Project description:Mosquitoes account for a significant burden of morbidity and mortality globally. Despite evidence of (1) imminent anthropogenic climate and environmental changes, (2) vector-pathogen spatio-temporal dynamics and (3) emerging and re-emerging mosquito borne infections, public knowledge on mosquito bio-ecology remain scant. In particular, knowledge, attitude and practices (KAPs) on mosquitoes are often neglected despite otherwise expensive remedial efforts against consequent infections and other indirect effects associated with disease burden. To gather baseline KAPs that identify gaps for optimising vector-borne disease control, we surveyed communities across endemic and non-endemic malaria sub-districts (Botswana). The study revealed limited knowledge of mosquitoes and their infections uniformly across endemic and non-endemic areas. In addition, a significant proportion of respondents were concerned about mosquito burdens, although their level of personal, indoor and environmental protection practices varied significantly across sub-districts. Given the limited knowledge displayed by the communities, this study facilitates bridging KAP gaps to minimise disease burdens by strengthening public education. Furthermore, it provides a baseline for future studies in mosquito bio-ecology and desirable control practices across differential spheres of the rural-urban lifestyle, with implications for enhanced livelihoods as a consequence of improved public health.

Project description:Using human evaluation of 100,000 words spread across 24 corpora in 10 languages diverse in origin and culture, we present evidence of a deep imprint of human sociality in language, observing that (i) the words of natural human language possess a universal positivity bias, (ii) the estimated emotional content of words is consistent between languages under translation, and (iii) this positivity bias is strongly independent of frequency of word use. Alongside these general regularities, we describe interlanguage variations in the emotional spectrum of languages that allow us to rank corpora. We also show how our word evaluations can be used to construct physical-like instruments for both real-time and offline measurement of the emotional content of large-scale texts.

Project description:Emotions play an important role in human communication, and the daily-life interactions of young children often include situations that require the verbalization of emotional states with verbal means, e.g., with emotion terms. Through them, one can express own emotional states and those of others. Thus, the acquisition of emotion terms allows children to participate more intensively in social contexts - a basic requirement for learning new words and for elaborating socio-emotional skills. However, little is known about how children acquire and process this specific word category, which is positioned between concrete and abstract words. In particular, the influence of valence on emotion word processing during childhood has not been sufficiently investigated. Previous research points to an advantage of positive words over negative and neutral words in word processing. While previous studies found valence effects to be influenced by factors such as arousal, frequency, concreteness, and task, it is still unclear if and how valence effects are also modified by age. The present study compares the performance of children aged from 5 to 12 years and adults in two experimental tasks: lexical decision (word or pseudoword) and emotional categorization (positive or negative). Stimuli consisted of 48 German emotion terms (24 positive and 24 negative) matched for arousal, concreteness, age of acquisition, word class, word length, morphological complexity, frequency, and neighborhood density. Results from both tasks reveal two developmental trends: First, with increasing age children responded faster and more correctly, suggesting that emotion vocabulary gradually becomes more stable and differentiated during middle childhood. Second, the influence of valence varied with age: younger children (5- and 6-year-olds) showed significantly higher performance levels for positive emotion terms compared to negative emotion terms, whereas older children and adults did not. This age-related valence effect in emotion word processing will be discussed with respect to linguistic and methodological aspects.

Project description:BackgroundDuring the COVID-19 pandemic, the Ministry of Health asked Singapore's private general practitioners (GPs) to perform swab testing in their clinics, but some GPs had concerns about swabber protection. Our aim was to develop a swabbing booth to address these concerns.MethodsWe developed a prototype with potential GP users using a human-centred design approach and piloted it with 10 GP clinics. The pilot was then extended to 170 GP clinics around Singapore. These GPs were then surveyed on user satisfaction.ResultsNinety-three GPs (54%) responded. The majority (75%) practiced in public residential estates in small practices (mean 1.95 doctors). 86% requested the booth to enhance swabber protection. 74% "would recommend" or "would strongly recommend" the booth to colleagues. 79% continue to use the booth to conduct swab tests. 92% liked that it offered swabber protection. 71% liked that the booth created a separate space for swabbing and 64% liked its ease of disinfection. 47% started swabbing only after receiving the booth and 58% said the booth was "important" or "very important" to their decision to participate in swab testing. However, 34% disliked that it took up too much space and the most frequently critiqued area was the gloves.ConclusionThe human-centred design approach generated a product that had high user satisfaction, addressed GPs' concerns of swabber protection and increased GPs' participation in swab testing. The booth may be useful where GPs are concerned about swabber protection and space is limited.

