Complete mitochondrial genome of Dahlica (Dahlica) ochrostigma Roh and Byun, 2018 (Lepidoptera: Psychidae).

ABSTRACT: We, herein, report the complete mitochondrial genome of Dahlica (Dahlica) ochrostigma. This species' genome has a total length of 15,429?bp (GenBank accession number: MK890245), consisting of 13 protein-coding genes, 22 tRNA genes, two rRNA genes, and an A?+?T rich control region. The nucleotide composition is 39.1% T, 42.8% A, 11.1% C, and 7.0% G. This is the first report of a complete mitochondrial genome of the subfamily Naryciinae, and this mitogenomic sequence can be used as a reference for phylogenetic studies on the family Psychidae.

Project description:We report the mitochondrial genome (mitogenome) of a case-making moth Acanthopsyche nigraplaga Wileman, 1911 (Lepidoptera: Psychidae). The 15,704 bp long complete mitogenome comprises a typical set of genes [13 protein-coding genes (PCGs), 2 rRNA genes, and 22 tRNA genes] and one major non-coding, A + T-rich region, with an arrangement identical to that observed in most lepidopteran mitogenomes. Twelve of the 13 PCGs of the A. nigraplaga mitogenome initiate with a typical ATN start codon, however COI contains the atypical CGA start codon that is common for lepidopteran COI genes. A phylogenetic analysis using concatenated nucleotide sequences of the 13 PCGs and 2 rRNA genes using the Bayesian inference method fully resolved A. nigraplaga in a monophyletic clade within the Psychidae. Acanthopsyche nigraplaga was situated in a sister position to Eumeta variegata and Mahasena oolona with high nodal support. As more mitogenome sequences are available further scrutinized analysis for the superfamily Tineoidea including Psychidae will be possible.

Project description:The complete mitochondrial genome of Eumeta variegate, largest bagworm moth in Japan, has been sequenced using a nanopore sequencer as a single long read. The genome has a total length of 16,601 bp, consisting of 13 protein-coding genes, 20 tRNA, 2 rRNA genes, and an AT-rich control region. The nucleotide composition was extremely AT-rich, with 42.4% A, 40.4% T, 6.67% G, and 10.6% C. This is the second report of a complete mitochondrial genome of Psychidae, and the sequence information together with a phylogenetic analysis would provide a reference data in the future studies of Lepidoptera and Psychidae.

Project description:The mitochondrial genome of Mahasena colona Sonan has been sequenced and annotated completely. The entire genome is 16,119?bp in length with an A?+?T content of 82.85% (GenBank accession No. KY856825). The tea bagworm mt genome encodes all 37 genes that are typically found in animal mt genomes, consists of 13 protein-coding genes, 2 ribosomal RNA genes, and 22 transfer RNA genes. Within the mt genome of M. colona, there are six gene reading frame overlaps. The gene order is consistent with other sequenced mt genome of moths and butterflies in Ditrysia. The mt genome of M. colona contains a 728?bp A?+?T-rich region with a high A?+?T content of 97.66%.

Project description:The complete mitochondrial genome of the female-wingless bagworm moth, Eumeta variegata Snellen, 1879 (Lepidoptera: Psychidae), is 15,660 base pairs (bp) and contains a typical set of genes (13 protein-coding genes [PCGs], 2 rRNA genes, and 22 tRNA genes) and one non-coding region, with an arrangement identical to that observed in most lepidopteran genomes. Twelve PCGs contained the typical ATN start codon, whereas COI had the atypical CGA codon, which is frequently detected in the start region of the lepidopteran COI. The A?+?T-rich region was unusually short with only 94?bp. A recent report of the same species originating from Japan revealed a lack of trnE and trnF and a 1,118?bp long A?+?T-rich region. Phylogenetic analyses with concatenated sequences of the 13 PCGs and two rRNA genes using the Bayesian inference method placed E. variegata in Psychidae, as a sister to a within-familial species, Mahasena colona, with the highest nodal support (Bayesian posterior probability?=?1).

Project description:The mitochondrial (mt) genome of Eumeta variegata Snellen (Psychidae) has been sequenced and annotated. The mt genome has a total length of 15,793 bp, consisting of 13 protein-coding genes, 22 tRNA, two rRNA genes, and an AT-rich control region (GenBank accession no. MN242985). The nucleotide composition was extremely AT-rich, and the AT content is 81.57%. The gene order is consistent with other sequenced mt genome of moths and butterflies from Ditrysia. The sequence similarity of E. variegate mt genomes between the specimen of China and South Korea is 98.38%, whereas the similarity between the specimen of China and Japan is 90.61%. The sequence of PCGs and rRNAs among different specimens are similar, and many differences are detected at the region of A + T-rich region and the tRNA block 'ARNS1EF'.

