The complete mitochondrial genome of Chrysomya nigripes (Diptera: Calliphoridae).

ABSTRACT: Chrysomya nigripes (Diptera: Calliphoridae) is a blow fly species of forensic importance. Here we demonstrated the complete mitochondrial genome of this species for the first time. Phylogenetic analyses indicated that entire mitochondrial genome sequences can provide more useful information for distinguishing C. nigripes from the other species.

Project description:Chrysomya phaonis (Seguy, 1928) is one of the blowflies of great medical and forensic importance. In this paper, we report that the entire genome of mitochondrial DNA of C. phaonis is 15,831 bp in length, which consists of 39 genes including13 protein-coding genes, 24 tRNA genes, 2 mitochondrial ribosomal RNA genes, and a 992 bp non-coding A + T-rich region. The overall base compositions of A, G, C, and T are 38.79%, 9.75%, 14.15%, and 37.31%, respectively. We provide the first complete mitochondrial genome of C. phaonis, and should provide useful information for phylogenetic and species identification for C. phaonis.

The complete mitochondrial genome of Lucilia shenyangensis (Diptera: Calliphoridae).

Project description:Lucilia shenyangensis Fan, 1965 (Diptera: Calliphoridae) is of potential importance in epidemiology, veterinary medicine, and forensic entomology due to their necrophilous habit and behaviors associated with mammals. In this study, we report the complete mitochondrial genome (mitogenome) of L. shenyangensis. The mitogenome is 14,989 bp in length, comprising 13 protein-coding genes (PCGs), two ribosomal RNAs (rRNAs), 22 transfer RNAs (tRNAs), and a non-coding control region. The arrangement of genes is identical to that of the ancestral metazoan. Nucleotide composition revealed a high A/T bias, accounting for 76.50% total mitogenome nucleotides (A 39.2%, G 9.6%, C 14.0%, T 37.3%). Phylogenetic analysis indicated that L. shenyangensis was clear separated from other blow flies and emerged as the sister lineage to the rest species from genus Lucilia (L. illustris, L. sericata, L. coeruleiviridis, and L. porphyrina). The mitogenome data of L. shenyangensis could facilitate further evolutionary genetic researches on blow flies.

The complete nucleotide sequence of the mitochondrial genome of Calliphora chinghaiensis (Diptera: Calliphoridae).

Project description:The complete mitogenome genome of Calliphora chinghaiensis (Diptera: Calliphoridae) is determined in this study. Mitochondrion of C. chinghaiensis is 15,269?bp long. It forms a circular DNA molecule that includes 37 typical animal mitochondrial genes and an A?+?T-rich region. All protein-coding genes are initiated by ATN start codon, except for cox1, which uses TCG as its start codon. 11 protein-coding genes stop with the termination codon TAN, while other protein-coding genes use CTT and AGT (cytb) as termination codon, respectively. Furthermore, the largest non-coding A?+?T-rich region with a length of 442?bp is at the end of rrns. The mitochondrial genome of C. chinghaiensis has been completely sequenced for the first time in this study.

Project description:In the present study, the complete mitochondrial genome of a blowfly Calliphora nigribarbis has been sequenced and analyzed. The length of complete the mitochondrial genome is 16,279?bp, with 39.50% A, 13.20% C, 9.30% G, and 38.0% T nucleotide distribution. The complete mitochondrial genome consists of 13 protein-coding genes, 22 transfer RNAs, and 2 ribosomal RNAs likewise the most metazoan mitochondrial genomes. Furthermore, phylogenetic relationships of C. nigribarbis in the subfamily Calliphorinae investigated. The results suggested that C. vomitoria is the most related species to C. nigribarbis and the genus Calliphora is not monophyletic. This study provides the first complete mitochondrial genome sequence for the species.

Characterization of the complete mitochondrial genome of Triceratopyga calliphoroides (Rohdendorf, 1931) (Insecta: Diptera: Calliphoridae).

Project description:Triceratopyga calliphoroides is a blowfly species which is the only member of the genus Triceratopyga. Because of their forensic importance, we sequenced the complete mitogenome of the T. calliphoroides and analyzed phylogenetic relationships. According to data, it has the longest mitogenome in the family with 16,529?bp in length. In the phylogenetic tree, T. calliphoroides were positioned in the subfamily Calliphorinae, and the closest species is Caliphora vomitoria. This is the first complete mitogenome record for the species.

Phylogenetic insights based on the first complete mitochondrial genome of Isomyia nebulosa (Diptera: Calliphoridae).

