During an ongoing virome metagenomics project we identified hepatitis C virus (HCV) in a pool of recently blood-fed common bed bug (<i>Cimex lectularius</i>) nymphs sampled from domestic residences in Europe. Additional PCR and genomic analysis revealed that the virus was a member of HCV genotype 3A, one of the most prevalent genotypes in Europe. Project description:Hepatitis C Virus (HCV) infection is a global public health problem affecting over 160 million individuals worldwide. Its symptoms include chronic hepatitis, liver cirrhosis and hepatocellular carcinoma. HCV is an enveloped RNA virus mainly targeting liver cells and for which the initiation of infection occurs through a complex multistep process involving a series of specific cellular entry factors. This process is likely mediated through the formation of a tightly orchestrated complex of HCV entry factors at the plasma membrane. Among HCV entry factors, the tetraspanin CD81 is one of the best characterized and it is undoubtedly a key player in the HCV lifecycle. In this review, we detail the current knowledge on the involvement of CD81 in the HCV lifecycle, as well as in the immune response to HCV infection.

Project description:Although not presently implicated as a vector of human pathogens, the common bed bug, Cimex lectularius, has been suspected of carrying human pathogens because of its close association with humans and its obligate hematophagy. Recently, we characterized the vectorial competence of C. lectularius for the parasite Trypanosoma cruzi, the causative agent of Chagas disease. We observed that C. lectularius can acquire T. cruzi infection when fed on T. cruzi-carrying mice, and subsequently transmit T. cruzi to uninfected mice. This led us to ask why has C. lectularius not been implicated in the transmission of T. cruzi outside of the laboratory? We hypothesized that T. cruzi reduces C. lectularius fitness (i.e., survival and/or reproduction) as an explanation for why C. lectularius does not to transmit T. cruzi in natural settings. We tested this hypothesis by comparing the survival and reproduction of uninfected and T. cruzi-infected C. lectularius. We observed that T. cruzi had a variable effect on C. lectularius survival and reproduction. There were negligible differences between treatments in juveniles. Infected adult females tended to live longer and produce more eggs. However, no effect was consistent, and infected bugs showed more variation in survival and reproduction metrics than control bugs. We did not observe any negative effects of T. cruzi infection on C. lectularius survival or reproduction, suggesting that decreased fitness in T. cruzi-infected C. lectularius is not why bed bugs have not been observed to transmit T. cruzi in natural settings.

Project description:The common bed bug Cimex lectularius is a temporary ectoparasite on humans and currently resurgent in many developed countries. The ability of bed bugs to detect human odorants in the environment is critical for their host-seeking behavior. This study deciphered the chemical basis of host detection by investigating the neuronal response of olfactory sensilla to 104 human odorants using single sensillum recording and characterized the electro-physiological responses of bed bug odorant receptors to human odorants with the Xenopus expression system. The results showed that the D type of olfactory sensilla play a predominant role in detecting the human odorants tested. Different human odorants elicited different neuronal responses with different firing frequencies and temporal dynamics. Particularly, aldehydes and alcohols are the most effective stimuli in triggering strong response while none of the carboxylic acids showed a strong stimulation. Functional characterization of two bed bug odorant receptors and co-receptors in response to human odorants revealed their specific responses to the aldehyde human odorants. Taken together, the findings of this study not only provide exciting new insights into the human odorant detection of bed bugs, but also offer valuable information for developing new reagents (attractants or repellents) for the bed bug control.

