Thermoplasmonic neural chip platform for in situ manipulation of neuronal connections in vitro.

ABSTRACT: Cultured neuronal networks with a controlled structure have been widely studied as an in vitro model system to investigate the relationship between network structure and function. However, most cell culture techniques lack the ability to control network structures during cell cultivation, making it difficult to assess functional changes induced by specific structural changes. In this study, we present an in situ manipulation platform based on gold-nanorod-mediated thermoplasmonics to interrogate an in vitro network model. We find that it is possible to induce new neurite outgrowths, eliminate interconnecting neurites, and estimate functional relationships in matured neuronal networks. This method is expected to be useful for studying functional dynamics of neural networks under controlled structural changes.

A micro-fabricated in vitro complex neuronal circuit platform

Project description:Abstract Developments in micro-manufacture as well as biofabrication technologies are driving our ability to create complex tissue models such as ‘organ-on-a-chip’ devices. The complexity of neural tissue, however, requires precisely specific cellular connectivity across many neuronal populations, and thus there have been limited reports of complex ‘brain-on-a-chip’ technologies modelling specific cellular circuit function. Here we describe the development of a model of in vitro brain circuitry designed to accurately reproduce part of the complex circuitry involved in neurodegenerative diseases; using segregated co-culture of specific basal ganglia (BG) neuronal subtypes to model central nervous system circuitry. Lithographic methods and chemical modification were used to form structured micro-channels, which were populated by specifically cultured neuronal sub-types to represent parts of the inter-communicating neural circuit. Cell morphological assessment and immunostaining showed connectivity, which was supported by electrophysiology measurements. Electrical activity of cells was measured using patch-clamp, showing voltage dependant Na+ and K+ currents, and blocking of Na+ current by TTX, and calcium imaging showing TTX-sensitive slow Ca2+ oscillations resulting from action potentials. Monitoring cells across connected ports post-TTX addition demonstrated both upstream and downstream changes in activity, indicating network connectivity. The model developed herein provides a platform technology that could be used to better understand neurological function and dysfunction, contributing to a growing urgency for better treatments of neurodegenerative disease. We anticipate the use of this advancing technology for the assessment of pharmaceutical and cellular therapies as a means of pre-clinical assessment, and further for the advancement of neural engineering approaches for tissue engineering.

Vascularization of iNSC spheroid in a 3D spheroid-on-a-chip platform enhances neural maturation.

Project description:In vitro platforms for studying the human brain have been developed, and brain organoids derived from stem cells have been studied. However, current organoid models lack three-dimensional (3D) vascular networks, limiting organoid proliferation, differentiation, and apoptosis. In this study, we created a 3D model of vascularized spheroid cells using an injection-molded microfluidic chip. We cocultured spheroids derived from induced neural stem cells (iNSCs) with perfusable blood vessels. Gene expression analysis and immunostaining revealed that the vascular network greatly enhanced spheroid differentiation and reduced apoptosis. This platform can be used to further study the functional and structural interactions between blood vessels and neural spheroids, and ultimately to simulate brain development and disease.

Photothermally triggered actuation of hybrid materials as a new platform for in vitro cell manipulation.

Project description:Mechanical forces in the cell's natural environment have a crucial impact on growth, differentiation and behaviour. Few areas of biology can be understood without taking into account how both individual cells and cell networks sense and transduce physical stresses. However, the field is currently held back by the limitations of the available methods to apply physiologically relevant stress profiles on cells, particularly with sub-cellular resolution, in controlled in vitro experiments. Here we report a new type of active cell culture material that allows highly localized, directional and reversible deformation of the cell growth substrate, with control at scales ranging from the entire surface to the subcellular, and response times on the order of seconds. These capabilities are not matched by any other method, and this versatile material has the potential to bridge the performance gap between the existing single cell micro-manipulation and 2D cell sheet mechanical stimulation techniques.

Plasmon-driven nanowire actuators for on-chip manipulation.

Project description:Chemically synthesized metal nanowires are promising building blocks for next-generation photonic integrated circuits, but technological implementation in monolithic integration will be severely hampered by the lack of controllable and precise manipulation approaches, due to the strong adhesion of nanowires to substrates in non-liquid environments. Here, we demonstrate this obstacle can be removed by our proposed earthworm-like peristaltic crawling motion mechanism, based on the synergistic expansion, friction, and contraction in plasmon-driven metal nanowires in non-liquid environments. The evanescently excited surface plasmon greatly enhances the heating effect in metal nanowires, thereby generating surface acoustic waves to drive the nanowires crawling along silica microfibres. Advantages include sub-nanometer positioning accuracy, low actuation power, and self-parallel parking. We further demonstrate on-chip manipulations including transporting, positioning, orientation, and sorting, with on-situ operation, high selectivity, and great versatility. Our work paves the way to realize full co-integration of various functionalized photonic components on single chips.

Metamaterial-enabled arbitrary on-chip spatial mode manipulation.

