Project description:The physical-chemical properties of activated sludge, such as flocculating ability, hydrophobicity, surface charge, settleability, dewaterability and bacteria extracellular polymer substances (EPS), play vital roles in the normal operation of wastewater treatment plants (WWTPs). The nanoparticles released from commercial products will enter WWTPs and can induce potential adverse effects on activated sludge. This paper focused on the effects of copper nanoparticles (CuNPs) on these specific physical-chemical properties of activated sludge. It was found that most of these properties were unaffected by the exposure to lower CuNPs concentration (5 ppm), but different observation were made at higher CuNPs concentrations (30 and 50 ppm). At the higher CuNPs concentrations, the sludge surface charge increased and the hydrophobicity decreased, which were attributed to more Cu2+ ions released from the CuNPs. The carbohydrate content of EPS was enhanced to defense the toxicity of CuNPs. The flocculating ability was found to be deteriorated due to the increased cell surface charge, the decreased hydrophobicity, and the damaged cell membrane. The worsened flocculating ability made the sludge flocs more dispersed, which further increased the toxicity of the CuNPs by increasing the availability of the CuNPs to the bacteria present in the sludge. Further investigation indicated that the phosphorus removal efficiency decreased at higher CuNPs concentrations, which was consistent with the deteriorated physical-chemical properties of activated sludge. It seems that the physical-chemical properties can be used as an indicator for determining CuNPs toxicity to the bacteria in activated sludge. This work is important because bacteria toxicity effects to the activated sludge caused by nanoparticles may lead to the deteriorated treatment efficiency of wastewater treatment, and it is therefore necessary to find an easy way to indicate this toxicity.

Project description:We recently reported a novel synthesis of copper nanoparticles from copper sulphate utilizing the charge-compensatory effect of ionic liquid [bmim]BF(4) and ethylene glycol. The nanoparticles were characterized and found to be stable for one year. Here we hypothesize that the stabilized nanoparticles should be able to catalyze one-pot multicomponent organic reactions. We show that the nanoparticles catalyzed Biginelli reaction at room temperature to give the product 3,4-dihydopyrimidinone (>90% yield in ~15 minutes) from aldehydes, ?-diketoester (ethylacetoacetate) and urea (or thiourea). ). Remarkably, such high yields and rapid kinetics was found to be independent of the electronic density on the reactant aryl-aldehyde. This was probably because even the surface-active particles reacted faster in the presence of ionic liquid as compared to conventional methods. The heterocyclic dihydropyrimidinones (DHPMs) and their derivatives are widely used in natural and synthetic organic chemistry due to their wide spectrum of biological and therapeutic properties (resulting from their antibacterial, antiviral, antitumor and anti-inflammatory activities. Our method has an easy work-up procedure and the nanoparticles could be recycled with minimal loss of efficiency.

Project description:Plasmon-mediated shape transformation from quasi-spherical silver nanoparticles (AgNPs) to silver nanoprisms (AgNPrs) and decahedral silver nanoparticles (D-AgNPs) under irradiation of blue LEDs (λ = 456 ± 12 nm, 80 mW/cm2) was studied at temperatures ranging between 60, 40, 30, 20, 10, and 0 °C. It was found that reaction temperature affected transformation rates and influenced the morphology distribution of final products. The major products synthesized at temperatures between 60 °C and 0 °C were AgNPrs and D-AgNPs, respectively. The D-AgNPs synthesized at such low temperatures are unstable and become blunt when light irradiation is removed after the photochemical synthesis. These blunt nanoparticles with pentagonal multiple-twinned structures can be further used as the seeds to reconstruct complete D-AgNPs after irradiating blue LEDs at various bath temperatures. Our results showed that these rebuilt D-AgNPs are much more stable when at higher bath temperatures. Furthermore, the rebuilt D-AgNPs (edge lengths ~41 nm) can grow into larger D-AgNPs (edge lengths ~53 nm) after the irradiation of green LEDs. Surface-enhanced Raman spectra of CV in AgNP colloids showed that D-AgNP colloids have better SERS enhancements factors than AgNPrs.

