Project description:The distribution of Triplophysa yarkandensis is restricted to Xinjiang's Tarim River basin. We collected 119 T. yarkandensis samples from nine geographic populations in the Tarim River basin and utilized the RAD-seq method for SNP genotyping. In this study, a total of 164.81 Gb bases were generated with the Illumina platform, and 129,873 candidate SNPs were obtained with the Stacks pipeline for population genetic analyses. High levels of genetic diversity were detected among nine populations. The AMOVA results showed that the majority of genetic variations originated from among populations (FST = 0.67), and the pairwise FST values ranged from 0.4579 to 0.8736, indicating high levels of genetic differentiation among these populations. The discriminate analysis of principal components (DAPCs) and neighbor joining (NJ) tree revealed that the nine populations could be separated into two clusters (i.e., south and north populations), and modest genetic differentiation between south and north populations was observed, while the individuals from several populations were not clustered together by geographical location. The evidence of two genetic boundaries between south and north populations (except TTM) was supported by barrier analysis. The Bayesian skyline plotting indicated that T. yarkandensis populations in the Tarim River basin had not experienced genetic bottlenecks, and the effective population size remained stable. This study first clarified the genetic diversity and differentiation of T. yarkandensis populations in the Tarim River basin, and it provided valuable molecular data for conservation and management of natural populations.

Project description:The Tibetan Plateau uplift and Cenozoic global cooling are thought to induce enhanced aridification in the Asian interior. Although the onset of Asian desertification is proposed to have started in the earliest Miocene, prevailing desert environment in the Tarim Basin, currently providing much of the Asian eolian dust sources, is only a geologically recent phenomenon. Here we report episodic occurrences of lacustrine environments during the Late Miocene and investigate how the episodic lakes vanished in the basin. Our oxygen isotopic (δ(18)O) record demonstrates that before the prevailing desert environment, episodic changes frequently alternating between lacustrine and fluvial-eolian environments can be linked to orbital variations. Wetter lacustrine phases generally corresponded to periods of high eccentricity and possibly high obliquity, and vice versa, suggesting a temperature control on the regional moisture level on orbital timescales. Boron isotopic (δ(11)B) and δ(18)O records, together with other geochemical indicators, consistently show that the episodic lakes finally dried up at ∼4.9 million years ago (Ma), permanently and irreversibly. Although the episodic occurrences of lakes appear to be linked to orbitally induced global climatic changes, the plateau (Tibetan, Pamir, and Tianshan) uplift was primarily responsible for the final vanishing of the episodic lakes in the Tarim Basin, occurring at a relatively warm, stable climate period.

Project description:Livestock husbandry is vital to economy of the Tarim Basin, Xinjiang Autonomous Region, China. However, there have been few surveys of the distribution of ixodid ticks (Acari: Ixodidae) and tick-borne pathogens affecting domestic animals at these locations. In this study, 3,916 adult ixodid ticks infesting domestic animals were collected from 23 sampling sites during 2012-2016. Ticks were identified to species based on morphology, and the identification was confirmed based on mitochondrial 16S and 12S rRNA sequences. Ten tick species belonging to 4 genera were identified, including Rhipicephalus turanicus, Hyalomma anatolicum, Rh. bursa, H. asiaticum asiaticum, and Rh. sanguineus. DNA sequences of Rickettsia spp. (spotted fever group) and Anaplasma spp. were detected in these ticks. Phylogenetic analyses revealed possible existence of undescribed Babesia spp. and Borrelia spp. This study illustrates potential threat to domestic animals and humans from tick-borne pathogens.

