The complete mitochondrial genome of Plagiodera versicolora (Laicharting)(Coleoptera: Chrysomelidae).

ABSTRACT: Plagiodera versicolora (Laicharting) is a leaf-eating pest widely distributed in the world. In this study, the first complete mitochondrial genome of P. Versicolora (Laicharting) was assembled and analyzed. The complete mitochondrial genome of P. Versicolora (Laicharting) is 16,857 bp with 22.39% GC containing, 13 protein-coding genes, 22 transfer RNA (tRNA), 2 ribosomal RNA (rRNA), as well as an AT-rich region. Phylogenomic analysis indicated that P. Versicolora (Laicharting) is sister to Chrysomela populiThis study provides useful information for the identification of this species and the study of genetic evolution with other species of Chrysomelidae.

Bacteria-Mediated RNA Interference for Management of Plagiodera versicolora (Coleoptera: Chrysomelidae).

Project description:RNA interference (RNAi) has emerged as a novel and feasible strategy for pest management. Methods for cost-effective production and stable delivery of double-stranded RNA (dsRNA) to the target insects are crucial for the wide application of RNAi for pest control. In this study, we tested the expression of dsRNA in RNaseIII-deficient Escherichia coli HT115 which was then fed to Plagiodera versicolora larvae, an insect pest of Salicaceae plants worldwide. By targeting six potential genes, including actin (ACT), signal recognition particle protein 54k (SRP54), heat shock protein 70 (HSC70), shibire (SHI), cactus (CACT), and soluble N-ethylmaleimide-sensitive fusion attachment proteins (SNAP), we found that feeding bacteria-expressed dsRNA successfully triggered the silencing of the five target genes tested and the suppression of ACT and SRP54 genes caused significant mortality. Our results suggest that the oral delivery of bacteria-expressed dsRNA is a potential alternative for the control of P. versicolora, and that ACT and SRP54 genes are the potent targets.

The complete mitochondrial genome of Psylliodes balyi Jacoby (Coleoptera: Chrysomelidae).

Project description:The complete mitogenome of Psylliodes balyi Jacoby (GenBank accession number MT644112) is 14,561 bp in length, and contains 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), 2 ribosomal RNA genes, and a putative control region. The gene content and orientation of P. balyi were identical to other beetle mitogenomes. ATT, ATA, and ATG were initiation codons and TAA, TAG, and T were termination codons. All the 22 tRNAs have the typical cloverleaf secondary structure, except for trnS1 which lacked the dihydrouracil (DHU) arm. The phylogenetic relationship based on the neighbor-joining method showed that P. balyi is closely related to Agasicles hygrophila, which agrees with the conventional classification.

The complete mitochondrial genome of Downesia tarsata (Coleoptera: Chrysomelidae: Cassidinae).

Project description:Downesia species are leaf-mining beetles mainly feed on Poaceae plants in the tropical and subtropical areas in Asia. In this study, we firstly sequenced and reported the complete mitochondrial genome for the genus. The complete mitogenome of Downesia tarsata is 18,557 bp in length, including 13 protein-coding genes (PCG), 22 transfer RNA (tRNA), two ribosomal RNA (rRNA), and one AT-rich region. Phylogenomic analysis indicated that D. tarsata is closely related to Agonita chinensis, and the two species belong to the same tribe of Gonophorini. The complete mitochondrial genome of D. tarsata could help clarify the phylogenetic relationship among Cassidinae species.

Project description:In this study, the first complete mitochondrial genome of Chrysolina aeruginosa Fald was assembled and analyzed. The total length of this mitochondrial genome is 16,335 base pairs. It consists of 13 protein-coding genes, 22 transfer RNAs, two ribosomal RNAs, and an AT-rich region. Phylogenomic analysis indicated that C. aeruginosa Fald is sister to Chrysodinopsis sp. This study provides new molecular data for the further taxonomic and phylogenetic studies of the Chrysomelidae of Coleoptera.

The complete mitochondrial genome of Monolepta hieroglyphica (Motschulsky) (Coleoptera: Chrysomelidae).

