Glycoconjugates via phosphorus ylides.

ABSTRACT: A facile, high yielding access to rare chimeric compounds combining phosphorus ylides with complex glycosyl formamides is described. We determined x-ray structures gaining structural insight into this compounds class. In addition, data mining of similar compounds deposited within the Cambridge Structural Database was performed. These derivatives could be used either as synthetic intermediates via the ylide functionalization and glyco chemical biology synthons or improving the pharmacokinetic properties of a potential bioactive molecule, exploiting the glycosyl moiety.

Accessing Cationic α-Silylated and α-Germylated Phosphorus Ylides.

Project description:The synthesis and full characterization of α-silylated (α-SiCPs; 1-7) and α-germylated (α-GeCPs; 11-13) phosphorus ylides bearing one chloride substituent R3 PC(R1 )E(Cl)R2 2 (R=Ph; R1 =Me, Et, Ph; R2 =Me, Et, iPr, Mes; E=Si, Ge) is presented. The molecular structures were determined by X-ray diffraction studies. The title compounds were applied in halide abstraction studies in order to access cationic species. The reaction of Ph3 PC(Me)Si(Cl)Me2 (1) with Na[B(C6 F5 )4 ] furnished the dimeric phosphonium-like dication [Ph3 PC(Me)SiMe2 ]2 [B(C6 F5 )4 ]2 (8). The highly reactive, mesityl- or iPr-substituted cationic species [Ph3 PC(Me)SiMes2 ][B(C6 F5 )4 ] (9) and [Ph3 PC(Et)SiiPr2 ][B(C6 F5 )4 ] (10) could be characterized by NMR spectroscopy. Carrying out the halide abstraction reaction in the sterically demanding ether iPr2 O afforded the protonated α-SiCP [Ph3 PCH(Et)Si(Cl)iPr2 ][B(C6 F5 )4 ] (6 dec) by sodium-mediated basic ether decomposition, whereas successfully synthesized [Ph3 PC(Et)SiiPr2 ][B(C6 F5 )4 ] (10) readily cleaves the F-C bond in fluorobenzene. Thus, the ambiphilic character of α-SiCPs is clearly demonstrated. The less reactive germanium analogue [Ph3 PC(Me)GeMes2 ][B{3,5-(CF3 )2 C6 H3 }4 ] (14) was obtained by treating 11 with Na[B{3,5-(CF3 )2 C6 H3 }4 ] and fully characterized including by X-ray diffraction analysis. Structural parameters indicate a strong CYlide -Ge interaction with high double bond character, and consequently the C-E (E=Si, Ge) bonds in 9, 10 and 14 were analyzed with NBO and AIM methods.

Ylide-Functionalization via Metalated Ylides: Synthesis and Structural Properties.

Project description:The ?-metallated ylides [Ph3P-C-Z]-M+ (with Z=SO2Tol or CN and M=Na or K) were used as versatile nucleophiles for the facile access to ylide-substituted compounds. Halogenations, alkylations, carbonylations and functionalization reactions with main group element halides were easily accomplished by simple trapping reactions with the appropriate electrophiles. X-ray crystallographic studies of all compounds - including the first structures of ?-fluorinated P-ylides - showed remarkable differences in the ylide backbone depending on the substituents. In the fluorinated compounds, a change from a fully planar to a pyramidalized ylidic carbon centre was observed despite the strongly anion-stabilizing ability of the yldiide substituent. ?-Donation from the ylide substituent also resulted in geometric restrictions depending on the steric and electronic properties of the introduced substituents.

Stereoselective Synthesis of Selenium-Containing Glycoconjugates via the Mitsunobu Reaction.

Project description:A simple and efficient route for the synthesis of new glycoconjugates has been developed. The approach acts as a model for a mini-library of compounds with a deoxy-selenosugar core joined to a polyphenolic moiety with well-known antioxidant properties. An unexpected stereocontrol detected in the Mitsunobu key reaction led to the most attractive product showing a natural d-configuration. Thus, we were able to obtain the target molecules from the commercially available d-ribose via a shorter and convenient sequence of reactions.

Diversity enhances agricultural productivity via rhizosphere phosphorus facilitation on phosphorus-deficient soils.

