Transcranial photoacoustic computed tomography based on a layered back-projection method.

ABSTRACT: A major challenge of transcranial human brain photoacoustic computed tomography (PACT) is correcting for the acoustic aberration induced by the skull. Here, we present a modified universal back-projection (UBP) method, termed layered UBP (L-UBP), that can de-aberrate the transcranial PA signals by accommodating the skull heterogeneity into conventional UBP. In L-UBP, the acoustic medium is divided into multiple layers: the acoustic coupling fluid layer between the skull and detectors, the skull layer, and the brain tissue layer, which are assigned different acoustic properties. The transmission coefficients and wave conversion are considered at the fluid-skull and skull-tissue interfaces. Simulations of transcranial PACT using L-UBP were conducted to validate the method. Ex vivo experiments with a newly developed three-dimensional PACT system with 1-MHz center frequency demonstrated that L-UBP can substantially improve the image quality compared to conventional UBP.

Dichroism-sensitive photoacoustic computed tomography.

Project description:Photoacoustic computed tomography (PACT), a fast-developing modality for deep tissue imaging, images the spatial distribution of optical absorption. PACT usually treats the absorption coefficient as a scalar. However, the absorption coefficients of many biological tissues exhibit an anisotropic property, known as dichroism or diattenuation, which depends on molecular conformation and structural alignment. Here we present a novel imaging method called dichroism-sensitive PACT (DS-PACT), which measures both the amplitude of tissue's dichroism and the orientation of the optic axis of uniaxial dichroic tissue. By modulating the polarization of linearly polarized light and measuring the alternating signals through lock-in detection, DS-PACT can boost dichroic signals from biological tissues. To validate the proposed approach, we experimentally demonstrated the performance of DS-PACT by imaging plastic polarizers and ex vivo bovine tendons deep inside scattering media. We successfully detected the orientation of the optic axis of uniaxial dichroic materials, even at a depth of 4.5 transport mean free paths. We anticipate that the proposed method will extend the capability of PACT to imaging tissue absorption anisotropy.

Photoacoustic computed tomography of human extremities.

Project description:We present a method of imaging angiographic structures in human extremities, including hands, arms, legs, and feet, using a newly developed photoacoustic computed tomography (PACT) system. The system features deep penetration (1.8 cm in muscular tissues) with high spatial and temporal resolutions. A volumetric image is acquired within 5 to 15 s while each cross sectional image is acquired within 100???s. Therefore, we see no blurring from motion in the imaging plane. Longitudinal and latitudinal cross-sectional images of a healthy volunteer clearly show the vascular network of each appendage and highlight the system's ability to image major and minor vasculatures, without the use of an external contrast or ionizing radiation. We also track heartbeat-induced arterial movement at a two-dimensional frame rate of 10 Hz. This work substantiates the idea that PACT could be used as a noninvasive method for imaging human vasculatures.

Adaptive dual-speed ultrasound and photoacoustic computed tomography.

Project description:Full-ring dual-modal ultrasound and photoacoustic computed tomography has unique advantages of nearly isotropic spatial resolution, complementary contrast, deep penetration, and full-view detection. However, the imaging quality may be deteriorated by the inaccurate sound speed estimation. Automatic determining and compensation for sound speed has been a long-standing problem in image reconstruction. Here, we present new adaptive dual-speed ultrasound and photoacoustic computed tomography (ADS-USPACT) to address this challenge. The system features full-view coverage (360°), high-speed dual-modal imaging (10-Hz), automated dual sound speed correction, and synergistic high imaging quality. To correct the sound speed, we develop a two-compartment method that can automatically segment the sample boundary and search for the optimal sound speed based on the rich ultrasonic pulse-echo signals. The method does not require the operator's intervention. We validate this technique in numerical simulation, phantom study, and in vivo experiments. The ADS-USPACT represents significant progress in dual-modal imaging.

Single-breath-hold photoacoustic computed tomography of the breast.

