NALCN, a sodium leak channel expressed mainly in the central nervous system, is responsible for the resting Na<sup>+</sup> permeability that controls neuronal excitability. Dysfunctions of the NALCN channelosome, NALCN with several auxiliary subunits, are associated with a variety of human diseases.
