The complete mitochondrial genome of Kuhl's pipistrelle, Pipistrellus kuhlii (Chiroptera: Vespertilionidae).

ABSTRACT: The Kuhl's pipistrelle (Pipistrellus kuhlii) is a small, vespertilionid bat species, with a large range extending from the Iberian Peninsula into the Near East and the Arabian Peninsula. In this study, we determine for the first time the complete mitogenome of this species. The P. kuhlii mitogenome is 16,991 base pairs long with 37 genes and 1 control region, showing conserved gene content and order with other vertebrate mitogenomes. The length of the 22 tRNA genes ranges between 60?bp (tRNA-Ser) and 75?bp (tRNA-Leu). The D-loop region is 1553?bp long with low CG content (39.8%).

Unusual Pipistrelle: Taxonomic Position of the Malayan Noctule (Pipistrellus stenopterus; Vespertilionidae; Chiroptera).

Project description:Sergei V. Kruskop, Evgeniya N. Solovyeva, and Anna D. Kaznadzey (2018) Despite huge progress in the systematics of bats and, in particular, of the Vespertilionidae family in latest years, the taxonomic position of a number of remarkable bat species has been uncertain until now, partly because of limits in acceptable comparative material. Researchers have previously placed the Malayan noctule, Pipistrellus stenopterus, into Nyctalus, because of similar body shape and proportions, or into Pipistrellus, based on karyological analysis. This study reassesses Pipistrellus stenopterus using available collection material and compares it to various members of Nyctalus and Pipistrellus, as well as with some other related and similar genera, based on respective morphological and molecular genetic features. This species demonstrates vast morphological peculiarities compared to other Pipistrellus-like bats. Nonetheless, both mitochondrial and nuclear genetic markers unequivocally place it close to other Asian pipistrelles, most probably in a sister position to the "javanicus" species group. We propose establishing P. stenopterus as a separate subgenus, Alionoctula. Our results also confirm that Pipistrellus is paraphyletic in its current state, and we suggest that further studies explore its internal taxonomy and limits.

Rickettsiae in the common pipistrelle Pipistrellus pipistrellus (Chiroptera: Vespertilionidae) and the bat soft tick Argas vespertilionis (Ixodida: Argasidae).

Project description:BACKGROUND:Increasing molecular evidence supports that bats and/or their ectoparasites may harbor vector-borne bacteria, such as bartonellae and borreliae. However, the simultaneous occurrence of rickettsiae in bats and bat ticks has been poorly studied. METHODS:In this study, 54 bat carcasses and their infesting soft ticks (n = 67) were collected in Shihezi City, northwestern China. The heart, liver, spleen, lung, kidney, small intestine and large intestine of bats were dissected, followed by DNA extraction. Soft ticks were identified both morphologically and molecularly. All samples were examined for the presence of rickettsiae by amplifying four genetic markers (17-kDa, gltA, ompA and ompB). RESULTS:All bats were identified as Pipistrellus pipistrellus, and their ticks as Argas vespertilionis. Molecular analyses showed that DNA of Rickettsia parkeri, R. lusitaniae, R. slovaca and R. raoultii was present in bat organs/tissues. In addition, nine of the 67 bat soft ticks (13.43%) were positive for R. raoultii (n = 5) and R. rickettsii (n = 4). In the phylogenetic analysis, these bat-associated rickettsiae clustered together with conspecific sequences reported from other host and tick species, confirming the above results. CONCLUSIONS:To the best of our knowledge, DNA of R. parkeri, R. slovaca and R. raoultii was detected for the first time in bat organs/tissues. This is also the first molecular evidence for the presence of R. raoultii and R. rickettsii in bat ticks. To our knowledge, R. parkeri was not known to occur in Asia. Our results highlight the need to assess rickettsial agents in a broader range of bat species and associated tick species.

In the control region of R. yunnanensis, the sequence of 5'-CAACGTATACACG-3' repeats 18 times. Phylogenetic analyses indicate that R. yunnanensis is a sister clade to ((Rhinolophus sinicus sinicus?+?R. sinicus sinicus)?+?(R. macrotis?+?(R. pumilus?+?R. monoceros))).