Project description:Metal nanoparticle synthesis is an interesting area in nanotechnology due to their remarkable optical, magnetic, electrical, catalytic and biomedical properties, but there needs to develop clean, non-toxic and environmental friendly methods for the synthesis and assembly of nanoparticles. Biological agents in the form of microbes have emerged up as efficient candidates for nanoparticle synthesis due to their extreme versatility to synthesize diverse nanoparticles with varying size and shape. In the present study, an eco favorable method for the biosynthesis of silver nanoparticles using marine bacterial isolate has been attempted. Very interestingly, molecular identification proved it as a strain of Ochrobactrum anhtropi. In addition, the isolate was found to have the potential to form silver nanoparticles intracellularly at room temperature within 24 h. The biosynthesized silver nanoparticles were characterized by UV-Vis spectroscopy, transmission electron microscope (TEM) and scanning electron microscope (SEM). The UV-visible spectrum of the aqueous medium containing silver nanoparticles showed a peak at 450 nm corresponding to the plasmon absorbance of silver nanoparticles. The SEM and TEM micrographs revealed that the synthesized silver nanoparticles were spherical in shape with a size range from 38 nm - 85 nm. The silver nanoparticles synthesized by the isolate were also used to explore its antibacterial potential against pathogens like Salmonella Typhi, Salmonella Paratyphi, Vibrio cholerae and Staphylococcus aureus.

Project description:Polyester (PET) fabrics are widely applied in functional textiles due to their outstanding properties such as high strength, dimensional stability, high melting point, low cost, recyclability, and flexibility. Nevertheless, the lack of polar groups in the PET structure makes its coloration and functionalization difficult. The present work reports the one-step in situ synthesis of copper nanoparticles (CuNPs) onto the PET fabric employing sodium hypophosphate and ascorbic acid as reducing and stabilizing agents, at acidic (pH 2) and alkaline pH (pH 11). This synthesis (i) used safer reagents when compared with traditional chemicals for CuNP production, (ii) was performed at a moderate temperature (85 °C), and (iii) used no protective inert gas. The dielectric barrier discharge (DBD) plasma was used as an environmentally friendly method for the surface functionalization of PET to enhance the adhesion of CuNPs. The size of the CuNPs in an alkaline reaction (76-156 nm for not treated and 93.4-123 nm for DBD plasma-treated samples) was found to be smaller than their size in acidic media (118-310 nm for not treated and 249-500 nm for DBD plasma-treated samples), where the DBD plasma treatment promoted some agglomeration. In acidic medium, metallic copper was obtained, and a reddish color became noticeable in the textile. In alkaline medium, copper(I) oxide (Cu2O) was detected, and the PET samples exhibited a yellow color. The PET samples with CuNPs presented improved ultraviolet protection factor values. Finally, a minimal concentration of copper salt was studied to obtain the optimized antibacterial effect against Staphylococcus aureus and Escherichia coli. The functionalized samples showed strong antibacterial efficacy using low-concentration solutions in the in situ synthesis (2.0 mM of copper salt) and even after five washing cycles. The DBD plasma treatment improved the antibacterial action of the samples prepared in the alkaline medium.

Project description:Biosynthesis of copper oxide nanoparticles (CuONPs) in a cost-effective and eco-friendly way has gained its importance. CuONPs has been prepared from copper sulfate by using Adiantum lunulatum whole plant extract. CuONPs have been characterized by X-ray diffraction, Fourier transform infrared spectroscopic, transmission electron microscope, etc. Mono-disperse, spherical, pure, and highly stable CuONPs have formed with an average diameter of 6.5 ± 1.5 nm. Biosynthesized CuONPs at different concentrations were applied to seeds of Lens culinaris. Physiological characteristics were investigated in the germinated seeds. Roots obtained from the seeds treated with 0.025 mgmL-1 concentration of CuONPs showed highest activity of different defence enzymes and total phenolics. However, at higher concentration it becomes close to control. It showed gradual increase of antioxidative enzymes, in accordance with the increasing dose of CuONPs. Likewise, lipid peroxidation and proline content gradually increased with the increasing concentration. Reactive oxygen species and nitric oxide generation was also altered due to CuONPs treatment indicating stress signal transduction. Finally, this study provides a new approach of the production of valuable CuONPs, is a unique, economical, and handy tool for large scale saleable production which can also be used as a potent plant defence booster instead of other commercial uses.