Project description:Monolepta hieroglyphica (Motschulsky) (Coleoptera: Chrysomelidae) is an important agricultural insect pest. In this study, the complete mitochondrial genome of M. hieroglyphica (GenBank accession number MW732714) was sequenced using Illumina HiSeq X Ten. The mitogenome was 16,213 bp long and comprised 13 protein-coding genes (PCGs), two ribosomal RNA genes (rRNAs), 22 transfer RNA genes (tRNAs) and a putative control region (CR). The nucleotide composition of the M. hieroglyphica mitochondrial genome was significantly biased (A, G, C and T accounted for 41.04%, 8.01%, 11.76% and 39.18%, respectively) with 80.23% A + T content. Two rRNAs were located between tRNA-Leu and the CR, separated by tRNA-Val. The CR, located between 12 s rRNA and tRNA-Ile, was 1,661 bp long. The length of the 22 tRNAs ranged from 61 to 71 bp. Phylogenetic analyses of 29 Chrysomelidae-Galerucinae species based on 13 mitochondrial protein-coding genes reconstructed using Bayesian 3.2.0 showed that the M. hieroglyphica mitogenome was clustered with the existing three different species of the Monolepta genus mitogenomes in a monophyletic manner. The M. hieroglyphica mitogenome provides an important data resource for further studies and contributes to our understanding of the phylogeny of this group.

Complete mitochondrial genome of a leaf beetle, Callispa bowringi (Coleoptera: Chrysomelidae).

Project description:The complete circular mitochondrial genome of Callispa bowringi was 17,060 bp in length, including two ribosomal RNA genes, 22 transfer RNAs, 13 protein-coding genes (PCGs) and one 2246-bp non-coding AT-rich region. All 22 tRNA genes displayed a typical clover-leaf structure except for tRNASer (AGN). All 13 PCGs initiated with ATN codons. Only three PCGs used the incomplete stop codons "TA" or "T", while ten PCGs terminated with typical stop codons "TAA" and "TGA". Phylogenetic analysis based on 13 PCGs of Chrysomelidae mitogenomes showed that C. bowringi was closely related to Agonita chinensis and Rhadinosa nigrocyanea.

Complete mitochondrial genome of coconut hispine beetle Brontispa longissima (Coleoptera: Chrysomelidae: Cassidinae).

Project description:In the present study, we determined complete mitogenome of Brontispa longissima, the first for the tribe Cryptonychini. This mitochondrial genome contains 15,696 bp, with an A+T content of 72.2% (GenBank accession no. MN052901). All of the 22 tRNA genes displayed a typical clover-leaf structure, with the exception of tRNASer (TCT). 12 PCGs were initiated by ATN codons, and ND1 started with TTG. Eight PCGs used the typical stop codon 'TAA' and 'TGA', while five PCGs terminated with incomplete stop codons "T". Phylogenetic analyses performed using the mitogenomes for the B. longissima and other seven species from subfamily Cassidinae.

Complete mitochondrial genome of a leaf-mining beetle, Podagricomela nigricollis (Coleoptera: Chrysomelidae).

Project description:Podagricomela nigricollis is a citrus pest that distributes in South China. Currently, there was no complete mitochondrial genome of Podagricomela species available in GenBank. Here, we reported the complete circular mitogenome of P. nigricollis. It had a total length of 16,756 bp, including 13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes, and one A + T-rich region. Among the 13 PCGs, only four (NAD5, NAD4, NAD4l, NAD1) located on the L-strand, whereas the other nine (NAD2, COX1, COX2, ATP8, ATP6, COX3, NAD3, NAD6, COB) located on the H-strand. Phylogenetic analysis using nucleotide sequences of the 13 PCGs indicated that P. nigricollis were clustered with six Galerucinae species, which was consistent with previous morphological classification.

Phylogenetic analysis based on 13 PCGs of Chrysomelidae mitogenomes showed that A. chinensis was closely related to Cassida viridis.

In this study, the complete mitochondrial genome of Colasposoma dauricum auripenne (Motschlsky, 1860) (Coleoptera: Chrysomelidae: Eumolpinae) has been sequenced. The mitogenome length is 15,614 bp (GenBank: MW528208), which includes 13 typical insect protein-coding genes (PCGs), 2 ribosomal RNA genes (rRNAs), and 22 transfer RNA genes (tRNAs), and it contains 78.6% A + T and 21.4% G + C. All PCGs start with ATD (ATT, ATA, ATG) codon and stop with termination codon TAA, TAG and single T. The phylogenetic tree showed that Colasposoma had been cluster together, and form a sister group relationship with Pseudocolaspis. The complete mitogenome of C. dauricum auripenne would help understand Chrysomelidae evolution.
