Chromosomal and reproductive features of some Oriental and Australasian scale insects (Homoptera, Coccinea).

ABSTRACT: Fourteen species of scale insects from the families Margarodidae s.l., Pseudococcidae, Eriococcidae, and Coccidae were investigated for the first time in respect to karyotypes, genetic systems, modes of reproduction and general anatomy of the female reproductive system. One of the studied species, Steatococcus samaraius Morrison, 1927, showed hermaphroditic reproduction of the female-like specimens, the other species demonstrated bisexual reproduction with a peculiar "Lecanoid" heterochromatinization of the paternal set of chromosomes in male embryos or thelytocous parthenogenesis. Antonina parazonata Williams, 2004 and Saccharolecanium krugeri (Zehntner, 1897) are recorded here for the first time from Thailand, Antonina vietnamensis Williams, 2004 and Geococcus satellitum Williams, 2004 - for the first time from Laos.

General trends of chromosomal evolution in Aphidococca (Insecta, Homoptera, Aphidinea + Coccinea).

Project description:Parallel trends of chromosomal evolution in Aphidococca are discussed, based on the catalogue of chromosomal numbers and genetic systems of scale insects by Gavrilov (2007) and the new catalogue for aphids provided in the present paper. To date chromosome numbers have been reported for 482 species of scale insects and for 1039 species of aphids, thus respectively comprising about 6% and 24% of the total number of species. Such characters as low modal numbers of chromosomes, heterochromatinization of part of chromosomes, production of only two sperm instead of four from each primary spermatocyte, physiological sex determination, "larval" meiosis, wide distribution of parthenogenesis and chromosomal races are considered as a result of homologous parallel changes of the initial genotype of Aphidococca ancestors. From a cytogenetic point of view, these characters separate Aphidococca from all other groups of Paraneoptera insects and in this sense can be considered as additional taxonomic characters. In contrast to available paleontological data the authors doubt that Coccinea with their very diverse (and partly primitive) genetic systems may have originated later then Aphidinea with their very specialised and unified genetic system.

A contribution to the taxonomy, cytogenetics and reproductive biology of the genus Aclerda Signoret (Homoptera, Coccinea, Aclerdidae).

Project description:A new species of scale insects, Aclerda pseudozoysiae sp. n., is described and illustrated. The karyotypes and some aspects of reproductive biology and cytogenetics of the new species species and Aclerda takahashiiKuwana, 1932 were studied, representing the first data for the genus Aclerda Signoret, 1874 and the family Aclerdidae as a whole. Aclerda pseudozoysiae sp. n. has 2n=16, bisexual reproduction, and heterochromatinization of one haploid set of chromosomes in male stages of the life cycle, matching either a Lecanoid or a Comstockioid genetic system. Aclerda takahashii demonstrates 2n=18 and unusual type of parthenogenesis with diploid and haploid embryos (inside each gravid female) without heterochromatinization. Both species are ovoviviparous; all stages of embryonic development occur inside the mother's body.

Cytogenetic and taxonomic studies of some legless mealybugs (Homoptera, Coccinea, Pseudococcidae).

Project description:A new monotypic genus and species, Komodesia circuliplurimagen. et sp. n., from Flores Is. (Indonesia) and the new species, Antonina diversiglandulosasp. n., from Southern Thailand are described and illustrated. Chromosomes of these species and also the ones of Antonina purpurea Signoret, 1872 and Antonina thaiensis Takahashi, 1942 are studied for the first time: 2n = 30, 20, 12 and 22+Bs correspondingly; the male embryos of all four species demonstrate Lecanoid paternal heterochromatinization of one haploid set of chromosomes. The karyotypes of three widely distributed species, Antonina pretiosa Ferris, 1953, Antonina graminis (Maskell, 1897) and Chaetococcus bambusae (Maskell, 1893), are studied based on material from other regions in comparison with previously published data. Photographs of the karyotypes are provided for the first time for all seven species. The terminological problems connected with the identification and naming of the three scale insect genetic systems, Lecanoid, Comstockioid and Diaspidoid, are discussed.

Positive selection in glycolysis among Australasian stick insects.

