Impact of Stress on Health and Final Weight in Fattening Lambs.

ABSTRACT: In order to determine whether the stress level had any influence on the health and final weight of Rasa Aragonesa fattening lambs, stress markers were measured throughout the fattening period in 80 feedlot lambs (group F) and in 40 lambs finished on the farm of birth (group C). The highest values of the stress indicators-neutrophil-to-lymphocyte-ratio (N/L), non-esterified fatty acids (NEFA), serum cortisol and fecal cortisol metabolites (FCM)-were recorded after the road transportation of group F to the feedlot. No differences between the groups were identified at the end of the adaptation period, but at the end of the fattening, statistically significant higher values for NEFA and serum cortisol were determined in group F. However, statistically significant differences between the groups were not found in the percentage of lambs with clinical signs of illness and ovine respiratory complex (ORC) lesions or in the final weight of the lambs. Independent of the location at which the lambs were fattened, those with FCM values at the beginning of the study (at the time of weaning) in the highest quartile developed a higher percentage of clinical signs of illness (45.5% vs. 32.1%, p > 0.05) and ORC lesions (45.5% vs. 10.7%, p < 0.01), and their average final weight was lower (24.36 kg vs. 27.40 kg, p = 0.001) than those with values in the lowest quartile. Stress experienced by lambs prior to finishing seems to be relevant for their further development, and FCM concentration at the time of weaning could be used as an indicator of health and productive performance of the lambs during the fattening period, regardless of finishing location.

Project description:The increasing world population is driving demand for improved efficiency of feed resources of livestock. However, the molecular mechanisms behind various feed efficiency traits and their regulation by nutrition remain poorly understood. Here, we aimed to identify differentially expressed (DE) genes in the liver tissues of fattening Merino lambs and differences in metabolites accumulated in plasma to identify modified metabolic pathways as a consequence of milk restriction during the suckling period. Twenty-four male Merino lambs (4.81 ± 0.256 kg) were divided into 2 groups (n = 12 per dietary treatment). The first group (ad libitum, ADL) was kept permanently with the dams, whereas the other group (restricted, RES) was milk restricted. When they reached 15 kg of live body weight (LBW), all the animals were offered the same complete pelleted diet at the same level (35 g DM/kg LBW per day) to ensure no differences in dry matter intake. All the lambs were harvested when they reached 27 kg of LBW. For transcriptomic analysis, 4 liver samples from each group (8 samples in total) were selected for RNA sequencing (RNA-seq), and plasma samples from all animals (24 samples in total) were used to perform a nontargeted metabolomic analysis on a hybrid quadrupole-time-of-flight mass spectrometer coupled to an ultra-high performance liquid chromatographic system (UHPLC/QTOF-MS). Thirty-eight DE annotated genes were identified by RNA-seq, with 23 DE genes being down-regulated and 15 up-regulated in the liver of RES lambs relative to the ADL group (P < 0.10). Also, the metabolomic assay identified 38 differentially accumulated compounds (P < 0.10). In general, those genes and pathways involved in protein synthesis or protease inhibitors were down-regulated in the RES group, whereas those related to proteolytic degradation were up-regulated, thus suggesting a higher catabolism of proteins in these lambs. RES lambs showed over-expression of xenobiotic metabolism pathways, whereas those genes related to β-oxidation of fatty acids were down-regulated. According to the data obtained, early feed restriction during the suckling period of Merino lambs promoted long-term effects on both the hepatic transcriptomic profile and plasma metabolic profile, which might have modified fatty acids metabolism, catabolism of proteins, and detoxification of xenobiotics, thus reducing feed efficiency during the fattening period.

Project description:Several ruminal cellulolytic bacteria species are sensitive to pH and could therefore be used as biomarkers to determine the risk of sub-acute ruminal acidosis (SARA) in finishing lambs. This study compared a 2-4 h post feeding ruminal pH measurement to abundances of the ruminal pH-sensitive bacteria to evaluate the risk of SARA in a herd of 120 finishing lambs. The lambs were reared in individual units for 50 days. Ruminal fluid was collected by use of an orogastric tube on day 51 2-4 h after feeding. Although the lambs were fed an identical diet, they responded differently in the abundances of four ruminal pH sensitive cellulolytic bacteria (Ruminococcus albus, Ruminococcus flavefaciens and Fibrobacter succinogenes and Butyrivibrio fibrisolvens). Lambs with the most or the least cellulolytic bacteria were then classified as either lower SARA risk (LSR, n = 10) or higher SARA risk group (HSR, n = 10), respectively. Data showed that the ruminal pH and VFA profiles were uncorrelated with the number of cellulolytic bacteria (P > 0.050). Lambs with the HSR showed lower ruminal pH (P = 0.013) and acetate to propionate ratio (P = 0.018), higher concentrations of lactate (P = 0.035) and proportion of propionate (P = 0.033) compared to those with the LSR. The DMI and ADG did not differ in LSR and HSR lambs (P > 0.050). A diversity analysis revealed significantly lower diversity in HSR lambs than in LSR (Simpson index, P = 0.004). The relative abundances of the phyla Bacteroidetes, Fibrobacteres, Verruomicrobia, and Proteobacteria were higher in LSR lambs than in HSR (P < 0.050). The abundances of several phyla including Firmicutes, Tenericutes and Actinobacteria were higher in the HSR than in the LSR group (P < 0.050). The bacterial communities of the LSR and HSR clustered separately in rumen based on the Unifrac distances, indicating distinct bacteria communities at OTU level between the LSR and HSR lambs. Overall, there was no correlation between 2 and 4 h post-feeding ruminal pH and the abundance of pH-sensitive bacteria and the amount of these bacteria could be used as a potential biomarker of SARA in lamb herd.

