Georgian cattle, sheep, goats: are they of Near-Eastern origins?

ABSTRACT: The main aim of this research was to establish the nucleotide sequence of the highly variable region of the D loop of the mitochondrial DNA of some Georgian domestic animal species (cattle, goat, sheep) as well as their phylogenetic position among the worldwide set of domestic animals. In this study, a total of 5 haplogroups (T - 5; T3 - 7; T1 - 1; T2 - 2; T5 - 2) in 17 Georgian Mountain cattle (GMC), 4 haplogroups (A - 15; A2a1 - 3; A1a - 1; A6 - 3) in 22 Georgian goats and 3 haplogroups (A - 10; B - 16; C -15) in 41 Georgian sheeps (15 Imeretian and 26 Tushetian) were detected. This study represents the first attempt of Genetic study of native Georgian livestocks. The GMC, Georgian (Megrelian) goat, Georgian (Imeretian and Tushetian) sheep mitogenomes were grouped phylogenetically in the haplogroups indicating the closeness to the Near Eastern animals.

Serological and Molecular Characterization of Mycobacterium avium Subsp. paratuberculosis (MAP) from Sheep, Goats, Cattle and Camels in the Eastern Province, Saudi Arabia.

Project description:The objectives of the present study were to characterize Mycobacterium avium subsp. paratuberculosis (MAP) infection using serological and molecular tools and investigate the distribution and molecular characterization of MAP strains (cattle (C) and sheep (S) types) in sheep, goat, cattle, and camel herds in Eastern Province, Saudi Arabia. Serum and fecal samples were collected from all animals aged >2 years old in 31 herds (sheep = 8, goats = 6, cattle = 8 and camels = 9) from January to December 2019. Serum samples were tested by ELISA for the detection of MAP antibodies. Fecal samples were tested by PCR for the detection of MAP IS900 gene and the identification of MAP strains. MAP antibodies were detected in 19 (61.3%) herds. At the animal level, antibodies against MAP were detected in 43 (19.5%) sheep, 21 (17.1%) goats, 13 (19.7%) cattle and 22 (9.1%) camels. The IS900 gene of MAP was detected in 23 (74.2%) herds and was directly amplified from fecal samples of 59 (26.8%) sheep, 34 (27.6%) goats, 20 (30.3%) cattle and 36 (15.0%) camels. The S-type was the most prevalent MAP type identified in 15 herds, and all were identified as type-I, while the C-type was identified in only 8 herds. The IS900 sequences revealed genetic differences among the MAP isolates recovered from sheep, goats, cattle and camels. Results from the present study show that MAP was prevalent and confirm the distribution of different MAP strains in sheep, goat, cattle and camel herds in Eastern Province, Saudi Arabia.

First molecular characterization of poxviruses in cattle, sheep, and goats in Botswana.

Project description:BackgroundPoxviruses within the Capripoxvirus, Orthopoxvirus, and Parapoxvirus genera can infect livestock, with the two former having zoonotic importance. In addition, they induce similar clinical symptoms in common host species, creating a challenge for diagnosis. Although endemic in the country, poxvirus infections of small ruminants and cattle have received little attention in Botswana, with no prior use of molecular tools to diagnose and characterize the pathogens.MethodsA high-resolution melting (HRM) assay was used to detect and differentiate poxviruses in skin biopsy and skin scab samples from four cattle, one sheep, and one goat. Molecular characterization of capripoxviruses and parapoxviruses was undertaken by sequence analysis of RPO30 and GPCR genes.ResultsThe HRM assay revealed lumpy skin disease virus (LSDV) in three cattle samples, pseudocowpox virus (PCPV) in one cattle sample, and orf virus (ORFV) in one goat and one sheep sample. The phylogenetic analyses, based on the RPO30 and GPCR multiple sequence alignments showed that the LSDV sequences of Botswana were similar to common LSDV field isolates encountered in Africa, Asia, and Europe. The Botswana PCPV presented unique features and clustered between camel and cattle PCPV isolates. The Botswana ORFV sequence isolated from goat differed from the ORFV sequence isolated from sheep.ConclusionsThis study is the first report on the genetic characterization of poxvirus diseases circulating in cattle, goats, and sheep in Botswana. It shows the importance of molecular methods to differentially diagnose poxvirus diseases of ruminants.

Seroprevalence of peste des petits ruminants in sheep and goats in Eastern India.