The complete mitochondrial genome of Rondotia menciana (Lepidoptera: Bombycidae).

Project description:The mulberry white caterpillar, Rondotia menciana Moore (Lepidoptera: Bombycidae) is a species with closest relationship with Bombyx mori and Bombyx mandarina, and the genetic information of R. menciana is important for understanding the diversity of the Bombycidae. In this study, the mitochondrial genome (mitogenome) of R. menciana was amplified by polymerase chain reaction and sequenced. The mitogenome of R. menciana was determined to be 15,301 bp, including 13 protein-coding genes (PCGs), 2 ribosomal RNA genes, 22 transfer RNA genes, and an AT-rich region. The A+T content (78.87%) was lower than that observed for other Bombycidae insects. All PCGs were initiated by ATN codons and terminated with the canonical stop codons, except for coxII, which was terminated by a single T. All the tRNA genes displayed a typical clover-leaf structure of mitochondrial tRNA. The length of AT-rich region (360 bp) of R. menciana mitogenome is shorter than that of other Bombycidae species. Phylogenetic analysis showed that the R. menciana was clustered on one branch with B. mori and B. mandarina from Bombycidae.

The complete mitochondrial genome of Leucoptera malifoliella Costa (Lepidoptera: Lyonetiidae).

Project description:The mitochondrial genome (mitogenome) of Leucoptera malifoliella (=L. scitella) (Lepidoptera: Lyonetiidae) was sequenced. The size was 15,646 bp with gene content and order the same as those of other lepidopterans. The nucleotide composition of L. malifoliella mitogenome is highly A+T biased (82.57%), ranked just below Coreana raphaelis (82.66%) (Lepidoptera: Lycaenidae). All protein-coding genes (PCGs) start with the typical ATN codon except for the cox1 gene, which uses CGA as the initiation codon. Nine PCGs have the common stop codon TAA, four PCGs have the common stop codon T as incomplete stop codons, and nad4l and nad6 have TAG as the stop codon. Cloverleaf secondary structures were inferred for 22 tRNA genes, but trnS1(AGN) was found to lack the DHU stem. The secondary structure of rrnL and rrnS is generally similar to other lepidopterans but with some minor differences. The A+T-rich region includes the motif ATAGA, but the poly (T) stretch is replaced by a stem-loop structure, which may have a similar function to the poly (T) stretch. Finally, there are three long repeat (154 bp) sequences followed by one short repeat (56 bp) with four (TA)(n) intervals, and a 10-bp poly-A is present upstream of trnM. Phylogenetic analysis shows that the position of Yponomeutoidea, as represented by L. malifoliella, is the same as traditional classifications. Yponomeutoidea is the sister to the other lepidopteran superfamilies covered in the present study.

It is 15,562 bp in length, and contains 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), 2 ribosomal RNA genes (rRNAs), and a 612?bp D-Loop. All PCGs start with ATN codon except for COI gene, which uses CGA as the initiation codon. Nine of 13 PCGs use a typical stop codon of TAA, the rest use incomplete stop codon of T or TA. Phylogenetic analysis of P. dolosana with other 17 leaf rollers is conducted with neighbor-joining method, the result is consistent with the conventional classification.

We sequenced 15,346-bp long complete mitochondrial genome (mitogenome) of the species, which consists of a typical set of genes (13 protein-coding genes, 2 rRNA genes, and 22 tRNA genes) and one major non-coding A?+?T-rich region. The A. iphiata mitogenome harbored the gene order tRNAMet, tRNAIle, and tRNAGln between the A?+?T-rich region and ND2 that is found in most lepidopteran mitogenomes. Bayesian inference (BI) and maximum likelihood (ML) phylogeny, using 13 protein-coding genes (PCGs) and 2 rRNAs showed that A. iphiata was placed as a sister to Geometridae with the highest nodal support (Bayesian posterior probabilities for BI = 1.00 and Bootstrap support for ML = 100).

In this study, we describe the genomic features of the mitogenome sequences of the insects. The mitogenome of C. flavescens is 15,406?bp long consisting a typical set of genes (13 protein-coding genes, 22 tRNA genes, and 2 rRNA genes) and one major 415?bp non-coding A?+?T-rich region. All PCGs of C. flavescens start with ATN codons and end with TAA codons. The gene arrangement of C. flavescens mitogenome is same to Monema flavescens while the intergenic spacers and overlaps are different. The 415?bp A?+?T-rich region contains a conserved ATAGA motif followed a poly-T stretch.
