Project description:We assembled one mitochondrial genome of Pseudachorutes palmiensis from Illumina sequencing data. The circularized mitochondrial assembly is 17,110 bp in length, including 13 protein-coding genes, 22 transfer RNA genes, and two ribosomal RNA genes, which showed the typical insect mitochondrial gene composition, but had different order with most springtails. The overall base composition is 33.5% for A, 31.7% for T, 22.2% for C, and 12.5% for G. A phylogeny of 10 collembolans showed P. palmiensi was clustered within Neanuridae.

Project description:BACKGROUND: Mitogenomics data, i.e. complete mitochondrial genome sequences, are popular molecular markers used for phylogenetic, phylogeographic and ecological studies in different animal lineages. Their comparative analysis has been used to shed light on the evolutionary history of given taxa and on the molecular processes that regulate the evolution of the mitochondrial genome. A considerable literature is available in the fields of invertebrate biochemical and ecophysiological adaptation to extreme environmental conditions, exemplified by those of the Antarctic. Nevertheless, limited molecular data are available from terrestrial Antarctic species, and this study represents the first attempt towards the description of a mitochondrial genome from one of the most widespread and common collembolan species of Antarctica. RESULTS: In this study we describe the mitochondrial genome of the Antarctic collembolan Cryptopygus antarcticus Willem, 1901. The genome contains the standard set of 37 genes usually present in animal mtDNAs and a large non-coding fragment putatively corresponding to the region (A+T-rich) responsible for the control of replication and transcription. All genes are arranged in the gene order typical of Pancrustacea. Three additional short non-coding regions are present at gene junctions. Two of these are located in positions of abrupt shift of the coding polarity of genes oriented on opposite strands suggesting a role in the attenuation of the polycistronic mRNA transcription(s). In addition, remnants of an additional copy of trnL(uag) are present between trnS(uga) and nad1. Nucleotide composition is biased towards a high A% and T% (A+T = 70.9%), as typically found in hexapod mtDNAs. There is also a significant strand asymmetry, with the J-strand being more abundant in A and C. Within the A+T-rich region, some short sequence fragments appear to be similar (in position and primary sequence) to those involved in the origin of the N-strand replication of the Drosophila mtDNA. CONCLUSION: The mitochondrial genome of C. antarcticus shares several features with other pancrustacean genomes, although the presence of unusual non-coding regions is also suggestive of molecular rearrangements that probably occurred before the differentiation of major collembolan families. Closer examination of gene boundaries also confirms previous observations on the presence of unusual start and stop codons, and suggests a role for tRNA secondary structures as potential cleavage signals involved in the maturation of the primary transcript. Sequences potentially involved in the regulation of replication/transcription are present both in the A+T-rich region and in other areas of the genome. Their position is similar to that observed in a limited number of insect species, suggesting unique replication/transcription mechanisms for basal and derived hexapod lineages. This initial description and characterization of the mitochondrial genome of C. antarcticus will constitute the essential foundation prerequisite for investigations of the evolutionary history of one of the most speciose collembolan genera present in Antarctica and other localities of the Southern Hemisphere.

Project description:The paper is devoted to a taxonomic revision of the genus Sensillonychiurus Pomorski & Sveenkova, 2006. Five new species of this genus, i.e. Sensillonychiurus mirussp. n., Sensillonychiurus taimyrensissp. n., Sensillonychiurus vegaesp. n., Sensillonychiurus vitimicussp. n., and Sensillonychiurus amuricussp. n., as well as three new species of the related genus Allonychiurus Yoshii, 1995, i.e. Allonychiurus subvolinensissp. n., Allonychiurus elikoniussp. n., and Allonychiurus unisetosussp. n. are being described from various regions of Eurasia. The diagnoses of both genera are amended to include described species. Two genera, Tantulonychiurus Pomorski, 1996 and Thibaudichiurus Weiner, 1996, are treated as junior synonyms of the genus Allonychiurus. Agraphorura eisi (Rusek, 1976) is transferred to Sensillonychiurus; Tantulonychiurus volinensis (Szeptycki, 1964) and Tantulonychiurus asiaticus Babenko, 2007 to Allonychiurus. A review of morphological peculiarities of Sensillonychiurus is performed, comparisons with the other genera of Thalassaphorurini given, and a key to the known species provided.

