Realization of pitch-rotational torque wrench in two-beam optical tweezers.

ABSTRACT: 3D Pitch (out-of-plane) rotational motion has been generated in spherical particles by maneuvering the laser spots of holographic optical tweezers. However, since the spherical particles, which are required to minimise drag are perfectly isotropic, a controllable torque cannot be applied with it. It remains free to spin about any axis even after moving the tweezers beams. It is here that we trap birefringent particles of about 3 μm diameter in two tweezers beams and then change the depth of one of the beam foci controllably to generate a pitch rotational torque-wrench and avoid the free spinning of the particle. We also detect the rotation with newly developed pitch motion detection technique and apply controlled torques on the particle.

Multiplexed vortex beam-based optical tweezers generated with spiral phase mask.

Project description:The design and implementation of a multiplexed spiral phase mask in an experimental optical tweezers setup are presented. This diffractive optical element allows the generation of multiple concentric vortex beams with independent topological charges and without amplitude modulation. The generalization of the phase mask for multiple concentric vortices is also shown. The design for a phase mask of two multiplexed vortices with different topological charges is developed. We experimentally show the transfer of angular momentum to the optically trapped microparticles by enabling nearly independent orbiting dynamics around the optical axis within each vortex. The angular velocity of the confined particles versus the optical power in the focal region is also discussed for different combinations of topological charges.

Accounting for polarization in the calibration of a donut beam axial optical tweezers.

Project description:Advances in light shaping techniques are leading to new tools for optical trapping and micromanipulation. For example, optical tweezers made from Laguerre-Gaussian or donut beams display an increased axial trap strength and can impart angular momentum to rotate a specimen. However, the application of donut beam optical tweezers to precision, biophysical measurements remains limited due to a lack of methods for calibrating such devices sufficiently. For instance, one notable complication, not present when trapping with a Gaussian beam, is that the polarization of the trap light can significantly affect the tweezers' strength as well as the location of the trap. In this article, we show how to precisely calibrate the axial trap strength as a function of height above the coverslip surface while accounting for focal shifts in the trap position arising from radiation pressure, mismatches in the index of refraction, and polarization induced intensity variations. This provides a foundation for implementing a donut beam optical tweezers capable of applying precise axial forces.

Magnetic tweezers measurement of single molecule torque.

Project description:Torsional stress in linear biopolymers such as DNA and chromatin has important consequences for nanoscale biological processes. We have developed a new method to directly measure torque on single molecules. Using a cylindrical magnet, we manipulate a novel probe consisting of a nanorod with a 0.1 microm ferromagnetic segment coupled to a magnetic bead. We achieve controlled introduction of turns into the molecule and precise measurement of torque and molecule extension as a function of the number of turns at low pulling force. We show torque measurement of single DNA molecules and demonstrate for the first time measurements of single chromatin fibers.

Teleportation-based realization of an optical quantum two-qubit entangling gate.

Project description:In recent years, there has been heightened interest in quantum teleportation, which allows for the transfer of unknown quantum states over arbitrary distances. Quantum teleportation not only serves as an essential ingredient in long-distance quantum communication, but also provides enabling technologies for practical quantum computation. Of particular interest is the scheme proposed by D. Gottesman and I. L. Chuang [(1999) Nature 402:390-393], showing that quantum gates can be implemented by teleporting qubits with the help of some special entangled states. Therefore, the construction of a quantum computer can be simply based on some multiparticle entangled states, Bell-state measurements, and single-qubit operations. The feasibility of this scheme relaxes experimental constraints on realizing universal quantum computation. Using two different methods, we demonstrate the smallest nontrivial module in such a scheme--a teleportation-based quantum entangling gate for two different photonic qubits. One uses a high-fidelity six-photon interferometer to realize controlled-NOT gates, and the other uses four-photon hyperentanglement to realize controlled-Phase gates. The results clearly demonstrate the working principles and the entangling capability of the gates. Our experiment represents an important step toward the realization of practical quantum computers and could lead to many further applications in linear optics quantum information processing.

Marangoni effect visualized in two-dimensions Optical tweezers for gas bubbles.

Project description:In the report we demonstrate how, using laser light, effectively trap gas bubbles and transport them through a liquid phase to a desired destination by shifting the laser beam position. The physics underlying the effect is complex but quite general as it comes from the limited to two-dimension, well-known, Marangoni effect. The experimental microscope-based system consists of a thin layer of liquid placed between two glass plates containing a dye dissolved in a solvent and a laser light beam that is strongly absorbed by the dye. This point-like heat source locally changes surface tension of nearby liquid-air interface. Because of temperature gradients a photo-triggered Marangoni flows are induced leading to self-amplification of the effect and formation of large-scale whirls. The interface is bending toward beam position allowing formation of a gas bubble upon suitable beam steering. Using various techniques (employing luminescent particles or liquid crystals), we visualize liquid flows propelled by the tangential to interface forces. This helped us to understand the physics of the phenomenon and analyze accompanying effects leading to gas bubble trapping. The manipulation of sessile droplets moving on the glass surface induced via controlled with laser light interface bending (i.e. "droplet catapult") is demonstrated as well.

Negative optical torque.

