<i>Sapajus flavius</i> has been listed on the International Union for Conservation of Nature (IUCN) Red List of Threatened Species. The complete mitochondrial genome sequence of <i>Sapajus flavius</i> is presented here first, sequenced by next-generation sequencing (NGS). The <i>Sapajus flavius</i> mitogenome is 16,543 bp long, contains 13 protein-coding genes (PCGs), 2 rRNA genes (12S rRNA and 16S rRNA), 22 transfer RNA (tRNA) genes, and one control region (D-loop). Project description:The phylogenetic relationships of primates have been extensively investigated, but key issues remain unresolved. Complete mitochondrial genome (mitogenome) data have many advantages in phylogenetic analyses, but such data are available for only 46 primate species. In this work, we determined the complete mitogenome sequence of the black-capped capuchin (Cebus apella). The genome was 16,538 bp in size and consisted of 13 protein-coding genes, 22 tRNAs, two rRNAs and a control region. The genome organization, nucleotide composition and codon usage did not differ significantly from those of other primates. The control region contained several distinct repeat motifs, including a putative termination-associated sequence (TAS) and several conserved sequence blocks (CSB-F, E, D, C, B and 1). Among the protein-coding genes, the COII gene had lower nonsynonymous and synonymous substitutions rates while the ATP8 and ND4 genes had higher rates. A phylogenetic analysis using Maximum likelihood and Bayesian methods and the complete mitogenome data for platyrrhine species confirmed the basal position of the Callicebinae and the sister relationship between Atelinae and Cebidae, as well as the sister relationship between Aotinae (Aotus) and Cebinae (Cebus/Saimiri) in Cebidae. These conclusions agreed with the most recent molecular phylogenetic investigations on primates. This work provides a framework for the use of complete mitogenome information in phylogenetic analyses of the Platyrrhini and primates in general.

Project description:In humans, facial symmetry has been linked to an individual's genetic quality, and facial symmetry has a small yet significant effect on ratings of facial attractiveness. The same evolutionary processes underlying these phenomena may also convey a selective advantage to symmetrical individuals of other primate species, yet to date, few studies have examined sensitivity to facial symmetry in nonhuman primates. Here we presented images of symmetrical and asymmetrical human and monkey faces to tufted capuchin monkeys (Sapajus apella) and hypothesized that capuchins would visually prefer symmetrical faces of opposite-sex conspecifics. Instead, we found that male capuchins preferentially attended to symmetrical male conspecific faces, whereas female capuchins did not appear to discriminate between symmetrical and asymmetrical faces. These results suggest that male capuchin monkeys may use facial symmetry to judge male quality in intramale competition. (PsycINFO Database Record

Project description:The present study evaluated the efficacy of an adapted version of the Mollon-Reffin test for the behavioral investigation of color vision in capuchin monkeys. Ten tufted capuchin monkeys (Sapajus spp., formerly referred to as Cebus apella) had their DNA analyzed and were characterized as the following: one trichromat female, seven deuteranope dichromats (six males and one female), and two protanope males, one of which was identified as an "ML protanope." For their behavioral characterization, all of the subjects were tested at three regions of the Commission International de l'Eclairage (CIE) 1976 u'v' diagram, with each test consisting of 20 chromatic variation vectors that were radially distributed around the chromaticity point set as the test background. The phenotypes inferred from the behavioral data were in complete agreement with those predicted from the genetic analysis, with the threshold distribution clearly differentiating between trichromats and dichromats and the estimated confusion lines characteristically converging for deuteranopes and the "classic" protanope. The discrimination pattern of the ML protanope was intermediate between protan and deutan, with confusion lines horizontally oriented and parallel to each other. The observed phenotypic differentiation confirmed the efficacy of the Mollon-Reffin test paradigm as a useful tool for evaluating color discrimination in nonhuman primates. Especially noteworthy was the demonstration of behavioral segregation between the "classic" and "ML" protanopes, suggesting identifiable behavioral consequences of even slight variations in the spectral sensitivity of M/L photopigments in dichromats.

