Complete mitochondrial genome of Sesia siningensis (Lepidoptera: Sesiidae).

ABSTRACT: Sesia siningensis is an important trunk borer of poplar and is widely distributed in China. Here, the complete mitochondrial genome of S. siningensis was sequenced. The circle genome of the clearwing moth is 15,454 bp in length. There are 38 sequence elements including 13 protein coding genes, 22 tRNA genes, 2 rRNA genes, and a control region. The order of most elements was consistent with that of Chilo suppressalis, with the exception of one tRNA gene. As the first reported mitochondrial genome in the Sesiidae family, it will provide useful information to the development and application of better markers and primers in the molecular taxonomy of this family.

Project description:The mulberry white caterpillar, Rondotia menciana Moore (Lepidoptera: Bombycidae) is a species with closest relationship with Bombyx mori and Bombyx mandarina, and the genetic information of R. menciana is important for understanding the diversity of the Bombycidae. In this study, the mitochondrial genome (mitogenome) of R. menciana was amplified by polymerase chain reaction and sequenced. The mitogenome of R. menciana was determined to be 15,301 bp, including 13 protein-coding genes (PCGs), 2 ribosomal RNA genes, 22 transfer RNA genes, and an AT-rich region. The A+T content (78.87%) was lower than that observed for other Bombycidae insects. All PCGs were initiated by ATN codons and terminated with the canonical stop codons, except for coxII, which was terminated by a single T. All the tRNA genes displayed a typical clover-leaf structure of mitochondrial tRNA. The length of AT-rich region (360 bp) of R. menciana mitogenome is shorter than that of other Bombycidae species. Phylogenetic analysis showed that the R. menciana was clustered on one branch with B. mori and B. mandarina from Bombycidae.

The complete mitochondrial genome of Leucoptera malifoliella Costa (Lepidoptera: Lyonetiidae).

Project description:The mitochondrial genome (mitogenome) of Leucoptera malifoliella (=L. scitella) (Lepidoptera: Lyonetiidae) was sequenced. The size was 15,646 bp with gene content and order the same as those of other lepidopterans. The nucleotide composition of L. malifoliella mitogenome is highly A+T biased (82.57%), ranked just below Coreana raphaelis (82.66%) (Lepidoptera: Lycaenidae). All protein-coding genes (PCGs) start with the typical ATN codon except for the cox1 gene, which uses CGA as the initiation codon. Nine PCGs have the common stop codon TAA, four PCGs have the common stop codon T as incomplete stop codons, and nad4l and nad6 have TAG as the stop codon. Cloverleaf secondary structures were inferred for 22 tRNA genes, but trnS1(AGN) was found to lack the DHU stem. The secondary structure of rrnL and rrnS is generally similar to other lepidopterans but with some minor differences. The A+T-rich region includes the motif ATAGA, but the poly (T) stretch is replaced by a stem-loop structure, which may have a similar function to the poly (T) stretch. Finally, there are three long repeat (154 bp) sequences followed by one short repeat (56 bp) with four (TA)(n) intervals, and a 10-bp poly-A is present upstream of trnM. Phylogenetic analysis shows that the position of Yponomeutoidea, as represented by L. malifoliella, is the same as traditional classifications. Yponomeutoidea is the sister to the other lepidopteran superfamilies covered in the present study.

Project description:We describe the complete mitochondrial genome of Phiaris dolosana. It is 15,562 bp in length, and contains 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), 2 ribosomal RNA genes (rRNAs), and a 612?bp D-Loop. All PCGs start with ATN codon except for COI gene, which uses CGA as the initiation codon. Nine of 13 PCGs use a typical stop codon of TAA, the rest use incomplete stop codon of T or TA. Phylogenetic analysis of P. dolosana with other 17 leaf rollers is conducted with neighbor-joining method, the result is consistent with the conventional classification.

Complete mitochondrial genome of Cnidocampa flavescens (Lepidoptera: Limacodidae).

Project description:Cnidocampa flavescens, lives in Nepal, Bhutan, China, Far East of Russia, Korea, and Japan, belongs to the Lepidoptera family Limacodidae. In this study, we describe the genomic features of the mitogenome sequences of the insects. The mitogenome of C. flavescens is 15,406?bp long consisting a typical set of genes (13 protein-coding genes, 22 tRNA genes, and 2 rRNA genes) and one major 415?bp non-coding A?+?T-rich region. All PCGs of C. flavescens start with ATN codons and end with TAA codons. The gene arrangement of C. flavescens mitogenome is same to Monema flavescens while the intergenic spacers and overlaps are different. The 415?bp A?+?T-rich region contains a conserved ATAGA motif followed a poly-T stretch.

