Multiple Coexisting Species and the First Known Case of a Cheater in Epicephala (Gracillariidae) Associated with a Species of Glochidion (Phyllanthaceae) in Tropical Asia.

ABSTRACT: Glochidion plants and Epicephala moths played different roles and kept the balance in the mutualism. We studied the four coexisting Epicephala species on Glochidion sphaerogynum in detail and reconstructed the phylogenic tree of 40 Gracillariidae species. The results showed that one of them (Epicephala impolliniferens) did not pollinate G. sphaerogynum, because of lacking the specialized structure of carrying pollen. These results suggested that E. impolliniferens acted as a 'cheater' in the system. The phylogenetic analyses suggested that E. impolliniferens derived from a pollinating species, and had secondarily gave up the ability to pollinate. This is a typical phenomenon of mutualism reversal. The phenomenon exhibits the co-evolutionary diversification under selection pressures.

The latter two species are also associated with Glochidionwrightii. Photographs of adults and genital structures are provided.

Diffuse coevolution between two Epicephala species (Gracillariidae) and two Breynia species (Phyllanthaceae).

Project description:The diffuse coevolution between two moth species (Epicephala lativalvaris and E. mirivalvata) and two plant species (Breynia fruticosa and B. rostrata) is reported based on field observations and indoor experiments conducted in Hainan and Fujian, China. Study results showed that the two Epicephala species jointly pollinated the two Breynia species, which led to a unique obligate pollination mutualism of two-to-two species specificity. A single Epicephala larva exclusively fed on seeds of host plants and developed to maturity by consuming all six seeds of each fruit, whereas a fraction of intact fruits were left to ensure the reproduction of plants within the whole population. Larvae of the two Epicephala species are competitive for resources; the population of E. mirivalvata is much smaller than that of E. lativalvaris, which has resulted from the differences in the female ovipositor structures and oviposition mode. The life history of Epicephala species highly coincides with the phenology of Breynia plants, and different phenology of B. fruticosa resulted in the different life history of the two Epicephala species in Hainan and Fujian. The natural hybridization of two host plants, possibly induced by the alternate pollination of two Epicephala species, is briefly discussed.

Diversity and species-specificity of brood pollination of leafflower trees (Phyllanthaceae: Glochidion) by leafflower moths (Lepidoptera: Epicephala) in tropical Southeast Asia (Cambodia).

Project description:Glochidion (Phyllanthaceae; leafflower trees) is a genus of trees which is widely reported to be pollinated by leafflower moths (Gracillariidae: Epicephala) in temperate and subtropical Asia, Australia, and the Pacific islands. However, the pollination ecology of Glochidion is not well described from tropical Asia, the region where it is most species-rich at both local (≤9 spp.) and regional (~200 spp.) scales. Here we report investigations of pollination biology and species-specificity of five Glochidion species in tropical Southeast Asia (Cambodia). Through nocturnal observations and fruit dissections, we find that at least three and likely five Glochidion species in Cambodia are pollinated by seed-parasitic leafflower moths. We find no evidence that any of these leafflower moths are non-mutualistic parasites, despite known examples of such parasites of this mutualism elsewhere in Asia. While the presence of a single larva in a fruit results in only a fraction of seeds being consumed, the presence of more than one larva per fruit-a frequent occurrence in some species-can result in almost all seeds within the fruit being infested. Multilocus phylogenetic analysis indicates that there are five different minimally monophyletic leafflower moth clades, each of which pollinates a unique Glochidion host species. Our results indicate that in its center of diversity in tropical Asia this system is an obligate pollination mutualism as previously described at the global margins of its distribution. These findings provide insights into the processes that generate and maintain biodiversity and maintain mutualism stability in plant-insect interactions in this biodiversity hotspot.

Revision of the Japanese species of Epicephala Meyrick with descriptions of seven new species (Lepidoptera, Gracillariidae).

Project description:Epicephala moths are involved in obligate mutualisms with their Phyllanthaceae hosts, in which the female moths assure pollination and, in return, their progeny develop by consuming the seeds. Ecological, molecular and geographical data suggest that the genus includes several hundred species, but the majority remains to be formally described. Here we revise the Japanese species of Epicephala Meyrick, 1880. In addition to two previously named species, seven species are newly described: Epicephala anthophilia sp. n., Epicephala lanceolatella sp. n., Epicephala perplexa sp. n., Epicephala obovatella sp. n., Epicephala corruptrix sp. n., Epicephala parasitica sp. n. and Epicephala nudilingua sp. n. The first four are species involved in obligate pollination mutualism, while the fifth is a pollinating seed parasite and the last two are derived non-pollinating seed parasites of herbaceous Phyllanthus. Each of the nine Japanese Epicephela species is specialized to a single plant species in the genera Glochidion, Breynia or Phyllanthus, except for Epicephala obovatella and Epicephala corruptrix that each utilizes two closely related Glochidion species. Considerable variations are found in pollination and oviposition behaviors among species, which are reflected in their proboscis and ovipositor morphologies, respectively. Molecular phylogeny indicated that there have been repeated transitions in oviposition mode during the diversification of Epicephala, which were accompanied by changes in ovipositor morphology, as suggested by a correlation analysis. Keys to species are provided.

