Facile Fabrication of Oxygen-Releasing Tannylated Calcium Peroxide Nanoparticles.

ABSTRACT: This study reports a new approach for the facile fabrication of calcium peroxide (CaO2) nanoparticles using tannic acid (TA) as the coordinate bridge between calcium ions. Tannylated-CaO2 (TA/CaO2) nanoparticles were prepared by reacting calcium chloride (CaCl2) with hydrogen peroxide (H2O2) in ethanol containing ammonia and different amounts of TA (10, 25, and 50 mg). The prepared TA/CaO2 aggregates consisted of nanoparticles 25-31 nm in size. The nanoparticles prepared using 10 mg of TA in the precursor solution exhibited the highest efficiency for oxygen generation. Moreover, the oxygen generation from TA (10 mg)/CaO2 nanoparticles was higher in an acidic environment.

The Oxygen-Generating Calcium Peroxide-Modified Magnetic Nanoparticles Attenuate Hypoxia-Induced Chemoresistance in Triple-Negative Breast Cancer.

Project description:Cancer response to chemotherapy is regulated not only by intrinsic sensitivity of cancer cells but also by tumor microenvironment. Tumor hypoxia, a condition of low oxygen level in solid tumors, is known to increase the resistance of cancer cells to chemotherapy. Triple-negative breast cancer (TNBC) is the most aggressive subtype of breast cancer. Due to lack of target in TNBC, chemotherapy is the only approved systemic treatment. We evaluated the effect of hypoxia on chemotherapy resistance in TNBC in a series of in vitro and in vivo experiments. Furthermore, we synthesized the calcium peroxide-modified magnetic nanoparticles (CaO2-MNPs) with the function of oxygen generation to improve and enhance the therapeutic efficiency of doxorubicin treatment in the hypoxia microenvironment of TNBC. The results of gene set enrichment analysis (GSEA) software showed that the hypoxia and autophagy gene sets are significantly enriched in TNBC patients. We found that the chemical hypoxia stabilized the expression of hypoxia-inducible factor 1α (HIF-1α) protein and increased doxorubicin resistance in TNBC cells. Moreover, hypoxia inhibited the induction of apoptosis and autophagy by doxorubicin. In addition, CaO2-MNPs promoted ubiquitination and protein degradation of HIF-1α. Furthermore, CaO2-MNPs inhibited autophagy and induced apoptosis in TNBC cells. Our animal studies with an orthotopic mouse model showed that CaO2-MNPs in combination with doxorubicin exhibited a stronger tumor-suppressive effect on TNBC, compared to the doxorubicin treatment alone. Our findings suggest that combined with CaO2-MNPs and doxorubicin attenuates HIF-1α expression to improve the efficiency of chemotherapy in TNBC.

Tin Oxide (SnO2) Nanoparticles: Facile Fabrication, Characterization, and Application in UV Photodetectors.

Project description:Tin oxide (SnO2) nanomaterials are of great interest in many fields such as catalytic, electrochemical, and biomedical applications, due to their low cost, suitable stability characteristics, high photosensitivity, etc. In this contribution, SnO2 NPs were facilely fabricated by calcination of tin (II) oxalate in air, followed by a liquid-phase exfoliation (LPE) method. Size-selected SnO2 NPs were easily obtained using a liquid cascade centrifugation (LCC) technique. The as-obtained SnO2 NPs displayed strong absorption in the UV region (~300 nm) and exhibited narrower absorption characteristics with a decrease in NP size. The as-fabricated SnO2 NPs were, for the first time, directly deposited onto a poly(ethylene terephthalate) (PET) film with a regular Ag lattice to fabricate a flexible working electrode for a photoelectrochemical (PEC)-type photodetector. The results demonstrated that the SnO2-NP-based electrode showed the strongest photoresponse signal in an alkaline electrolyte compared with those in neutral and acidic electrolytes. The maximum photocurrent density reached 14.0 μA cm-2, significantly outperforming black phosphorus nanosheets and black phosphorus analogue nanomaterials such as tin (II) sulfide nanosheets and tellurene. The as-fabricated SnO2 NPs with relatively larger size had better self-powered photoresponse performance. In addition, the as-fabricated SnO2-NP-based PEC photodetector exhibited strong cycling stability for on/off switching behavior under ambient conditions. It is anticipated that SnO2 nanostructures, as building blocks, can offer diverse availabilities for high-performance self-powered optoelectronic devices to realize a carbon-neutral or carbon-free environment.

