Mitochondrial genome of a medicinal beetle Blaps rhynchopetera (Coleoptera, Tenebrionidae) and phylogenetic analysis.

ABSTRACT: Blaps rhynchopetera is a species of folk medicinal beetle that has been used for a long time in Southwest China. The complete mitogenome of the beetle (GenBank accession number MK854717) is 16,149?bp in size, including 13 protein-coding, 22 transfer RNAs, two ribosomal RNAs genes, and a noncoding D-loop region. The D-loop of 1255?bp length is located between rRNA-S and tRNA^Ile. The overall base composition of B. rhynchopetera is 41.58% for A, 10.31% for G, 31.77% for T, and 16.34% for C, with a high AT bias of 73.35%. The present data could contribute to detailed phylogeographic analysis of this valuable medicinal insect.

Project description:The complete mitochondrial genome of Blaps rynchopetera Fairmaire (GenBank accession number MN267802) from Dali city consists of a circular DNA molecule of 16,766?bp (with 72.01% A?+?T content), which is longer than that of the mitogenome of B. rynchopetera from Kunming. The mitogenome comprises 13 protein-coding, 22 tRNA, and 2 rDNA genes. The protein-coding genes have typical ATN (Met) initiation codons, and are terminated by typical TAN stop codons. A phylogenetic tree generated by the neighbour-joining method showed that B. rynchopetera is closely related to Platydema sp. PLA01, which has the potential for medicinal development.

Biochemical and histological alterations induced by nickel oxide nanoparticles in the ground beetle Blaps polychresta (Forskl, 1775) (Coleoptera: Tenebrionidae).

Project description:The present study evaluates the effect of nickel oxide nanoparticles on some biochemical parameters and midgut tissues in the ground beetle Blaps polychresta as an indicator organism for nanotoxicity. Serial doses of the NiO-NPs colloid (0.01, 0.02, 0.03, 0.04, 0.05, and 0.06 mg/g) were prepared for injecting into the adult beetles. Insect survival was reported daily for 30 days, and the sublethal dose of 0.02 mg/g NiO-NPs was selected for the tested parameters. After the treatment, nickel was detected in the midgut tissues by X-ray microanalysis. The treated group demonstrated a significant increase in aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities when compared to the untreated group. However, the treated group demonstrated a significant decrease in ascorbate peroxidase (APOX) activity when compared to the untreated group. Histological and ultrastructural changes in the midgut tissues of treated and untreated beetles were also observed. The current findings provide a precedent for describing the physiological and histological changes caused by NiO-NPs in the ground beetle B. polychresta.

Complete mitochondrial genome of Morphostenophanes yunnanus (Zhou, 2020) (Insecta: Coleoptera: Tenebrionidae) and phylogenetic analysis.

Project description:Morphostenophanes yunnanus (Zhou, 2020) is widely distributed from central to eastern Yunnan with distinct geographical variations in morphology. Beetles were collected in Manwan Town, and a mitochondrial genome sequence (GenBank accession number MZ298928) of this species was sequenced using the MGI-SEQ 2000 platform, assembled using NOVOPlasty v4.3.1, and characterized. The mitogenome was a circular DNA molecule of 15,690 bp with 64.710% AT content, which comprised 13 protein-coding genes, 22 tRNA genes, two rRNA genes, and one control region. The protein-coding genes showed the typical ATN (Met) and TTG (Met) start codons, except nad1 and cox1 (TTG as start codon), and were terminated by typical TAN stop codons. The maximum-likelihood polygenetic tree was generated using protein sequences of thirteen protein-coding regions of seventeen mitogenomes with mtREV + G + F + I with 1000 replicates under the Bayesian information criterion using MEGA 11, which showed that M. yunnanus was the most closely related to M. sinicus. This study provides essential genetic and molecular data for phylogenetic analyses of the genus Morphostenophanes.

The complete mitochondrial genomes of four lagriine species (Coleoptera, Tenebrionidae) and phylogenetic relationships within Tenebrionidae.

Project description:It is common to use whole mitochondrial genomes to analyze phylogenetic relationships among insects. In this study, seven mitogenomes of Tenebrionidae are newly sequenced and annotated. Among them, four species (Cerogira janthinipennis (Fairmaire, 1886), Luprops yunnanus (Fairmaire, 1887), Anaedus unidentasus Wang & Ren, 2007, and Spinolyprops cribricollis Schawaller, 2012) represent the subfamily Lagriinae. In this subfamily, the mitogenomes of the tribes Goniaderini (A. unidentasus) and Lupropini (L. yunnanus and S. cribricollis) were first reported; they were found to be 15,328-16,437 bp in length and encode 37 typical mitochondrial genes (13 PCGs, 2 rRNAs, 22 tRNAs, and a single noncoding control region). Most protein-coding genes in these mitogenomes have typical ATN start codons and TAR or an incomplete stop codon T-. In these four lagriine species, F, L2, I, and N are the most frequently used amino acids. In the 13 PCGs, the gene atp8 (Pi = 0.978) was the most diverse nucleotide, while cox1 was the most conserved gene with the lowest value (Pi = 0.211). The phylogenetic results suggest that Pimelinae, Lagriinae, Blaptinae, Stenochiinae, and Alleculinae are monophyletic, Diaperinae is paraphyletic, and Tenebrioninae appears polyphyletic. In Lagriinae, the tribe Lupropini appears paraphyletic because Spinolyprops is clustered with Anaedus in Goniaderini. These mitogenomic data provide important molecular data for the phylogeny of Tenebrionidae.

