The complete mitochondrial genome of Appias albina (Lepidoptera: Pieridae) and phylogenetic analysis.

ABSTRACT: The complete mitochondrial genome of Appias albina (Lepidoptera: Pieridae) was firstly sequenced and characterized in our study. The total length of mitogenome is 15,193 bp and contains 13 protein-coding genes, 2 ribosomal RNA genes, 22 transfer RNA genes, and 1 A + T-rich region. The overall nucleotide composition was 38.4% of A, 8.0% of G, 40.9% of T, and 12.4% of C. Phylogenetic tree was reconstructed using Bayesian Inference (BI) to validate the taxonomic status of A. albina, exhibiting the close relationship with Appias remedios.

Project description:The complete mitochondrial genome of Pontia edusa was sequenced and analyzed in the study. The length of the complete mitogenome is 15,125 bp, including 37 genes and a control region. Twelve genes start with typical ATN, but COI gene initiate with a CGA codon. Twelve of 13 PCGs have a complete stop codon TAA or TAG except for COI has an incomplete stop codon T--. Twenty-one of the 22 tRNAs have a typical clover-leaf secondary structure. The tRNASer(AGN) gene lacked the DHU loop. Bayesian analyses highly support the monophyly of Pieridae. In Pieridae, P. edusa is subordinate to the Pierinae clade.

Phylogenetic relationship and characterization of the complete mitochondrial genome of Colias fieldii (Lepidoptera: Papilionoidea: Pieridae).

Project description:Colias fieldii is a common seen diurnal butterflies in the fields and widely distributed in many provinces of China. In this study, we sequenced and analyzed the complete mitochondrial genome (mitogenome) of C. fieldii. This mitogenome was 15,150 bp long and encoded 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and two ribosomal RNA unit genes (rRNAs). The overall base composition of the mitogenome was estimated to be A 39.8%, T 41.2%, C 11.4% and G 7.6%, with a high A + T content of 81.0%. Except for cox1 started with CGA, all other PCGs started with the standard ATN codons (seven ATG, four ATT and one ATC). Most of the PCGs terminated with the stop codon TAA or TAG, whereas cox1, cox2, nad5 and nad4 end with the incomplete codon T--. Phylogenetic analysis showed that C. fieldii is indeed the sister species of Colias erate with a high support value. All seven Coliadinae species formed one clade and was sister to Pierinae butterflies. Within Coliadinae, the relationships (Eurema + (Gonepteryx + (Catopsilia + Colias))) were highly supported.

The complete mitochondrial genome of Aporia crataegi L. (Lepidoptera: Pieridae).

Project description:The complete mitochondrial genome of Aporia crataegi is 15,147 bp long, and consists of 13 protein-coding genes (PCGs), 2 ribosomal RNA genes, 22 transfer RNA (tRNA) genes, and a putative control region (GenBank accession No. MN371463). The nucleotide composition is significantly biased (A, G, C, and T is 39.66%, 7.30%, 11.41%, and 41.63%, respectively) with A + T contents of 81.29%. All PCGs are initiated by ATG, ATT, and ATC codons. Seven PCGs use a common stop codon of TAA, whereas the remaining six terminated with a single T. The phylogenetic relationships based on maximum-likelihood phylogenetic tree method showed that A. crataegi is closely related to Aporia bieti, Mesapia peloria, and Aporia martineti.

Complete mitochondrial genome of Kentrochrysalis streckeri (Lepidoptera: Sphingidae) and phylogenetic analysis.

Project description:In this study, we sequenced the complete mitogenome of Kentrochrysalis streckeri (Staudinger, 1880). The complete mitogenome sequence of K. streckeri is circular, 15,253 bp in size and contains 13 protein-coding genes (PCGs), two ribosomal RNA (rRNA) genes, 22 transfer RNA (tRNA) genes, and a control region (CR). Nucleotide composition was A + T biased, and all the PCGs exhibited a positive AT-skew, which was reflected in the nucleotide composition, codon, and amino acid usage. Most PCGs start with ATG or ATT and stop with TAA. However, COX1 gene starts with CGA and three genes (COX1, COX2, NAD5) use the incomplete stop codon T. Phylogenetic analyses showed that the relationship (K. streckeri+((Manduca sexta+Sphinx morio)+(Psilogramma increta+(Psilogramma menephron+Notonagemia analis scribae)))).

Complete mitochondrial genome of the little-known regional endemic Aporia hastata (Oberthur, 1892) (Lepidoptera: Pieridae).

Project description:The present study reports the mitochondrial genome of A. hastata (Oberthür, 1892), a little-known Aporia species endemic to the southern margin of the Hengduan Mountains in Yunnan Province. This genome is circular, 15,148 bp in length, and consists of 13 PCGs, 22 tRNAs, and two rRNAs. The Bayesian phylogenetic tree clusters A. hastata with other Aporia taxa inside tribe Pierini Duponchel, [1835]. The findings of this study add valuable new information to the genus Aporia and are beneficial to a better understanding of phylogeography of these butterflies.

The complete mitochondrial genome of Dudusa sphingiformis (Lepidoptera: Notodontidae) and phylogenetic analysis.