Facile Fabrication of Fluorescent Inorganic Nanoparticles with Diverse Shapes for Cell Imaging.

Project description:In the present work, we describe a facile and general method of fabricating fluorescent inorganic nanoparticles with diverse shapes for cell imaging application. The hematite (α-Fe₂O₃) nanoparticles (HNPs) with three different shapes (i.e., spindle shape, ellipsoidal shape and quasi-spherical shape) were first prepared as model systems in consideration of good biocompatibility and the controllable morphology of α-Fe₂O₃. Three fluorescent HNPs with different shapes were readily achieved via one-pot sol-gel reaction of AIE luminogen-functionalized siloxane (AIEgen-Si(OCH₃)₃) and TEOS in the presence of PVP-stabilized HNPs. Due to the fluorescence originating from the thin AIEgens-contained SiO₂ shell around the HNPs, their photoluminescent intensities can be tuned by changing the concentrations of TEOS and AIEgen-Si(OCH₃)₃ in feed prior to the sol-gel reaction. When the as-prepared fluorescent products were dispersed in water, they gave intense green light emission upon excitation at 360 nm with relatively high fluorescence quantum yield. Further, fluorescent HNPs exhibited low cytotoxicity and excellent photostability and, thus, were used as optical probes to preliminarily explore the effect of nanoparticle shapes on their cellular uptake behaviors. This work should open a facile way to prepare various fluorescent inorganic nanoparticles with specific morphology for various biological applications.

Facile Fabrication of Nickel Supported on Reduced Graphene Oxide Composite for Oxygen Reduction Reaction.

Project description:Due to the depletion of fossil fuels, the demand for renewable energy has increased, thus stimulating the development of novel materials for energy conversion devices such as fuel cells. In this work, nickel nanoparticles loaded on reduced graphene oxide (Ni/rGO) with small size and good dispersibility were successfully prepared by controlling the pyrolysis temperature of the precursor at 450 °C, assisted by a microwave-assisted hydrothermal method, and exhibited enhanced electrocatalytic activity towards oxygen reduction reaction (ORR). Additionally, the electron enrichment on Ni NPs was due to charge transfer from the rGO support to metal nickel, as evidenced by both experimental and theoretical studies. Metal-support interactions between nickel and the rGO support also facilitated charge transfer, contributing to the enhanced ORR performance of the composite material. DFT calculations revealed that the first step (from O2 to HOO*) was the rate-determining step with an RDS energy barrier lower than that of the Pt(111), indicating favorable ORR kinetics. The HOO* intermediates can be transferred onto rGO by the solid-phase spillover effect, which reduces the chemical adsorption on the nickel surface, thereby allowing continuous regeneration of active nickel sites. The HO2- intermediates generated on the surface of rGO by 2e- reduction can also efficiently diffuse towards the nearby Ni surface or the interface of Ni/rGO, where they can be further rapidly reduced to OH-. This mechanism acts as the pseudo-four-electron path on the RRDE. Furthermore, Ni/rGO-450 demonstrated superior stability, methanol tolerance, and durability compared to a 20 wt% Pt/C catalyst, making it a cost-effective alternative to conventional noble metal ORR catalysts for fuel cells or metal-air batteries.

Facile and Green Fabrication of Carrageenan-Silver Nanoparticles for Colorimetric Determination of Cu2+ and S2.

