Highly sensitive fluorometric method based on nitrogen-doped carbon dot clusters for tartrazine determination in cookies samples.

ABSTRACT: Nitrogen-doped carbon nanodots (CDs) were prepared via the solvothermal method, using urea and triethylene glycol as the starting materials. The as-prepared CDs had individual diameters of approximately 100 nm and were in clusters of different sizes. The surface composition and optical properties of the as-prepared CDs were characterized. They exhibited multicolor emission properties in the visible range when excited with a wide wavelength range. The aqueous solution of the CDs was used in highly sensitive tartrazine determination. The fluorescence quenching of the CDs was in a linear relationship with the concentrations of tartrazine in the range of 0.5-30.0 ?M. The detection limit of the assay was 0.18 ?M. Acceptable recovery results were obtained via spike-recovery experiments on cookie samples.

Copper Nanoparticle and Nitrogen Doped Graphite Oxide Based Biosensor for the Sensitive Determination of Glucose.

Project description:Copper nanoparticles with the diameter of 50 ± 20 nm decorated nitrogen doped graphite oxide (NGO) have been prepared through a simple single step carbonization method using copper metal-organic framework (MOF), [Cu₂(BDC)₂(DABCO)] (where BDC is 1,4-benzenedicarboxylate, and DABCO is 1,4-Diazabicyclo[2.2.2]octane) as precursor. The surface morphology, porosity, surface area and elemental composition of CuNPs/NGO were characterized by various techniques. The as-synthesized CuNPs/NGO nanomaterials were coated on commercially available disposable screen-printed carbon electrode for the sensitive determination of glucose. We find that the modified electrode can detect glucose between 1 μM and 1803 μM (linear range) with good sensitivity (2500 μA mM−1 cm−2). Our glucose sensor also possesses low limits of detection (0.44 μM) towards glucose determination. The highly selective nature of the fabricated electrode was clearly visible from the selectivity studies. The practicability of CuNPs/NGO modified electrode has been validated in the human serum samples. The storage stability along with better repeatability and reproducibility results additionally substantiate the superior electrocatalytic activity of our constructed sensor towards glucose.

Au-Nitrogen-Doped Graphene Quantum Dot Composites as "On-Off" Nanosensors for Sensitive Photo-Electrochemical Detection of Caffeic Acid.

Project description:Herein, gold-nitrogen-doped graphene quantum dot (Au/NGQD) composite modified electrodes were fabricated and applied as "on-off" nanosensors for the photo-electrochemical (PEC) detection of caffeic acid under visible-light irradiation. An effective and simple strategy was established for the preparation of Au/NGQD composites by hydrothermal and calcination methods. Owing to the quantum confinement effect of NGQDs, the local surface plasmon resonance (LSPR) effect of Au nanoparticles (NPs), and the synergistic effect between Au and NGQDs NPs, the Au/NGQDs showed excellent PEC performance, with wide linear concentration ranges (0.11 to 30.25 ?M and 30.25 to 280.25 ?M), a low detection limit (0.03 ?M), excellent sensitivity, and high stability. The present study may provide an advanced strategy for the simple design of Au/NGQD composites to allow their effective application for selective and sensitive sensing of small biological molecules.

Green Preparation of Fluorescent Nitrogen-Doped Carbon Quantum Dots for Sensitive Detection of Oxytetracycline in Environmental Samples.

Project description:Nitrogen-doped carbon quantum dots (N-CQDs) with strong fluorescence were prepared by a one-step hydrothermal method using natural biomass waste. Two efficient fluorescent probes were constructed for selective and sensitive detection of oxytetracycline (OTC). The synthesized N-CQDs were characterized by UV-visible absorption spectra, fluorescence spectra, Fourier transform infrared spectroscopy (FT-IR), X-ray photon spectroscopy (XPS), atomic force microscopy (AFM), and high-resolution transmission electron microscopy (HRTEM), which proved that the synthesized N-CQDs surface were functionalized and had stable fluorescence performance. The basis of N-CQDs detection of OTC was discussed, and various reaction conditions were studied. Under optimized conditions, orange peel carbon quantum dots (ON-CQDs) and watermelon peel carbon quantum dots (WN-CQDs) have a good linear relationship with OTC concentrations in the range of 2-100 µmol L-1 and 0.25-100 µmol L-1, respectively. ON-CQDs and WN-CQDs were both successfully applied in detecting the OTC in pretreated tap water, lake water, and soil, with the recovery rate at 91.724-103.206%, and the relative standard deviation was less than 5.35%. The results showed that the proposed N-CQDs proved to be green and simple, greatly reducing the detection time for OTC in the determination environment.