MN306531) using next-generation sequencing (NGS) method. The mitogenome is 15,842?bp in length, consisting of 13 protein-coding genes (PCGs), two ribosomal RNA genes (rRNAs), 22 transfer RNA genes (tRNAs), and one non-coding control region. The overall nucleotide composition was 41.6% A, 31.1% T, 16.9% C, and 10.4% G, with 72.7% AT, respectively. The gene arrangement is consistent with the typical insect mitochondrial genome. Phylogenetic analysis revealed that C. vittiger clustered into a clade with Chiagosnius sulcicollis with high bootstrap support.

Project description:BackgroundThe stag beetle Lucanus cervus (Coleoptera: Lucanidae) is widely distributed in Europe. Habitat loss and fragmentation has led to significant reductions in numbers of this species. In this study, we sequenced the complete mitochondrial genome of L. cervus and reconstructed phylogenetic relationships among Lucanidae using complete mitochondrial genome sequences.MethodsRaw data sequences were generated by the next generation sequencing using Illumina platform from genomic DNA of L. cervus. The mitochondrial genome was assembled by IDBA and annotated by MITOS. The aligned sequences of mitochondrial genes were partitioned using PartitionFinder 2. Phylogenetic relationships among 19 stag beetle species were constructed using Maximum Likelihood (ML) method implemented in IQ-TREE web server and Bayesian method implemented in PhyloBayes MPI 1.5a. Three scarab beetles were used as outgroups.ResultsThe complete mitochondrial genome of L. cervus is 20,109 bp in length, comprising 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNAs and a control region. The A + T content is 69.93% for the majority strand. All protein-coding genes start with the typical ATN initiation codons except for cox1, which uses AAT. Phylogenetic analyses based on ML and Bayesian methods shown consistent topologies among Lucanidae.

Project description:Larvae of click beetles are the most important soil-dwelling agricultural pests and are abundant throughout the world. Agrypnus sp. belongs to the subfamily Agrypninae of Elateridae. Here, we sequenced and annotated the complete mitochondrial genome (mitogenome) of Agrypnus sp., the new representative of the mitogenome of the subfamily. This mitogenome was 16,056 bp long and encoded 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and two ribosomal RNA unit genes (rRNAs). Gene order was conserved and identical to most other previously sequenced Elateridae. The nucleotide composition biases towards A and T, which together made up 70.8% of the entirety. Phylogenetic analysis based on 13 PCGs sequences showed that Agrypnus sp. got together with the same subfamily species Pyrophorus divergens, Pyrearinus termitilluminans, Ignelater luminosus, and Hapsodrilus ignifer with high support value.

Characterization of the Complete Mitochondrial Genome of a Flea Beetle Luperomorpha xanthodera (Coleoptera: Chrysomelidae: Galerucinae) and Phylogenetic Analysis.

Project description:In this study, the mitochondrial genome of Luperomorpha xanthodera was assembled and annotated, which is a circular DNA molecule including 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, 2 ribosomal RNA genes (12S rRNA and 16S rRNA), and 1388 bp non-coding regions (A + T rich region), measuring 16,021 bp in length. The nucleotide composition of the mitochondrial genome is 41.3% adenine (A), 38.7% thymine (T), 8.4% guanine (G), and 11.6% cytosine (C). Most of the protein-coding genes presented a typical ATN start codon (ATA, ATT, ATC, ATG), except for ND1, which showed the start codon TTG. Three-quarters of the protein-coding genes showed the complete stop codon TAR (TAA, TAG), except the genes COI, COII, ND4, and ND5, which showed incomplete stop codons (T- or TA-). All the tRNA genes have the typical clover-leaf structure, except tRNASer1 (AGN), which has a missing dihydrouridine arm (DHU). The phylogenetic results determined by both maximum likelihood and Bayesian inference methods consistently supported the monophyly of the subfamily Galerucinae and revealed that the subtribe Luperina and genus Monolepta are polyphyletic groups. Meanwhile, the classification status of the genus Luperomorpha is controversial.

Project description:The complete circular mitochondrial genome (mitogenome) of Rhadinosa nigrocyanea was 17,965?bp in length, which contained 2 ribosomal RNA genes, 22 transfer RNAs, 13 protein-coding genes (PCGs) and 1 non-coding AT-rich region with the length of 3002?bp. All of the 22 tRNA genes displayed a typical clover-leaf structure, with the exception of tRNASer (TCT). 12 PCGs were initiated by ATN codons, except for ND1 started with TTG. Only six PCGs used the typical stop codon 'TAA' and 'TGA', while seven PCGs terminated with incomplete stop codons (TA or T). Phylogenetic analysis showed that R. nigrocyanea grouped with Cassidinae species, sister to Clytrinae?+?Cryptocephalinae.