Project description:In the present work, silver nanoparticles (AgNPs) were prepared by a simple and green method using carrageenan as reducing and capping agent. The as-synthesized carrageenan-AgNPs was demonstrated as an effective duel colorimetric sensing for selective and sensitive recognition of Cu2+ and S2-, which could be used to detect these ions with naked eyes. In addition, the possible sensing mechanism was that Cu2+ ions caused serious aggregation of carrageenan-AgNPs, which led to the color change of carrageenan-AgNPs. AgNPs were etched by S2- forming Ag2S, which played an important role in the determination of S2- ions. Furthermore, it has been successfully applied to the determination of Cu2+ and S2- in tap water and lake water, showing its great potential for the analysis of environmental water samples.

Polymeric Composite Dressings Containing Calcium-Releasing Nanoparticles Accelerate Wound Healing in Diabetic Mice.

Project description:Objective: Wound healing is a complex process that involves the interaction between different cell types and bioactive factors. Impaired wound healing is characterized by a loss in synchronization of these interactions, resulting in nonhealing chronic wounds. Chronic wounds are a socioeconomic burden, one of the most prominent clinical manifestations of diabetes, however, they lack satisfactory treatment options. The objective of this study was to develop polymeric composites that deliver ions having wound healing properties and evaluate its performance using a pressure ulcer model in diabetic mice. Approach: To develop a polymeric composite wound dressing containing ion-releasing nanoparticles for chronic wound healing. This composite was chemically and physically characterized and evaluated using a pressure ulcer wound model in diabetic (db/db) mice to explore their potential as novel wound dressing. Results: This dressing exhibits a controlled ion release and a good in vitro bioactivity. The polymeric composite dressing treatment stimulates angiogenesis, collagen synthesis, granulation tissue formation, and accelerates wound closure of ischemic wounds created in diabetic mice. In addition, the performance of the newly designed composite is remarkably better than a commercially available dressing frequently used for the treatment of low-exuding chronic wounds. Innovation: The developed nanoplatforms are cell- and growth factor free and control the host microenvironment resulting in enhanced wound healing. These nanoplatforms are available by cost-effective synthesis with a defined composition, offering an additional advantage in potential clinical application. Conclusion: Based on the obtained results, these polymeric composites offer an optimum approach for chronic wound healing without adding cells or external biological factors.

Facile Fabrication of Platinum-Cobalt Alloy Nanoparticles with Enhanced Electrocatalytic Activity for a Methanol Oxidation Reaction.

Project description:Decreasing the cost associated with platinum-based catalysts along with improving their catalytic properties is a major challenge for commercial direct methanol fuel cells. In this work, a simple and facile strategy was developed for the more efficient preparation of multi-walled carbon nanotube (MWCNT) -supported Pt/CoPt composite nanoparticles (NPs) via solution plasma sputtering with subsequent thermal annealing. Quite different from general wet synthesis methods, Pt/CoPt composite NPs were directly derived from metal wire electrodes without any additions. The obtained Pt/CoPt/MWCNTs composite catalysts exhibited tremendous improvement in the electro-oxidation of methanol in acidic media with mass activities of 1719 mA mg-1Pt. This value is much higher than that of previous reports of Pt-Co alloy and commercial Pt/C (3.16 times) because of the many active sites and clean surface of the catalysts. The catalysts showed good stability due to the special synergistic effects of the CoPt alloy. Pt/CoPt/MWCNTs can be used as a promising catalyst for direct methanol fuel cells. In addition, this solution plasma sputtering-assisted synthesis method introduces a general and feasible route for the synthesis of binary alloys.

Facile Fabrication of Protein-Macrocycle Frameworks.

