Thermoreversible Polymer Gels in DMF Formed from Charge- and Crystallization-Induced Assembly.

ABSTRACT: Polymer organogels formed through dynamic interactions are interesting for various applications. The fabrication of polymer organogels in polar solvents through ionic interaction is rare, although such organogels in non-polar organic solvents have been well studied. Herein, polymer organogels in a polar solvent N,N-dimethyl formamide (DMF) were fabricated from a triblock copolymer, poly(4-vinyl pyridine)-block-poly(ethylene glycol)-block-poly(4-vinyl pyridine) (4VPm-EGn-4VPm), and a fluorinated surfactant, perfluorooctanoic acid (PFOA), and their microphase separation and properties were studied. Ordered microphase separation and the crystalline structures were revealed by small-angle X-ray scattering (SAXS) and wide-angle X-ray scattering (WAXS), respectively. All the 4VPm-EGn-4VPm/PFOA organogels are sensitive to temperature, and the ratio of PFOA to pyridine groups reversibly. The polymer organogels are also responsive to triethylamine and triethylammonium acetate.

Amylomaltase of Pyrobaculum aerophilum IM2 produces thermoreversible starch gels.

Project description:Amylomaltases are 4-alpha-glucanotransferases (EC 2.4.1.25) of glycoside hydrolase family 77 that transfer alpha-1,4-linked glucans to another acceptor, which can be the 4-OH group of an alpha-1,4-linked glucan or glucose. The amylomaltase-encoding gene (PAE1209) from the hyperthermophilic archaeon Pyrobaculum aerophilum IM2 was cloned and expressed in Escherichia coli, and the gene product (PyAMase) was characterized. PyAMase displays optimal activity at pH 6.7 and 95 degrees C and is the most thermostable amylomaltase described to date. The thermostability of PyAMase was reduced in the presence of 2 mM dithiothreitol, which agreed with the identification of two possible cysteine disulfide bridges in a three-dimensional model of PyAMase. The kinetics for the disproportionation of malto-oligosaccharides, inhibition by acarbose, and binding mode of the substrates in the active site were determined. Acting on gelatinized food-grade potato starch, PyAMase produced a thermoreversible starch product with gelatin-like properties. This thermoreversible gel has potential applications in the food industry. This is the first report on an archaeal amylomaltase.

Crystallization-induced mechanofluorescence for visualization of polymer crystallization.

Project description:The growth of lamellar crystals has been studied in particular for spherulites in polymeric materials. Even though such spherulitic structures and their growth are of crucial importance for the mechanical and optical properties of the resulting polymeric materials, several issues regarding the residual stress remain unresolved in the wider context of crystal growth. To gain further insight into micro-mechanical forces during the crystallization process of lamellar crystals in polymeric materials, herein, we introduce tetraarylsuccinonitrile (TASN), which generates relatively stable radicals with yellow fluorescence upon homolytic cleavage at the central C-C bond in response to mechanical stress, into crystalline polymers. The obtained crystalline polymers with TASN at the center of the polymer chain allow not only to visualize the stress arising from micro-mechanical forces during polymer crystallization via fluorescence microscopy but also to evaluate the micro-mechanical forces upon growing polymer lamellar crystals by electron paramagnetic resonance, which is able to detect the radicals generated during polymer crystallization.

Pharmacokinetics and Pharmacodynamics Evaluation of Tramadol in Thermoreversible Gels.

