Here we report on chelating ligands for Signal Amplification By Reversible Exchange (SABRE) catalysts that permit hyperpolarisation on otherwise sterically hindered substrates. We demonstrate <sup>1</sup>H enhancements of ∼100-fold over 8.5 T thermal for 2-substituted pyridines, and smaller, yet significant enhancements for provitamin B<sub>6</sub> and caffeine. We also show <sup>15</sup>N-enhancements of ∼1000-fold and <sup>19</sup>F-enhancements of 30-fold. ...[more]

Project description:Reductive dehalogenases are responsible for the reductive cleavage of carbon-halogen bonds during organohalide respiration. A variety of mechanisms have been proposed for these cobalamin and [4Fe-4S] containing enzymes, including organocobalt, radical, or cobalt-halide adduct based catalysis. The latter was proposed for the oxygen-tolerant Nitratireductor pacificus pht-3B catabolic reductive dehalogenase (NpRdhA). Here, we present the first substrate bound NpRdhA crystal structures, confirming a direct cobalt-halogen interaction is established and providing a rationale for substrate preference. Product formation is observed in crystallo due to X-ray photoreduction. Protein engineering enables rational alteration of substrate preference, providing a future blue print for the application of this and related enzymes in bioremediation.

Project description:Nuclear spin hyperpolarization through signal amplification by reversible exchange (SABRE), the non-hydrogenative version of para-hydrogen induced polarization, is demonstrated to enhance sensitivity for the detection of biomacromolecular interactions. A target ligand for the enzyme trypsin includes the binding motif for the protein, and at a distant location a heterocyclic nitrogen atom for interacting with a SABRE polarization transfer catalyst. This molecule, 4-amidinopyridine, is hyperpolarized with 50% para-hydrogen to yield enhancement values ranging from -87 and -34 in the ortho and meta positions of the heterocyclic nitrogen, to -230 and -110, for different solution conditions. Ligand binding is identified by flow-NMR, in a two-step process that separately optimizes the polarization transfer in methanol while detecting the interaction in a predominantly aqueous medium. A single scan Carr-Purcell-Meiboom-Gill (CPMG) experiment identifies binding by the change in R 2 relaxation rate. The SABRE hyperpolarization technique provides a cost effective means to enhance NMR of biological systems, for the identification of protein-ligand interactions and other applications.

Project description:Discoveries about the cultures and cultural capacities of the great apes have played a leading role in the recognition emerging in recent decades that cultural inheritance can be a significant factor in the lives not only of humans but also of nonhuman animals. This prominence derives in part from these primates being those with whom we share the most recent common ancestry, thus offering clues to the origins of our own thoroughgoing reliance on cumulative cultural achievements. In addition, the intense research focus on these species has spawned an unprecedented diversity of complementary methodological approaches, the results of which suggest that cultural phenomena pervade the lives of these apes, with potentially major implications for their broader evolutionary biology. Here I review what this extremely broad array of observational and experimental methodologies has taught us about the cultural lives of chimpanzees, gorillas, and orangutans and consider the ways in which this knowledge extends our wider understanding of primate biology and the processes of adaptation and evolution that shape it. I address these issues first by evaluating the extent to which the results of cultural inheritance echo a suite of core principles that underlie organic Darwinian evolution but also extend them in new ways and then by assessing the principal causal interactions between the primary, genetically based organic processes of evolution and the secondary system of cultural inheritance that is based on social learning from others.

Project description:Methionine sulfoxide reductase A (MsrA) enzymes have recently found applications as nonoxidative biocatalysts in the enantioselective kinetic resolution of racemic sulfoxides. This work describes the identification of selective and robust MsrA biocatalysts able to catalyze the enantioselective reduction of a variety of aromatic and aliphatic chiral sulfoxides at 8-64 mM concentration with high yields and excellent ees (up to 99%). Moreover, with the aim to expand the substrate scope of MsrA biocatalysts, a library of mutant enzymes has been designed via rational mutagenesis utilizing in silico docking, molecular dynamics, and structural nuclear magnetic resonance (NMR) studies. The mutant enzyme MsrA33 was found to catalyze the kinetic resolution of bulky sulfoxide substrates bearing non-methyl substituents on the sulfur atom with ees up to 99%, overcoming a significant limitation of the currently available MsrA biocatalysts.

Project description:The phytohormone (+)-7-iso-jasmonoyl-L-isoleucine (JA-Ile) is a major regulator of developmental and stress responses in plants. The perception of JA-Ile involves the formation of a ternary complex with the F-box protein COI1 and a member of the JAZ family of co-repressors, which leads to JAZ degradation. Coronatine (COR) is a bacterial phytotoxin that functionally mimics JA-Ile and interacts with the co-receptor COI1-JAZ complex with higher affinity than JA-Ile. Based on the crystal structure of the co-receptor, we designed ligand derivatives that spatially impede the interaction of the co-receptor proteins and, therefore, should act as competitive antagonists of COR or JA-Ile. One derivative, Coronatine O-methyloxime (COR-MO), shows a strong activity preventing COI-JAZ interaction and the subsequent JAZ degradation. COR-MO efficiently reverts the effects of JA-Ile or COR treatments on JA-mediated responses, such as anthocyanin accumulation, root-growth inhibition and gene expression in Arabidopsis plants. Moreover, it potentiates plant resistance preventing the effect of bacterially produced COR during Pseudomonas syringae infections in different plant species. In addition to the utility of COR-MO for Plant Biology research, our results underscore its biotechnological and agronomical potential for a safer and sustainable agriculture. Project description:Biomedically important histone lysine acetyltransferase KAT8 catalyses the acetyl coenzyme A-dependent acetylation of lysine on histone and other proteins. Here, we explore the ability of human KAT8 to catalyse the acetylation of histone H4 peptides possessing lysine and its analogues at position 16 (H4K16). Our synthetic and enzymatic studies on chemically and structurally diverse lysine mimics demonstrate that KAT8 also has a capacity to acetylate selected lysine analogues that possess subtle changes on the side chain and main chain. Overall, this work highlights that KAT8 has a broader substrate scope beyond natural lysine, and contributes to the design of new chemical probes targeting KAT8 and other members of the histone lysine acetyltransferase (KAT) family.

