Novel species of Pestalotiopsis fungi on Dracaena from Thailand.

ABSTRACT: A survey of the diversity and distribution of microfungi on Dracaena leaf litter in Songkhla Province (Thailand) yielded two collections of pestalotiopsis-like fungi. Analyses of a combined ITS, TEF1-α and TUB2 sequence data matrix were applied to infer the phylogenetic position of these new isolates in Pestalotiopsis. The phylogenies indicated that these two isolates were monophyletic and constituted a distinct lineage that perceived a taxonomic novelty in Pestalotiopsis. This clade shared a close phylogenetic affinity with P. adusta, P. krabiensis, P. pandanicola and P. papuana. The comparison of morphological features with the phylogenetically closely related taxa are given and the new species is introduced as Pestalotiopsis dracaenicola sp. nov. with comprehensive descriptions and illustrations herein.

Project description:A morphologically distinct element of the group of Dracaena species from Thailand and Burma with undifferentiated leaf sheaths, no leaf blade central costa, free tepals and free thickened filaments known as Chan nuu or Chan pha krai in Thai is shown to be a distinct species, Dracaena kaweesakii Wilkin & Suksathan based on habit, leaf base and margin, inflorescence axis indumentum and floral characters. It is described and illustrated. Ecological and conservation status assessment information are provided.

Three Novel Entomopathogenic Fungi From China and Thailand.

Project description:Entomopathogenic fungi are ubiquitous in tropical rainforests and feature a high level of diversity. This group of fungi not only has important ecological value but also medicinal value. Nevertheless, they are often ignored, and many unknown species have yet to be discovered and described. The present study aims to contribute to the taxonomical and phylogenetic understanding of the genus Paraisaria by describing three new species collected from Guizhou and Yunnan Provinces in China and Krabi Province in Thailand. The three novel species named Paraisaria alba, P. arcta, and P. rosea share similar morphologies as those in the genus Paraisaria, containing solitary, simple, fleshy stroma, completely immersed perithecia and cylindrical asci with thickened caps and filiform ascospores that often disarticulate at maturity. Phylogenetic analyses of combined LSU, SSU, TEF1-α, RPB1, RPB2, and ITS sequence data confirm their placement in the genus Paraisaria. In this study, the three entomopathogenic taxa are comprehensively described with color photographs and phylogenetic analyses. A synopsis table and a key to all treated species of Paraisaria are also included.

Novel Mucor species (Mucoromycetes, Mucoraceae) from northern Thailand

Project description:Abstract Mucor species are common fast-growing fungi found in soil. Two new species of Mucor and one new geographical record of M.nederlandicus were collected from northern Thailand and are described in this study. Evidence from morphophysiological data and phylogenetic analysis supports the introduction of the new taxa. Phylogenetic analysis based on the internal transcribed spacer (ITS) and large subunit of the nuclear ribosomal RNA (LSU) data showed that the new isolates cluster distinctly from other Mucor species with high or maximum bootstrap support. Mucoraseptatophorus is characterized by aseptate sporangiophores, globose columella, resistant and deliquescent sporangia, has sympodial, and monopodial branches and shows growth at 37 °C. It also differs from M.irregularis in having smaller sporangiospores, and larger sporangia. Mucorchiangraiensis has subglobose or slightly elongated globose columella, produces hyaline sporangiospores, and resistant and deliquescent sporangia. Furthermore, this species has wider sporangiophore, smaller sporangia and lower growth than M.nederlandicus. A detailed description of the species and illustrations are provided for the novel species.

Disseminated infection caused by novel species of Microsporidium, Thailand.

Project description:We describe a case of microsporidial myositis in a healthy man from Thailand. The small subunit rRNA sequence of this microsporidium is novel and has a close phylogenetic relationship with Endoreticulatus, a genus of lepidopteran microsporidia. Myositis could be caused by more genera of microsporidia than previously known.

A Novel Hypovirus Species From Xylariaceae Fungi Infecting Avocado.

Project description:The white rot root disease caused by Rosellinia necatrix is a major concern for avocado cultivation in Spain. Healthy escapes of avocado trees surrounded by diseased trees prompted us to hypothesize the presence of hypovirulent R. necatrix due to mycovirus infections. Recently, we reported the presence of another fungal species, Entoleuca sp., belonging to the Xylariaceae, that was also found in healthy avocado trees and frequently co-infecting the same roots than R. necatrix. We investigated the presence of mycoviruses that might explain the hypovirulence. For that, we performed deep sequencing of dsRNAs from two isolates of Entoleuca sp. that revealed the simultaneous infection of several mycoviruses, not described previously. In this work, we report a new member of the Hypoviridae, tentatively named Entoleuca hypovirus 1 (EnHV1). The complete genome sequence was obtained for two EnHV1 strains, which lengths resulted to be 14,958 and 14,984 nt, respectively, excluding the poly(A) tails. The genome shows two ORFs separated by a 32-nt inter-ORF, and both 5'- and 3'-UTRs longer than any other hypovirus reported to date. The analysis of virus-derived siRNA populations obtained from Entoleuca sp. demonstrated antiviral silencing activity in this fungus. We screened a collection of Entoleuca sp. and R. necatrix isolates and found that EnHV1 was present in both fungal species. A genetic population analysis of EnHV1 strains revealed the presence of two main clades, each of them including members from both Entoleuca sp. and R. necatrix, which suggests intra- and interspecific virus transmission in the field. Several attempts failed to cure Entoleuca sp. from EnHV1. However, all Entoleuca sp. isolates collected from avocado, whether harboring the virus or not, showed hypovirulence. Conversely, all R. necatrix isolates were pathogenic to that crop, regardless of being infected by EnHV1.