Project description:We evaluated pharmacokinetics (PK) and pharmacodynamics (PD) induced by new formulations of tramadol (TR) in thermoreversible gels. The poloxamer- (PL-) tramadol systems were prepared by direct dispersion of the drug in solutions with PL 407 and PL 188. The evaluated formulations were as follows: F1: TR 2% in aqueous solution and F2: PL 407 (20%) + PL 188 (10%) + TR 2%; F3: PL 407 (25%) + PL 188 (5%) + TR 2%; F4: PL 407 (20%) + TR 2%. New Zealand White rabbits were divided into four groups (n = 6) and treated by subcutaneous route with F1, F2, F3, or F4 (10 μg·kg-1). PK evaluation used TR and M1 plasma levels. PD evaluation was performed with the measurement of both pupils' diameters. F2 showed higher TR plasma concentration after 180 minutes and presented lower M1 concentrations at almost all evaluated periods. Areas under the curve (ASC0-480 and ASC0-∞ ) and clearance of F2 presented differences compared to F1. F2 presented significant correlation (Pearson correlation) between the enhancement of TR and M1 concentrations and the decrease of pupil size (miosis). Thus, F2 was effective in altering pharmacokinetics and pharmacodynamics effects of TR.

Anion-Induced Reversible Actuation of Squaramide-Crosslinked Polymer Gels.

Project description:Supramolecular anion binding to squaramide crosslinkers in poly(N,N-dimethylacrylamide) gel networks enhances swelling and allows reversible chemically driven actuation. The volume swelling ratio of the gels is shown to depend on both the type of anion and its concentration. 1H NMR and UV-vis titrations with the squaramide crosslinkers reveal a relationship between anion binding affinity and the concentration-dependent swelling behavior. Gel swelling is shown to be reversible, and by embedding a solid support into rod-shaped gels, soft actuators are fabricated that undergo forward and backward bending motion in response to changing anion concentration. The swelling and bending process, which is accompanied by intense green coloration of the gel, is achieved by using only low amounts of crosslinker. This macroscopic actuation achieved by anion binding to specific molecular entities in the polymer network will open new opportunities in the field of chemically responsive materials.

Spray coverage analysis of topical sprays formed by cold thermoreversible hydrogels.

Project description:ObjectiveSprayable hydrogel formulations are promising topical treatments for skin wounds due to their ability to reduce application pain, prolong drug release, and provide moisture to promote skin healing. These viscoelastic materials, however, present challenges in spray ability which can be overcome using a thermoreversible hydrogels sprayed as lower viscosity liquids at cooler temperatures. The purpose of this research was to evaluate the impact of thermoreversible hydrogel formulation and device characteristics on topical spray patterns and to develop metrics to accurately describe surface coverage.MethodsCold solutions of Pluronic F127 were prepared at 15, 17, and 20% (w/w) and tested to determine their rheological properties. Formulations were sprayed from hand-held atomizing pump dispersers under cold conditions and two distinct areas of their spray patterns analyzed: the concentrated core and the full spray pattern. Traditional analysis of spray patterns was conducted to determine major and minor axes, ovality, and total area. In addition, new scripts were developed to evaluate the concentrated core.ResultsThe full spray pattern analysis quantified the total area over which the spray would extend a flat surface, while the concentrated core analysis quantified the continuous region where a drug dose would be concentrated. The combination of formulation viscosity, sprayer nozzle, and spray distance produced spray patterns from highly concentrated to highly dispersed. These parameters can be controlled to generate desired hydrogel spray patterns for application on skin surfaces.ConclusionThe developed metrics provide a basis for topical spray analysis that can inform future product performance.

Thermoreversible crystallization-driven aggregation of diblock copolymer nanoparticles in mineral oil.