Project description:The seroprevalence study of peste des petits ruminants (PPR) in small ruminants in Bihar and Odisha states in the Eastern region of India was carried out. A total of 1836 serum samples were collected from sheep (n = 648) and goats (n = 1188) from various epidemiological units (n = 112) in these states by a two-stage sampling plan during April 2017-March 2018. These samples were tested for the detection of virus antibodies by PPR competitive ELISA kit. The results revealed that the seroprevalence of PPR in sheep and goats in Bihar and Odisha states was 30.91% and 54.20%, respectively. Further, the chi-square analysis showed that the association exists between the presence of PPR virus antibodies in the goats (χ2 = 93.28, p < 0.01) and between the states (χ2 = 82.61, p < 0.01). This cross-sectional serosurvey also infers that the sheep and goats in most of the epi-units (n = 87) had < 70% of PPR virus antibodies prevalence. This warrants the intensive continuous mass vaccination program for a few more years to achieve the desired level of population immunity (epidemiological units protection level) and active surveillance to make these states free from PPR in the Eastern region of India.

Understanding Circular RNAs in Health, Welfare, and Productive Traits of Cattle, Goats, and Sheep.

Project description:Circular RNAs (circRNAs) are unique noncoding RNA molecules, notable for their covalent closed-loop structures, which play a crucial role in regulating gene expression across a variety of biological processes. This review comprehensively synthesizes the existing knowledge of circRNAs in three key livestock species: Bos taurus (cattle), Ovis aries (sheep), and Capra hircus (goats). It focuses on their functional importance and emerging potential as biomarkers for disease detection, stress response, and overall physiological health. Specifically, it delves into the expression and functionality of circRNAs in these species, paying special attention to traits critical to livestock productivity such as milk production, meat quality, muscle development, wool production, immune responses, etc. We also address the current challenges faced in circRNA research, including the need for standardized methodologies and broader studies. By providing insights into the molecular mechanisms regulated by circRNAs, this review underscores their scientific and economic relevance in the livestock industry. The potential of circRNAs to improve animal health management and the quality of animal-derived products aligns with growing consumer concerns for animal welfare and sustainability. Thus, this paper aims to guide future research directions while supporting the development of innovative strategies in livestock management and breeding.

Global distribution data for cattle, buffaloes, horses, sheep, goats, pigs, chickens and ducks in 2010.

Project description:Global data sets on the geographic distribution of livestock are essential for diverse applications in agricultural socio-economics, food security, environmental impact assessment and epidemiology. We present a new version of the Gridded Livestock of the World (GLW 3) database, reflecting the most recently compiled and harmonized subnational livestock distribution data for 2010. GLW 3 provides global population densities of cattle, buffaloes, horses, sheep, goats, pigs, chickens and ducks in each land pixel at a spatial resolution of 0.083333 decimal degrees (approximately 10 km at the equator). They are accompanied by detailed metadata on the year, spatial resolution and source of the input census data. Two versions of each species distribution are produced. In the first version, livestock numbers are disaggregated within census polygons according to weights established by statistical models using high resolution spatial covariates (dasymetric weighting). In the second version, animal numbers are distributed homogeneously with equal densities within their census polygons (areal weighting) to provide spatial data layers free of any assumptions linking them to other spatial variables.

Tracking the Near Eastern origins and European dispersal of the western house mouse.

Project description:The house mouse (Mus musculus) represents the extreme of globalization of invasive mammals. However, the timing and basis of its origin and early phases of dispersal remain poorly documented. To track its synanthropisation and subsequent invasive spread during the develoment of complex human societies, we analyzed 829 Mus specimens from 43 archaeological contexts in Southwestern Asia and Southeastern Europe, between 40,000 and 3,000?cal. BP, combining geometric morphometrics numerical taxonomy, ancient mitochondrial DNA and direct radiocarbon dating. We found that large late hunter-gatherer sedentary settlements in the Levant, c. 14,500?cal. BP, promoted the commensal behaviour of the house mouse, which probably led the commensal pathway to cat domestication. House mouse invasive spread was then fostered through the emergence of agriculture throughout the Near East 12,000 years ago. Stowaway transport of house mice to Cyprus can be inferred as early as 10,800 years ago. However, the house mouse invasion of Europe did not happen until the development of proto urbanism and exchange networks - 6,500 years ago in Eastern Europe and 4000 years ago in Southern Europe - which in turn may have driven the first human mediated dispersal of cats in Europe.

The first complete mitogenomes and phylogeny of Georgian Mountain Cattle.