Project description:Intercropping, which grows at least two crop species on the same pieces of land at the same time, can increase grain yields greatly. Legume-grass intercrops are known to overyield because of legume nitrogen fixation. However, many agricultural soils are deficient in phosphorus. Here we show that a new mechanism of overyielding, in which phosphorus mobilized by one crop species increases the growth of a second crop species grown in alternate rows, led to large yield increases on phosphorus-deficient soils. In 4 years of field experiments, maize (Zea mays L.) overyielded by 43% and faba bean (Vicia faba L.) overyielded by 26% when intercropped on a low-phosphorus but high-nitrogen soil. We found that overyielding of maize was attributable to below-ground interactions between faba bean and maize in another field experiment. Intercropping with faba bean improved maize grain yield significantly and above-ground biomass marginally significantly, compared with maize grown with wheat, at lower rates of P fertilizer application (<75 kg of P(2)O(5) per hectare), and not significantly at high rate of P application (>112.5 kg of P(2)O(5) per hectare). By using permeable and impermeable root barriers, we found that maize overyielding resulted from its uptake of phosphorus mobilized by the acidification of the rhizosphere via faba bean root release of organic acids and protons. Faba bean overyielded because its growth season and rooting depth differed from maize. The large increase in yields from intercropping on low-phosphorus soils is likely to be especially important on heavily weathered soils.

From Ylides to Doubly Yldiide-Bridged Iron(II) High Spin Dimers via Self-Protolysis.

Project description:A synthetic strategy for the preparation of novel doubly yldiide bridged iron(II) high spin dimers ([(?2-C)FeL]2, L = N(SiMe3)2, Mesityl) has been developed. This includes the synthesis of ylide-iron(II) monomers [(Ylide)FeL2] via adduct formation. Subsequent self-protolysis at elevated temperatures by in situ deprotonation of the ylide ligands results in a dimerization reaction forming the desired bridging ?2-C yldiide ligands in [(?2-C)FeL]2. The comprehensive structural and electronic analysis of dimers [(?2-C)FeL]2, including NMR, Mössbauer, and X-ray spectroscopy, as well as X-ray crystallography, SQUID, and DFT calculations, confirm their high-spin FeII configurations. Interestingly, the Fe2C2 cores display very acute Fe-C-Fe angles (averaged: 78.6(2)°) resulting in short Fe···Fe distances (averaged: 2.588(2) Å). A remarkably strong antiferromagnetic coupling between the Fe centers has been identified. Strongly polarized Fe-C bonds are observed where the negative charge is mostly centered at the ?2-C yldiide ligands.

Synthesis of (18)F-Arenes from Spirocyclic Iodonium(III) Ylides via Continuous-Flow Microfluidics.

Project description:Spirocyclic hypervalent iodine(III) ylides have proven to be synthetically versatile precursors for efficient radiolabelling of a diverse range of non-activated (hetero)arenes, highly functionalised small molecules, building blocks and radiopharmaceuticals from [(18)F]fluoride ion. Herein, we report the implementation of these reactions onto a continuous-flow microfluidic platform, thereby offering an alterative and automated synthetic procedure of a radiopharmaceutical, 3-[(18)F]fluoro-5-[(pyridin-3-yl)ethynyl]benzonitrile ([(18)F]FPEB) and a routinely used building block for click-radiochemistry, 4-[(18)F]fluorobenzyl azide. This new protocol was applied to the synthesis of [(18)F]FPEB (radiochemical conversion (RCC) = 68 ± 5%) and 4-[(18)F]fluorobenzyl azide (RCC=68 ± 5%; isolated radiochemical yield = 24±0%). We anticipate that the high throughput microfluidic platform will accelerate the discovery and applications of (18)F-labelled building blocks and labelled compounds prepared by iodonium ylide precursors as well as the production of radiotracers for preclinical imaging studies.

Pyruvate Substitutions on Glycoconjugates.