Project description:The objective of this study was to describe the anatomic and histologic features of the Sapajus sp. eye, comparing similarities and differences of humans and other species of non-human primates for biomedical research purposes. Computed tomography (CT) of adnexa, eye and orbit live animal, as well as formolized pieces of the same structures of Sapajus sp. for anatomical and histological study were also performed. The anatomical description of the eye and adnexa was performed using the techniques of topographic dissection and exenteration. Histological fragments were fixated in buffered formalin 10%, processed by the routine paraffin inclusion technique, stained with hematoxylin-eosin and special stains. CT scan evaluation showed no differences between the live animal and the formolized head on identification of visual apparatus structures. Anatomic and histologic evaluation revealed rounded orbit, absence of the supraorbital foramen and frontal notch, little exposure of the sclera, with slight pigmentation of the exposed area and marked pigmentation at the sclerocorneal junction. Masson's Trichrome revealed the Meibomian glands, the corneal epithelium and Bowman's membrane; in the choroid, melanocytes and Bruch's membrane were observed; and in the retina, cones and rods as well as, optic nerve, the lamina cribrosa of the nerve fibers bundles. Toluidine blue highlighted the membranes: Bowman, Descemet and the endothelium; in the choroid: melanocytes; and in the retina: nuclear layers and retinal pigment epithelium. In view of the observed results Sapajus sp. is an important experimental model for research in the ophthalmology field, which has been shown due to the high similarity of its anatomical and histological structures with the human species.

Project description:Most experimental paradigms to study visual cognition in humans and non-human species are based on discrimination tasks involving the choice between two or more visual stimuli. To this end, different types of stimuli and procedures for stimuli presentation are used, which highlights the necessity to compare data obtained with different methods. The present study assessed whether, and to what extent, capuchin monkeys' ability to solve a size discrimination problem is influenced by the type of procedure used to present the problem. Capuchins' ability to generalise knowledge across different tasks was also evaluated. We trained eight adult tufted capuchin monkeys to select the larger of two stimuli of the same shape and different sizes by using pairs of food items (Experiment 1), computer images (Experiment 1) and objects (Experiment 2). Our results indicated that monkeys achieved the learning criterion faster with food stimuli compared to both images and objects. They also required consistently fewer trials with objects than with images. Moreover, female capuchins had higher levels of acquisition accuracy with food stimuli than with images. Finally, capuchins did not immediately transfer the solution of the problem acquired in one task condition to the other conditions. Overall, these findings suggest that--even in relatively simple visual discrimination problems where a single perceptual dimension (i.e., size) has to be judged--learning speed strongly depends on the mode of presentation.

Project description:We report the first observation of probe tool use by a wild adult female bearded capuchin (Sapajus libidniosus), at Serra da Capivara National Park (SCNP), Brazil. This individual used several stick tools and one grass stem to probe her nostrils, usually triggering a sneeze reaction, and also used stick tools to probe her teeth or gum. Both of these behaviours were accompanied by inspection and licking of the tool following use. We have termed these self-directed actions nasal probe and toothpick, and neither has been previously reported in wild capuchins. While stick tool use is common among foraging male capuchins at SCNP, the novel and at present idiosyncratic activities performed by the female monkey add to the known behavioural repertoire for this species.

Project description:Capuchin monkeys (Sapajus spp.) in captive settings frequently manipulate and throw objects. In the wild, they may push or drop stones and sticks toward targets during inter- or intraspecific threat displays. In addition, female capuchin monkeys exhibit a broad repertoire of behaviors during their proceptive period, including facial expressions, vocalizations, stereotyped body postures, and touch-and-run behavior. This study reports stone throwing as a newly-described communicative behavior during the proceptive display of females in a group of bearded capuchin monkeys (S. libidinosus) in Serra da Capivara National Park, Brazil. During a two-year study, three females from one group were seen throwing stones at males during their proceptive phase. After this period, three other females in the same group exhibited the same behavior. Although it may be possible that this pattern is the result of several independent innovations by each female, the apparent absence of this behavior in other groups leads us to suggest that we have documented the diffusion of a new behavioral trait or tradition within this capuchin social group.