Project description:The Georgian Mountain Cattle (GMC) (Species: Bos taurus Linnaeus, 1758 - aurochs, domesticated cattle, domestic cattle (feral), Aurochs, Subspecies: Bos taurus taurus Linnaeus, 1758) is a local breed from Georgia. It is well adapted to the harsh mountain conditions of the Caucasus, resistant to different pathogens and able to provide milk during the lowest feed rations. In this study, we report for the first time the complete mitochondrial genome sequence of GMC. We selected the five unique control region mitochondrial haplotypes of GMC and sequenced for the complete mitogenome, using Illumina MiSeq platform. The results of our research showed a total of 59 substitutions and seven indels, in comparison to the Bovine Reference Sequence (BRS; V00654), with 15 SNPs never observed before. The complete mitochondrial DNA (mtDNA) phylogenetic analyses revealed hitherto unknown haplotypes falling outside the known taurine diversity. Four mitogenomes fell within haplogroup T (sub-lineages T1, T3, and T5), while one belonged to haplogroup Q (branch Q1). The combination of our results with precision agriculture holds great promises for the identification of genetic variants economically affecting important traits of GMC cattle.

Polychlorinated biphenyl and polybrominated diphenyl ether profiles in serum from cattle, sheep, and goats across California.

Project description:It has been previously been shown by our lab and others that persistent organic pollutants, such as polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs), are contaminants in milk produced for human consumption. To further this research we determined the concentration of 21 PCB and 14 PBDE congeners in livestock serum, mainly bovine, across California. Congeners were extracted from serum using solid phase extraction (SPE), cleaned up by silica cartridge and quantified using gas chromatography-triple quadruple mass spectrometry. We detected significant differences among species and the production class of cattle (beef or dairy). The sum of all 21 PCB congeners (?PCBs) in caprine and ovine sera had a mean value of 9.26 and 9.13 ng/mL, respectively, compared to 3.98 ng/mL in bovine sera. The mean value for the sum of all 14 PBDE congeners (?PBDEs) in caprine and ovine sera was 2.82 and 2.39 ng/mL, respectively, compared to 0.91 ng/mL in bovine sera. Mean ?PCBs in dairy cattle was 5.92 ng/mL compared to 2.70 ng/mL in beef cattle. Mean ?PBDEs in dairy cattle was 1.33 ng/mL compared to 0.70 ng/mL in beef cattle. There were no regional differences in the ?PCBs or ?PBDEs in cattle distributed across California. These results highlight the fact that livestock are still being exposed to these pollutants yet little is known about where this exposure may be coming from.

Safety and efficacy of four-segmented Rift Valley fever virus in young sheep, goats and cattle.

Project description:Rift Valley fever virus (RVFV) is a mosquito-borne bunyavirus that causes severe and recurrent outbreaks on the African continent and the Arabian Peninsula and continues to expand its habitat. RVFV induces severe disease in newborns and abortion in pregnant ruminants. The viral genome consists of a small (S), medium (M) and large (L) RNA segment of negative polarity. The M segment encodes a glycoprotein precursor protein that is co-translationally cleaved into the two structural glycoproteins Gn and Gc, which are involved in receptor attachment and cell entry. We previously constructed a four-segmented RVFV (RVFV-4s) by splitting the M genome segment into two M-type segments encoding either Gn or Gc. RVFV-4s replicates efficiently in cell culture but was shown to be completely avirulent in mice, lambs and pregnant ewes. Here, we show that a RVFV-4s candidate vaccine for veterinary use (vRVFV-4s) does not disseminate in vaccinated animals, is not shed or spread to the environment and does not revert to virulence. Furthermore, a single vaccination of lambs, goat kids and calves was shown to induce protective immunity against a homologous challenge. Finally, the vaccine was shown to provide full protection against a genetically distinct RVFV strain. Altogether, we demonstrate that vRVFV-4s optimally combines efficacy with safety, holding great promise as a next-generation RVF vaccine.

Genotyping Characterization of Toxoplasma gondii in Cattle, Sheep, Goats and Swine from the North of Portugal.

Project description:BackgroundEpidemiological investigations on Toxoplasma gondii infection have found a significant association between human toxoplasmosis and consumption of raw or undercooked meat. The present study aimed to characterize genotypes of T. gondii in 20 cattle, 40 sheep, 15 goats and 16 pigs from the North of Portugal.MethodsNested PCR amplified the surface antigen 2 (SAG2) gene. Sequencing analysis was performed in order to assess the prevalence of SAG2 type strains (I, II and III).ResultsThree and 4 strains of SAG2 type II were identified in heart samples of cattle and sheep, respectively. Three SAG2 type II strains were detected in brain, diaphragm and heart of 3 pigs. Three strains detected in heart samples of 3 goats belonged to SAG2 types I or II; with the same result being observed in heart samples from 2 sheep and in 2 brain and 1 heart samples from 3 pigs.ConclusionSAG2 type II has been shown for the first time to infect cattle in North of Portugal. In addition, SAG2 type II has also been confirmed as the predominant strain in sheep and pigs in the same region. This is the first molecular report of T. gondii in goats from Portugal.