Project description:Self-positivity bias is one of the well-studied psychological phenomena, however, little is known about the bias in the specific dimension on social interaction, which we called herein interpersonal self-positivity bias-people tend to evaluate themselves more positively on social interactions, prefer to be included rather than to be excluded by others. In the present study, we used a modified self-reference task associated with N400 to verify such bias and explore whether impoverished social interaction (loneliness) could modulate it. Findings showed that exclusion verbs elicited larger N400 amplitudes than inclusion verbs, suggesting that most people have interpersonal self-positivity bias. However, loneliness was significantly correlated with N400 effect, showing those with high scores of loneliness had smaller differences in the N400 than those with lower scores. These findings indicated impoverished social interaction weakens interpersonal self-positivity bias; however, the underlying mechanisms need to be explored in future research.

Project description:The negativity bias has been shown in many fields, including in face processing. We assume that this bias stems from the potential threat inlayed in the stimuli (e.g., negative moral behaviors) in previous studies. In the present study, we conducted one behavioral and one event-related potentials (ERPs) experiments to test whether the positivity bias rather than negativity bias will arise when participants process information whose negative aspect involves no threat, i.e., the ability information. In both experiments, participants first completed a valence rating (negative-to-positive) of neutral facial expressions. Further, in the learning period, participants associated the neutral faces with high-ability, low-ability, or control sentences. Finally, participants rated these facial expressions again. Results of the behavioral experiment showed that compared with pre-learning, the expressions of the faces associated with high ability sentences were classified as more positive in the post-learning expression rating task, and the faces associated with low ability sentences were evaluated as more negative. Meanwhile, the change in the high-ability group was greater than that of the low-ability group. The ERP data showed that the faces associated with high-ability sentences elicited a larger early posterior negativity, an ERP component considered to reflect early sensory processing of the emotional stimuli, than the faces associated with control sentences. However, no such effect was found in faces associated with low-ability sentences. To conclude, high ability sentences exerted stronger influence on expression perception than did low ability ones. Thus, we found a positivity bias in this ability-related facial perceptual task. Our findings demonstrate an effect of valenced ability information on face perception, thereby adding to the evidence on the opinion that person-related knowledge can influence face processing. What's more, the positivity bias in non-threatening surroundings increases scope for studies on processing bias.

Project description:Background. Health and social management of the SARS-CoV-2 epidemic, responsible for the COVID-19 disease, requires both screening tools and diagnostic procedures. Reliable screening tests aim at identifying (truely) infectious individuals that can spread the viral infection and therefore are essential for tracing and harnessing the epidemic diffusion. Instead, diagnostic tests should supplement clinical and radiological findings, thus helping in establishing the diagnosis. Several analytical assays, mostly using RT-PCR-based technologies, have become commercially available for healthcare workers and clinical laboratories. However, such tests showed some critical limitations, given that a relevant number of both false-positive and false-negative cases have been so far reported. Moreover, those analytical techniques demonstrated to be significantly influenced by pre-analytical biases, while the sensitivity showed a dramatic time dependency. Aim. Herein, we critically investigate limits and perspectives of currently available RT-PCR techniques, especially when referring to the required performances in providing reliable epidemiological and clinical information. Key Concepts. Current data cast doubt on the use of RT-PCR swabs as a screening procedure for tracing the evolution of the current SARS-COV-2 pandemic. Indeed, the huge number of both false-positive and false-negative results deprives the trustworthiness of decision making based on those data. Therefore, we should refine current available analytical tests to quickly identify individuals able to really transmit the virus, with the aim to control and prevent large outbreaks.

