Hybrid 3D printed-paper microfluidics.

ABSTRACT: 3D printed and paper-based microfluidics are promising formats for applications that require portable miniaturized fluid handling such as point-of-care testing. These two formats deployed in isolation, however, have inherent limitations that hamper their capabilities and versatility. Here, we present the convergence of 3D printed and paper formats into hybrid devices that overcome many of these limitations, while capitalizing on their respective strengths. Hybrid channels were fabricated with no specialized equipment except a commercial 3D printer. Finger-operated reservoirs and valves capable of fully-reversible dispensation and actuation were designed for intuitive operation without equipment or training. Components were then integrated into a versatile multicomponent device capable of dynamic fluid pathing. These results are an early demonstration of how 3D printed and paper microfluidics can be hybridized into versatile lab-on-chip devices.

Optical Approach to Resin Formulation for 3D Printed Microfluidics.

Project description:Microfluidics imposes different requirements on 3D printing compared to many applications because the critical features for microfluidics consist of internal microvoids. Resins for general 3D printing applications, however, are not necessarily formulated to meet the requirements of microfluidics and minimize the size of fabricated voids. In this paper we use an optical approach to guide custom formulation of resins to minimize the cross sectional size of fabricated flow channels as exemplars of such voids. We focus on stereolithgraphy (SL) 3D printing with Digital Light Processing (DLP) based on a micromirror array and use a commercially available 3D printer. We develop a mathematical model for the optical dose delivered through the thickness of a 3D printed part, including the effect of voids. We find that there is a fundamental trade-off between the homogeneity of the optical dose within individual layers and how far the critical dose penetrates into a flow channel during fabrication. We also experimentally investigate the practical limits of flow channel miniaturization given the optical properties of a resin and find that the minimum flow channel height is ~3.5-5.5ha where ha is the optical penetration depth of the resin, and that the minimum width is 4 pixels in the build plane. We also show that the ratio of the build layer thickness to ha should be in the range 0.3-1.0 to obtain the minimum flow channel height for a given resin. The minimum flow channel size that we demonstrate for a custom resin is 60 ?m × 108 ?m for a 10 ?m build layer thickness. This work lays the foundation for 3D printing of <100 ?m microfluidic features.

Automated 4-Sample Protein Immunoassays using 3D-Printed Microfluidics.

Project description:Low cost, miniaturized assay platforms that work with small sample volumes, high sensitivity and rapid detection will have high value in future biomolecular diagnostics. Herein we report an automated, 3D printed electrochemiluminescent (ECL) immunoarray integrated with a nanostructured pyrolytic graphite sheet (PGS) microwell chip configured to detect 2 proteins simultaneously from complex liquid samples with high sensitivity and selectivity. Assays are done in 18 min at cost of < $1.00 using 1-2 microliters of sample. 3D printed microfluidic array design integrates reagent and sample chambers with rapid ECL detection. A commercial programmable syringe pump used with a preset program allows pump to pause and resume reagent delivery as required for completion of the sandwich immunoassays. Nanostructured surfaces feature antibody-decorated single wall carbon nanotube forests on PGS chip microwells, and sensitivity is amplified via massively labeled RuBPY-silica nanoparticles for detection. Prostate specific antigen (PSA) and prostate specific membrane antigen (PSMA) were measured simultaneously from human serum on the immunoarray with detection limits 150 fg mL-1 for PSA and 230 fg mL-1 for PSMA, with dynamic ranges up to 5 ng mL-1. Validation of the immunoarray by measuring these proteins in human serum showed good correlation with single protein ELISA. These 3D printed platforms can be easily adapted to multiple applications and configurable CAD files for the immunoarray can be downloaded from our lab's website.

3D printed self-supporting elastomeric structures for multifunctional microfluidics.

