Transcriptome-wide identification of WRKY family genes and their expression profiling toward salicylic acid in Camellia japonica.

ABSTRACT: The ornamental plant Camellia japonica is widely distributed worldwide and is susceptible to various environmental stresses. The WRKY transcription factor (TF) is an important node of plant tolerance. However, WRKY TFs from C. japonica have not been reported yet. In this study, 48 CjWRKYs, namely, CjWRKY1 to CjWRKY48, were identified. Protein structure analysis revealed that CjWRKY proteins contain a highly conserved motif (WRKYGQK) and two variant motifs (WRKYGKK and WRKYGRK). Phylogenetic analysis indicated that the 48 CjWRKYs can be divided into three groups, which are further classified into six subgroups, namely, I-C, II-a, II-b, II-c, II-e, and III, which contain 10, 6, 8, 13, 7, and 4 members, respectively. The expression patterns of 15 CjWRKYs under salicylic acid (SA) treatment were investigated by real-time quantitative PCR (qRT-PCR). Results showed that the 15 CjWRKYs could be induced by SA treatment. This study is the first to screen CjWRKYs and identify the expression profile of CjWRKYs under SA treatment and provides a theoretical basis for analyzing the function of CjWRKY genes to SA stress tolerance in C. japonica.

Project description:Camellia oleifera Abel. is an economically important woody edible-oil species that is mainly cultivated in hilly areas of South China. The phosphorus (P) deficiency in the acidic soils poses severe challenges for the growth and productivity of C. oleifera. WRKY transcription factors (TFs) have been proven to play important roles in biological processes and plant responses to various biotic/abiotic stresses, including P deficiency tolerance. In this study, 89 WRKY proteins with conserved domain were identified from the C. oleifera diploid genome and divided into three groups, with group II further classified into five subgroups based on the phylogenetic relationships. WRKY variants and mutations were detected in the gene structure and conserved motifs of CoWRKYs. Segmental duplication events were considered as the primary driver in the expanding process of WRKY gene family in C. oleifera. Based on transcriptomic analysis of two C. oleifera varieties characterized with different P deficiency tolerances, 32 CoWRKY genes exhibited divergent expression patterns in response to P deficiency stress. qRT-PCR analysis demonstrated that CoWRKY11, -14, -20, -29 and -56 had higher positive impact on P-efficient CL40 variety compared with P-inefficient CL3 variety. Similar expression trends of these CoWRKY genes were further observed under P deficiency with longer treatment period of 120d. The result indicated the expression sensitivity of CoWRKYs on the P-efficient variety and the C. oleifera cultivar specificity on the P deficiency tolerance. Tissue expression difference showed CoWRKYs may play a crucial role in the transportation and recycling P in leaves by affecting diverse metabolic pathways. The available evidences in the study conclusively shed light on the evolution of the CoWRKY genes in C. oleifera genome and provided a valuable resource for further investigation of functional characterization of WRKY genes involved to enhance the P deficiency tolerance in C. oleifera.

Project description:BackgroundCucumber (Cucumis sativus L.) is an economically important vegetable crop species. However, it is susceptible to various abiotic and biotic stresses. WRKY transcription factors play important roles in plant growth and development, particularly in the plant response to biotic and abiotic stresses. However, little is known about the expression pattern of WRKY genes under different stresses in cucumber.ResultsIn the present study, an analysis of the new assembly of the cucumber genome (v3.0) allowed the identification of 61 cucumber WRKY genes. Phylogenetic and synteny analyses were performed using related species to investigate the evolution of the cucumber WRKY genes. The 61 CsWRKYs were classified into three main groups, within which the gene structure and motif compositions were conserved. Tissue expression profiles of the WRKY genes demonstrated that 24 CsWRKY genes showed constitutive expression (FPKM > 1 in all samples), and some WRKY genes showed organ-specific expression, suggesting that these WRKYs might be important for plant growth and organ development in cucumber. Importantly, analysis of the CsWRKY gene expression patterns revealed that five CsWRKY genes strongly responded to both salt and heat stresses, 12 genes were observed to be expressed in response to infection from downy mildew and powdery mildew, and three CsWRKY genes simultaneously responded to all treatments analysed. Some CsWRKY genes were observed to be induced/repressed at different times after abiotic or biotic stress treatment, demonstrating that cucumber WRKY genes might play different roles during different stress responses and that their expression patterns vary in response to stresses.ConclusionsSixty-one WRKY genes were identified in cucumber, and insight into their classification, evolution, and expression patterns was gained in this study. Responses to different abiotic and biotic stresses in cucumber were also investigated. Our results provide a better understanding of the function of CsWRKY genes in improving abiotic and biotic stress resistance in cucumber.

Project description:Camellia (Camellia japonica) is a semi-shaded plant that is highly vulnerable to heat stress. To investigate the mechanisms underlying heat stress in C. japonica, two C. japonica cultivars, "Xiaotaohong" and "Zhuapolian", which exhibit significant differences in heat tolerance, were selected from four common cultivars. The selection methods included phenotypic observations and physiological index detection, including relative electric conductivity (REC), malondialdehyde (MDA) content, superoxide dismutase (SOD) enzyme activity, relative water content (RWC), and chlorophyll content. RNA-seq analysis yielded 980 million reads and identified 68,455 differentially expressed genes (DEGs) in the two C. japonica cultivars during heat stress compared to the control samples. Totals of 12,565 and 16,046 DEGs were differentially expressed at 16 h and 32 h, respectively, in "Xiaotaohong" during heat stress. In "Zhuapolian", 40,280 and 37,539 DEGs were found at 16 h and 32 h, respectively. KEGG enrichment analysis revealed that both cultivars were enriched in the "plant hormone signal transduction" and "circadian rhythm" pathways at two stages, indicating the critical role these pathways play in the heat stress response. The differences in the tolerance between the two cultivars are likely linked to pathways such as "plant hormone signal transduction", "photosynthesis", and "circadian rhythm". Some members of heat shock proteins (HSPs) are associated with the heat stress response. It is speculated that transcription factor families contributing to the tolerance differences include AP2/ERF, C3H, bHLH, bZIP, and MYB-related with a small number of heat shock factors (HSFs) also induced by the stress. In conclusion, these results reveal the changes in the physiological indices and molecular networks of two C. japonica cultivars under heat stress. This study lays the foundation for the breeding of superior heat-resistant C. japonica cultivars and for further molecular research.
