The complete mitochondrial genome sequence of Lactarius trivialis (Russulalles, Basidiomycota).

ABSTRACT: Lactarius trivialis is a very common species and widely distributes in north temperate. In this study, a complete mitogenome of L. trivialis was assembled and annotated. The whole mitogenome of L. trivialis was a circular molecule with 42,366 bp in length, encoding 44 genes as follows: 19 coding genes, two rRNAs, and 23 tRNAs. The contents of four bases in mtDNA were A (39.65%), T (38.82%), C (11.30%), and G (21.53%), respectively. Phylogenetic analysis recovered that it is nested with other Lactarius spp. in the order Russulalles.

Project description:In the present study, the complete mitochondrial DNA sequence of the milkfish Lactarius lactarius is determined. The full-length of the mitochondrial genome consists of a 16,552 bp fragment, with the base composition of A (28.24%), C (29.82%), G (15.96%), and T (25.98%) with a high A + T content (54.22%). The base compositions present clearly the A-T skew, which is most obvious in the control region and protein-coding genes. It includes 2 ribosomal RNA (rRNA) genes, 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, and a major non-coding control region (D-loop region). Furthermore, the composition and order of these genes are identical to most of other vertebrates. All the protein initiation codons are ATG, except that COX1 and ATP6 begin with GTG. All the protein termination codons are TAA, except ND3 and ND6 end with TAG, and COXII, ND4, Cytb finish only with incomplete T. Furthermore, the phylogenetic analysis base on 13 concatenated PCGs amino acid datasets among the 14 species, suggesting that high value support for the following sister clade with Toxotes chatareus. Our findings provide useful information for phylogenetic and evolutionary research of Perciformes species.

The complete mitochondrial genome sequence of the medicinal fungus Inonotus obliquus (Hymenochaetaceae, Basidiomycota).

Project description:Inonotus obliquus is a medicinal fungus in the family Hymenochaetaceae and commonly known as chaga. The sclerotium of this fungus has been used as a traditional medicine for long time. In this study, we present the mitochondrial genome sequence of I. obliquus. The mitochondrial DNA (mtDNA) is 119,110 base pairs in length and contained genes for 58 Open reading frames, 2 ribosomal RNAs, and 30 transfer RNAs. Consequently performed phylogenetic analysis indicates that this fungus is closely related to Sanghuangporus sanghuang which belongs to the same family Hymenochaetaceae. We first reported about the complete mitochondrial genome of fungi belonging to the genus Inonotus.

Project description:Sanghuang is a polypore mushroom, which has been widely used in oriental medicine. Since recent molecular phylogenetic studies elucidated its species delimitation, Sanghaungporus sanghuang became the official name of this fungus. In this study, the complete sequence of the mitochondrial DNA of S. sanghuang was determined. The whole genome was 112,060?bp containing 14 proteins, 2 ribosomal RNA subunits, and 45 transfer RNAs. The overall GC content of the genome was 23.21%. A neighbour-joining tree based on atp6 sequence data showed its close relationship with the species of Ganoderma and Trametes.

The complete mitochondrial genome of Hericium coralloides (Hericiaceae, Basidiomycota).

Project description:In this study, the complete mitochondrial genome of the medicinal mushroom Hericium coralloides (Hericiaceae, Basidiomycota) was sequenced. This mitochondrial genome is 72,961 bp in length and consisted of 14 protein-coding genes, 21 hypothetical open reading frames, 2 ribosomal RNA subunits and 27 transfer RNAs. The overall nucleotide composition of is 41.33% A, 40.71% T, 9.06% C and 8.90% G, with GC content of 17.96%. A phylogenetic tree with the complete mitochondrial genome sequences of Hericium coralloides together with 9 other affinis mushrooms was constructed. The newly achieved mitochondrial genome sequence seem to be useful for addressing taxonomic issues and studying related evolution events, which would contribute to enrich the fungal mitochondrial genome resource and promote the biological research.

Project description:In this study, the complete mitochondrial genome of Sanghuangporus vaninii Zhehuang-1 was determined. Genomic DNA samples individually collected from a population in southeastern China (Qiandaohu, Zhejiang Province, N29°44'24'', E118°52'48'') were sequenced with an Illumina NovaSeq 6000. The complete mitochondrial genome was 97,345 bp in length and consisted of 22 protein-coding genes (PCGs), 29 tRNAs, 3 ribosomal RNAs, and a control region. Sanghuangporus vaninii has a mitochondrial gene arrangement that is similar to that of S. sanghuang. Phylogenetic analysis performed using ML methods based on the complete mitogenome sequence showed that S. vaninii is a member of the Polyporaceae. The complete mitogenome sequence provides important data for further study of the Inonotus linteus complex.

Complete mitochondrial genome of the aluminum-tolerant fungus Rhodotorula taiwanensis RS1 and comparative analysis of Basidiomycota mitochondrial genomes.

Project description:The complete mitochondrial genome of Rhodotorula taiwanensis RS1, an aluminum-tolerant Basidiomycota fungus, was determined and compared with the known mitochondrial genomes of 12 Basidiomycota species. The mitochondrial genome of R. taiwanensis RS1 is a circular DNA molecule of 40,392 bp and encodes the typical 15 mitochondrial proteins, 23 tRNAs, and small and large rRNAs as well as 10 intronic open reading frames. These genes are apparently transcribed in two directions and do not show syntenies in gene order with other investigated Basidiomycota species. The average G+C content (41%) of the mitochondrial genome of R. taiwanensis RS1 is the highest among the Basidiomycota species. Two introns were detected in the sequence of the atp9 gene of R. taiwanensis RS1, but not in that of other Basidiomycota species. Rhodotorula taiwanensis is the first species of the genus Rhodotorula whose full mitochondrial genome has been sequenced; and the data presented here supply valuable information for understanding the evolution of fungal mitochondrial genomes and researching the mechanism of aluminum tolerance in microorganisms.
