The complete chloroplast genome of endangered Zhangjiajie sage Salvia daiguii Y. K. Wei & Y. B. Huang (Lamiaceae).

ABSTRACT: The complete chloroplast genome of Zhangjiajie sage, Salvia daiguii, was assembled in this study. The genome is 151,434 bp in length and contained 134 encoded genes in total, including 88 protein-coding genes, eight ribosomal RNA genes, and 37 transfer RNA genes. The result of phylogenetic analysis based on 17 chloroplast genomes revealed that S. daiguii is clustered with Salvia miltiorrhiza in Lamiaceae.

The genome is 151,473 bp in length and contained 129 encoded genes in total, including 84 protein-coding genes, eight ribosomal RNA genes, and 37 transfer RNA genes. The result of phylogenetic analysis based on 15 chloroplast genomes revealed that S. yangii is closely related to common sage (Salvia officinalis) in Lamiaceae.

Characterization of the complete chloroplast genome of Salvia leucantha (Lamiaceae).

Project description:Salvia leucantha (Lamiaceae) is an important horticultural plant with great ornamental and economic value. Here, we report the complete chloroplast genome of this species. The chloroplast genome was determined to be 151021 bp and the GC contents was 38.0%. The sequence includes a large single copy (LSC) region of 82,262 bp, a small single copy (SSC) region of 17,537 bp, and two separated inverted regions of 25,611 bp each. It contains 130 unique genes, including 85 protein-coding genes, 37 tRNA genes, and 8 rRNA genes. Based on the chloroplast genomes data of 26 species in Salvia, our result indicated that S. leucantha, S. tiliifolia, S. hispanica, and S. splendens formed one clade with Bootstrap = 100%. The four species belong to Salvia subgenus Calosphace, and S. leucantha was closely related to Salvia tiliifolia and Salvia hispanica. This result will facilitate population, genetic diversity and phylogenetic studies of S. leucantha.

The complete chloroplast genome of Salvia liguliloba Y. Z. Sun (Lamiaceae).

Project description:Salvia liguliloba Y. Z. Sun is a plant species endemic to the Tianmu Mountains. In this study, we assembled the complete chloroplast genome of S. liguliloba. The chloroplast genome of S. liguliloba was 151,490 bp with quadripartite structure in length, which contained 124 encoded genes, including 79 protein-coding genes, eight ribosomal RNA genes, and 37 transfer RNA genes. Our phylogenetic analysis result based on 54 chloroplast genomes revealed that S. liguliloba was closely related to S. miltiorrhiza according to the current sampling extent in Lamiaceae.

The complete chloroplast (cp) genome of S. tiliifolia was 150,836 bp in length, which contained a large single-copy (LSC) region of 82,129 bp, a small single-copy (SSC) region of 17,533 bp, and a pair of inverted repeats (IRs) region of 25,587 bp each. The genome harbored 130 genes, including 85 protein coding genes, 37 tRNA genes, and 8 ribosomal RNA genes. The overall GC content was 37.99%. Phylogenetic analysis indicated that S. tiliifolia is closely related to the species of Salvia chanryoenica.

Project description:Salvia karwinskii Benth. 1835 is a perennial herb in the Lamiaceae family native in Mexico and Central America. The complete chloroplast (cp) genome of S. karwinskii was sequenced using the Illumina platform and assembled for the first time. The complete plastid genome of S. karwinskii was 150,907 bp in length including a large single-copy (LSC) region of 82,205 bp, a small single-copy (SSC) region of 17,538 bp, and a pair of inverted repeat (IR) regions of 25,582 bp. The total GC content of this genome was 38.05%, and that of LSC, SSC, and IR regions was 36.22%, 31.77%, and 43.14%, respectively. The cp genome contained 114 unique genes, including 80 protein-coding genes, 30 tRNA genes, and four rRNA genes. The maximum-likelihood phylogenetic tree was constructed with 38 complete cp genomes, supporting a close relationship between S. karwinskii and a 10 species lineage, all of which belong to the subg. Calosphace of Salvia. The cp genome of S. karwinskii provides a foundation for further studies on genetic diversity and improving the classification system of Salvia.

The total GC content of this genome was 38.04%, and that of LSC, SSC and IR regions was 36.21%, 31.80% and 43.13%, respectively. A total of 114 unique genes of this genome have been annotated, including 80 protein-coding genes, 30 transfer RNA genes, and four ribosomal RNA genes. The maximum likelihood phylogenetic tree was constructed with 51 complete chloroplast genomes, illustrating the close relationship of S. oxyphora to the Brazil’s native medicinal species S. splendens. The chloroplast genome of S. oxyphora provides a foundation for further studies on the adaptive evolution and genetic diversity of the genus Salvia.

Project description:Salvia yunnanensis is a medicinal plant commonly used in the southwest of China. In this study, we sequenced the complete chloroplast (cp) genome sequence of S. yunnanensis to investigate its phylogenetic relationship in the family Lamiaceae. The total length of the cp genome was 151,338 bp, with 38.0% overall GC content and exhibited typical quadripartite structure, a pair of IRs (inverted repeats) of 25,578 bp each were separated by a small single-copy (SSC) region of 17,564 bp and a large single-copy (LSC) region of 82,618 bp. The cp genome contained 114 genes, including 80 protein coding genes, 30 tRNA genes, and 4 rRNA genes. The phylogenetic analysis indicated S. yunnanensis was closely related to S. miltiorrhiza, which afforded a scientific evidence that S. yunnanensis could be used as original species of Radix et Rhizoma Saliviae Miltiorrhizae (Danshen).

Extracellular localization of the diterpene sclareol in clary sage (Salvia sclarea L., Lamiaceae).

Project description:Sclareol is a high-value natural product obtained by solid/liquid extraction of clary sage (Salvia sclarea L.) inflorescences. Because processes of excretion and accumulation of this labdane diterpene are unknown, the aim of this work was to gain knowledge on its sites of accumulation in planta. Samples were collected in natura or during different steps of the industrial process of extraction (steam distillation and solid/liquid extraction). Samples were then analysed with a combination of complementary analytical techniques (gas chromatography coupled to a mass spectrometer, polarized light microscopy, environmental scanning electron microscopy, two-photon fluorescence microscopy, second harmonic generation microscopy). According to the literature, it is hypothesized that sclareol is localized in oil pockets of secretory trichomes. This study demonstrates that this is not the case and that sclareol accumulates in a crystalline epicuticular form, mostly on calyces.