Project description:The use of parahydrogen based hyperpolarisation in NMR is becoming more widespread due to the rapidly expanding range of suitable target molecules and low-cost of parahydrogen production. Hyperpolarisation via SABRE catalysis employs a metal complex to transfer polarisation from parahydrogen into a substrate whilst they are bound. In this paper we present a quantitative study of substrate-iridium ligation effects by reference to the substrates 4-chloropyridine (A), 4-pyridinecarboxaldehyde methyl hemiacetal (B), 4-methylpyridine (C) and 4-methoxypyridine (D), and evaluate the role they play in the SABRE catalysis. Substrates whose substituents enable stronger associations yield slower substrate dissociation rates (k d). A series of variable temperature studies link these exchange rates to optimal SABRE performance and reveal the critical impact of NMR relaxation times (T 1). Longer catalyst residence times are shown to result in shorter substrate T 1 values in solution as binding to iridium promotes relaxation thereby not only reducing SABRE efficiency but decreasing the overall level of achieved hyperpolarisation. Based on these data, a route to achieve more optimal SABRE performance is defined.

Project description:Choice of social group can affect the likelihood of survivorship and reproduction for social species. By joining larger social groups-shoals-small freshwater fish like the mosquitofish Gambusia affinis can reduce predation risk and forage more efficiently. We tested shoal choice in mosquitofish to determine whether such choices are economically rational, i.e. consistent and optimal. Although many studies of decision-making assume rational choice, irrational decision-making is common and occurs across contexts. We tested rationality of shoaling decisions by testing the constant ratio rule, which states that the relative preference for two options should not change in the presence of a third option. Female mosquitofish upheld this rule when tested for shoal preference based on group size. Our results contrast with other studies showing violations of the constant ratio rule in foraging and mate choice decision-making contexts. These results suggest that decisions that immediately influence survivorship or decision-making along a single dimension may favour rational decision-making.

Project description:In mammals, ovarian folliculogenesis leading to the ovulation of completely mature oocytes is a long and complex process that is regulated at different levels. The mechanisms that underlie the selection of one or several dominant follicles as well as the regulation of the number of ovulating follicles are largely unknown. In this project, we proposed to study the genetic determinism that underlies the difference of ovulation rate between species (cattle and pigs). Towards this purpose, we made a comparative transcriptomics study on granulosa cells. Pig and cattle comparison was achieved by a transcriptome analysis with a 9K nylon pig microarray (GPL3729) on granulosa cells from either small healthy antral follicles (SHF) or large healthy antral follicles (LHF). The images were quantified using AGscan software and the data were managed with BASE software. Statistical analysis was performed using R software. Transcriptomic analysis on pig and cattle evidenced 252 differentially expressed genes (FDR5%) between the two follicle classes and/or the two species. This research project which implicated three laboratories from INRA: « Laboratoire de Génétique Cellulaire » (UMR444-LGC) , « Station d’Amélioration Génétique des Animaux » (UR 631-SAGA) and « Physiologie des Comportements et de la reproduction » (UMR 85-PRC) benefited from both European funding through SABRE project and French ANR funding through GenOvul project. Project description:Caged complexes can provide impressive selective catalysts. Due to the complex shapes of such caged catalysts, however, the level of selectivity control of a single substrate cannot be extrapolated to other substrates. Herein, the substrate scope using 41 terminal alkene substrates is investigated in the hydroformylation reaction with an encapsulated rhodium catalyst [Rh(H)(CO)3(P(mPy3(ZnTPP)3))] (CAT1). For all substrates, the amount of branched products formed was higher with CAT1 than with the unencapsulated reference catalyst [Rh(H)(CO)2(P(mPy3))2] (CAT2) (linear/branched ratio between 2.14 and 0.12 for CAT1 and linear/branched ratio between 6.22 and 0.59 for CAT2). Interestingly, the level of cage induced selectivity depends strongly on the substrate structure that is converted. Analysis of the substrate scope combined with DFT calculations suggests that noncovalent interactions between the substrate moieties and cage walls play a key role in controlling the regioselectivity. Consequently, these supramolecular interactions were further optimized by replacing the ZnTPP building block with a zinc porphyrin analog that contained OiPr substituents on the meta position of the aryl rings. The resulting caged catalyst, CAT4, converted substrates with even higher branched selectivity.