Project description:BackgroundSpecies of the fungal genus Arthrinium (Sordariomycetes, Amphisphaeriales, Apiosporaceae) are often found on bamboo in Asia. They are endophytes, saprobes and important plant pathogens. The genus Arthrinium currently contains 92 species and is widely distributed in North and South America, Europe, Africa, Asia and Oceania.New informationIn this study, a new species, Arthrinium bambusicola sp. nov., is described and illustrated. The new taxon is characterised by oval to broadly or irregularly round, medium brown, multi-guttulate to roughened, granular conidia, with finely pale slits in the outer edges. Arthrinium bambusicola can be distinguished from the closest related species A. gutiae by its conidial characteristics. Phylogenetic analyses of a four-locus dataset (ITS, LSU, TEF1, TUB2) confirm that A. bambusicola is a distinct new species.

Two new species of Hymenochaetaceae on Dracaena cambodiana from tropical China.

Project description:Two new wood-rotting fungi in the family Hymenochaetaceae, Fulvifomes dracaenicola sp. nov. and Hymenochaete dracaenicola sp. nov., are described and illustrated from tropical China based on morphological characteristics and molecular data. It is worth to mention that both of them grow on Dracaena cambodiana which is a kind of angiosperm tree distributed in tropical regions. F. dracaenicola is characterised by perennial, pileate, triquetrous basidioma with yellowish brown fresh pores which becoming honey yellow with silk sheening upon drying, a dimitic hyphal system in trama and monomitic in context, and subglobose basidiospores measuring 4.8-5 × 4-4.1 μm. H. dracaenicola is characterised by annual, resupinate basidioma with a clay buff hymenophore, a dimitic hyphal system, absence of tomentum and cortex, presence of subulate setae, absence of cystidia, presence of cystidioles and simple hyphidia, and oblong ellipsoid basidiospores measuring 5.2-5.8 × 2.5-2.8 µm. The phylogenetic analyses based on ITS + nLSU rDNA sequences confirm the placement of two new species respectively in Fulvifomes and Hymenochaete. Phylogenetically closely related species to the two new species are discussed.

Pestalotiopsis revisited.

Project description:Species of Pestalotiopsis occur commonly as plant pathogens, and represent a fungal group known to produce a wide range of chemically novel, diverse metabolites. In the present study, we investigated 91 Pestalotiopsis isolates from the CBS-KNAW Fungal Biodiversity Centre (CBS) culture collection. The phylogeny of the Amphisphaeriaceae was constructed based on analysis of 28S nrRNA gene (LSU) sequence data, and taxonomic changes are proposed to reflect more natural groupings. We combined morphological and DNA data, and segregated two novel genera from Pestalotiopsis, namely Neopestalotiopsis and Pseudopestalotiopsis. The three genera are easily distinguishable on the basis of their conidiogenous cells and colour of their median conidial cells. We coupled morphological and combined sequence data of internal transcribed spacer (ITS), partial ?-tubulin (TUB) and partial translation elongation factor 1-alpha (TEF) gene regions, which revealed 30 clades in Neopestalotiopsis and 43 clades in Pestalotiopsis. Based on these data, 11 new species are introduced in Neopestalotiopsis, 24 in Pestalotiopsis, and two in Pseudopestalotiopsis. Several new combinations are proposed to emend monophyly of Neopestalotiopsis, Pestalotiopsis and Pseudopestalotiopsis.

Pestalotiopsis kaki sp. nov., a Novel Species Isolated from Persimmon Tree (Diospyros kaki) Bark in Korea.

Project description:During the screening of Korean microflora, a fungal strain (KNU-PT-1804) belonging to the genus Pestalotiopsis was isolated from persimmon tree (Diospyros kaki) bark collected from North Gyeongsang Province, Korea. The strain, KNU-PT-1804, produced smaller conidia compared with related species P. kenyana, P. neglecta, and P. telopeae. The novelty of the strain was confirmed based on phylogenetic analysis using molecular datasets of internal transcribed spacer (ITS) regions, β-tubulin (TUB2), and translation elongation factor 1-alpha (TEF1α) genes. Molecular phylogeny strongly supports that the strain is distinct from previously known Pestalotiopsis species, and we proposed the novel species, Pestalotiopsis kaki sp. nov., and provide a detailed description and illustration.

Novel genera and species of coniothyrium-like fungi in Montagnulaceae (Ascomycota).

Project description:Based on analyses of concatenated internal transcribed spacer regions of the nrDNA operon (ITS), large subunit rDNA (LSU), ?-actin and ?-tubulin gene sequences the taxonomy of coniothyrium-like fungi belonging in the family Montagnulaceae, order Pleosporales, was re-assessed. Two new genera are proposed, Alloconiothyrium, to accommodate A. aptrootii sp. nov., and Dendrothyrium for D. longisporum sp. nov. and D. variisporum sp. nov. One new species is described in Paraconiothyrium, viz. Parac. archidendri sp. nov., while two species so far classified in Paraconiothyrium are transferred to Paraphaeosphaeria, viz. Paraph. minitans comb. nov. and Paraph. sporulosa comb. nov. In Paraphaeosphaeria five new species are described based on asexual morphs, viz. Paraph. arecacearum sp. nov., Paraph. neglecta sp. nov., Paraph. sardoa sp. nov., Paraph. verruculosa sp. nov., and Paraph. viridescens sp. nov. Macro- and micromorphological characteristics are fully described.
