Depicting the RNA Virome of Hematophagous Arthropods from Belgrade, Serbia.

ABSTRACT: Hematophagous arthropods are important vectors for zoonotic pathogens. To date, a huge number of viruses have been identified in these arthropods, with a considerable proportion of them being human pathogens. However, the viromes of hematophagous arthropods are still largely unresearched. In this study, a number of arthropods were collected from Belgrade, Serbia including mosquitoes, ticks and bedbugs. The viromes of these arthropods were identified and characterized using Illumina MiSeq sequencing. In total, 21 viruses belonging to 11 families were characterized, with 11 of them representing novel species. These results may contribute to our knowledge of RNA viruses in arthropods and the discovery of novel human pathogens.

Project description:This paper presents the results of a research study where ground penetrating radar (GPR) was successfully used to reveal the remains of the Württemberg-Stambol Gate in the subsurface of Republic Square, in Belgrade, Serbia. GPR investigations were carried out in the context of renovation works in the square, which involved rearranging traffic control, expanding the pedestrian zone, renewing the surface layer, and valorising existing archaeological structures. The presence of the gate remains was suggested by historical documents and information from previous restoration works. A pulsed radar unit was used for the survey, with antennas having 200- and 400-MHz central frequencies. Data were recorded over a grid and two three-dimensional models were built, one for each set of antennas. The grid was the same for both sets of antennas, therefore the two models could be compared. Several horizontal cross sections of the models were plotted, corresponding to different depths; these images were carefully examined and interpreted, paying particular attention to signatures that could originate from the sought archaeological structures. Reflections coming from the gate remains were identified in both models, in the same region of the survey area and at the same depth; the geometry, size, and layout of the gate columns, as well as of other construction elements belonging to the gate, were determined with very good accuracy. Based on the GPR findings, archaeological excavation works were carried out in the region where the foundation remains were estimated to be. The presence of the remains was confirmed, with various columns and side walls. This case study demonstrates and further corroborates the effectiveness and reliability of GPR for the non-invasive prospection of archaeological structures hidden in the heterogeneous subsurface of urban environments. In the opinion of the authors, GPR should be incorporated as a routine field procedure in construction and renovation projects involving historical cities.

Project description:Bloodsucking arthropods are a rich source of salivary molecules (sialogenins) which inhibit platelet aggregation, neutrophil function and angiogenesis. Here we review the literature on salivary disintegrins and their targets. Disintegrins were first discovered in snake venoms, and were instrumental in our understanding of integrin function and also for the development of anti-thrombotic drugs. In hematophagous animals, most disintegrins described so far have been discovered in the salivary gland of ticks and leeches. A limited number have also been found in hookworms and horseflies, and none identified in mosquitoes or sand flies. The vast majority of salivary disintegrins reported display a RGD motif and were described as platelet aggregation inhibitors, and few others as negative modulator of neutrophil or endothelial cell functions. This notably low number of reported disintegrins is certainly an underestimation of the actual complexity of this family of proteins in hematophagous secretions. Therefore an algorithm was created in order to identify the tripeptide motifs RGD, KGD, VGD, MLD, KTS, RTS, WGD, or RED (flanked by cysteines) in sialogenins deposited in GenBank database. The search included sequences from various blood-sucking animals such as ticks (e.g., Ixodes sp., Argas sp., Rhipicephalus sp., Amblyommasp.), tabanids (e.g., Tabanus sp.), bugs (e.g., Triatoma sp., Rhodnius prolixus), mosquitoes (e.g., Anopheles sp., Aedes sp., Culex sp.), sand flies (e.g., Lutzomyia sp., Phlebotomus sp.), leeches (e.g., Macrobdella sp., Placobdella sp.) and worms (e.g., Ancylostoma sp.). This approach allowed the identification of a remarkably high number of novel putative sialogenins with tripeptide motifs typical of disintegrins (>450 sequences) whose biological activity remains to be verified. This database is accessible online as a hyperlinked worksheet and displays biochemical, taxonomic, and gene ontology aspects for each putative disintegrin.