Project description:BACKGROUND: The glycolytic pathway is central to cellular energy production. Selection on individual enzymes within glycolysis, particularly phosphoglucose isomerase (Pgi), has been associated with metabolic performance in numerous organisms. Nonetheless, how whole energy-producing pathways evolve to allow organisms to thrive in different environments and adopt new lifestyles remains little explored. The Lanceocercata radiation of Australasian stick insects includes transitions from tropical to temperate climates, lowland to alpine habitats, and winged to wingless forms. This permits a broad investigation to determine which steps within glycolysis and what sites within enzymes are the targets of positive selection. To address these questions we obtained transcript sequences from seven core glycolysis enzymes, including two Pgi paralogues, from 29 Lanceocercata species. RESULTS: Using maximum likelihood methods a signature of positive selection was inferred in two core glycolysis enzymes. Pgi and Glyceraldehyde 3-phosphate dehydrogenase (Gaphd) genes both encode enzymes linking glycolysis to the pentose phosphate pathway. Positive selection among Pgi paralogues and orthologues predominately targets amino acids with residues exposed to the protein's surface, where changes in physical properties may alter enzyme performance. CONCLUSION: Our results suggest that, for Lancerocercata stick insects, adaptation to new stressful lifestyles requires a balance between maintaining cellular energy production, efficiently exploiting different energy storage pools and compensating for stress-induced oxidative damage.

Protomedetera, a new genus from the Oriental and Australasian realms (Diptera, Dolichopodidae, Medeterinae).

Project description:Protomedeteragen. n. (Diptera: Dolichopodidae), a new genus of the subfamily Medeterinae, is described from the Oriental and Australasian realms based on four new species. Protomedetera singaporensis Grootaert & Tang, sp. n. is designated as type of the new genus. The genus is peculiar because of the small body size, the small globular first flagellomere (postpedicel), the simple male genitalia with indistinct or small epandrial lobe and half-hidden cercus. The following four new species are described and illustrated: P. biconvexasp. n., P. bisetasp. n., P. glabrasp. n., and P. singaporensissp. n. A key to the species of the new genus is provided.

Reproductive trade-offs maintain bract color polymorphism in Scarlet Indian paintbrush (Castilleja coccinea).

Project description:Populations of scarlet Indian paintbrush (Castilleja coccinea) in the Midwestern United States exhibit a bract color polymorphism, with each population having predominantly yellow or scarlet bracts. We investigated a possible mechanism for this maintenance of bract color polymorphism in C. coccinea by conducting hand-pollination experiments in two nearby populations, one predominantly yellow and one predominantly scarlet. The hand-pollination treatments were either self-pollination or cross pollination using pollen from within and between populations. Both color morphs were used as pollen donors for the within and between crosses. We found that both color morphs of C. coccinea were self-compatible. When the scarlet morph was the maternal plant it had higher seed set. When pollinators were excluded, the yellow morph outperformed the scarlet morph in fruit set and seed set. The apparent trade-offs between a higher reproductive output in the scarlet morph and a reproductive assurance advantage in the yellow morph may explain the maintenance of the polymorphism in C. coccinea. While many previous studies have provided evidence for pollinator preference playing a role in floral color polymorphism, the results of the current study indicate that reproductive assurance, which would be important for fluctuations in pollinator abundance or colonizing new areas, may act as a selective agent to maintain such polymorphisms.

Hydrocarbon divergence and reproductive isolation in Timema stick insects.

Project description:BackgroundIndividuals commonly prefer certain trait values over others when choosing their mates. If such preferences diverge between populations, they can generate behavioral reproductive isolation and thereby contribute to speciation. Reproductive isolation in insects often involves chemical communication, and cuticular hydrocarbons, in particular, serve as mate recognition signals in many species. We combined data on female cuticular hydrocarbons, interspecific mating propensity, and phylogenetics to evaluate the role of cuticular hydrocarbons in diversification of Timema walking-sticks.ResultsHydrocarbon profiles differed substantially among the nine analyzed species, as well as between partially reproductively-isolated T. cristinae populations adapted to different host plants. In no-choice trials, mating was more likely between species with similar than divergent hydrocarbon profiles, even after correcting for genetic divergences. The macroevolution of hydrocarbon profiles, along a Timema species phylogeny, fits best with a punctuated model of phenotypic change concentrated around speciation events, consistent with change driven by selection during the evolution of reproductive isolation.ConclusionAltogether, our data indicate that cuticular hydrocarbon profiles vary among Timema species and populations, and that most evolutionary change in hydrocarbon profiles occurs in association with speciation events. Similarities in hydrocarbon profiles between species are correlated with interspecific mating propensities, suggesting a role for cuticular hydrocarbon profiles in mate choice and speciation in the genus Timema.