Project description:A series of trace compounds (diamondoids, ethanodiamondoids, and thiadiamondoids) were detected through two-dimensional gas chromatography/time-of-flight mass spectrometry (GC × GC-TOFMS) analysis of Ordovician condensate samples from the Tazhong area. Gas chromatography-mass spectrometry (GC-MS) analysis showed that the biomarker parameters are less effective for high-maturity oils. Carbon isotope and geochemical features suggested that the gas is a high-temperature cracking gas when its temperature is higher than 170 °C. The H2S content is 8.27%, suggesting that it is affected by thermochemical sulfate reduction (TSR). However, the geological analysis indicated that the Ordovician reservoirs do not satisfy the conditions for TSR. The high-maturity oil in the Ordovician reservoirs may generate diamondoids and ethanodiamondoids when cracking, while TSR and severe cracking occur in deep Cambrian source rocks and produce a large number of diamondoids, ethanodiamondoids, organic sulfur compounds (OSCs), etc. The secondary geochemical products that are carried up by the dry gas and migrate upward through faults and are enriched in Ordovician crude oil reservoirs. On this basis, we proposed that the condensate presented was formed by the mixing of dry gas from Cambrian oil that was altered by cracking and TSR into Ordovician in situ slightly cracked oil, therefore speculating that the favorable reservoir-seal assemblages in this area may contain abundant oil and gas resources. Consequently, improved knowledge of secondary alteration effects on the reservoir and underground fluids is vital for oil and gas prediction and exploration development in the next step.

Project description:The Tarim River Basin in Xinjiang, China, has a typical desert riparian forest ecosystem. Analysis of the resilience of this type of ecosystem under extreme drought conditions and ecological rehabilitation projects could provide a theoretical basis for understanding ecosystem stability and resistance, and provide new ecological rehabilitation measures to improve ecosystem resilience. We employed a quantitative framework to assess net primary productivity (NPP) resilience, emphasizing four aspects of NPP dynamics: NPP, NPP stability, NPP resistance, and maximum NPP potential. We compared ecosystem resilience across four time periods: before the implementation of ecological rehabilitation projects (1990-2000), during construction and partial implementation of ecological rehabilitation projects (2001-2012), during the initial project stage of ecological rehabilitation (2013-2015), and during the late project stage of ecological rehabilitation (2016-2018). There are three main finding of this research. (1) Mean NPP was increased significantly from 2013 and was decreased from 2016, especially in the main stream of the Tarim River and in the basins of eight of its nine tributary rivers. (2) Ecosystem resilience in 2013-2018 was greater than in 1990-2012, with the greatest NPP stability, mean NPP and NPP resistance, especially in part one of the river basin (the Aksu River, the Weigan-Kuche River, the Dina River, the Kaidu-Konqi River, and the main stream of the Tarim River). Ecosystem resilience in 2001-2012 was lowest when compared to 1990-2000 and 2013-2018, with lowest mean NPP, NPP stability, NPP resistance and maximum NPP potential, particularly in part two of the river basin (the Kashigr River, the Yarkand River and the Hotan River basins). Therefore, part one was most affected by ecological restoration projects. When 2013-2018 was divided into two distinct stages, 2013-2015 and 2016-2018, resilience in the latter stage was the lowest, with lowest mean NPP, NPP resistance and maximum NPP potential, especially in the main stream of the Tarim River. This may be due to unreasonable water conveyance in 2014-2015. (3) Ecological resilience has increased significantly in 2013-2015 after the implementation of ecological water transfer projects, river regulation, and natural vegetation enclosure projects. Ecosystem resilience could continue to increase even more in the future with the continued implementation of reasonable ecological water transfer projects.

Project description:Argasid ticks (Acari: Argasidae) carry and transmit a variety of pathogens of animals and humans, including viruses, bacteria and parasites. There are several studies reporting ixodid ticks (Acari: Ixodidae) and associated tick-borne pathogens in Xinjiang, China. However, little is known about the argasid ticks and argasid tick-associated pathogens in this area. In this study, a total of 3829 adult argasid ticks infesting livestock were collected at 12 sampling sites of 10 counties in the Peripheral Oases, which carry 90% of the livestock and humans population, around the Tarim Basin (southern Xinjiang) from 2013 to 2016. Tick specimens were identified to two species from different genera by morphology and sequences of mitochondrial 16S rRNA and 12S rRNA were derived to confirm the species designation. The results showed that the dominant argasid ticks infesting livestock in southern Xinjiang were Ornithodoros lahorensis (87.86%, 3364/3829). Ornithodoros lahorensis was distributed widely and were collected from 10 counties of southern Xinjiang. Argas japonicus was collected from Xinjiang for the first time. In addition, we screened these ticks for tick-associated pathogens and showed the presence of DNA sequences of Rickettsia spp. of Spotted fever group and Anaplasma spp. in the argasid ticks. This finding suggests the potential role for Argas japonicus as a vector of pathogens to livestock and humans.

