altered excitatory-inhibitory balance of visual neural circuits. By contrast, the lower parietal increase and the missing downregulation in auditory regions suggest a reduced influence of the visual system on multisensory and the other sensory systems after restoring sight in congenitally blind individuals. These results demonstrate a crucial dependence of visual and multisensory neural system functioning on visual experience during a sensitive phase in human brain development.

Project description:Visual phenomena demonstrating striking perceptual disappearances of salient stimuli have fascinated researchers because of their utility in identifying neural processes that underlie subjective visibility and invisibility. Motion-induced blindness (MIB) is appealing for such purposes because it, like a class of ostensibly related paradigms such as binocular rivalry, features periods of unequivocal subjective disappearances despite constant physical stimulation. It remains unclear, however, exactly how the mechanisms that cause MIB are related to subjectively observed fluctuations in visual awareness. To address this question, we used continuous flash suppression (CFS) to present the MIB stimulus outside visual awareness. Results indicated that MIB occasionally reappeared from suppression with its salient yellow target absent. To quantify this observation, we measured reaction times (RTs) to detect the yellow dot target following visible or perceptually suppressed MIB and indeed found no difference in RTs between these conditions. We also provide evidence that MIB fluctuations can occur without attention. In sum, these experiments indicate that MIB fluctuations are effectively changes in stimulus strength, which under typical conditions result in unmistakable subjective disappearances, but are not inherently fluctuations in stimulus visibility. More broadly, these results challenge the assumed privileged link between bistable stimuli and visual awareness.

Project description:Complex neural circuits in the mammalian brain develop through a combination of genetic instruction and activity-dependent refinement. The relative role of these factors and the form of neuronal activity responsible for circuit development is a matter of significant debate. In the mammalian visual system, retinal ganglion cell projections to the brain are mapped with respect to retinotopic location and eye of origin. We manipulated the pattern of spontaneous retinal waves present during development without changing overall activity levels through the transgenic expression of ?2-nicotinic acetylcholine receptors in retinal ganglion cells of mice. We used this manipulation to demonstrate that spontaneous retinal activity is not just permissive, but instructive in the emergence of eye-specific segregation and retinotopic refinement in the mouse visual system. This suggests that specific patterns of spontaneous activity throughout the developing brain are essential in the emergence of specific and distinct patterns of neuronal connectivity.

Project description:Temporal recalibration (TR) may arise to realign asynchronous stimuli after exposure to a short, constant delay between voluntary movement and sensory stimulus. The objective of this study was to determine if awareness of the temporal lag between a motor response (i.e., a keypress) and a sensory event (i.e., a visual flash) is necessary for TR to occur. We further investigated whether manipulating the required motor and perceptual judgment tasks modified the influence of awareness on TR. Participants (n = 48) were randomly divided between two groups (Group 1: Aware and Group 2: Unaware). The Aware group was told of the temporal lag between their keypress and visual flash at the beginning of the experiment, whereas the Unaware group was not. All participants completed eight blocks of trials, in which the motor task (single or repetitive tap), perceptual judgment task (judging the temporal order of the keypress in relation to the visual flash or judging whether the two stimuli were simultaneous or not), and fixed temporal lag between keypress and visual flash (0 or 100 ms) varied. TR was determined by comparing judgments between corresponding blocks of trials in which the temporal lag was 0 or 100 ms. Results revealed that both the Aware and Unaware groups demonstrated a similar magnitude of TR across all motor and perceptual judgment tasks, such that the magnitude of TR did not vary between Aware and Unaware participants. These results suggest that awareness of a temporal lag does not influence the magnitude of TR achieved and that motor and perceptual judgment task demands do not modulate the influence of awareness on TR.

Project description:Incorporation of new neurons enables plasticity and repair of circuits in the adult brain. Adult neurogenesis is a key feature of the mammalian olfactory system, with new olfactory sensory neurons (OSNs) wiring into highly organized olfactory bulb (OB) circuits throughout life. However, neither when new postnatally generated OSNs first form synapses nor whether OSNs retain the capacity for synaptogenesis once mature, is known. Therefore, how integration of adult-born OSNs may contribute to lifelong OB plasticity is unclear. Here, we use a combination of electron microscopy, optogenetic activation and in vivo time-lapse imaging to show that newly generated OSNs form highly dynamic synapses and are capable of eliciting robust stimulus-locked firing of neurons in the mouse OB. Furthermore, we demonstrate that mature OSN axons undergo continuous activity-dependent synaptic remodelling that persists into adulthood. OSN synaptogenesis, therefore, provides a sustained potential for OB plasticity and repair that is much faster than OSN replacement alone.

Project description:ObjectiveTo investigate the spatial distribution characteristics of alterations in spontaneous brain activity in severe traumatic brain injury (sTBI) patients with disorders of consciousness (DOC), based on the mesocircuit theoretical framework, and to establish models for predicting recovery of consciousness.MethodsResting-state functional magnetic resonance imaging was employed to measure the mean fractional amplitude of low-frequency fluctuations (mfALFF) in sTBI patients with DOC and healthy controls, identifying differential brain regions for conducting gene and functional decoding analyses. Patients were classified into wake and DOC groups according to Extended Glasgow Outcome Score at 6 months. Furthermore, predictive models for consciousness recovery were developed using Nomogram and Linear Support Vector Machine (LSVM) based on mfALFF.ResultsIn total, 28 sTBI patients with DOC and 30 healthy controls were included, with no significant baseline differences between groups (P > 0.05). The results revealed increased mfALFF of subcortical Ascending Reticular Activating System and decreased cortical mfALFF (default mode network) in DOC patients within the framework of the mesocircuit model (FDR_P < 0.001, Clusters > 100). The study identified 2080 differentially expressed genes associated with reduced brain activity regions, indicating mechanisms involving synaptic function, the oxytocin signaling pathway, and GABAergic processes in DOC formation. In addition, significantly higher mfALFF values were observed in the left angular gyrus, supramarginal gyrus, and inferior parietal lobule of DOC group compared to the wake group (AlphaSim_P < 0.01, Cluster > 19). The Nomogram prediction model highlighted the pivotal role of these regions' activity levels in prognosis (AUC = 0.90). Validation using LSVM demonstrated robust predictive performance with an AUC of 0.90 and positive predictive values of 80% for wake and 83% for DOC.ConclusionsThis study offered crucial insights underlying DOC in sTBI patients, demonstrating the dissociation between cortical and subcortical brain activities. The findings supported the use of mfALFF as a robust and non-invasive biomarker for evaluating brain function and predicting recovery outcomes.