Project description:We have developed a single-breath-hold photoacoustic computed tomography (SBH-PACT) system to reveal detailed angiographic structures in human breasts. SBH-PACT features a deep penetration depth (4 cm in vivo) with high spatial and temporal resolutions (255 µm in-plane resolution and a 10 Hz 2D frame rate). By scanning the entire breast within a single breath hold (~15 s), a volumetric image can be acquired and subsequently reconstructed utilizing 3D back-projection with negligible breathing-induced motion artifacts. SBH-PACT clearly reveals tumors by observing higher blood vessel densities associated with tumors at high spatial resolution, showing early promise for high sensitivity in radiographically dense breasts. In addition to blood vessel imaging, the high imaging speed enables dynamic studies, such as photoacoustic elastography, which identifies tumors by showing less compliance. We imaged breast cancer patients with breast sizes ranging from B cup to DD cup, and skin pigmentations ranging from light to dark. SBH-PACT identified all the tumors without resorting to ionizing radiation or exogenous contrast, posing no health risks.

Retrospective respiration-gated whole-body photoacoustic computed tomography of mice.

Project description:Photoacoustic tomography (PAT) is an emerging technique that has a great potential for preclinical whole-body imaging. To date, most whole-body PAT systems require multiple laser shots to generate one cross-sectional image, yielding a frame rate of <1??Hz. Because a mouse breathes at up to 3 Hz, without proper gating mechanisms, acquired images are susceptible to motion artifacts. Here, we introduce, for the first time to our knowledge, retrospective respiratory gating for whole-body photoacoustic computed tomography. This new method involves simultaneous capturing of the animal's respiratory waveform during photoacoustic data acquisition. The recorded photoacoustic signals are sorted and clustered according to the respiratory phase, and an image of the animal at each respiratory phase is reconstructed subsequently from the corresponding cluster. The new method was tested in a ring-shaped confocal photoacoustic computed tomography system with a hardware-limited frame rate of 0.625 Hz. After respiratory gating, we observed sharper vascular and anatomical images at different positions of the animal body. The entire breathing cycle can also be visualized at 20??frames/cycle.

Dry coupling for whole-body small-animal photoacoustic computed tomography.

Project description:We have enhanced photoacoustic computed tomography with dry acoustic coupling that eliminates water immersion anxiety and wrinkling of the animal and facilitates incorporating complementary modalities and procedures. The dry acoustic coupler is made of a tubular elastic membrane enclosed by a closed transparent water tank. The tubular membrane ensures water-free contact with the animal, and the closed water tank allows pressurization for animal stabilization. The dry coupler was tested using a whole-body small-animal ring-shaped photoacoustic computed tomography system. Dry coupling was found to provide image quality comparable to that of conventional water coupling.

Adaptive polarization photoacoustic computed tomography for biological anisotropic tissue imaging.

Project description:Most photoacoustic computed tomography (PACT) systems usually ignore the anisotropy of the tissue absorption coefficient, which will lead to the lack of information in reconstructed images. In this work, the effect is addressed of the possible optical absorption anisotropy of tissue on PACT images. The functional relationship is derived between the photoacoustic response and the polarization angle of the excitation light. An adaptive polarized light photoacoustic imaging (AP-PACT) approach is proposed and shown to make up for the lack of imaging information and achieve optimal image contrast when imaging samples with anisotropic optical absorption, by utilizing the standard deviation of photoacoustic response as the feedback signal in an adaptive data acquisition process. The method is implemented both on phantom and in vitro experiments, which show that AP-PACT can recover anisotropic absorption-related information from reconstructed images and thus significantly improve their quality.

Massively parallel functional photoacoustic computed tomography of the human brain.