Project description:A poly(behenyl methacrylate)37 (PBeMA37) macromolecular chain transfer agent is utilized for the reversible addition-fragmentation chain transfer (RAFT) dispersion polymerization of benzyl methacrylate (BzMA) directly in mineral oil at 90 °C. Polymerization-induced self-assembly (PISA) occurs under these conditions, yielding a series of sterically-stabilized PBeMA37-PBzMA x diblock copolymer spheres of tunable diameter as confirmed by dynamic light scattering (DLS) and transmission electron microscopy (TEM) studies. Rheological studies indicate that a relatively transparent, free-flowing, concentrated dispersion of non-interacting 32 nm PBeMA37-PBzMA100 spheres at 50 °C forms a turbid, paste-like dispersion on cooling to 20 °C. Turbidimetry and differential scanning calorimetry (DSC) studies conducted on solutions of PBeMA37 homopolymer in mineral oil suggest that this switchable colloidal stability is linked to crystallization-induced phase separation exhibited by this stabilizer block. Indeed, variable-temperature small-angle X-ray scattering (SAXS) indicates that a loose mass fractal network of strongly interacting spheres is formed on cooling to 20 °C, which accounts for this thermoreversible sol-gel transition. Moreover, SAXS, DSC and wide-angle X-ray scattering (WAXS) analyses indicate that the behenyl (C22H45) side-chains first form crystalline domains comprising adjacent stabilizer chains within individual spherical nanoparticles, with subsequent crystallization between neighboring nanoparticles leading to the formation of the mass fractal aggregates.

Photoluminescent coordination polymer bulk glasses and laser-induced crystallization.

Project description:We synthesized luminescent coordination polymer glasses composed of d10 metal cyanides and triphenylphosphine through melt-quenching and mechanical milling protocols. Synchrotron X-ray total scattering measurements and solid-state NMR revealed their one-dimensional chain structures and high structural dynamics. Thermodynamic and photoluminescence properties were tunable by the combination of heterometallic ions (Ag+, Au+, and Cu+) in the structures. The glasses are moldable and thermally stable, and over centimeter-sized glass monoliths were fabricated by the hot-press technique. They showed high transparency over 80% from the visible to near-infrared region and strong green emission at room temperature. Furthermore, the glass-to-crystal transformation was demonstrated by laser irradiation through the photothermal effect of the glasses.

Molecular recognition and self-assembly special feature: Supramolecular polymer formed by reversible self-assembly of tetrakisporphyrin.

Project description:S-shaped tetrakisporphyrin 2 forms supramolecular polymeric assemblies via a complementary affinity of its bisporphyrin units in solution. The self-association constant determined by applying the isodesmic model is >10(6) L mol(-1), which suggests that a sizable polymer forms at millimolar concentrations at room temperature. The electron deficient aromatic guest (TNF) binds within the molecular clefts provided by the bisporphyrin units via a charge-transfer interaction. This guest complexation completely disrupts supramolecular polymeric assembly. The long, fibrous fragments of the polymeric assemblies were characterized by atomic-force microscopy imaging of a film cast on a mica surface. The polymeric assemblies have lengths of >1mum and show a coiled structure with a higher level of organization. The approach discussed in this report concerning the quick preparation of supramolecular polymeric assemblies driven by noncovalent forces sets the stage for the preparation of a previously undescribed class of macromolecular porphyrin architectures.

Counting primary loops in polymer gels.

Project description:Much of our fundamental knowledge related to polymer networks is built on an assumption of ideal end-linked network structure. Real networks invariably possess topological imperfections that negatively affect mechanical properties; modifications of classical network theories have been developed to account for these defects. Despite decades of effort, there are no known experimental protocols for precise quantification of even the simplest topological network imperfections: primary loops. Here we present a simple conceptual framework that enables primary loop quantification in polymeric materials. We apply this framework to measure the fraction of primary loop junctions in trifunctional PEG-based hydrogels. We anticipate that the concepts described here will open new avenues of theoretical and experimental research related to polymer network structure.

Colloid-in-liquid crystal gels formed via spinodal decomposition.

Project description:We report that colloid-in-liquid crystal (CLC) gels can be formed via a two-step process that involves spinodal decomposition of a dispersion of colloidal particles in an isotropic phase of mesogens followed by nucleation of nematic domains within the colloidal network defined by the spinodal process. This pathway contrasts to previously reported routes leading to the formation of CLC gels, which have involved entanglement of defects or exclusion of particles from growing nematic domains. The new route provides the basis of simple design rules that enable control of the microstructure and dynamic mechanical properties of the gels.