Project description:Capuchin monkeys have rich social relationships and from very young ages they participate in complex interactions with members of their group. Lipsmacking behaviour, which involves at least two individuals in socially mediated interactions, may tell about processes that maintain, accentuate or attenuate emotional exchanges in monkeys. Lipsmacking is a facial expression associated with the establishment and maintenance of affiliative interactions, following under the 'emotional regulation' umbrella, which accounts for the ability to manage behavioural responses. We investigated behaviours related to the emitter and to the receiver (infant) of lipsmacking to answer the question of how lipsmacking occurs. In capuchin monkeys, lipsmacking has been previously understood solely as a face-to-face interaction. Our data show that emitters are engaged with infants, looking longer towards their face and seeking eye contact during the display. However, receivers spend most of the time looking away from the emitter and stay in no contact for nearly half of the time. From naturalistic observations of wild infant capuchin monkeys from Brazil we found that lipsmacking is not restricted to mutual gaze, meaning there are other mechanisms in place than previously known. Our results open paths to new insights about the evolution of socio-emotional displays in primates.

Project description:Abstract Objectives Foods that are geometrically and mechanically challenging to eat have been associated with specializations in feeding behavior and craniodental morphology across primates, and many of these foods are embedded, requiring a variety of positional behaviors during feeding. However, variation in positional behaviors in response to food properties is not well understood. Here, we examine differences in feeding postural behaviors across feeding events in relation to substrate and food geometric and material properties in a species of extractive foragers, bearded capuchins (Sapajus libidinosus). Methods and materials We coded over 1400 co‐occurring postural and feeding behaviors, their durations, and relative sizes of substrate and food from videos recorded at Fazenda Boa Vista in Gilbués, Piauí, Brazil. Food material properties were measured from foods collected at the time of the video recordings. Results Our results suggest that bearded capuchin feeding postures significantly differ across the feeding sequence, with substrate size, and between foods of high and low toughness and elastic modulus. Feeding postures were less variable for highly mechanically challenging foods. Food size also had a significant effect on postural behaviors. Large foods were more likely to be associated with suspended postures and small foods with sitting and squatting. Feeding postural behaviors were best explained by a combination of substrate and food variables. Conclusions Our results indicate that food geometric and mechanical properties have a significant influence on feeding postural behaviors in bearded capuchins. We posit that feeding postural behaviors reflect a combination of substrate variables and food properties, and large, mechanically challenging foods have a limiting effect on postural variation.

Project description:Humans can use hand tools smoothly and effectively in varying circumstances; in other words, skillfully. A few other species of primates crack encased foods using hammer tools and anvils. Are they skilled? Positioning the food on the anvil so that it does not fall off when struck is a component of skilled cracking. We discovered that bearded capuchin monkeys deliberately place palm nuts in a relatively stable position on the anvil before striking them. In the first experiment, we marked the meridians of palm nuts where they stopped when rolled on a flat surface ("Stop meridian"). We videotaped monkeys as they cracked these nuts on an anvil. In playback we coded the position of the Stop meridian prior to each strike. Monkeys typically knocked the nuts on the anvil a few times before releasing them in a pit. They positioned the nuts so that the Stop meridian was within 30 degrees of vertical with respect to gravity more often than expected, and the nuts rarely moved after the monkeys released them. In the second experiment, 14 blindfolded people (7 men) asked to position marked nuts on an anvil as if to crack them reliably placed them with the Stop meridian in the same position as the monkeys did. In the third experiment, two people judged that palm nuts are most bilaterally symmetric along a meridian on, or close to, the Stop meridian. Thus the monkeys reliably placed the more symmetrical side of the nuts against the side of the pit, and the nuts reliably remained stationary when released. Monkeys apparently used information gained from knocking the nut to achieve this position. Thus, monkeys place the nuts skillfully, strategically managing the fit between the variable nuts and pits in the anvil, and skilled placement depends upon information generated by manual action.
