The complete mitochondrial genome of Agelena silvatica (Araneae: Agelenidae).

ABSTRACT: The complete mitochondrial genome sequence of a funnel-web weaving spider, Agelena silvatica (Agelenidae), was obtained using long PCR and conventional PCR methods. The circular genome is 14,776?bp in length. It contains 13 protein-coding genes, 22 transfer RNAs, 2 ribosomal RNAs, and a control region. The A?+?T content (74.46%) of the all base composition is higher than G?+?C content (25.54%). Bayesian and maximum likelihood phylogenetic analyses demonstrate that Agelena silvatica is the sister clade of the water spider, Argyroneta aquatica.

The length of 13 PCGs is 10,782 bp in total, encoding 3638 amino acids. The phylogenetic analyses upon mitogenomes proves its relatively basal position within Lycosidae. It would provide further evolutionary research data for the wolf spider family Lycosidae.

Project description:Trichonephila clavipes (Linnaeus, 1767) is known as a golden silk orb-weaver and belongs to the family Araneidae. T. clavipes is one of the few spider species whose genome has been reported and model organism for a molecular biology. Here, we present the complete mitochondrial genome sequence (mtDNA) of T. clavipes. The sequence was obtained using a long-read Nanopore technology and corrected with an Illumina technology. The circular genome is 14,902 bp in length, and the AT content was 77.21%. The T. clavipes mitochondrial genome contains 13 protein-coding genes (PCGs), 22 tRNA genes, and 2 rRNA genes. The majority of PCGs were found on the heavy strain.

The complete mitochondrial genome sequence of Desis martensi (Araneae: Desidae).

Project description:The complete mitochondrial genome sequence of Desis martensi (L. Koch, 1872) was reported. In this study, we sequenced, assembled and annotated the mitochondrial genome of Desis martensi using next-generation sequencing (NGS). The sequence was 14,662 base pairs (bp) in length and consisted of 37 mitochondrial genes (13 protein-coding genes, 22 transfer RNAs, two ribosomal RNA genes). The overall base composition of the genome showed slightly A + T bias, AT content (77.2%) higher than GC content (22.9%). The phylogenetic analyses based on 13 protein-coding genes indicated that the family Desidae belonged to the Retrolateral Tibial Apophysis (RTA) clade in Araneae.

First Cytogenetic Analysis of Eratigena agrestris (Araneae: Agelenidae) From Turkey.

Project description:In this study, chromosomal characteristics of Eratigena agrestris (Agelenidae) were investigated for the first time. Karyotype features including diploid chromosome number, sex chromosome system, chromosome morphology, and meiotic behavior were obtained from specimens collected in two localities of Mediterranean region. A spreading method including dissection, hypotonization, fixation, and staining was used to prepare the chromosome slides. In a total, 10 adult males were used due to having high numbers of dividing cells. Cytogenetical results showed that the diploid chromosome number and sex chromosome system was 2n? = 42 (X1X20). The sex chromosomes were identified tentatively. All chromosomes were telocentric. Relative chromosome lengths of autosomal pairs ranged between 5.65 and 3.32%, and relative chromosome lengths of X1 and X2 were 5.33 and 4.19%, respectively. In the first meiotic division stages, bivalents usually had one chiasma, but some had two chiasmata. At the end of the meiosis, two kinds of nuclei, with or without sex chromosomes, have occurred. These results contribute to a better characterization of the Agelenidae cytogenetic.

Complete mitochondrial genome of Pisaura bicornis (Araneae: Pisauridae) and its phylogenetic implications.