Project description:Microfluidic devices fabricated via soft lithography have demonstrated compelling applications such as lab-on-a-chip diagnostics, DNA microarrays, and cell-based assays. These technologies could be further developed by directly integrating microfluidics with electronic sensors and curvilinear substrates as well as improved automation for higher throughput. Current additive manufacturing methods, such as stereolithography and multi-jet printing, tend to contaminate substrates with uncured resins or supporting materials during printing. Here, we present a printing methodology based on precisely extruding viscoelastic inks into self-supporting microchannels and chambers without requiring sacrificial materials. We demonstrate that, in the submillimeter regime, the yield strength of the as-extruded silicone ink is sufficient to prevent creep within a certain angular range. Printing toolpaths are specifically designed to realize leakage-free connections between channels and chambers, T-shaped intersections, and overlapping channels. The self-supporting microfluidic structures enable the automatable fabrication of multifunctional devices, including multimaterial mixers, microfluidic-integrated sensors, automation components, and 3D microfluidics.

Engineering of Removing Sacrificial Materials in 3D-Printed Microfluidics.

Project description:Three-dimensional (3D) printing will create a revolution in the field of microfluidics due to fabricating truly three-dimensional channels in a single step. During the 3D-printing process, sacrificial materials are usually needed to fulfill channels inside and support the printed chip outside. Removing sacrificial materials after printing is obviously crucial for applying these 3D printed chips to microfluidics. However, there are few standard methods to address this issue. In this paper, engineering techniques of removing outer and inner sacrificial materials were studied. Meanwhile, quantification methods of removal efficiency for outer and inner sacrificial materials were proposed, respectively. For outer sacrificial materials, a hot bath in vegetable oil can remove 89.9% ± 0.1% of sacrificial materials, which is better than mechanics removal, hot oven heating, and an ethanol bath. For inner sacrificial materials, injecting 70 °C vegetable oil for 720 min is an optimized approach because of the uniformly high transmittance (93.8% ± 6.8%) and no obvious deformation. For the industrialization of microfluidics, the cost-effective removing time is around 10 min, which considers the balance between time cost and chip transmittance. The optimized approach and quantification methods presented in this paper show general engineering sacrificial materials removal techniques, which promote removing sacrificial materials from 3D-printed microfluidics chips and take 3D printing a step further in microfluidic applications.

Monolithic multilayer microfluidics via sacrificial molding of 3D-printed isomalt.

Project description:Here we demonstrate a method for creating multilayer or 3D microfluidics by casting a curable resin around a water-soluble, freestanding sacrificial mold. We use a purpose-built 3D printer to pattern self-supporting filaments of the sugar alcohol isomalt, which we then back-fill with a transparent epoxy resin. Dissolving the sacrificial mold leaves a network of cylindrical channels as well as input and output ports. We use this technique to fabricate a combinatorial mixer capable of producing 8 combinations of two fluids in ratios ranging from 1?:?100 to 100?:?1. This approach allows rapid iteration on microfluidic chip design and enables the use of geometry and materials not accessible using conventional soft lithography. The ability to precisely pattern round channels in all three dimensions in hard and soft media may prove enabling for many organ-on-chip systems.

Simple and Versatile 3D Printed Microfluidics Using Fused Filament Fabrication.

Project description:The uptake of microfluidics by the wider scientific community has been limited by the fabrication barrier created by the skills and equipment required for the production of traditional microfluidic devices. Here we present simple 3D printed microfluidic devices using an inexpensive and readily accessible printer with commercially available printer materials. We demonstrate that previously reported limitations of transparency and fidelity have been overcome, whilst devices capable of operating at pressures in excess of 2000 kPa illustrate that leakage issues have also been resolved. The utility of the 3D printed microfluidic devices is illustrated by encapsulating dental pulp stem cells within alginate droplets; cell viability assays show the vast majority of cells remain live, and device transparency is sufficient for single cell imaging. The accessibility of these devices is further enhanced through fabrication of integrated ports and by the introduction of a Lego®-like modular system facilitating rapid prototyping whilst offering the potential for novices to build microfluidic systems from a database of microfluidic components.

PDMS-Parylene Hybrid, Flexible Microfluidics for Real-Time Modulation of 3D Helical Inertial Microfluidics.