Fungal Associates of Soft Scale Insects (Coccomorpha: Coccidae).

Project description:Ophiocordyceps fungi are commonly known as virulent, specialized entomopathogens; however, recent studies indicate that fungi belonging to the Ophiocordycypitaceae family may also reside in symbiotic interaction with their host insect. In this paper, we demonstrate that Ophiocordyceps fungi may be obligatory symbionts of sap-sucking hemipterans. We investigated the symbiotic systems of eight Polish species of scale insects of Coccidae family: Parthenolecanium corni, Parthenolecanium fletcheri, Parthenolecanium pomeranicum, Psilococcus ruber, Sphaerolecanium prunasti, Eriopeltis festucae, Lecanopsis formicarum and Eulecanium tiliae. Our histological, ultrastructural and molecular analyses showed that all these species host fungal symbionts in the fat body cells. Analyses of ITS2 and Beta-tubulin gene sequences, as well as fluorescence in situ hybridization, confirmed that they should all be classified to the genus Ophiocordyceps. The essential role of the fungal symbionts observed in the biology of the soft scale insects examined was confirmed by their transovarial transmission between generations. In this paper, the consecutive stages of fungal symbiont transmission were analyzed under TEM for the first time.

Large-scale analysis reveals that the genome features of simple sequence repeats are generally conserved at the family level in insects.

Project description:Simple sequence repeats (SSR), also called microsatellites, have been widely used as genetic markers, and have been extensively studied in some model insects. At present, the genomes of more than 100 insect species are available. However, the features of SSRs in most insect genomes remain largely unknown.We identified 15.01 million SSRs across 136 insect genomes. The number of identified SSRs was positively associated with genome size in insects, but the frequency and density per megabase of genomes were not. Most insect SSRs (56.2-93.1%) were perfect (no mismatch). Imperfect (at least one mismatch) SSRs (average length 22-73 bp) were longer than perfect SSRs (16-30 bp). The most abundant insect SSRs were the di- and trinucleotide types, which accounted for 27.2% and 22.0% of all SSRs, respectively. On average, 59.1%, 36.8%, and 3.7% of insect SSRs were located in intergenic, intronic, and exonic regions, respectively. The percentages of various types of SSRs were similar among insects from the same family. However, they were dissimilar among insects from different families within orders. We carried out a phylogenetic analysis using the SSR frequencies. Species from the same family were generally clustered together in the evolutionary tree. However, insects from the same order but not in the same family did not cluster together. These results indicated that although SSRs undergo rapid expansions and contractions in different populations of the same species, the general genomic features of insect SSRs remain conserved at the family level.Millions of insect SSRs were identified and their genome features were analyzed. Most insect SSRs were perfect and were located in intergenic regions. We presented evidence that the variance of insect SSRs accumulated after the differentiation of insect families.

Nonadaptive host-use specificity in tropical armored scale insects.

Project description:Most herbivorous insects are diet specialists in spite of the apparent advantages of being a generalist. This conundrum might be explained by fitness trade-offs on alternative host plants, yet the evidence of such trade-offs has been elusive. Another hypothesis is that specialization is nonadaptive, evolving through neutral population-genetic processes and within the bounds of historical constraints. Here, we report on a striking lack of evidence for the adaptiveness of specificity in tropical canopy communities of armored scale insects. We find evidence of pervasive diet specialization, and find that host use is phylogenetically conservative, but also find that more-specialized species occur on fewer of their potential hosts than do less-specialized species, and are no more abundant where they do occur. Of course local communities might not reflect regional diversity patterns. But based on our samples, comprising hundreds of species of hosts and armored scale insects at two widely separated sites, more-specialized species do not appear to outperform more generalist species.
