Isolation and Identification of Naegleria Species in Irrigation Channels for Recreational Use in Mexicali Valley, Mexico.

ABSTRACT: Members of the genus Naegleria are free-living amoebae that are widely distributed in water and soil environments. Moreover, Naegleria fowleri is a pathogenic amoeba species that causes a fatal disease in the central nervous system known as primary amoebic meningoencephalitis (PAM) in humans. Since most reported infections due to N. fowleri are reported in recreational waters worldwide, this study was aimed to describe the presence of these amoebic genus in Mexicali Valley irrigation channels of recreational use. A total of nine water samples were collected and processed by triplicate, in nine different sites of the Valley. After filtering and culturing the samples, plates were examined, and the observed amoebae were morphologically identified at the genus level. In addition, the pathogenicity of these amoebic isolates was checked, and molecular characterization was performed by PCR/sequencing. The results revealed the presence of Naegleria spp. in all the channels sampled. Finally, molecular identification confirmed the presence of five different species of Naegleria: N. fowleri, N. australiensis, N. gruberi, N. clarki and N. pagei. The presence of these protists, particularly N. fowleri, should be considered as a potential human health risk in the region.

Project description:The electrical response of plants to environmental stimuli can be measured and quantitatively related to the intensity of several stimulating sources, like temperature, solar radiation, soil water content, evapotranspiration rates, sap flow and dendrometric cycles. These relations can be used to assess the influence of different environmental situations on soil water availability to plants, defined as a steady state condition between leaf transpirative flow and soil water flow to plant roots. A restricted soil water flow due to soil dryness can trigger water stress in plants, if the atmospheric evaporative demand is high, causing partial stomata closure as a physiological response to avoid plant dehydration; water stressed and unstressed plants manifest a differential electrical response. Real time plant electrical response measurements can anticipate actions that prevent the plant reaching actual stress conditions, optimizing stomata gas exchange and photosynthetic rates. An electrophysiological sensor developed in this work, allows remote real-time recording information on plant electrical potential (EP) in the field, which is highly related to EP measurements obtained with a laboratory Keithley voltmeter sensor used in an highly controlled experimental setup. Our electrophysiological sensor is a wireless, autonomous devise, which transmits EP information via Internet to a data server. Using both types of sensors (EP electrodes with a Keithley voltmeter and the electrophysiological sensor), we measured in real time the electrical responses of Persea americana and Prunus domestica plants, to induced water deficits. The differential response for 2 scenarios: irrigation and water restriction is identified by a progressive change in slope on the daily maximal and minimal electric signal values in stressed plants, and a zero-slope for similar signals for well-watered plants. Results show a correspondence between measured signals obtained by our electrophysiological sensor and the EP electrodes connected to the Keithley voltmeter in each irrigation stage. Also, both sensors show a daily cyclical signal (circadian cycle).

Project description:BackgroundReusing wastewater for irrigation is a longstanding practice that enhances crop yields and improves climate resilience. Without treatment, however, wastewater contains harmful pathogens and chemicals. Reuse of untreated wastewater has been shown to be harmful to the health of nearby communities, but the routes of exposure are unknown and do not appear to be occupational. Some routes occur throughout entire communities, such as food contamination. Other routes may be spatially dependent, such as spread by domestic animals or through aerosolization.ObjectivesTo examine whether those wastewater exposure routes with a spatial dependency affect health, we estimated the risks of diarrheal disease in children under age 5 associated with living near wastewater canals, while adjusting for potential individual- and household-level confounders.MethodsWe conducted three surveys over 1 y in the Mezquital Valley, Mexico, to measure diarrhea in children. The distance between each participating household and a wastewater canal was measured using GPS coordinates. The association between proximity and diarrhea was estimated with a multilevel logistic regression model accounting for spatial autocorrelation.ResultsA total of 564 households completed one to three surveys, resulting in 1,856 survey observations of 646 children. Children living 100m from a canal had 45% lower odds of diarrhea than those living within 10m of a canal, and children living 1000m away had 70% lower odds of diarrhea [100m vs. 10m adjusted odds ratio (OR)=0.55, 95% credible interval (CI): 0.33, 0.91; 1000m vs. 10m adjusted OR=0.30, 95% CI: 0.11, 0.82].DiscussionThe estimated decline in diarrheal prevalence with household distance from a canal persisted after controlling for occupational exposure. Identifying the specific routes of exposure that drive this relationship will help identify which interventions, such as upstream treatment, can reduce health risks for entire communities where wastewater exposure occurs. https://doi.org/10.1289/EHP6443.