Project description:Blood-oxygen-level-dependent (BOLD) functional magnetic resonance imaging of the human brain requires bulky equipment for the generation of magnetic fields. Photoacoustic computed tomography obviates the need for magnetic fields by using light and sound to measure deoxyhaemoglobin and oxyhaemoglobin concentrations to then quantify oxygen saturation and blood volumes. Yet, the available imaging speeds, fields of view (FOV), sensitivities and penetration depths have been insufficient for functional imaging of the human brain. Here, we show that massively parallel ultrasonic transducers arranged hemispherically around the human head can produce tomographic images of the brain with a 10-cm-diameter FOV and spatial and temporal resolutions of 350 µm and 2 s, respectively. In patients who had a hemicraniectomy, a comparison of functional photoacoustic computed tomography and 7 T BOLD functional magnetic resonance imaging showed a strong spatial correspondence in the same FOV and a high temporal correlation between BOLD signals and photoacoustic signals, with the latter enabling faster detection of functional activation. Our findings establish the use of photoacoustic computed tomography for human brain imaging.

Sparsity-based method for ring artifact elimination in computed tomography.

Project description:Ring artifact elimination is one of the popular problems in computed tomography (CT). It appears in the reconstructed image in the form of bright or dark patterns of concentric circles. In this paper, based on the compressed sensing theory, we propose a method for eliminating the ring artifact during the image reconstruction. The proposed method is based on representing the projection data by a sum of two components. The first component contains ideal correct values, while the latter contains imperfect error values causing the ring artifact. We propose to minimize some sparsity-induced norms corresponding to the imperfect error components to effectively eliminate the ring artifact. In particular, we investigate the effect of using different sparse models, i.e. different sparsity-induced norms, on the accuracy of the ring artifact correction. The proposed cost function is optimized using an iterative algorithm derived from the alternative direction method of multipliers. Moreover, we propose improved versions of the proposed algorithms by incorporating a smoothing penalty function into the cost function. We also introduce angular constrained forms of the proposed algorithms by considering a special case as follows. The imperfect error values are constant over all the projection angles, as in the case where the source of ring artifact is the non-uniform sensitivity of the detector. Real data and simulation studies were performed to evaluate the proposed algorithms. Results demonstrate that the proposed algorithms with incorporating smoothing penalty and their angular constrained forms are effective in ring artifact elimination.

ScatterNet for projection-based 4D cone-beam computed tomography intensity correction of lung cancer patients.

Project description:Background and purpose: In radiotherapy, dose calculations based on 4D cone beam CTs (4DCBCTs) require image intensity corrections. This retrospective study compared the dose calculation accuracy of a deep learning, projection-based scatter correction workflow (ScatterNet), to slower workflows: conventional 4D projection-based scatter correction (CBCTcor) and a deformable image registration (DIR)-based method (4DvCT). Materials and methods: For 26 lung cancer patients, planning CTs (pCTs), 4DCTs and CBCT projections were available. ScatterNet was trained with pairs of raw and corrected CBCT projections. Corrected projections from ScatterNet and the conventional workflow were reconstructed using MA-ROOSTER, yielding 4DCBCTSN and 4DCBCTcor. The 4DvCT was generated by 4DCT to 4DCBCT DIR, as part of the 4DCBCTcor workflow. Robust intensity modulated proton therapy treatment plans were created on free-breathing pCTs. 4DCBCTSN was compared to 4DCBCTcor and the 4DvCT in terms of image quality and dose calculation accuracy (dose-volume-histogram parameters and 3%/3mm gamma analysis). Results: 4DCBCTSN resulted in an average mean absolute error of 87HU and 102HU when compared to 4DCBCTcor and 4DvCT respectively. High agreement was observed in targets with median dose differences of 0.4Gy (4DCBCTSN-4DCBCTcor) and 0.3Gy (4DCBCTSN-4DvCT). The gamma analysis showed high average 3%/3mm pass rates of 96% for both 4DCBCTSN vs. 4DCBCTcor and 4DCBCTSN vs. 4DvCT. Conclusions: Accurate 4D dose calculations are feasible for lung cancer patients using ScatterNet for 4DCBCT correction. Average scatter correction times could be reduced from 10min (4DCBCTcor) to 3.9s, showing the clinical suitability of the proposed deep learning-based method.