Project description:Introduction:Copper oxide nanoparticles (CuO-NPs) are widely used as feed additives for livestock and poultry and implicated in many biomedical applications; however, overload of copper NPs induces various toxicological changes and dysfunction of animal's organs. Thus, this study was designed to evaluate the comparative toxicological effects of biologically and chemically synthesized CuO-NPs on mice. Methods:Transmission electron microscopy (TEM), X-ray diffraction (XRD) and Fourier-transform infrared spectroscopy (FT-IR) were used to characterize the sizes, shapes and functional groups of CuO-NPs. Forty-five mice were randomly allocated into three groups. Control group received distilled water. The second group was administered a single dose of biologically synthesized CuO-NPs (500 mg/kg bw) orally. The third group was administered a single dose of chemically synthesized CuO-NPs (500 mg/kg bw) orally. Results:TEM revealed that biologically synthesized NPs were spherical in shape, whereas chemically synthesized NPs were spherical or elongated in shape. XRD showed that the size of biologically synthesized NPs ranged from 4.14 to 12.82 nm and that of chemically synthesized NPs ranged from 4.06 to 26.82 nm. FT-IR spectroscopy indicated that the peaks appeared between 779 cm-1 and 425 cm-1 in biologically synthesized NPs and between 858 cm-1 and 524 cm-1 in chemically synthesized NPs were for Cu-O nanostructure. Four mice died due to administration of biologically synthesized CuO-NPs. Both biologically and chemically synthesized CuO-NPs induced leukocytosis, elevated serum activities of alanine aminotransferase and aspartate aminotransferase and serum levels of urea and creatinine and increased P53 mRNA and caspase-3 protein expressions in hepatic tissues. Moreover, CuO-NPs induced degenerative and necrotized changes in hepatic, renal and splenic tissues. Biochemical, apoptotic and pathological changes were more serious in mice administered with biologically synthesized CuO-NPs. Conclusion:This study indicated that a high dose of biologically and chemically synthesized CuO-NPs induced adverse effects on hepatic, renal and splenic tissues. At the same dose level, the biologically synthesized CuO-NPs evoked more potent toxic effects than the chemically synthesized CuO-NPs.

Project description:Background:Nanoparticles (NPs) have been emerging as potential players in modern medicine with clinical applications ranging from therapeutic purposes to antimicrobial agents. However, before applications in medical agents, some in vitro studies should be done to explore their biological responses. Aim:In this study, protein binding, anticancer and antibacterial activates of zero valent iron (ZVFe) were explored. Materials and methods:ZVFe nanoparticles were synthesized and fully characterized by X-ray diffraction, field-emission scanning electron microscope, and dynamic light scattering analyses. Afterward, the interaction of ZVFe NPs with human serum albumin (HSA) was examined using a range of techniques including intrinsic fluorescence, circular dichroism, and UV-visible spectroscopic methods. Molecular docking study was run to determine the kind of interaction between ZVFe NPs and HSA. The anticancer influence of ZVFe NPs on SH-SY5Y was examined by MTT and flow cytometry analysis, whereas human white blood cells were used as the control cell. Also, the antibacterial effect of ZVFe NPs was examined on Pseudomonas aeruginosa (ATCC 27853), Escherichia coli (ATCC 25922), and Staphylococcus aureus (ATCC 25923). Results:X-ray diffraction, transmission electron microscope, and dynamic light scattering analyses verified the synthesis of ZVFe NPs in a nanosized diameter. Fluorescence spectroscopy analysis showed that ZVFe NPs spontaneously formed a complex with HSA through hydrogen bonds and van der Waals interactions. Also, circular dichroism spectroscopy study revealed that ZVFe NPs did not change the secondary structure of HSA. Moreover, UV-visible data presented that melting temperature (Tm) of HSA in the absence and presence of ZVFe NPs was almost identical. Molecular dynamic study also showed that ZVFe NP came into contact with polar residues on the surface of HSA molecule. Cellular assays showed that ZVFe NPs can induce cell mortality in a dose-dependent manner against SH-SY5Y cells, whereas these NPs did not trigger significant cell mortality against normal white bloods in the concentration range studied (1-100 µg/mL). Antibacterial assays showed a noteworthy inhibition on both bacterial strains. Conclusion:In conclusion, it was revealed that ZVFe NPs did not induce a substantial influence on the structure of protein and cytotoxicity against normal cell, whereas they derived significant anticancer and antibacterial effects.

