The complete mitochondrial genome of oil palm pollinating weevil, Elaeidobius kamerunicus Faust. (Coleoptera : Curculionidae).

ABSTRACT: Elaeidobius kamerunicus is the most important insect pollinator in oil palm plantations. In this study, the mitochondrial genome (mitogenome) of E. kamerunicus (17.729 bp), a member of the Curculionidae family, will be reported. The mitogenome consisted of 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), 2 ribosomal RNA genes (rRNAs), and a putative control region (CR). Phylogenetic analysis based on 13 protein-coding genes (PCGs) using maximum Likelihood (ML) methods indicated that E. kamerunicus belongs to the Curculionidae family. This mitochondrial genome provides essential information for understanding genetic populations, phylogenetics, molecular evolution, and other biological applications in this species.

Project description:Elaeidobius kamerunicus Faust. (Coleoptera: Curculionidae) is an essential insect pollinator in oil palm plantations. Recently, researches have been undertaken to improve pollination efficiency using this species. A fundamental understanding of the genes related to this pollinator behavior is necessary to achieve this goal. Here, we present the draft genome sequence, annotation, and simple sequence repeat (SSR) marker data for this pollinator. In total, 34.97 Gb of sequence data from one male individual (monoisolate) were obtained using Illumina short-read platform NextSeq 500. The draft genome assembly was found to be 269.79 Mb and about 59.9% of completeness based on Benchmarking Universal Single-Copy Orthologs (BUSCO) assessment. Functional gene annotation predicted about 26.566 genes. Also, a total of 281.668 putative SSR markers were identified. This draft genome sequence is a valuable resource for understanding the population genetics, phylogenetics, dispersal patterns, and behavior of this species.

Project description:The olive weevil Dyscerus cribripennis (Coleoptera: Curculionidae) is an uncontrollable noxious insect to Olea europaea. The 15,977 bp complete mitochondrial genome of D. cribripennis contained 13 protein-coding genes (PCGs), 2 ribosomal RNA genes (rRNAs), 21 transfer RNA genes (tRNAs), and a control region (GenBank accession number MW023069). The trnI was not found in the D. cribripennis mitogenome. The phylogenetic analysis based on mitogenomes showed that D. cribripennis is closed related with Hylobitelus xiaoi.

The phylogenetic analysis based on mitogenomes showed that A. cribratus is the sister group of Molytinae.

The complete mitochondrial genome of the tea weevil, Myllocerinus aurolineatus (Coleoptera: Curculionidae).

Project description:The tea weevil, Myllocerinus aurolineatus (Voss), is a serious pest of tea plants. We have obtained and annotated the complete mitochondrial genome of M. aurolineatus (GenBank accession No. MH197100). The entire mt genome is 17,762 bp long with an A + T content of 75.45%. The mt genome of M. aurolineatus encodes all 37 genes that are typically found in animal mt genomes, consists of 13 protein-coding genes, 2 ribosomal RNA and 22 transfer RNA genes. The gene order is consistent with other weevil mt genomes in Entiminae, within a typical gene order of "RANSEF". Phylogenetic analysis was performed using 13 protein-coding genes among 18 weevils showed that M. aurolineatus is closely related to another Entiminae species, Sympiezomias velatus.

Project description:The complete mitochondrial genome (mitogenome) of Sitophilus zeamais was determined by an Illumina platform. This mitogenome was 18,531 bp in length, containing 13 protein coding genes, 2 rRNA genes, 22 tRNA genes that is typical among curculionoidea. Stop codon was incomplete for ND4 gene and ND5. The non-coding intergenic regions have increased in size to 1033 bp due to expansion of tandem repeat arrays. Phylogenetic analysis on mitogenome of S. zeamais would further contribute to resolve phylogenetic position and interrelationships of Sitophilus.

The complete mitochondrial genome of a walnut weevil, Alcidodes juglans Chao (Coleoptera: Curculionidae).