Project description:The complete mitochondrial genome of Pisaura bicornis (GenBank accession number MN296112) is 15,281?bp in length and contains 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), 2 ribosomal RNA genes, and a putative control region. The overall base composition of this genome is A (35.65%), C (8.12%), G (13.47%) and T (42.76%), demonstrating an obvious bias of high AT content (73.2%). ATT, ATA, TTG were initiation codons and TAA, TAG, and T were termination codons. Ten tRNAs (trnD, trnF, trnG, trnH, trnK, trnP, trnR, trnT, trnY, and trnE) lacked the T?C arm stem, while three tRNAs (trnA, trnS1 , and trnS2 ) lost the dihydrouracil (DHU) arm. Project description:The complete mitochondrial genome of Argiope ocula (GenBank accession number MN331657) is a circular molecule of 14,079 bp in length, comprising of 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), two ribosomal RNA genes, and a control region. Ten intergenic spacer regions and 19 reading frame overlaps were found in the mitogenome of A. ocula. Ten tRNAs (trnD, trnF, trnK, trnR, trnT, trnW, trnE, trnG, trnL1 , and trnM) lacked the TΨC arm stem, whereas three tRNAs (trnA, trnS1 , and trnS2 ) lost the dihydrouracil (DHU) arm. Phylogenetic analysis suggested that A. ocula had the closest evolutionary relationship with A. perforata.

Seven tRNAs (trnY, trnC, trnG, trnN, trnH, trnP, and trnV) lacked the TΨC arm stem, while two tRNAs (trnS1 and trnS2 ) lost the dihydrouracil (DHU) arm. Phylogenetic analysis based on 13 PCGs indicated that P. cavaleriei was closely related to Cheliceroides longipalpis, and clustered within Salticidae clade.

The complete mitochondrial genome of Atypus karschi (Araneae, Atypidae) with phylogenetic consideration.

Project description:The complete mitochondrial genome sequence of Atypus karschi has a circular genome of 14,149 bp, comprised of 13 protein-coding genes, two rRNA genes, 22 tRNA genes, and a control region. The nucleotide composition is 35.82% of T, 35.13% of A, 17.19% of G, and 9.16% of C. Most genes are encoded on the heavy strand except seven tRNA genes (Leu, Phe, His, Pro, Leu, Ile, Gln), four protein-coding genes (nad5, nad4, nad4l, nad1), and 16S-rRNA on the light strand. Most protein-coding genes start with TTG, ATT or ATA initiation codon except cox1, cox1's start codon cannot be determined, and three types of inferred termination codons are TAA, TAG, and an incomplete stop codon. There are four intergenic spacers and 25 gene overlaps. The phylogenetic analysis shows that A. karschi has closer genetic relationship with Cyriopagopus schmidti (von Wirth, 1991) and Phyxioschema suthepium (Raven & Schwendinger, 1898) with high bootstrap support.

The Complete Mitochondrial Genome of two Tetragnatha Spiders (Araneae: Tetragnathidae): Severe Truncation of tRNAs and Novel Gene Rearrangements in Araneae.

Project description:Mitogenomes can provide information for phylogenetic analysis and evolutionary biology. The Araneae is one of the largest orders of Arachnida with great economic importance. In order to develop mitogenome data for this significant group, we determined the complete mitogenomes of two long jawed spiders Tetragnatha maxillosa and T. nitens and performed the comparative analysis with previously published spider mitogenomes. The circular mitogenomes are 14578 bp long with A+T content of 74.5% in T. maxillosa and 14639 bp long with A+T content of 74.3% in T. nitens, respectively. Both the mitogenomes contain a standard set of 37 genes and an A+T-rich region with the same gene orientation as the other spider mitogenomes, with the exception of the different gene order by the rearrangement of two tRNAs (trnW and trnG). Most of the tRNAs lose T?C arm stems and have unpaired amino acid acceptor arms. As interesting features, both trnS(AGN) and trnS(UCN) lack the dihydrouracil (DHU) arm and long tandem repeat units are presented in the A+T-rich region of both the spider mitogenomes. The phylogenetic relationships of 23 spider mitogenomes based on the concatenated nucleotides sequences of 13 protein-coding genes indicated that the mitogenome sequences could be useful in resolving higher-level relationship of Araneae. The molecular information acquired from the results of this study should be very useful for future researches on mitogenomic evolution and genetic diversities in spiders.