Project description:Dudusa sphingiformis is an important lepidopteran pest widely distributed in tropical and subtropical zones of Asia. In this paper, the complete mitochondrial genome (mitogenome) of D. sphingiformis was determined by next-generation sequencing. The mitogenome was 15,806 bp in length, comprising 13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes, and an AT-rich control region (D-loop). The gene arrangement of this mitogenome was identical to that of the previous studies of Notodontidae moths. Almost all the PCGs initiated with typical ATN codons, except for cox1 with CGA. Among them, nine PCGs terminated with TAA or TAG, while other four PCGs (cox1, cox2, nad5, and nad4) with incomplete stop codon T. All the 22 tRNAs had the typical cloverleaf structure, except for trnS1, whose dihydrouridine (DHU) arm forms a simple loop. Phylogenetic analysis based on the concatenated nucleotide sequences of 13 PCGs indicated that D. sphingiformis was more closely related to other species of family Notodontidae, forming a monophyletic group, with well-resolved relationships among five family of Noctuoidea.

The Complete Mitochondrial Genome of Brachmia macroscopa (Lepidoptera: Gelechiidae) and Its Related Phylogenetic Analysis.

Project description:The sweet potato leaf folder, Brachmia macroscopa, is an important pest in China. The complete mitogenome, which consists of 13 protein-coding genes (PCGs), 22 transfer RNA genes, two ribosomal RNA genes, and an A + T-rich region, was sequenced and found to be 15,394 bp in length (GeneBank no. KT354968). The gene order and orientation of the B. macroscopa mitogenome were similar to those of other sequenced lepidopteran species. All of the PCGs started with ATN as the canonical start codon except for cox1, which started with CGA. In regard to stop codons, most PCGs stopped at TAA except for cox2, which stopped at TA, and nad4, which stopped at a single T. Thirteen PCGs of the available species (33 taxa) were used to demonstrate phylogenetic relationships. The ditrysian cluster was supported as a monophyletic clade at high levels by using maximum likelihood and Bayesian methods. The apoditrysian group, covering the Gelechioidea, formed a monophyletic clade with a bootstrap value of 88% and a posterior probability of 1.00. The superfamily Gelechioidea was supported as a monophyletic lineage by a posterior probability of 1.00.

Characterization and Phylogenetic Analysis of the Complete Mitochondrial Genome of Laelia suffusa (Lepidoptera: Erebidae, Lymantriinae).

Project description:In this study, the complete mitochondrial genome of a white tussock moth, Laelia suffusa (Walker, 1855) (Lepidoptera: Erebidae, Lymantriinae), was sequenced and annotated. The genome sequence was 15,502 bp in length and comprised 13 PCGs, 2 rRNAs, 22 tRNAs, and a single noncoding control region (CR). The nucleotide composition of the genome was highly A + T biased, accounting for 79.04% of the whole genome and with a slightly positive AT skewness (0.015). Comparing the gene order with the basal species of Lepidoptera, a typical trnM rearrangement was detected in the mitogenome of L. suffusa. Besides, the trnM rearrangement was found at the head of trnI and trnQ, rather than at the back. The 13 PCGs used ATN as their start codons, except for the cox1 which used CGA. Out of the 22 tRNAs, only 1 tRNA (trnS1) failed to fold in a typical cloverleaf secondary structure. The conserved motif 'ATAGA + poly-T' was detected at the start of the control region which was similar to other Lepidoptera species. In total, 10 overlapping regions and 19 intergenic spacers were identified, ranging from 1 to 41 and 2 to 73 bp, respectively. Phylogenetic analysis showed that Lymantriinae was a monophyletic group with a high support value and L. suffusa was closely related to tribe Orgyiini (Erebidae, Lymantriinae). Moreover, the phylogenetic relationship of Noctuoidea (Lepidoptera) species was reconstructed using two datasets (13 PCGs and 37 genes) and these supported the topology of (Notodontidae + (Erebidae + (Nolidae + (Euteliidae + Noctuidae)))).

Determining the complete mitochondrial genome of Tethea albicostata (Lepidoptera: Drepanidae) and phylogenetic analysis.

Project description:Tethea albicostata is a widely distributed insect species in northern and central China. To date, few studies have been conducted on this species, with the exception of morphological taxonomy studies. Here, we report the complete mitochondrial genome of T. albicostata collected in China. The circular-mapping mitogenome is 15,308 bp in length, with an overall A + T content of 80.52%, encoding 2 ribosomal RNA genes, 22 transfer RNA genes, and 13 protein-coding genes. The gene arrangement and components of T. albicostata are identical to those of most other Lepidopteran insects. Phylogenetic analysis based on mitogenomes showed that T. albicostata is grouped with Drepana pallida, which belongs to the same family as Drepanidae. The family Drepanidae formed a separate branch from other families in the phylogenetic tree. This study determined the second mitochondrial genome of the Drepanidae species.

Complete mitochondrial genome of banana skipper Erionota torus Evans (Lepidoptera: Hesperiidae) and phylogenetic analysis.

Project description:Erionota torus (Evans, 1941) is a banana pest and is mainly distributed in Southeast Asia and the Pacific regions. The complete mitogenome of E. torus (GenBank accession number MW586888) is 15,987 bp in size, including 13 protein-coding genes, 22 transfer RNAs, 2 ribosomal RNAs genes, and a noncoding A + T-rich region. The A + T-rich region is located between 12S rRNA and tRNAMet . The base composition of the whole E. torus mitogenome is 39.68% for A, 7.30% for G, 41.55% for T, and 11.47% for C, with a high AT content of 81.23%. The phylogeny analysis indicated that E. torus had a close relationship with Notocrypta curvifascia. The present data could contribute to the further detailed phylogeographic analysis and provide a comprehensive control strategy for this banana pest.
