Remediation of heavy metal contamination of sediments and soils using ligand-coated dense nanoparticles.

ABSTRACT: Sediment and soil contamination with toxic heavy metals, including cadmium (Cd2+) and lead (Pb2+), represents a major long-term remediation challenge. Resuspension of contaminated sediments into the water column, or the uptake of toxic metals from top soil, can lead to exposure of aquatic or terrestrial organisms, followed by bioconcentration, bioaccumulation and biomagnification, which may pose a threat to public health. We have developed a novel nanoscale engineered material, namely ligand-coated dense nanoparticles (Ligand DNPs), which contain a dense WO3 nanoparticle core and a shell functionalized with a metal-binding organic ligand (EDTA), to effectively sequester heavy metal ions deeper into the soil and sediments. We demonstrate that one application of Ligand DNPs can remove from 60% to almost 80% of the Cd and Pb in two different soil matrices, driving these metal ions deeper into the sediment or soil column via gravity, and making them less bioavailable. Ligand DNPs can provide a relatively fast, convenient, and efficient in-situ approach for the remediation of sediments and soils contaminated with heavy metals.

Project description:BackgroundIncreased growth of industrial activities, especially in urban centers, is one of the main sources of toxic substances in Nigeria. The level of these impacts is not well known. Soil and sediment samples from one such industrial area were examined for their mineralogical composition and heavy metals contents in order to assess the level of contamination and potential ecological risk status.MethodsMineralogical composition of the media and their heavy metals concentrations were determined using X-ray diffractometry and inductively coupled plasma-mass spectrometry methods, respectively. Ecological risk assessment was carried out using single (contamination factor, geo-accumulation index, enrichment factor) and multi-elemental (contamination degree, pollution index and modified pollution index) standard indices.ResultsThe average heavy metal concentrations in soils and sediments followed the order magnesium (Mn) > chromium (Cr) > lead (Pb) > copper (Cu) > cadmium (Cd) > cobalt (Co) > nickel (Ni), with corresponding values for soils and sediments of 324.3, 79.9, 66.1, 40.7, 14.3, 9.1, 6.8 mg kg-1 and 266.8, 78.6, 40.6, 39.8, 12.9, 8.4, 4.6 mg kg-1, respectively. Principal component (PC) analysis of the results indicated three main sources of metals (industrial, vehicular activities and geogenic input). Evaluated contamination factor (Cf), enrichment factor (Ef) and geo-accumulation index (Igeo) revealed very high contamination for Pb, Cd and Cu in all of the samples, with calculated pollution index (PI) and modified pollution index (MPI) revealing that all the samples were severely polluted. Calculated potential ecological risk factor (ERi) within the industrial area demonstrated a strong potential ecological risk for Cd, Pb and Cu.ConclusionsActivities in the industrial area have affected the quality of the analyzed environmental media, with possible detrimental health consequences. Regular environmental monitoring of the industrial area and the formulation of appropriate policies that support reduction of contamination are strongly recommended. Project description:Soil microorganisms play a crucial role in remediating contaminated soils in modern ecosystems. However, the potential of combining microorganisms with legumes to enhance the remediation of heavy metal-contaminated soils remains unexplored. To investigate this, we isolated and purified a highly efficient cadmium and lead-tolerant strain. Through soil-cultivated pot experiments with two leguminous plants (Robinia pseudoacacia L. and Sophora xanthantha), we studied the effects of applying this microbial agent on plant nutrient uptake of soil nutrients, heavy metal accumulation, and the dynamics of heavy metal content. Additionally, we examined the response characteristics of inter-root microbial and bacterial communities. The results demonstrated that microorganisms screened from heavy metal-contaminated soil environments exhibited strong survival and adaptability in heavy metal solutions. The use of the Serratia marcescens WZ14 strain-phytoremediation significantly increased the soil's ammonium nitrogen (AN) and organic carbon (OC) contents compared to monoculture. In addition, the lead (Pb) and cadmium (Cd) contents of the soil significantly decreased after combined remediation than those of the soil before potting. However, the remediation effects on Pb- and Cd-contaminated soils differed between the two legumes following the Serratia marcescens WZ14 inoculation. The combined restoration altered the composition of the plant inter-rhizosphere bacterial community, with the increase in the relative abundance of both Proteobacteria and Firmicutes. Overall, the combined remediation using the tolerant strain WZ14 with legumes proved advantageous. It effectively reduced the heavy metal content of the soil, minimized the risk of heavy metal migration, and enhanced heavy metal uptake, accumulation, and translocation in the legumes of S. xanthantha and R. pseudoacacia. Additionally, it improved the adaptability and resistance of both legumes, leading to an overall improvement in the soil's environmental quality. These studies can offer primary data and technical support for remediating and treating Cd and Pb in soils, as well as rehabilitating mining sites.