Project description:The walnut weevil, Alcidodes juglans Chao (Coleoptera: Curculionidae), is an important agricultural pest and distributed widely in China. The complete mitochondrial genome of A. juglans is 15,638 bp long, and consists of 13 protein-coding genes (PCGs), two ribosomal RNA genes, 21 transfer RNA (tRNA) genes and a putative control region (GenBank accession No. MH819192). The trnI gene has not been observed in the A. juglans mitogenome. The nucleotide composition is significantly biased (A, G, C, and T was 38.35%, 10.02%, 14.96%, and 36.67%, respectively) with A + T contents of 75.02%. All of the 21 tRNAs have the typical cloverleaf structure, with an exception for trnS1 (AGN). All PCGs are initiated by ATN codons, except for cox1 with AAT instead. Ten PCGs use a common stop codon of TAA or TAG, whereas the remaining three were terminated with a single T. The phylogenetic relationships based on neighbour-joining method showed that A. juglans is closely related to Naupactus xanthographus, which is in accordance with the traditional morphological classification.

Tracking outbreak populations of the pepper weevil Anthonomus eugenii (Coleoptera; Curculionidae) using complete mitochondrial genomes.

Project description:The pepper weevil, Anthonomus eugenii, is a major pest on Capsicum species. Apart from natural spread, there is a risk of spread via international pepper trade. In the Netherlands, a pepper weevil outbreak occurred in 2012 and affected six greenhouses producing different sweet pepper varieties. The following year, a pepper weevil outbreak occurred in Italy. To trace the origin of the Dutch outbreak and to establish if the Dutch and Italian outbreaks were linked, we determined the mitogenomes of A. eugenii specimens collected at outbreak locations, and compared these with specimens from the native area, and other areas where the pest was introduced either by natural dispersal or via trade. The circular 17,257 bp A. eugenii mitogenome comprises thirteen mitochondrial genes typically found in insect species. Intra-species variation of these mitochondrial genes revealed four main mitochondrial lineages encompassing 41 haplotypes. The highest diversity was observed for specimens from its presumed native area (i.e. Mexico). The Dutch outbreak specimens represented three highly similar haplotypes, suggesting a single introduction of the pest. The major Dutch haplotype was also found in two specimens from New Jersey. As the Netherlands does not have pepper trade with New Jersey, it is likely that the specimens sampled in New Jersey and those sampled in the Netherlands originate from a shared source that was not included in this study. In addition, our analysis shows that the Italian and Dutch outbreaks were not linked. The mitochondrial genome is a useful tool to trace outbreak populations and the methodology presented in this paper could prove valuable for other invasive pest species, such as the African fruit moth Thaumatotibia leucotreta and emerald ash borer Agrilus planipennis.

Project description:The complete mitochondrial genome of the maize weevil Sitophilus zeamais (GenBank accession number: MN905575) from Guizhou Province consists of a circular DNA molecule of 18,421 bp (with 75.69% A + T content), which is longer by 316 bp than that of the mitogenome of S. zeamais from Nigeria. The mitogenome comprises 13 protein-coding, 22 tRNA, and two rDNA genes. The protein-coding genes have typical ATN (Met) initiation codons and are terminated by typical TAN stop codons.

Project description:Niphades castanea Chao is an important insect pest on many plants which belong to genus Castanea. The complete mitochondrial genome of N. castanea was sequenced and analyzed. The phylogenetic relationships between N. castanea and other 41 species in the family Curculionidae were reconstructed using maximum likelihood (ML) methods based on the concatenated nucleotide sequences, the phylogenetic analysis showed that N. castanea is closely related to Hylobitelus xiaoi, which is in accordance with the traditional morphological classification.

The complete mitochondrial genome of Sympiezomias velatus (Coleoptera: Curculionidae).

Project description:Here, we determined the complete mitogenome sequence of Sympiezomias velatus (Coleoptera: Curculionidae: Entiminae). This mitogenome was 15,592 bp long with an A + T content of 74.1% and contains 13 protein-coding genes (PCGs), 21 transfer RNA genes (tRNAs), 2 ribosomal RNA unit genes and a large non-coding region (putative control region). The trnI gene was not found in the S. velatus mitogenome. The order and orientation of the mitochondrial genes were identical to the inferred ancestral arrangement of insects except for trnR which was changed to be adjacent the nad3 gene. All tRNAs had the typical cloverleaf structure, except for trnS1 which lacked the dihydrouridine arm. The Bayesian phylogenetic tree of 10 Entiminae species based on the concatenated nucleotide sequences of 13 PCGs strong supported a sister relationship of S. velatus and Barynotus obscures.