Project description:When a person makes a movement, a motor error is typically observed that then drives motor planning corrections on subsequent movements. This error correction, quantified as a trial-by-trial adaptation rate, provides insight into how the nervous system is operating, particularly regarding how much confidence a person places in different sources of information such as sensory feedback or motor command reproducibility. Traditional analysis has required carefully controlled laboratory conditions such as the application of perturbations or error clamping, limiting the usefulness of motor analysis in clinical and everyday environments. Here we focus on error adaptation during unperturbed and naturalistic movements. With increasing motor noise, we show that the conventional estimation of trial-by-trial adaptation increases, a counterintuitive finding that is the consequence of systematic bias in the estimate due to noise masking the learner's intention. We present an analytic solution relying on stochastic signal processing to reduce this effect of noise, producing an estimate of motor adaptation with reduced bias. The result is an improved estimate of trial-by-trial adaptation in a human learner compared to conventional methods. We demonstrate the effectiveness of the new method in analyzing simulated and empirical movement data under different noise conditions.

Project description:In this paper, we advocate and expand upon a previously described monitoring strategy for efficient and robust estimation of disease prevalence and case numbers within closed and enumerated populations such as schools, workplaces, or retirement communities. The proposed design relies largely on voluntary testing, which is notoriously biased (e.g., in the case of coronavirus disease 2019) due to nonrepresentative sampling. The approach yields unbiased and comparatively precise estimates with no assumptions about factors underlying selection of individuals for voluntary testing, building on the strength of what can be a small random sampling component. This component enables the use of a recently proposed "anchor stream" estimator, a well-calibrated alternative to classical capture-recapture (CRC) estimators based on 2 data streams. We show that this estimator is equivalent to a direct standardization based on "capture," that is, selection (or not) by the voluntary testing program, made possible by means of a key parameter identified by design. This equivalency simultaneously allows for novel 2-stream CRC-like estimation of general mean values (e.g., means of continuous variables like antibody levels or biomarkers). For inference, we propose adaptations of Bayesian credible intervals when estimating case counts and bootstrapping when estimating means of continuous variables. We use simulations to demonstrate significant precision benefits relative to random sampling alone.

Project description:Next-generation sequencing (NGS) enables clinical microbiology assays such as molecular typing of bacterial isolates which is now routinely applied for infection control and epidemiology. Additionally, feasibility for NGS-based identification of antimicrobial resistance (AMR) markers as well as genetic prediction of antibiotic susceptibility testing results has been demonstrated. Various bioinformatics approaches enabling NGS-based clinical microbiology assays exist, but standardized, computationally efficient and scalable sample-to-results workflows including validated quality control parameters are still lacking. Bioinformatics analysis workflows based on k-mers have been shown to allow for fast and efficient analysis of large genomics data sets as obtained from microbial sequencing applications. We here demonstrate applicability of k-mer based clinical microbiology assays for whole-genome sequencing (WGS) including variant calling, taxonomic identification, bacterial typing as well as AMR marker detection. The wet-lab and dry-lab workflows were developed and validated in line with Clinical Laboratory Improvement Act (CLIA) guidelines for laboratory-developed tests (LDTs) on multi-drug resistant ESKAPE pathogens. The developed k-mer based workflow demonstrated ?99.39% repeatability, ?99.09% reproducibility and ?99.76% accuracy for variant calling and applied assays as determined by intra-day and inter-day triplicate measurements. The limit of detection (LOD) across assays was found to be at 20× sequencing depth and 15× for AMR marker detection. Thorough benchmarking of the k-mer based workflow revealed analytical performance criteria are comparable to state-of-the-art alignment based workflows across clinical microbiology assays. Diagnostic sensitivity and specificity for multilocus sequence typing (MLST) and phylogenetic analysis were 100% for both approaches. For AMR marker detection, sensitivity and specificity were 95.29 and 99.78% for the k-mer based workflow as compared to 95.17 and 99.77% for the alignment-based approach. Summarizing, results illustrate that k-mer based analysis workflows enable a broad range of clinical microbiology assays, potentially not only for WGS-based typing and AMR gene detection but also genetic prediction of antibiotic susceptibility testing results.