Project description:Hepatitis C virus (HCV) is a major cause of chronic liver disease. However, little is known about the details of its assembly and secretion. Golgi-related proteins have been recently proven to have a key function in HCV secretion. Golgi protein 73 (GP73), a resident Golgi membrane protein, is a potential serum biomarker for the diagnosis of liver diseases and hepatocellular carcinoma. Previous studies have demonstrated the upregulation of GP73 in the liver samples and sera of HCV-infected patients. However, the function and regulatory mechanism of GP73 in HCV infection at the cellular level remain unknown. In this study, we examined the expression level of GP73 in HCV infected cells and its effect on HCV life cycle in cell culture systems. Both the protein expression and mRNA levels of GP73 significantly increased in HCV subgenomic replicon-harboring cells and HCV-infected cells, which imply that GP73 was upregulated by HCV infection. HCV production was significantly enhanced when GP73 was overexpressed, but dramatically inhibited when GP73 was silenced. However, the overexpression and knockdown of GP73 showed no evident effect on the entry, protein translation, RNA replication, and assembly of HCV, which indicates that GP73 enhanced the secretion process. Moreover, the coiled-coil domain of GP73 was required to increase HCV secretion. GP73 increased and interacted with apolipoprotein E, an identified host factor that assists in HCV secretion. These results demonstrate the critical function of GP73 in HCV secretion and provide new insights into the therapeutic design of antiviral strategies.

Project description:As one of the most notorious ectoparasites, bed bugs rely heavily on human or animal blood sources for survival, mating and reproduction. Chemoreception, mediated by the odorant receptors on the membrane of olfactory sensory neurons, plays a vital role in their host seeking and risk aversion processes. We investigated the responses of odorant receptors to a large spectrum of semiochemicals, including human odorants and plant-released volatiles and found that strong responses were sparse; aldehydes/ketones were the most efficient stimuli, while carboxylic acids and aliphatics/aromatics were comparatively less effective in eliciting responses from bed bug odorant receptors. In bed bugs, both the odorant identity and concentrations play important roles in determining the strength of these responses. The odor space constructed based on the responses from all the odorant receptors tested revealed that odorants within the same chemical group are widely dispersed while odorants from different groups are intermingled, suggesting the complexity of odorant encoding in the bed bug odorant receptors. This study provides a comprehensive picture of the olfactory coding mechanisms of bed bugs that will ultimately contribute to the design and development of novel olfactory-based strategies to reduce both the biting nuisance and disease transmission from bed bugs.

Project description:Bed bugs have reemerged recently as a serious and growing problem not only in North America but in many parts of the world. These insects have become the most challenging pest to control in urban environments. Residual insecticides are the most common methods used for bed bug control; however, insecticide resistance limits the efficacy of treatments. Desiccant dusts have emerged as a good option to provide a better residual effect for bed bug control. Several studies have focused on determining the efficacy of dust-based insecticides against bed bugs. However, behavioral responses of bed bugs to insecticide dusts could influence their efficacy. The behavioral responses of bed bugs to six insecticide dusts commonly used in the United States were evaluated with an advanced video tracking technique (Ethovision). Bed bugs took longer to make first contact with areas treated with the diatomaceous earth (DE)-based products MotherEarth D and Alpine than pyrethroid, pyrethrins or silica gel based products, DeltaDust, Tempo 1% Dust and CimeXa, respectively. Lower visitation rates of bed bugs were recorded for areas treated with MotherEarth D, Alpine and CimeXa than that of DeltaDust, Tempo 1% Dust, and Tri-Die Silica + Pyrethrum Dust. Bed bugs spent less time in areas treated with Tri-Die Dust, CimeXa, Alpine, and MotherEarth D than DeltaDust and Tempo 1% Dust, and they exhibited a reduction in locomotor parameters when crawling on areas treated with CimeXa and Alpine. The implications of these responses to bed bug control are discussed.

Project description:The common bed bug, Cimex lectularius harbors the endosymbiotic microorganism, Wolbachia (wCle), in a gonad-associated bacteriome as an obligate nutritional mutualist. The obligatory nature of this association suggests that all individuals in C. lectularius populations would be infected with wCle. However, studies spanning the past several decades have reported variation in both infection frequency and relative abundance of wCle in field-collected samples of bed bugs. Since the growth kinetics of wCle is poorly understood, the objective of this study was to quantify wCle over the life cycle of two strains of C. lectularius. Our results highlight that wCle is dynamic during bed bug development, changing relative to life stage, intermolt stage, and blood-fed status. These results suggest new hypotheses about the coordination of Wolbachia growth and regression with its host's physiology and endocrine events. The observed quantitative modulation of wCle during the bed bug life cycle and during periods of starvation may explain the disparities in wCle infections reported in field-collected C. lectularius.

