Protocol to Generate and Characterize Potent and Selective WNT Mimetic Molecules.

ABSTRACT: WNT signaling is crucial for embryonic development, adult tissue homeostasis, and injury repair. The poor biophysical characteristics of WNTs and their lack of receptor selectivity have hindered their use as research tools or potential therapeutics. We have developed a platform to generate potent, soluble, selective WNT mimetics with drug-like properties for both research and therapeutic applications. To help researchers adapt and expand on this platform, we describe these protocols and key considerations in generating and studying WNT mimetics. For complete details on the use and execution of this protocol, please refer to Chen et al., 2020.