Project description:Inertial microfluidics has drawn much attention for its applications for circulating tumor cell separations from blood. The fluid flows and the inertial particle focusing in inertial microfluidic systems are highly dependent on the channel geometry and structure. Flexible microfluidic systems can have adjustable 3D channel geometries by curving planar 2D channels into 3D structures, which will enable tunable inertial separation. We present a poly(dimethylsiloxane) (PDMS)-parylene hybrid thin-film microfluidic system that can provide high flexibility for 3D channel shaping while maintaining the channel cross-sectional shape. The PDMS-parylene hybrid microfluidic channels were fabricated by a molding and bonding technique using initiated chemical vapor deposition (iCVD) bonding. We constructed 3D helical inertial microfluidic channels by coiling a straight 2D channel and studied the inertial focusing while varying radius of curvature and Reynolds number. This thin film structure allows for high channel curvature and high Dean numbers which leads to faster inertial particle focusing and shorter channel lengths than 2D spiral channels. Most importantly, the focusing positions of particles and cells in the microchannel can be tuned in real time by simply modulating the channel curvature. The simple mechanical modulation of these 3D structure microfluidic systems is expected to provide unique advantages of convenient tuning of cell separation thresholds with a single device.

3D-printed microfluidics integrated with optical nanostructured porous aptasensors for protein detection.

Project description:Microfluidic integration of biosensors enables improved biosensing performance and sophisticated lab-on-a-chip platform design for numerous applications. While soft lithography and polydimethylsiloxane (PDMS)-based microfluidics are still considered the gold standard, 3D-printing has emerged as a promising fabrication alternative for microfluidic systems. Herein, a 3D-printed polyacrylate-based microfluidic platform is integrated for the first time with a label-free porous silicon (PSi)-based optical aptasensor via a facile bonding method. The latter utilizes a UV-curable adhesive as an intermediate layer, while preserving the delicate nanostructure of the porous regions within the microchannels. As a proof-of-concept, a generic model aptasensor for label-free detection of his-tagged proteins is constructed, characterized, and compared to non-microfluidic and PDMS-based microfluidic setups. Detection of the target protein is carried out by real-time monitoring reflectivity changes of the PSi, induced by the target binding to the immobilized aptamers within the porous nanostructure. The microfluidic integrated aptasensor has been successfully used for detection of a model target protein, in the range 0.25 to 18 μM, with a good selectivity and an improved limit of detection, when compared to a non-microfluidic biosensing platform (0.04 μM vs. 2.7 μM, respectively). Furthermore, a superior performance of the 3D-printed microfluidic aptasensor is obtained, compared to a conventional PDMS-based microfluidic platform with similar dimensions.

Implantable photonic neural probes with 3D-printed microfluidics and applications to uncaging.

Project description:Advances in chip-scale photonic-electronic integration are enabling a new generation of foundry-manufacturable implantable silicon neural probes incorporating nanophotonic waveguides and microelectrodes for optogenetic stimulation and electrophysiological recording in neuroscience research. Further extending neural probe functionalities with integrated microfluidics is a direct approach to achieve neurochemical injection and sampling capabilities. In this work, we use two-photon polymerization 3D printing to integrate microfluidic channels onto photonic neural probes, which include silicon nitride nanophotonic waveguides and grating emitters. The customizability of 3D printing enables a unique geometry of microfluidics that conforms to the shape of each neural probe, enabling integration of microfluidics with a variety of existing neural probes while avoiding the complexities of monolithic microfluidics integration. We demonstrate the photonic and fluidic functionalities of the neural probes via fluorescein injection in agarose gel and photoloysis of caged fluorescein in solution and in fixed brain tissue.

3D Printed Skin-Interfaced UV-Visible Hybrid Photodetectors.

Project description:Photodetectors that are intimately interfaced with human skin and measure real-time optical irradiance are appealing in the medical profiling of photosensitive diseases. Developing compliant devices for this purpose requires the fabrication of photodetectors with ultraviolet (UV)-enhanced broadband photoresponse and high mechanical flexibility, to ensure precise irradiance measurements across the spectral band critical to dermatological health when directly applied onto curved skin surfaces. Here, a fully 3D printed flexible UV-visible photodetector array is reported that incorporates a hybrid organic-inorganic material system and is integrated with a custom-built portable console to continuously monitor broadband irradiance in-situ. The active materials are formulated by doping polymeric photoactive materials with zinc oxide nanoparticles in order to improve the UV photoresponse and trigger a photomultiplication (PM) effect. The ability of a stand-alone skin-interfaced light intensity monitoring system to detect natural irradiance within the wavelength range of 310-650 nm for nearly 24 h is demonstrated.