Application of sulfur and nitrogen doped carbon quantum dots as sensitive fluorescent nanosensors for the determination of saxagliptin and gliclazide.

Project description:In this study, highly fluorescent sulfur and nitrogen doped carbon quantum dots (S,N-CQDs) were used as fluorescent nanosensors for direct spectrofluorimetric estimation of each of gliclazide (GLZ) and saxagliptin (SXG) without any pre-derivatization steps for the first time. S,N-CQDs were synthesized employing a simple hydrothermal technique using citric acid and thiosemicarbazide. The produced S,N-CQDs were characterized using different techniques including fluorescence emission spectroscopy, UV spectrophotometry, high-resolution transmission electron microscopy and FT-IR spectroscopy. Following excitation at 360 nm, S,N-CQDs exhibited a strong emission peak at 430 nm. The native fluorescence of S,N-CQDs was quantitatively enhanced by addition of increased concentrations of the studied drugs. The fluorescence enhancement of S,N-CQDs and the concentrations of the studied drugs revealed a wide linear relationship in the range of 30.0-500.0 µM and 75.0-600.0 µM with limits of detection of 5.0 and 10.15 µM for GLZ and SXG, respectively. The proposed method was efficiently used for determination of cited drugs in their commercial tablets with % recoveries ranging from 98.6% to 101.2% and low % relative standard deviation values (less than 2%). The mechanism of interaction between S,N-CQDs and the two drugs was studied. Validation of the proposed method was carried out in accordance with International Conference on Harmonization (ICH) guidelines.

Nitrogen-Doped Carbon Dots Encapsulated a Polyoxomolybdate-Based Coordination Polymer as a Sensitive Platform for Trace Tetracycline Determination in Water.

Project description:The requirement of simple, efficient and accurate detection of tetracycline (TC) in water environments poses new challenges for sensing platform development. Here, we report a simple method for TC sensing via fluorescence detection based on metal-organic coordination polymers (MOCPs, (4-Hap)4(Mo8O26)) coated with nitrogen-doped carbon dots (NCDs). These NCDs@(4-Hap)4(Mo8O26) composites showed excellent luminescence features of NCDs with stable bright-blue emission under UV light. The results of the sensing experiment showed that the fluorescence of NCDs@(4-Hap)4(Mo8O26) can be quenched by TC (166 µM) with 94.1% quenching efficiency via the inner filter effect (IFE) in a short time (10 s), with a detection limit (LOD) of 33.9 nM in a linear range of 8-107 µM. More significantly, NCDs@(4-Hap)4(Mo8O26) showed a high selectivity for TC sensing in the presence of anions and metal cations commonly found in water environments and can be reused in at least six cycles after washing with alcohol. The potential practicality of NCDs@(4-Hap)4(Mo8O26) was verified by sensing TC in real water samples with the standard addition method, and satisfactory recoveries from 91.95% to 104.72% were obtained.

Spectrophotometric and Fluorometric Methods for the Determination of Fe(III) Ions in Water and Pharmaceutical Samples.

Project description:Chemical sensors based on mesoporous silica nanotubes (MSNTs) for the quick detection of Fe(III) ions have been developed. The nanotubes' surface was chemically modified with phenolic groups by reaction of the silanol from the silica nanotubes surface with 3-aminopropyltriethoxysilane followed by reaction with 3-formylsalicylic acid (3-fsa) or 5-formylsalicylic acid (5-fsa) to produce the novel nanosensors. The color of the resultant 3-fsa-MSNT and 5-fsa-MSNT sensors changes once meeting a very low concentration of Fe(III) ions. Color changes can be seen by the naked eye and tracked with a smartphone or fluorometric or spectrophotometric techniques. Many experimental studies have been conducted to find out the optimum conditions for colorimetric and fluorometric determining of the Fe(III) ions by the two novel sensors. The response time, for the two sensors, that is necessary to achieve a steady spectroscopic signal was less than 15 s. The suggested methods were validated in terms of the lowest limit of detection (LOD), the lowest limit of quantification (LOQ), linearity, and precision according to International Conference on Harmonization (ICH) guidelines. The lowest limit of detection that was obtained from the spectrophotometric technique was 18 ppb for Fe(III) ions. In addition, the results showed that the two sensors can be used eight times after recycling using 0.1 M EDTA as eluent with high efficiency (90%). As a result, the two sensors were successfully used to determine Fe(III) in a variety of real samples (tap water, river water, seawater, and pharmaceutical samples) with great sensitivity and selectivity.

Photocatalytic Degradation of Organic Dyes and Antimicrobial Activities by Polyaniline-Nitrogen-Doped Carbon Dot Nanocomposite.