Project description:Glycoconjugates are the most diverse biomolecules of life. Mostly located at the cell surface, they translate into cell-specific "barcodes" and offer a vast repertoire of functions, including support of cellular physiology, lifestyle, and pathogenicity. Functions can be fine-tuned by non-carbohydrate modifications on the constituting monosaccharides. Among these modifications is pyruvylation, which is present either in enol or ketal form. The most commonly best-understood example of pyruvylation is enol-pyruvylation of N-acetylglucosamine, which occurs at an early stage in the biosynthesis of the bacterial cell wall component peptidoglycan. Ketal-pyruvylation, in contrast, is present in diverse classes of glycoconjugates, from bacteria to algae to yeast-but not in humans. Mild purification strategies preventing the loss of the acid-labile ketal-pyruvyl group have led to a collection of elucidated pyruvylated glycan structures. However, knowledge of involved pyruvyltransferases creating a ring structure on various monosaccharides is scarce, mainly due to the lack of knowledge of fingerprint motifs of these enzymes and the unavailability of genome sequences of the organisms undergoing pyruvylation. This review compiles the current information on the widespread but under-investigated ketal-pyruvylation of monosaccharides, starting with different classes of pyruvylated glycoconjugates and associated functions, leading to pyruvyltransferases, their specificity and sequence space, and insight into pyruvate analytics.

Bacterial cell-envelope glycoconjugates.

Project description:Prokaryotic glycosylation fulfills an important role in maintaining and protecting the structural integrity and function of the bacterial cell wall, as well as serving as a flexible adaption mechanism to evade environmental and host-induced pressure. The scope of bacterial and archaeal protein glycosylation has considerably expanded over the past decade(s), with numerous examples covering the glycosylation of flagella, pili, glycosylated enzymes, as well as surface-layer proteins. This article addresses structure, analysis, function, genetic basis, biosynthesis, and biomedical and biotechnological applications of cell-envelope glycoconjugates, S-layer glycoprotein glycans, and "nonclassical" secondary-cell wall polysaccharides. The latter group of polymers mediates the important attachment and regular orientation of the S-layer to the cell wall. The structures of these glycopolymers reveal an enormous diversity, resembling the structural variability of bacterial lipopolysaccharides and capsular polysaccharides. While most examples are presented for Gram-positive bacteria, the S-layer glycan of the Gram-negative pathogen Tannerella forsythia is also discussed. In addition, archaeal S-layer glycoproteins are briefly summarized.

Selective monophosphorylation of chitosan via phosphorus oxychloride.

Project description:Chitosan was selectively monophosphorylated via reaction with phosphorus oxychloride (POCl3) to enhance water solubility while avoiding polyphosphate formation. The use of POCl3 resulted in negligible product degradation (i.e., breakdown of O-glycosidic bonds) even after a 3 d reaction period (<5% weight loss). X-ray photoelectron spectroscopy (XPS) characterization of the POCl3-phosphorylated chitosan (P-chitosan) revealed a phosphorus to nitrogen (P/N) atomic ratio of 0.30. Phosphorus-31 nuclear magnetic resonance (31P NMR) spectroscopy verified the monophosphorylation of chitosan's primary and secondary alcohols, and primary amines. The calcium chelation efficiency for the phosphorylated product approached 0.05 mg Ca2+ per mg of P-chitosan as measured by inductively coupled plasma-optical emission spectrometry (ICP-OES), indicating improved chelation over native chitosan. This selective monophosphorylation approach proved useful for modifying other biopolymers, including cellulose and alginate.

Ring Expansion of Bicyclic Methyleneaziridines via Concerted, Near-Barrierless [2,3]-Stevens Rearrangements of Aziridinium Ylides.

Project description:The synthesis of densely functionalized azetidinesin a highly stereocontrolled manner is challenging, but interest in the bioactivities of these small heterocycles has stimulated methods for their preparation. We recently reported a one-carbon ring expansion of bicyclic methylene aziridines under dirhodium catalysis capable of delivering enantioenriched azetidines. This work explores this ring expansion using computational and experimental studies. DFT computations indicate that the reaction proceeds through formation of an aziridinium ylide, which is precisely poised for concerted, asynchronous ring-opening/closing to deliver the azetidines in a [2,3]-Stevens-type rearrangement. The concerted nature of this rearrangement is responsible for the stereospecificity of the reaction, where axial chirality from the initial allene substrate is transferred to the azetidine product with complete fidelity. The computed mechanistic pathway highlights the key roles of the olefin and the rigid structure of the methylene aziridine in differentiating our observed ring expansion from competing cheletropic elimination pathways noted with ylides derived from typical aziridines.