Project description:The Tarim Basin in western China, known for its amazingly well-preserved mummies, has been for thousands of years an important crossroad between the eastern and western parts of Eurasia. Despite its key position in communications and migration, and highly diverse peoples, languages and cultures, its prehistory is poorly understood. To shed light on the origin of the populations of the Tarim Basin, we analysed mitochondrial DNA polymorphisms in human skeletal remains excavated from the Xiaohe cemetery, used by the local community between 4000 and 3500 years before present, and possibly representing some of the earliest settlers.Xiaohe people carried a wide variety of maternal lineages, including West Eurasian lineages H, K, U5, U7, U2e, T, R*, East Eurasian lineages B, C4, C5, D, G2a and Indian lineage M5.Our results indicate that the people of the Tarim Basin had a diverse maternal ancestry, with origins in Europe, central/eastern Siberia and southern/western Asia. These findings, together with information on the cultural context of the Xiaohe cemetery, can be used to test contrasting hypotheses of route of settlement into the Tarim Basin.

Project description:The bacterial diversity in Populus euphratica stem storage liquid samples grown in Shaya County and Yuli County of the Tarim River Basin was investigated. A culture-dependent (dilution spread plate method) and culture-independent method (PCR-RFLP technique) were used to identify the endophytic bacteria community structure and composition in P. euphratica in Tarim River Basin. Sixty-six bacterial strains were isolated from P. euphratica stem storage liquid samples on three agar media. The 16S rDNA gene was amplified and sequenced using bacterial universal primers. Phylogenetic analysis showed that the 66 strains belonged to three phyla (Firmicutes, Actinomycetes, and Gamma-Proteobacteria) and included 16 genera and 29 species. Among them, Pseudomonas (27.27%) and Bacillus (19.69%) were the dominant isolates. CGM-17 was a potentially new species of Pantoea. Restriction fragment length polymorphism of 16S rDNA gene amplified by polymerase chain reaction (PCR-RFLP) revealed 48 operational taxonomic units (OTUs). Phylogenetic analysis indicated that the 48 OTUs belonged to Firmicutes, Actinobacteria, Proteobacteria (α-, β-, γ-subgroup), Bacteroidetes, and Verrucomicrobia. Gamma-Proteobacteria was the dominant group, similarly to the culture-dependent method, accounting for 53% of the entire bacterial clone library. Our results indicate that P. euphratica endophytic bacteria diversity in the Tarim River Basin was rich, and the resources of endophytic bacteria were high. They provide valuable reference data and species resources for screening indigenous and functional strains of endophytic bacteria in P. euphratica.

Project description:The complete mitochondrial genome of the microalgae Dunaliella salina strain SQ (GenBank accession number: KX641170) was de novo assembled and annotated using Illumina MiSeq sequencing data. The mitochondrial genome is 41,904 bp long with 31.85% GC content and contains 7 protein-coding genes, 16 introns, 3 ribosomal RNA genes and 3 transfer RNA genes. To date, only two complete mitochondrial genomes of Dunaliella salina strains have been reported, and this genome provides knowledge to the study of genetic variations and evolution of mitochondrial genomes of Dunaliella salina strains.

Project description:Microalgae have been identified to produce a plethora of bioactive compounds exerting growth stimulating effects on plants. The objective of this study was to investigate the plant-growth-promoting effects of three selected strains of eukaryotic green microalgae. The biostimulatory effects of two Chlorella species (MACC-360 and MACC-38) and a Chlamydomonas reinhardtii strain (cc124) were investigated in a Medicago truncatula model plant grown under controlled greenhouse conditions. The physiological responses of the M. truncatula A17 ecotype to algal biomass addition were characterized thoroughly. The plants were cultivated in pots containing a mixture of vermiculite and soil (1:3) layered with clay at the bottom. The application of live algae cells using the soil drench method significantly increased the plants' shoot length, leaf size, fresh weight, number of flowers and pigment content. For most of the parameters analyzed, the effects of treatment proved to be specific for the applied algae strains. Overall, Chlorella application led to more robust plants with increased fresh biomass, bigger leaves and more flowers/pods compared to the control and Chlamydomonas-treated samples receiving identical total nutrients.