Isolation and characterization of 11 microsatellite markers for Glochidion acuminatum (Phyllanthaceae).

Project description:UNLABELLED: PREMISE OF THE STUDY:Microsatellite markers were developed for Glochidion acuminatum (Phyllanthaceae) to investigate pollen dispersal distances and thereby to assess the effectiveness of specialized Epicephala moths as pollinators. • METHODS AND RESULTS:Using next-generation sequencing, 11 polymorphic microsatellite primer pairs were developed for G. acuminatum. The primer pairs were tested on 49 individuals from two populations in Japan. The number of alleles per locus ranged from two to 13, and the expected heterozygosity ranged from 0.12 to 0.85. Probability of identity for all loci combined was lower than 1.27 × 10(7). • CONCLUSIONS:The microsatellite markers developed in this study will be useful for evaluating the benefit of specialized Epicephala moth pollination to Glochidion plants.

Project description:Based on morphological studies performed on live plants in the field and specimens deposited in herbaria, a new species, Glochidion shanense Gang Yao & Shixiao Luo (Phyllantheae, Phyllanthaceae), is here described and illustrated. The species is morphologically most similar to G. ellipticum Wight, but can be distinguished from the latter by having hairy branchlets, longer pedicels, uniseriate and narrowly triangular sepals of female flowers, 4-5-locular ovaries, stout and cylindric persistent style on fruits.

Taxonomic revision of the genus Glochidion (Phyllanthaceae) in Taiwan, China.

Project description:A comprehensive taxonomic revision of the genus Glochidion J.R. Forst. & G. Forst. from Taiwan in China was carried out based on the examination of herbarium specimens and filed investigations. Eight species and three varieties are recognized, including a new species endemic to Taiwan, G. lanyuense Gang Yao & S.X. Luo, which is described and illustrated. Three names, viz. G. chademenosocarpum Hayata, G. kusukusense Hayata, and G. ovalifolium F.Y. Lu & Y.S. Hsu, are reduced to the new synonyms of G. rubrum Blume, G. lanceolatum Hayata, and G. ellipticum Wight, respectively. Two names, viz. G. lanceolatum Hayata and G. suishaense Hayata, are lectotypified here. A key to the Glochidion species in Taiwan is provided.

Simultaneous hyperaccumulation of nickel and cobalt in the tree Glochidion cf. sericeum (Phyllanthaceae): elemental distribution and chemical speciation.

Project description:Hyperaccumulation is generally highly specific for a single element, for example nickel (Ni). The recently-discovered hyperaccumulator Glochidion cf. sericeum (Phyllanthaceae) from Malaysia is unusual in that it simultaneously accumulates nickel and cobalt (Co) with up to 1500 μg g-1 foliar of both elements. We set out to determine whether distribution and associated ligands for Ni and Co complexation differ in this species. We postulated that Co hyperaccumulation coincides with Ni hyperaccumulation operating on similar physiological pathways. However, the ostensibly lower tolerance for Co at the cellular level results in the exudation of Co on the leaf surface in the form of lesions. The formation of such lesions is akin to phytotoxicity responses described for manganese (Mn). Hence, in contrast to Ni, which is stored principally inside the foliar epidermal cells, the accumulation response to Co consists of an extracellular mechanism. The chemical speciation of Ni and Co, in terms of the coordinating ligands involved and principal oxidation state, is similar and associated with carboxylic acids (citrate for Ni and tartrate or malate for Co) and the hydrated metal ion. Some oxidation to Co3+, presumably on the surface of leaves after exudation, was observed.

Complete plastome sequence of Bridelia tomentosa Blume (Phyllanthaceae): a medicinal shrub species in South Asia.

Project description:Bridelia tomentosa is a deciduous shrub in the family of Phyllanthaceae. It grows in the evergreen primary or secondary thickets or forests in the sea level from 1000 to 1500 m. It distributed in.south China (e.g., Fujian, Guangdong, Guangxi, Hainan etc) and other south Asian countries (e.g. Bangladesh, Bhutan, Cambodia etc). Here, we report and characterize the complete plastome of B. tomentosa. The complete plastome is of 149,958 bp in length with a typical structure and gene content of angiosperm plastome, including two inverted repeat (IRs) regions of 26,354 bp, a large single-copy (LSC) region of 81,355 bp and a small single-copy (SSC) region of 15,895 bp. The plastome contains 129 genes, consisting of 84 protein-coding genes, 37 tRNA genes, eight rRNA genes. The overall G/C content in the plastome of B. tomentosa is 36.0%. The complete plastome sequence of B. tomentosa will provide a useful resource for the conservation genetics of this species as well as for phylogenetic studies in Phyllanthaceae.