Project description:A distinctive feature of HCV is that its life cycle depends on lipoprotein metabolism. Viral morphogenesis and secretion follow the very low-density lipoprotein (VLDL) biogenesis pathway and, consequently, infectious HCV in the serum is associated with triglyceride-rich lipoproteins (TRL). Lipoprotein lipase (LPL) hydrolyzes TRL within chylomicrons and VLDL but, independently of its catalytic activity, it has a bridging activity, mediating the hepatic uptake of chylomicrons and VLDL remnants. We previously showed that exogenously added LPL increases HCV binding to hepatoma cells by acting as a bridge between virus-associated lipoproteins and cell surface heparan sulfate, while simultaneously decreasing infection levels. We show here that LPL efficiently inhibits cell infection with two HCV strains produced in hepatoma cells or in primary human hepatocytes transplanted into uPA-SCID mice with fully functional human ApoB-lipoprotein profiles. Viruses produced in vitro or in vivo were separated on iodixanol gradients into low and higher density populations, and the infection of Huh 7.5 cells by both virus populations was inhibited by LPL. The effect of LPL depended on its enzymatic activity. However, the lipase inhibitor tetrahydrolipstatin restored only a minor part of HCV infectivity, suggesting an important role of the LPL bridging function in the inhibition of infection. We followed HCV cell entry by immunoelectron microscopy with anti-envelope and anti-core antibodies. These analyses demonstrated the internalization of virus particles into hepatoma cells and their presence in intracellular vesicles and associated with lipid droplets. In the presence of LPL, HCV was retained at the cell surface. We conclude that LPL efficiently inhibits HCV infection by acting on TRL associated with HCV particles through mechanisms involving its lipolytic function, but mostly its bridging function. These mechanisms lead to immobilization of the virus at the cell surface. HCV-associated lipoproteins may therefore be a promising target for the development of new therapeutic approaches.

Project description:Hepatitis C virus (HCV)-specific CD8 cell exhaustion may represent a mechanism of HCV persistence. The inhibitory receptor PD-1 has been reported to be up-regulated in exhausted CD8 cells. Therefore, we studied PD-1 expression longitudinally during acute HCV infection. Most HCV-specific CD8 cells expressed PD-1 at the time of acute illness, irrespective of the final outcome. PD-1 expression declined with the acquisition of a memory phenotype and recovery of an efficient CD8 cell function in resolving HCV infections, whereas high levels were maintained when HCV persisted and HCV-specific CD8 cells remained dysfunctional. Blocking PD-1/PDL-1 interaction with an anti-PDL-1 antibody improved the capacity of expansion of virus-specific CD8 cells.

Project description:The insect male accessory gland (MAG) is an internal reproductive organ responsible for the synthesis and secretion of seminal fluid components, which play a pivotal role in the male reproductive strategy. In many species of insects, the effective ejaculation of the MAG products is essential for male reproduction. For this purpose, the fruit fly Drosophila has evolved binucleation in the MAG cells, which causes high plasticity of the glandular epithelium, leading to an increase in the volume of seminal fluid that is ejaculated. However, such a binucleation strategy has only been sporadically observed in Dipteran insects, including fruit flies. Here, we report the discovery of binucleation in the MAG of the common bed bug, Cimex lectularius, which belongs to hemimetabolous Hemiptera phylogenetically distant from holometabolous Diptera. In Cimex, the cell morphology and timing of synchrony during binucleation are quite different from those of Drosophila. Additionally, in Drosophila, the position of the two nuclei in the adult stage changes as a result of the mating history or the nutrient conditions; however, it remains stable in Cimex. These differences suggest that binucleation in the Cimex MAG plays a unique role in the male reproductive system that is distinct from that of Drosophila.