Project description:Precisely defined protein aggregates, as exemplified by crystals, have applications in functional materials. Consequently, engineered protein assembly is a rapidly growing field. Anionic calix[n]arenes are useful scaffolds that can mold to cationic proteins and induce oligomerization and assembly. Here, we describe protein-calixarene composites obtained via cocrystallization of commercially available sulfonato-calix[8]arene (sclx8) with the symmetric and "neutral" protein RSL. Cocrystallization occurred across a wide range of conditions and protein charge states, from pH 2.2-9.5, resulting in three crystal forms. Cationization of the protein surface at pH ∼ 4 drives calixarene complexation and yielded two types of porous frameworks with pore diameters >3 nm. Both types of framework provide evidence of protein encapsulation by the calixarene. Calixarene-masked proteins act as nodes within the frameworks, displaying octahedral-type coordination in one case. The other framework formed millimeter-scale crystals within hours, without the need for precipitants or specialized equipment. NMR experiments revealed macrocycle-modulated side chain pKa values and suggested a mechanism for pH-triggered assembly. The same low pH framework was generated at high pH with a permanently cationic arginine-enriched RSL variant. Finally, in addition to protein framework fabrication, sclx8 enables de novo structure determination.

Facile Fabrication of Hierarchical rGO/PANI@PtNi Nanocomposite via Microwave-Assisted Treatment for Non-Enzymatic Detection of Hydrogen Peroxide.

Project description:A hierarchical composite based on the modified reduced graphene oxide with platinum-nickel decorated polyaniline nano-spheres (rGO/PANI@PtNi) was facilely prepared via microwave-assisted self-reduction for an application in nonenzymatic hydrogen peroxide (H2O2) detection. Compared to the pristine rGO, the composite exhibited a much tougher surface due to the stacking of conductive PANI nano-spheres on rGO sheets, leading to good dispersion of PtNi nanoparticles and a large active area. Furthermore, the multi-valance Ni2+/3+ in the PtNi particles effectively promoted the catalytic property of Pt sites and facilitated a superior electrochemical performance of PtNi alloy than that of neat Pt. Owing to the synergistic effect of the improved electrical conductivity and the promoted electrocatalytical property, the modified glassy carbon electrode (GCE) with rGO/PANI@PtNi nanocomposite displayed an outstanding electrochemical sensitivity towards H2O2 with a fast response time (<2 s), a wide linear range (0.1-126.4 mM), a low detection limit (0.5 µM), as well as a long-life stability for one week without obvious degradation. This novel strategy opens a novel and promising approach to design high performance sensors for H2O2 detection.

Facile Osteoimmunomodulatory Nanoparticles Augment 3D-Printed Scaffold-Mediated Bone Regeneration

Project description:Regenerating critical-sized long bone defects poses substantial challenges due to limitations of autografts and processed allografts. Biomaterial scaffolds offer versatile alternatives, yet their effectiveness is often constrained by their limited innate osteoinductivity. While growth factors and cells can enhance osteoinduction, the inclusion of biologics in biomaterial scaffolds creates regulatory challenges for clinical translation. To address this, here we describe three-dimensional (3D) printed polycaprolactone (PCL) scaffolds for temporally controlled delivery of osteoimmunomodulatory amorphous calcium phosphate-chitosan nanoparticles (ACPC-NP). In vitro, the ACPC-NP exhibit concentration dependent effects on osteoblasts, monocytes, and osteoclasts. At increasing concentrations up to 500 μg/ml, these nanoparticles stimulate osteogenesis, modulate M2/M1 macrophage polarization, and inhibit osteoclast maturation and activity. Leveraging these concentration-dependent effects in vivo through temporally controlled release of ACPC-NP from 3D-printed PCL scaffolds, we observe the complete regeneration and the restoration of biomechanical strength of critically sized radial defects in rats. Such healing is absent in defects implanted with bare PCL scaffolds or those loaded with calcium-phosphate microparticles. The tunable osteoimmunomodulation by the NP underscore the translational potential of this technology to yield structurally sound and functionally robust bone regeneration outcomes.