Project description:In recent years, lignin has drawn increasing attention for different applications due to its intrinsic antibacterial and antioxidant properties, coupled with biodegradability and biocompatibility. However, chemical modification or combination with metals is usually required to increase its antimicrobial functionality and produce biobased added-value materials for applications wherein bacterial growth should be avoided, such as biomedical and food industries. In this work, a sonoenzymatic approach for the simultaneous functionalization and nanotransformation of lignin to prepare metal-free antibacterial phenolated lignin nanoparticles (PheLigNPs) is developed. The grafting of tannic acid, a natural phenolic compound, onto lignin was achieved by an environmentally friendly approach using laccase oxidation upon the application of high-intensity ultrasound to rearrange lignin into NPs. PheLigNPs presented higher antibacterial activity than nonfunctionalized LigNPs and phenolated lignin in the bulk form, indicating the contribution of both the phenolic content and the nanosize to the antibacterial activity. Studies on the antibacterial mode of action showed that bacteria in contact with the functionalized NPs presented decreased metabolic activity and high levels of reactive oxygen species (ROS). Moreover, PheLigNPs demonstrated affinity to the bacterial surface and the ability to cause membrane destabilization. Antimicrobial resistance studies showed that the NPs did not induce resistance in pathogenic bacteria, unlike traditional antibiotics.

Project description:Biosynthesis of zinc oxide nanoparticles has been reported using Cocos nucifera leaf (CNL) extract along with estimation of their antimicrobial potential before and after calcination using different micro-organisms. UV-visible spectra of ZnO nanoparticles showed absorption maxima at 383 nm and 363 nm, respectively, with 3.237 eV and 3.416 eV, respectively, as the corresponding band gap energies. FESEM and TEM images showed spherical morphologies of ZnO nanoparticles within the size range 109-215 nm. XRD analysis confirmed the formation of hexagonal wurtzite structures. ATR-IR spectra revealed the presence of stretching vibrations of N-H, O-H, C=C, C=O and NH2 groups along with C-H and N-H deformation involving biomolecules from CNL extract responsible for reduction and stabilization of ZnO nanoparticles. Uncalcinated ZnO nanoparticles displayed antibacterial activities only against S. aureus and P. aeruginosa whereas calcinated ZnO nanoparticles did not show antibacterial activities against E. coli, S. aureus, P. aeruginosa and B. subtilis. ZnO nanoparticles were not active against Penicillium spp., Fusarium oxysporum, Aspergillus flavus, Rhizoctonia solani as well as HCT-116 cancer cells before as well as after calcination. Antimicrobial nature and biocompatibility of ZnO nanoparticles were influenced by different parameters of the nanoparticles along with micro-organisms and the human cells. Non-antimicrobial properties of ZnO nanoparticles can be treated as a pre-requisite for its biocompatibility due to its inert nature. These ZnO nanoparticles can serve a dual purpose by facilitating use as antibacterial agent against susceptible micro-organisms as well as a biocompatible carrier molecule in drug delivery applications.Supplementary informationThe online version contains supplementary material available at 10.1007/s13205-022-03110-9.