Project description:Light carries angular momentum, and as such it can exert torques on material objects. Applications of these opto-mechanical effects were limited initially due to their smallness in magnitude, but later becomes powerful and versatile after the invention of laser. Novel and practical approaches for harvesting light for particle rotation have since been demonstrated, where the structure is always subjected to a positive optical torque along a certain axis if the incident angular momentum has a positive projection on the same axis. We report here an interesting phenomenon of "negative optical torque", meaning that incoming photons carrying angular momentum rotate an object in the opposite sense. Surprisingly this can be realized quite straightforwardly in simple planar structures. Field retardation is a necessary condition and discrete rotational symmetry of material object plays an important role. The optimal conditions are explored and explained.

Optical Tweezers Exploring Neuroscience.

Project description:Over the past decade, optical tweezers (OT) have been increasingly used in neuroscience for studies of molecules and neuronal dynamics, as well as for the study of model organisms as a whole. Compared to other areas of biology, it has taken much longer for OT to become an established tool in neuroscience. This is, in part, due to the complexity of the brain and the inherent difficulties in trapping individual molecules or manipulating cells located deep within biological tissue. Recent advances in OT, as well as parallel developments in imaging and adaptive optics, have significantly extended the capabilities of OT. In this review, we describe how OT became an established tool in neuroscience and we elaborate on possible future directions for the field. Rather than covering all applications of OT to neurons or related proteins and molecules, we focus our discussions on studies that provide crucial information to neuroscience, such as neuron dynamics, growth, and communication, as these studies have revealed meaningful information and provide direction for the field into the future.

Quantifying the influences of radiation therapy on deformability of human red blood cells by dual-beam optical tweezers.

Project description:Radiation therapy is widely used as a treatment tool for malignancies. However, radiation-related complications are still unavoidable risks for off-target cells. Little is known about radiation therapy's possible effects on mechanical features of the off-target cells such as human red blood cells (RBCs). RBCs are nucleus-free circulating cells that can deform without losing functionality in healthy conditions. Thus, to evaluate in vitro effects of radiation therapy on the healthy plasma membrane of cells, RBCs were selected as a primary test model. RBCs were exposed to clinically prescribed radiotherapy doses of 2 Gy, 12 Gy and, 25 Gy, and each radiotherapy dose group was compared to a non-irradiated group. Cells were characterized by stretching using dual-beam optical tweezers and compared using the resulting deformability index. The group receiving the highest radiation dose was found statistically distinguishable from the control group (DI0Gy = 0.33 ± 0.08), and revealed the highest deformability index (DI25Gy = 0.38 ± 0.11, p = 0.0068), while no significant differences were found for 2 Gy (DI2Gy = 0.33 ± 0.08, p = 0.9) and 12 Gy (DI12Gy = 0.31 ± 0.09, p = 0.2) dose groups. Based on these findings, we conclude that radiotherapy exposure may alter the deformability of red blood cells depending on the dose amount, and measurement of deformability index by dual-beam optical tweezers can serve as a sensitive biomarker to probe responses of cells to the radiotherapy.

On chip shapeable optical tweezers.

Project description:Particles manipulation with optical forces is known as optical tweezing. While tweezing in free space with laser beams was established in the 1980s, integrating the optical tweezers on a chip is a challenging task. Recent experiments with plasmonic nanoantennas, microring resonators, and photonic crystal nanocavities have demonstrated optical trapping. However, the optical field of a tweezer made of a single microscopic resonator cannot be shaped. So far, this prevents from optically driven micromanipulations. Here we propose an alternative approach where the shape of the optical trap can be tuned by the wavelength in coupled nanobeam cavities. Using these shapeable tweezers, we present micromanipulation of polystyrene microspheres trapped on a silicon chip. These results show that coupled nanobeam cavities are versatile building blocks for optical near-field engineering. They open the way to much complex integrated tweezers using networks of coupled nanobeam cavities for particles or bio-objects manipulation at a larger scale.

The elasticity of single titin molecules using a two-bead optical tweezers assay.

Project description:Titin is responsible for the passive elasticity of the muscle sarcomere. The mechanical properties of skeletal and cardiac muscle titin were characterized in single molecules using a novel dual optical tweezers assay. Antibody pairs were attached to beads and used to select the whole molecule, I-band, A-band, a tandem-immunoglobulin (Ig) segment, and the PEVK region. A construct from the PEVK region expressing >25% of the full-length skeletal muscle isoform was chemically conjugated to beads and similarly characterized. By elucidating the elasticity of the different regions, we showed directly for the first time, to our knowledge, that two entropic components act in series in the skeletal muscle titin I-band (confirming previous speculations), one associated with tandem-immunoglobulin domains and the other with the PEVK region, with persistence lengths of 2.9 nm and 0.76 nm, respectively (150 mM ionic strength, 22 degrees C). Novel findings were: the persistence length of the PEVK component rose (0.4-2.7 nm) with an increase in ionic strength (15-300 mM) and fell (3.0-0.3 nm) with a temperature increase (10-60 degrees C); stress-relaxation in 10-12-nm steps was observed in the PEVK construct and hysteresis in the native PEVK region. The region may not be a pure random coil, as previously thought, but contains structured elements, possibly with hydrophobic interactions.