Project description:There is a growing interest in the use of yeast (Saccharomyces cerevisiae) culture (YC) for the enhancement of growth performance and general animal health. Grain-based pelleted total mixed rations (TMR) are emerging in intensive sheep farming systems, but it is uncertain if the process of pelleting results in YC becoming ineffective. This study aimed to examine the effects of YC supplemented to pelleted TMR at two proportions of corn in the diet on animal performance, feed digestion, blood parameters, rumen fermentation, and microbial community in fattening lambs. A 2 × 2 factorial design was adopted with two experimental factors and two levels in each factor, resulting in four treatments: (1) low proportion of corn in the diet (LC; 350 g corn/kg diet) without YC, (2) LC with YC (5 g/kg diet), (3) high proportion of corn in the diet (HC; 600 g corn/kg diet) without YC, and (4) HC with YC. Fifty-six 3-month-old male F2 hybrids of thin-tailed sheep and Northeast fine-wool sheep with a liveweight of 19.9 ± 2.7 kg were randomly assigned to the four treatment groups with an equal number of animals in each group. The results showed that live yeast cells could not survive during pelleting, and thus, any biological effects of the YC were the result of feeding dead yeast and the metabolites of yeast fermentation rather than live yeast cells. The supplementation of YC resulted in 31.1 g/day more average daily gain regardless of the proportion of corn in the diet with unchanged feed intake during the 56-day growth measurement period. The digestibility of neutral detergent fibre and acid detergent fibre was increased, but the digestibility of dry matter, organic matter, and crude protein was not affected by YC. The supplementation of YC altered the rumen bacterial population and species, but the most abundant phyla Bacteroidetes, Firmicutes, and Proteobacteria remained unchanged. This study indicates that YC products can be supplemented to pelleted TMR for improved lamb growth performance, although live yeast cells are inactive after pelleting. The improved performance could be attributed to improved fibre digestibility.

Project description:Alternatives to antibiotics for improving productivity and maintaining the health of livestock health are urgently needed. The scope of this research was conducted to investigate the effects of two alternatives (Bacillus licheniformis and Saccharomyces cerevisiae) to monensin on growth performance, antioxidant capacity, immunity, ruminal fermentation and microbial diversity of fattening lambs. One hundred and sixty Dorper × Thin-tailed Han sheep (32 ± 3.45 kg BW) were randomly assigned into 5 treatments of n = 32 lambs/group. Lambs in the control group were fed a basal diet (NC) while the other four treatments were fed basal diets supplemented with monensin (PC), Bacillus licheniformis (BL), Saccharomyces cerevisiae (SC), and the combination of Bacillus licheniformis and Saccharomyces cerevisiae with protease (BS), respectively. The experiment lasted for 66 d. Feed intake was recorded every 2 d and lambs were weighed every 20 d. Ten lambs from each group were slaughtered at the end of the trial, and samples of serum and rumen fluid were collected. The results indicated that the dietary regimen did not affect the dry matter intake (DMI). The average daily gain (ADG) of BS treatment was significantly higher than NC group (P < 0.05). Compared with the NC treatment, the other four supplementation treatments increased the concentration of growth hormone (GH), insulin-like growth factor I (IGF-I) and insulin (INS) (P < 0.05). The malondialdehyde (MDA) and total antioxidant capacity (TAOC) showed no significant difference among the 5 treatments while the activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) of BS group was significantly increased (P < 0.05). The supplementation regimen decreased the concentration of ammonia Nitrogen (NH3-N) and increased the content of microbial crude proteins (MCP) (P < 0.05). The supplementation of antibiotics and probiotics reduced the concentrations of acetate and increased the concentrations of propionate (P < 0.05). The supplementation treatments increased the relative abundance of Lentisphaerae, Fibrobacteres and Tenericutes at the phylum level, whereas at the genus level, they increased the relative abundance of Fibrobacter (P < 0.05). Overall, this study confirmed the facilitating effect of B. licheniformis, S. cerevisiae and their compounds on growth performance, improve the antioxidant capacity and immune function, and beneficially manipulate ruminal fermentation and microbial diversity of fatting lambs.
