The complete mitochondrial genome of Neolissochilus benasi (Cypriniformes: Cyprinidae).

ABSTRACT: In this study, the complete mitochondrial genome of Neolissochilus benasi has been determined by polymerase chain reaction method for the first time. The overall base composition of N. benasi mitogenome is 31.8% for A, 27.4% for C, 15.9% for G and 25.0% for T. The percentage of G + C content is 41.3%. The mitogenome is a circular DNA molecule of 16 583 bp in length with a D-loop region and contains 22 transfer RNA (tRNA) genes, two ribosomal RNA (rRNA) genes and 13 protein-coding genes. The mitochondrial genome sequencing for N. benasi in this study provides important molecular data for further evolutionary analysis for Cyprinoidea.

Project description:In this paper, we first report the complete mitochondrial genome of Neolissochilus soroides. The main purpose of this study was to determine the mitochondrial genome and phylogenetic status of N. soroides. The length mitogenome was 16584 bp, containing 2 ribosomal RNA genes, 13 protein-coding genes, 22 transfer RNA genes, and 3 non-coding control regions. The genome showed a slight A + T bias (A + T = 56.47%). 12 genes (ND1, COX2, ATP6, ND4L, ND5, ND6, ND2, ATP8, ND3, ND4, Cytb, COX3) start with ATG codon, besides one gene (COX1) start with GTG codon. Six genes (ND1, COX1, ATP6, ND4L, ND5, ND6) end with a TAA codon, 3 genes (ND2, ATP8, ND3) end with a TAG codon, and four genes (COX2, ND4, Cytb, COX3) end with the TA or T codon. The phylogenetic analysis showed that N. soroides was closely related to N. hendersoni. The mitogenome could have important implications for phylogeny, population genetics, and conservation of the N. soroides.

Characterization of the complete mitochondrial genome of Neolissochilus hendersoni (Herre, 1940) (Cypriniformes: Cyprinidae).

Project description:The differentiation between Neolissochilus and Acrossocheilus species based only on morphology is ambiguous; however, phylogenetic analysis using their mitogenome sequences provides conclusive results. Here, the phylogenetic position of Neolissochilus hendersoni (Herre, 1940) was determined using its mitogenome data. Total DNA from N. hendersoni was sequenced using the Illumina NovaSeq6000 platform, and annotation of mitochondrial genes was performed using MITOS2. Phylogenetic trees were constructed using the complete mitogenomes of 16 fish species. The mitogenome of N. hendersoni was found to be 16584 bp long, containing two ribosomal RNA genes, 13 protein-coding genes, 22 transfer RNA genes, and three non-coding control regions. The genome showed a slight A + T bias (A + T = 56.46%). Most PCGs were found to be located on the L-strand. Results of the phylogenetic analysis showed that N. stracheyi is closely related to N. hendersoni. Our results will help to clarify the phylogenetic relationship between Neolissochilus and Acrossocheilus species.

The complete mitochondrial genome of Neolissochilus stracheyi (Osteichthyes:Cyprinidae).

Project description:Neolissochilus stracheyi Day 1871 is a rare specie of fish inhabit clear forest streams and rivers. In order to discuss the phylogenetic position of N. stracheyi, the mitochondrial genome was obtained by sequencing. The genome was 16,587 bp in length with an accession number OM203155. The AT contents were 56.59%. The location and composition of genes are consistent with published Cyprinids containing 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and 1 main non-coding regions. Sequence analysis showed that the mitochondrial genome of N. stracheyi has high sequence homology with other cyprinid fishes. Phylogenetic tree results showed that N. stracheyi is most closely related to Neolissochilus heterostomus. The mitochondrial sequence is of great significance for fish conservation, taxonomic status and resource exploitation.

Complete mitochondrial genome of Schizothorax davidi (Teleostei: Cypriniformes: Cyprinidae).

Project description:This study provides a mitochondrial complete genome of Schizothorax davidi. The complete genome is 16,576 bp in length with an A + T content of 54.9%, which contains 13 protein-coding genes, 22 tRNA genes, and 2 rRNA genes. The phylogenetic analysis indicated that S. davidi is closely related to Schizothorax lissolabiatus. These results contribute to explore the adaptation strategy of S. davidi on the conditions of Qinghai-Tibet Plateau.

Four nucleotide compositions and their relative proportions of the entire mitogenome was 27.14% C, 19.93% G, 29.58% A and 25.34% T. The complete mitochondrial genome of Leuciscus merzbacheri (Cypriniformes: Cyprinidae).

Project description:Leuciscus merzbacheri (Zugmayer, 1912) is a cyprinid fish endemic to China, with a distribution range limited to Xinjiang Province. As a landmark species in the Junggar Basin, L. merzbacheri is of considerable significance regarding our understanding of the adaptive evolution of salt and alkali tolerance. In this study, the complete mitochondrial sequence of L. merzbacheri was obtained for the first time by high-throughput sequencing. The circular mitogenome is 16,609 bp in length and contains the standard 37 genes, including 13 protein-coding, 22 transfer RNA, and 2 ribosomal RNA genes, which is similar to that of other fish. The mitogenome contents of A, T, C, and G were 27.9, 26.3, 27.1, and 18.7%, respectively. Phylogenetically, L. merzbacheri was located on a new branch near the base of the phylogenetic tree, thereby suggesting an early origin.

The complete mitochondrial genome of the Schizothorax curilabiatus (Cypriniformes: Cyprinidae).

Project description:The complete mitochondrial DNA sequence of Schizothorax curilabiatus was determined and analyzed. This mitochondrial genome was 16,578bp in length and showed significant AT bias (55.5% AT content, 44.5% GC content). Phylogenetic analysis showed that S. curilabiatus is more closely related to S. oconnori than to other species. The complete mitochondrial genome sequence provides an important dataset for the exploration of mitochondrial inheritance mechanism.

The complete mitochondrial genome of the Gymnocypris chui (Cypriniformes: Cyprinidae).

Project description:The Gymnocypris chui, a new recorded species in Lange Lake, was grouped into genus Gymnocypris in Schizothoracinae, and had the rare quantity and limited resources on biology and genetics, especially in the mitochondrion. In this study, the complete mitochondrial sequence of G. chui was assembled and phylogenetic relationships with other species in Cyprinidae were analyzed. The whole mitochondrial sequence was 16,864 bp in length, which contained two control regions (D-loop regions), two rRNA genes (12S and 16S rRNA), 13 protein-coding genes and 22 tRNA genes. The D-loop region was separated by tRNAPro . The 12S rRNA and 16S rRNA located between tRNAPhe and tRNALeu and were separated by tRNAVal . The 13 mRNAs had three start codons, five termination codons and four overlap regions. The 22 tRNA scattered among the whole mitochondrion, and they were range from 66 (tRNACys ) to 76 (tRNALys andtRNALeu ) in length. To further explore the phylogenetic relationship of the G. chui, we constructed the phylogenetic tree and verified that the G. chui was a part of genus Gymnocypris and had closer relationship with Gymnocypris dobula and was independent from other species of Schizothoracinae, Barbinae and Labeoninae in Cyprinidae. This study provided the valuable evidence on phylogenetic relationship of the G. chui at the molecular level and essential resource for further research on this species.