Project description:Nitrogen-doped carbon nanodots (N@CDs) were prepared by hydrothermal processing of bovine serum albumin (Mw: 69,324 with 607 amino acids). A polyaniline (PANI-N@CDs) nanocomposite was then synthesized by ultrasonication and used to degrade Congo red (CR), methylene blue (MB), Rhodamine B (RhB), and crystal violet (CV) four common organic dyes. The PANI-N@CD nanocomposite simultaneously adsorbed and concentrated the dye from the bulk solution and degraded the adsorbed dye, resulting in a high rate of dye degradation. The combination of holes (h+), hydroxyl (OH•), and O2•- was involved in the N@CD-mediated photocatalytic degradation of the dyes. Under visible light illumination at neutral pH, the PANI-N@CDs were proven as an efficient adsorbent and photocatalyst for the complete degradation of CR within 20 min. MB and RhB were also degraded but required longer treatment times. These findings supported the design of remediation processes for such dyes and predicted their fate in the environment. The nanocomposite also exhibited antimicrobial activities against Gram-negative bacterium E. coli and Gram-positive bacterium S. aureus.

Sensitive Determination of Proteolytic Proteoforms in Limited Microscale Proteome Samples.

Project description:Protein N termini unambiguously identify truncated, alternatively translated or modified proteoforms with distinct functions and reveal perturbations in disease. Selective enrichment of N-terminal peptides is necessary to achieve proteome-wide coverage for unbiased identification of site-specific regulatory proteolytic processing and protease substrates. However, many proteolytic processes are strictly confined in time and space and therefore can only be analyzed in minute samples that provide insufficient starting material for current enrichment protocols. Here we present High-efficiency Undecanal-based N Termini EnRichment (HUNTER), a robust, sensitive and scalable method for the analysis of previously inaccessible microscale samples. HUNTER achieved identification of >1000 N termini from as little as 2 μg raw HeLa cell lysate. Broad applicability is demonstrated by the first N-terminome analysis of sorted human primary immune cells and enriched mitochondrial fractions from pediatric cancer patients, as well as protease substrate identification from individual Arabidopsis thaliana wild type and Vacuolar Processing Enzyme-deficient mutant seedlings. We further implemented the workflow on a liquid handling system and demonstrate the feasibility of clinical degradomics by automated processing of liquid biopsies from pediatric cancer patients.

A highly sensitive dual-read assay using nitrogen-doped carbon dots for the quantitation of uric acid in human serum and urine samples.

Project description:A simple dual-read assay for uric acid (UA) was developed based on a combined ratiometric fluorescent and colorimetric strategy using nitrogen-doped carbon dots (N-CDs). The biosensor relies on the oxidation of UA by uricase to produce H2O2, which was then converted to •OH radicals by I-, resulting in the oxidation of o-phenylenediamine (OPD) to 2,3-diaminophenazine (DAP). In the presence of UA, the colorless biosensor system changed to yellow. Furthermore, the presence of DAP quenched the fluorescence emission of the N-CDs at 427 nm based on the inner filter effect (IFE). With increasing UA concentrations, the fluorescence intensity of the biosensor at 427 nm decreased but increased at 580 nm, demonstrating the ratiometric response. A strong linearity was observed between the fluorescence intensity ratio of DAP to N-CDs (I580/I427) and the corresponding UA concentration over the range 0.5-150 μM, and a limit of detection (S/N ratio of 3) of 0.06 μM was calculated. The dual-read assay was successfully employed in the quantitation of UA in human serum and urine samples, revealing its potential for measuring UA in clinical samples.

An Electrochemical Sensor Based on Carbon Paper Modified with Graphite Powder for Sensitive Determination of Sunset Yellow and Tartrazine in Drinks.

Project description:The paper describes the development of an electrochemical sensor to be used for the determination of synthetic food colorants such as Sunset Yellow FCF (SY) and Tartrazine (TZ). The sensor is a carbon paper (CP) electrode, manufactured by using hot lamination technology and volume modified with fine-grained graphite powder (GrP). The sensor (GrP/CP) was characterized by scanning electron microscopy, energy dispersive spectrometry, electrochemical impedance analysis, cyclic, linear sweep and differential pulse voltammetry. The mechanism of SY and TZ electrochemical oxidation on GrP/CP was studied. The developed sensor has good electron transfer characteristics and low electron resistance, high sensitivity and selectivity. Applying the differential pulse mode, linear dynamic ranges of 0.005-1.0 μM and 0.02-7.5 μM with limits of detection of 0.78 nM and 8.2 nM for SY and TZ, respectively, were obtained. The sensor was used to detect SY and TZ in non-alcoholic and alcoholic drinks. The results obtained from drink analysis prove good reproducibility (RSD ≤ 0.072) and accuracy (recovery 96-104%).
