By analyzing successive lifestyle stages of a model <i>Rhizobium-</i>legume symbiosis using mariner-based transposon insertion sequencing (INSeq), we have defined the genes required for rhizosphere growth, root colonization, bacterial infection, N<sub>2</sub>-fixing bacteroids, and release from legume (pea) nodules.