Project description:To investigate the phylogenetic position of Isomyia Walker, 1860, a genus that suffered from frequent revisions of the taxonomic status following the subfamily Rhiniinae (Diptera, Calliphoridae), we sequenced, assembled, annotated, and analyzed the first complete mitochondrial genome (mitogenome) of Isomyia nebulosa (Townsend, 1917) in this study. This mitogenome is 16,438 bp in length, with a standard set of 13 protein-coding genes (PCGs), 22 tRNAs, two rRNAs, and an A + T riched non-coding region without genetic rearrangement as most dipteran mitogenomes, but long intergenic nucleotides (IGNs) between trnQ and trnM are found. The phylogeny yielded by both Bayesian inference and maximum-likelihood analysis for all mitochondrial PCGs and rRNAs of 23 mitogenomes supports the monophyly of the family Calliphoridae and the subfamilies Calliphorinae, Chrysomyinae, and Luciliinae. In addition, I. nebulosa diverged anterior to the above-mentioned three calliphorid subfamilies with high genetic distances.

Project description:Calliphora vomitoria is a significant insect which belongs to the Calliphoridae family. In this study, the mitochondrial genome of C. vomitoria was completely sequenced for species identification. The entire mitogenome was 16,134 bp in length, composing of 13 protein-encoding genes, 22 transfer RNA genes and two ribosomal RNA genes, and then the array of the genes was similar to the other insects have discovered. The overall base compositions of A, G, C and T were 39.40%, 9.37%, 13.08% and 37.13% respectively. What is more, phylogenetic analyses tree indicated that entire mitochondria genome sequences of C. vomitoria had high degree of identification among the species listed in. We hope that the results from the present study will provide useful dipteran mitochondrial genomes information for the further studies on genetic structure and phylogenetic analyses of C. vomitoria in the species identifications.

The complete mitochondria genome of Aldrichina grahami (Diptera: Calliphoridae).

Project description:Aldrichina grahami is a significant medical and forensic insect belonging to the family calliphoridae. In this study, we present the complete mitochondrial genome of A. grahami for the first time. The mitogenome was 14 903 bp in length, consisting of 22 transfer RNA genes, 2 ribosomal RNA genes and 13 protein-encoding genes. The overall base compositions of A, G, C and T were 39.14, 9.59, 13.65 and 37.62%, respectively. Phylogenetic analysis is conducted using 12 mitochondrial genomes from two families. The monophyletic branches of the phylogenetic tree reveal that the composition of the mitochondrial genome, particularly the gene lengths, is very similar to that of another Calliphoridae fly, C. vicina, but the interspecific variations between them was 5.4%, which could distinguish these two species explicitly. Phylogenetic analyses indicated that phylogenetic analysis of whole mt genome sequences resulted in much stronger support for the identification of A. grahami between other species. This article aimed to enrich the dipteran mitochondrial genomes and provides the complete mitochondrial genome and phylogenetic analyses of A. grahami for species identifications forensic purpose.

The complete mitochondrial genome of the New Zealand parasitic blowfly Lucilia sericata (Insecta: Diptera: Calliphoridae).

Project description:In this study, the complete mitochondrial (mt) genome of the New Zealand parasitic blowfly Lucilia sericata (green bottle blowfly) field strain NZ_LucSer_NP was generated using next-generation sequencing technology. The length of complete the mt genome is 15,938 bp, with 39.4% A, 13.0% C, 9.3% G, and 38.2% T nucleotide distribution. The complete mt genome consists of 13 protein-coding genes (PCGs), two ribosomal RNAs, 22 transfer RNAs, and a 1124 bp non-coding region, similar to most metazoan mt genomes. Phylogenetic analysis showed that L. sericata NZ_LucSer_NP forms a monophyletic cluster with the remaining six Lucilia species and the Calliphoridae are polyphyletic. This study provides the first complete mt genome sequence for a L. sericata blowfly species derived from New Zealand to facilitate species identification and phylogenetic analysis.

Gene expression correlates of facultative predation in the blow fly Chrysomya rufifacies (Diptera: Calliphoridae).

Project description:Effects of intraguild predation (IGP) on omnivores and detritivores are relatively understudied when compared to work on predator guilds. Functional genetic work in IGP is even more limited, but its application can help answer a range of questions related to ultimate and proximate causes of this behavior. Here, we integrate behavioral assays and transcriptomic analysis of facultative predation in a blow fly (Diptera: Calliphoridae) to evaluate the prevalence, effect, and correlated gene expression of facultative predation by the invasive species Chrysomya rufifacies. Field work observing donated human cadavers indicated facultative predation by C. rufifacies on the native blow fly Cochliomyia macellaria was rare under undisturbed conditions, owing in part to spatial segregation between species. Laboratory assays under conditions of starvation showed predation had a direct fitness benefit (i.e., survival) to the predator. As a genome is not available for C. rufifacies, a de novo transcriptome was developed and annotated using sequence similarity to Drosophila melanogaster. Under a variety of assembly parameters, several genes were identified as being differentially expressed between predators and nonpredators of this species, including genes involved in cell-to-cell signaling, osmotic regulation, starvation responses, and dopamine regulation. Results of this work were integrated to develop a model of the processes and genetic regulation controlling facultative predation.