Project description:On-chip spatial mode operation, represented as mode-division multiplexing (MDM), can support high-capacity data communications and promise superior performance in various systems and numerous applications from optical sensing to nonlinear and quantum optics. However, the scalability of state-of-the-art mode manipulation techniques is significantly hindered not only by the particular mode-order-oriented design strategy but also by the inherent limitations of possibly achievable mode orders. Recently, metamaterials capable of providing subwavelength-scale control of optical wavefronts have emerged as an attractive alternative to manipulate guided modes with compact footprints and broadband functionalities. Herein, we propose a universal yet efficient design framework based on the topological metamaterial building block (BB), enabling the excitation of arbitrary high-order spatial modes in silicon waveguides. By simply programming the layout of multiple fully etched dielectric metamaterial perturbations with predefined mathematical formulas, arbitrary high-order mode conversion and mode exchange can be simultaneously realized with uniform and competitive performance. The extraordinary scalability of the metamaterial BB frame is experimentally benchmarked by a record high-order mode operator up to the twentieth. As a proof of conceptual application, an 8-mode MDM data transmission of 28-GBaud 16-QAM optical signals is also verified with an aggregate data rate of 813 Gb/s (7% FEC). This user-friendly metamaterial BB concept marks a quintessential breakthrough for comprehensive manipulation of spatial light on-chip by breaking the long-standing shackles on the scalability, which may open up fascinating opportunities for complex photonic functionalities previously inaccessible.

Development of input connections in neural cultures.

Project description:We introduce an approach for the quantitative assessment of the connectivity in neuronal cultures, based on the statistical mechanics of percolation on a graph. This allows us to monitor the development of the culture and to see the emergence of connectivity in the network. The culture becomes fully connected at a time equivalent to the expected time of birth. The spontaneous bursting activity that characterizes cultures develops in parallel with the connectivity. The average number of inputs per neuron can be quantitatively determined in units of m(0), the number of activated inputs needed to excite the neuron. For m(0) approximately 15 we find that hippocampal neurons have on average approximately 60-120 inputs, whereas cortical neurons have approximately 75-150, depending on neuronal density. The ratio of excitatory to inhibitory neurons is determined by using the GABA(A) antagonist bicuculine. This ratio changes during development and reaches the final value at day 7-8, coinciding with the expected time of the GABA switch. For hippocampal cultures the inhibitory cells comprise approximately 30% of the neurons in the culture whereas for cortical cultures they are approximately 20%. Such detailed global information on the connectivity of networks in neuronal cultures is at present inaccessible by any electrophysiological or other technique.

Physical connections between different SSVEP neural networks.

Project description:This work investigates the mechanism of the Steady-State Visual Evoked Potential (SSVEP). One theory suggests that different SSVEP neural networks exist whose strongest response are located in different frequency bands. This theory is based on the fact that there are similar SSVEP frequency-amplitude response curves in these bands. Previous studies that employed simultaneous stimuli of different frequencies illustrated that the distribution of these networks were similar, but did not discuss the physical connection between them. By comparing the SSVEP power and distribution under a single-eye stimulus and a simultaneous, dual-eye stimulus, this work demonstrates that the distributions of different SSVEP neural networks are similar to each other and that there should be physical overlapping between them. According to the band-pass filter theory in a signal transferring channel, which we propose in this work for the first time, there are different amounts of neurons that are involved under repetitive stimuli of different frequencies and that the response intensity of each neuron is similar to each other so that the total response (i.e., the SSVEP) that is observed from the scalp is different.

Engineering a 3D functional human peripheral nerve in vitro using the Nerve-on-a-Chip platform.

Project description:Development of "organ-on-a-chip" systems for neuroscience applications are lagging due in part to the structural complexity of the nervous system and limited access of human neuronal & glial cells. In addition, rates for animal models in translating to human success are significantly lower for neurodegenerative diseases. Thus, a preclinical in vitro human cell-based model capable of providing critical clinical metrics such as nerve conduction velocity and histomorphometry are necessary to improve prediction and translation of in vitro data to successful clinical trials. To answer this challenge, we present an in vitro biomimetic model of all-human peripheral nerve tissue capable of showing robust neurite outgrowth (~5 mm), myelination of hNs by primary human Schwann cells (~5%), and evaluation of nerve conduction velocity (0.13-0.28 m/sec), previously unrealized for any human cell-based in vitro system. To the best of our knowledge, this Human Nerve-on-a-chip (HNoaC) system is the first biomimetic microphysiological system of myelinated human peripheral nerve which can be used for evaluating electrophysiological and histological metrics, the gold-standard assessment techniques previously only possible with in vivo studies.

A reductionist metastasis-on-a-chip platform for in vitro tumor progression modeling and drug screening.

Project description:Current animal and 2-D cell culture models employed in metastasis research and drug discovery remain poor mimics of human cancer physiology. Here we describe a "metastasis-on-a-chip" system allowing real time tracking of fluorescent colon cancer cells migrating from hydrogel-fabricated gut constructs to downstream liver constructs within a circulatory fluidic device system that responds to environmental manipulation and drug treatment. Devices consist of two chambers in which gut and liver constructs are housed independently, but are connected in series via circulating fluid flow. Constructs were biofabricated with a hyaluronic acid-based hydrogel system, capable of a variety of customizations, inside of which representative host tissue cells were suspended and metastatic colon carcinoma tumor foci were created. The host tissue of the constructs expressed normal epithelial markers, which the tumor foci failed to express. Instead, tumor regions lost membrane-bound adhesion markers, and expressed mesenchymal and proliferative markers, suggesting a metastatic phenotype. Metastatic tumor foci grew in size, eventually disseminating from the intestine construct and entering circulation, subsequently reaching in the liver construct, thus mimicking some of the migratory events observed during metastasis. Lastly, we demonstrated the ability to manipulate the system, including chemically modulating the hydrogel system mechanical properties and administering chemotherapeutic agents, and evaluated the effects of these parameters on invasive tumor migration. These results describe the capability of this early stage metastasis-on-a-chip system to model several important characteristics of human metastasis, thereby demonstrating the potential of the platform for making meaningful advances in cancer investigation and drug discovery.