Project description:Anthyllis vulneraria is a legume associated with nitrogen-fixing rhizobia that together offer an adapted biological material for mine-soil phytostabilization by limiting metal pollution. To find rhizobia associated with Anthyllis at a given site, we evaluated the genetic and phenotypic properties of a collection of 137 rhizobia recovered from soils presenting contrasting metal levels. Zn-Pb mine soils largely contained metal-tolerant rhizobia belonging to Mesorhizobium metallidurans or to another sister metal-tolerant species. All of the metal-tolerant isolates harbored the cadA marker gene (encoding a metal-efflux PIB-type ATPase transporter). In contrast, metal-sensitive strains were taxonomically distinct from metal-tolerant populations and consisted of new Mesorhizobium genospecies. Based on the symbiotic nodA marker, the populations comprise two symbiovar assemblages (potentially related to Anthyllis or Lotus host preferences) according to soil geographic locations but independently of metal content. Multivariate analysis showed that soil Pb and Cd concentrations differentially impacted the rhizobial communities and that a rhizobial community found in one geographically distant site was highly divergent from the others. In conclusion, heavy metal levels in soils drive the taxonomic composition of Anthyllis-associated rhizobial populations according to their metal-tolerance phenotype but not their symbiotic nodA diversity. In addition to heavy metals, local soil physicochemical and topoclimatic conditions also impact the rhizobial beta diversity. Mesorhizobium communities were locally adapted and site specific, and their use is recommended for the success of phytostabilization strategies based on Mesorhizobium-legume vegetation. IMPORTANCE:Phytostabilization of toxic mine spoils limits heavy metal dispersion and environmental pollution by establishing a sustainable plant cover. This eco-friendly method is facilitated by the use of selected and adapted cover crop legumes living in symbiosis with rhizobia that can stimulate plant growth naturally through biological nitrogen fixation. We studied microsymbiont partners of a metal-tolerant legume, Anthyllis vulneraria, which is tolerant to very highly metal-polluted soils in mining and nonmining sites. Site-specific rhizobial communities were linked to taxonomic composition and metal tolerance capacity. The rhizobial species Mesorhizobium metallidurans was dominant in all Zn-Pb mines but one. It was not detected in unpolluted sites where other distinct Mesorhizobium species occur. Given the different soil conditions at the respective mining sites, including their heavy-metal contamination, revegetation strategies based on rhizobia adapting to local conditions are more likely to succeed over the long term compared to strategies based on introducing less-well-adapted strains.

Project description:Understanding the level of heavy metal contamination coupled with the assessment of environmental and human risks associated with mine waste dumpsites is an important step to initiating efficient measures for mine wasteland restoration, stabilization, and bioremediation. In the present study, concentration of the heavy metals; Copper (Cu), Cobalt (Co), Iron (Fe), Lead (Pb), Manganese (Mn), and Zinc (Zn) in soil from mine waste dumpsites around Kitwe (Sites: TD25 and TD26) and Mufulira (Site: TD10), Zambia, was assessed to determine the level of contamination, ecological risks, and progress made in reclamation. The mine waste dumpsites in the two towns are located in the vicinity of residential areas. Therefore, there is need to provide information for optimization of protocols for post-mining landscape in Zambia and elsewhere to limit soil, river, and groundwater contamination and to accelerate the restoration process . Mean values for soil pH, electrical conductivity, and organic matter varied between 5.9-8.4, 2534.8-538.6 μS/cm, and 0.90-2.75%, respectively. The mean concentrations of heavy metals of TD25, TD26, and TD10 decreased in order of Fe > Cu > Co > Mn > Pb > Zn across all sites. However, the order of overall degree of heavy metal contamination computed using control soil as a baseline in TD25, TD26, and TD10 was Cu > Co > Fe > Pb > Mn > Zn. The pollution load index was 0.355 at TD25, 0.329 at TD26, and 0.189 at TD10, indicating high soil pollution at TD25 and TD26. The Potential Ecological Risk Index for all heavy metals tested at TD25, TD26, and TD10 showed low ecological risk in the vicinity of the studied dumpsites. Furthermore, the present study also showed that the polluted soils around smelter sites and mine waste dumpsites are susceptible to dispersion by wind and water. Additionally, results from TD10 revealed that the initiated remediation of the tailings dam was somewhat successful. Finally, this study provided an updated status regarding the accumulation of heavy metals in mine waste dumpsites of Kitwe and Mufulira, Zambia and baseline information necessary to enhance post-mining landscape reclamation.
