Suitcase Lab: new, portable, and deployable equipment for rapid detection of specific harmful algae in Chilean coastal waters.

ABSTRACT: Phytoplankton blooms, including harmful algal blooms (HABs), have serious impacts on ecosystems, public health, and productivity activities. Rapid detection and monitoring of marine microalgae are important in predicting and managing HABs. We developed a toolkit, the Suitcase Lab, to detect harmful algae species in the field. We demonstrated the Suitcase Lab's capabilities for sampling, filtration, DNA extraction, and loop-mediated isothermal amplification (LAMP) detection in cultured Alexandrium catenella cells as well as Chilean coastal waters from four sites: Repollal, Isla García, Puerto Montt, and Metri. A LAMP assay using the Suitcase Lab in the field confirmed microscopic observations of A. catenella in samples from Repollal and Isla García. The Suitcase Lab allowed the rapid detection of A. catenella, within 2 h from the time of sampling, even at a single cell per milliliter concentrations, demonstrating its usefulness for quick and qualitative on-site diagnosis of target toxic algae species. This method is applicable not only to detecting harmful algae but also to other field studies that seek a rapid molecular diagnostic test.

Project description:Plastic is now a pervasive pollutant in all marine ecosystems. The microplastics and macroplastic debris were studied in three French Mediterranean coastal lagoons (Prevost, Biguglia and Diana lagoons), displaying different environmental characteristics. In addition, biofilm samples were analyzed over the seasons to quantify and identify microalgae communities colonizing macroplastics, and determine potentially harmful microorganisms. Results indicate low but highly variable concentrations of microplastics, in relation to the period and location of sampling. Micro-Raman spectroscopy analyses revealed that the majority of macroplastic debris corresponded to polyethylene (PE) and low-density polyethylene (LDPE), and to a far lesser extent to polypropylene (PP). The observations by Scanning Electron Microscopy of microalgae communities colonizing macroplastic debris demonstrated differences depending on the seasons, with higher amounts in spring and summer, but without any variation between lagoons and polymers. Among the Diatomophyceae, the most dominant genera were Amphora spp., Cocconeis spp., and Navicula spp.. Cyanobacteria and Dinophyceae such as Prorocentrum cordatum, a potentially toxic species, were also found sporadically. The use of Primer specific DNA amplification tools enabled us to detect potentially harmful microorganisms colonizing plastics, such as Alexandrium minutum or Vibrio spp. An additional in situ experiment performed over one year revealed an increase in the diversity of colonizing microalgae in relation to the duration of immersion for the three tested polymers PE, LDPE and polyethylene terephthalates (PET). Vibrio settled durably after two weeks of immersion, whatever the polymer. This study confirms that Mediterranean coastal lagoons are vulnerable to the presence of macroplastic debris that may passively host and transport various species, including some potentially harmful algal and bacterial microorganisms. Project description:Mount Xianbei is one of the largest shallow seamounts located in the middle of the South China Sea (SCS), which might play a role in shaping the biodiversity of surrounding continental coastal waters, particularly the diversity of phytoplankton species causing frequent harmful algal blooms (HABs) in northern SCS. However, the diversity, composition, and distribution of phytoplankton species in the seamount regions of Xianbei remain largely unexplored. In this study, samples around and outside the seamount regions were collected during a late summer cruise of 2021 to test whether seamounts play a role in HAB species propagation. In total, we identified 19 HAB species across all samples using the ASV-based DNA metabarcoding approach, 6 of which had not been reported previously in the SCS, suggesting a diverse HAB species in the SCS. Specifically, 16 HAB species were found in the seamount region of Xianbei, and 5 of them were also found in the coastal waters, indicating a close connection between seamount and coastal waters. This study was the first attempt to explore HAB species' spatial diversity and vertical distribution in the seamount region of Xianbei at single-nucleotide resolution, which provides a novel explanation for the coastal HAB occurrence in the northern SCS. IMPORTANCE There are a number of seamounts under the water of the South China Sea (SCS). The seamounts might play a role in shaping the biodiversity of surrounding continental coastal waters. However, there is no direct evidence revealing the relationship of the biodiversity of phytoplankton between seamounts and coastal waters in the SCS, especially those species having the potential to form harmful algal blooms (HABs). Some HAB species might proliferate in certain geographic locations, while others may be broadly distributed across oceanic provinces. In this study, we provided a detailed analysis of phytoplankton composition and molecular detection of HAB species from seamount to coastal waters in the SCS, which suggested a strong interaction in the HAB species between the two areas. This finding provides new insights into the diversity and distribution of HABs in seamounts and their role in shaping the composition and the occurrence of HABs in coastal water.