Project description:A checklist of Chinese Oligaphorurini is given. Two new Chinese species, Micraphorura changbaiensissp. n. and Oligaphorura pseudomontanasp. n., are described from Changbai Mountain Range. Micraphorura changbaiensis sp. n. has the same dorsal pseudocelli formula and number of papillae in Ant. III sensory organ as Micraphorura uralica, but they can be easily distinguished by number of chaetae in Ant. III sensory organ, ventral pseudocelli formula, ventral parapseudocelli formula, number of pseudocelli on subcoxa 1 of legs I-III, dorsal axial chaeta on Abd. V and number of chaetae on tibiotarsi. Oligaphorura pseudomontana sp. n. is very similar to the species Oligaphorura montana having an increased number of pseudocelli on body dorsally, well marked base of antenna with 1 pseudocellus and 3 pseudocelli outside, subcoxa 1 of legs I-III with 1 pseudocellus each, dorsally S-chaetae formula as 11/011/22211 from head to Abd. V, S-microchaeta present on Th. II-III, claw without inner teeth and with 1+1 lateral teeth, and unguiculus with basal lamella; but they can be separated easily by the number of pseudocelli on Abd. V and VI terga, parapseudocelli on the body, number of chaetae on Th. I tergum, and number of chaetae on tibiotarsi. A key to Chinese species of Oligaphorurini is provided in the present paper.

Project description:A checklist of onychiurid species from the Wanda Mountains in China is presented. Eighteen species belonging to twelve genera have been found, including two new species. Bionychiurus qinglongensis sp. n. can be easily distinguished from other known species of the genus by the absence of pseudocelli on Th. I tergum and fewer number of vesicles in postantennal organ. Onychiurus heilongjiangensis sp. n. is diagnosed by pseudocellar formulae as 32/133/33352 dorsally and 3/011/31120 ventrally, parapseudocellar formula as 0/000/111001+1(m), ratio of anal spine/unguis as 0.6, unguiculus without inner basal lamella, and male ventral organ absent.

Project description:Species delimitation remains a significant challenge when the diagnostic morphological characters are limited. Integrative taxonomy was applied to the genus Protaphorura (Collembola: Onychiuridae), which is one of most difficult soil animals to distinguish taxonomically. Three delimitation approaches (morphology, molecular markers and geography) were applied providing rigorous species validation criteria with an acceptably low error rate. Multiple molecular approaches, including distance- and evolutionary model-based methods, were used to determine species boundaries based on 144 standard barcode sequences. Twenty-two molecular putative species were consistently recovered across molecular and geographical analyses. Geographic criteria were was proved to be an efficient delimitation method for onychiurids. Further morphological examination, based on the combination of the number of pseudocelli, parapseudocelli and ventral mesothoracic chaetae, confirmed 18 taxa of 22 molecular units, with six of them described as new species. These characters were found to be of high taxonomical value. This study highlights the potential benefits of integrative taxonomy, particularly simultaneous use of molecular/geographical tools, as a powerful way of ascertaining the true diversity of the Onychiuridae. Our study also highlights that discovering new morphological characters remains central to achieving a full understanding of collembolan taxonomy.

Project description:A new collembolan species is described, Spinonychiurus sinensis sp. n., which has seven chaetae in the distal row of the tibiotarsi. It is placed in the genus Spinonychiurus due to two important characters: the two subsegments on Abd. III sternum and the absence of d0 on the head. This is the first report of the genus Spinonychiurus in China. The diagnosis of Spinonychiurus is broadened and the key to the world species is provided.