We described the complete mitogenome of M. intermedia, which is typical circular duplex molecules and 15,386 bp in length, containing the standard metazoan set of 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and an A + T-rich region with macro-repeat sequences. All the inferred tRNA secondary structures show the common cloverleaf pattern, with the exception of trnS1(AGN) which lacks the DHU arm. The M. intermedia mitochondrial genome has the same gene order with other lepidopterans.

Complete mitochondrial genome of Casmara patrona (Lepidoptera: Oecophoridae).

Project description:The complete mitochondrial genome of Casmara patrona (Lepidoptera: Oecophoridae) was sequenced for a future phylogenetic study of Lepidoptera. The circle genome of the moth is 15,393 bp in length with a pronounced base bias of A + T (79.3%), containing 13 protein-coding genes, 22 transfer RNAs, two ribosomal RNAs, and a putative control region. The coxI gene had a CGA start codon as most lepidopteran species, other PCGs use the typical ATN codons. All PCGs end with the complete stop codon TAA. Phylogenetic analyses showed that the monophyly of Oecophoridae was highly supported based on the concatenated sequence of the 13 PCGs. In addition, Oecophoridae and Xyloryctidae had the closest relationship.

Complete mitochondrial genome of Acanthopsyche nigraplaga (Lepidoptera: Psychidae).

Project description:We report the mitochondrial genome (mitogenome) of a case-making moth Acanthopsyche nigraplaga Wileman, 1911 (Lepidoptera: Psychidae). The 15,704 bp long complete mitogenome comprises a typical set of genes [13 protein-coding genes (PCGs), 2 rRNA genes, and 22 tRNA genes] and one major non-coding, A + T-rich region, with an arrangement identical to that observed in most lepidopteran mitogenomes. Twelve of the 13 PCGs of the A. nigraplaga mitogenome initiate with a typical ATN start codon, however COI contains the atypical CGA start codon that is common for lepidopteran COI genes. A phylogenetic analysis using concatenated nucleotide sequences of the 13 PCGs and 2 rRNA genes using the Bayesian inference method fully resolved A. nigraplaga in a monophyletic clade within the Psychidae. Acanthopsyche nigraplaga was situated in a sister position to Eumeta variegata and Mahasena oolona with high nodal support. As more mitogenome sequences are available further scrutinized analysis for the superfamily Tineoidea including Psychidae will be possible.

Complete mitochondrial genome of Parnassius glacialis (Lepidoptera: Papilionidae).

Project description:Parnassius glacialis is a butterfly species distributed in China, Korea, Japan. The complete P. glacialis mitochondrial genome was assembled using Illumina sequencing data. The mitogenome is 15,353 bp long and contains 13 protein-coding genes, 22 transfer RNA genes, and 2 ribosomal RNA genes. A phylogenetic analysis of P. glacialis and 14 related Papilionidae species indicated that P. glacialis is clustered with other Parnassius species. This study generated useful genetic information for future studies on the taxonomy, phylogeny, and evolution of Papilionidae species.

Complete mitochondrial genome of Scythris sinensis (Lepidoptera: Scythrididae).

Project description:The complete mitochondrial genome (mitogenome) of Scythris sinensis Felder & Rogenhofer, 1775 (Lepidoptera: Scythrididae) was 15,216 bp with a typical set of genes (13 protein-coding genes [PCGs], 2 rRNA genes, and 22 tRNA genes) and one non-coding region, with an arrangement identical to that observed in most lepidopteran genomes. Twelve PCGs had the typical ATN start codon, whereas COI had the atypical CGA codon that is frequently found in the start region of the lepidopteran COI. The 271-bp long A + T-rich region was the shortest among sequenced Gelechioidea, which ranged from 290 - 375 bp. Phylogenetic analyses with concatenated sequences of the 13 PCGs, two RNA genes, and 22 tRNA genes using the Bayesian inference (BI) method, placed S. sinensis in the Scythrididae, as a sister to the family Stathmopodidae. The nodal support for this sister relationship was the highest at Bayesian posterior probabilities = 1.