The complete mitochondrial genome of Tylonycteris fulvida (Chiroptera: Vespertilionidae) in South China

Project description:Abstract The complete mitochondrial genome of Tylonycteris fulvida (Peters, 1872) was obtained using high-throughput sequencing technology. The genome is a circular molecule of 16,621 bp length, containing 13 protein-coding genes, 2 rRNA genes, 22 tRNA genes, and a control region. A phylogenetic tree of 13 protein-coding genes was constructed using IQ-TREE. Our result suggests that T. fulvida cluster within Chiroptera and Fereuungulata. The complete mitochondrial genome sequence of T. fulvida will be helpful for future taxonomic and phylogenetic studies on Chiroptera.

The complete mitochondrial genome of the Bechstein's bat, Myotis bechsteinii (Chiroptera, Vespertilionidae).

Project description:In this study, we present the complete mitochondrial genome of the Bechstein's bat, Myotis bechsteinii. The mitogenome is 17,151 bp in length and is AT-rich with base composition A (27.8%), C (22%), G (16.1%), and T (34.1%). The mitogenome shows conserved gene content and order similar with other mammalian mitogenomes, being composed of 13 protein-coding genes, 2 ribosomal RNA genes, 22 transfer RNA genes, and one control region. The majority of genes are encoded on the H-Strand except for ND6 and 8 tRNAs as found in other bat species. The field identification of Myotis bechsteinii was confirmed by phylogenetic analyses using datasets comprising whole mitogenomes and COXI. This mitogenome is a resource for future studies of Myotis bats and other mammals.

Complete mitochondrial genome of the East Asian fish-eating bat: Myotis ricketti (Chiroptera, Vespertilionidae).

Project description:I report the complete mitochondrial genome (mitogenome) of the East Asian fish-eating bat (Myotis ricketti or M. pilosus), also known as the Rickett's big footed bat. The total length of the circular M. ricketti mitogenome is 17,098 base pairs, containing 13 protein-coding genes (PCGs), two ribosomal RNAs, 22 transfer RNAs, and a non-coding control region (D-loop region). The gene order and organisation of this mitogenome are similar to most of other determined vertebrate mitogenomes, with the nucleotide base composition of A 34.22%, T 30.32%, C 22.80%, and G 12.66%. Besides, the mitogenomic D-loop region contains 29 copies of a tandem repeat sequence of six nucleotides (CATACG). The phylogenetic analysis indicates that M. ricketti is closely related to M. macrodactylus and M. petax. This study will contribute to the investigations of phylogeny and evolution for Myotis and its relevant taxa.

Mitochondrial genome of the Harpiocephalus harpia (Chiroptera: Vespertilionidae) from China.

Project description:In this study, a complete mitochondrial genome of a female Harpiocephalus harpia from Zhejiang Province, China, was sequenced using Illumina Hiseq. The genome is ∼16,400 bp in length, containing 13 protein-coding genes, 2 ribosomal RNA genes, 22 transfer RNA genes, and a control region. Most of the genes were encoded on the H-strand, except for 8 tRNA and ND6 genes. Phylogenetic tree using maximum likelihood suggests H. harpia is sister taxon to Murina. The complete mitochondrial genome herein provides basic information for future taxonomic and phylogenetic studies.

The mitochondrial genome of Murina huttoni rubella (Chiroptera: Vespertilionidae) from China.

Project description:Murina huttoni rubella is a common Murina species in China, with a medium forearm length and reddish brown hairs. In this study, based on a male M. h. rubella individual from Jiangxi, China, its complete mitochondrial genome was sequenced and analyzed. The genome is 16,707?bp in length, including 22 tRNA genes, two rRNA genes, 13 protein-coding genes and a control region. The composition and arrangement of genes are similar to other bats. Phylogenetic trees that covered all released complete mitochondrial genome of bats were constructed using Bayesian Inference and maximum likelihood methods. Both phylogenetic results showed that M. h. rubella and M. ussuriensis have closer phylogenetic relationship. The complete mtDNA genome sequence of M. h. rubella would provide valuable information for solving taxonomic and phylogenetic problem in future.

A new genus of vespertilionid bat: the end of a long journey for Joffre's Pipistrelle (Chiroptera: Vespertilionidae).

Project description:Knowledge as to the taxonomic status of enigmatic bat species often is hindered by limited availability of specimens. This is particularly true for aerial-hawking bats that are difficult to catch. One such species, "Hypsugo" joffrei, was originally described in Nyctalus due to its long and slender wings, but subsequently transferred to Pipistrellus, and most recently to Hypsugo, on the basis of morphology. Analysis of newly available material, which more than doubles the known specimens of this taxon, demonstrates that it is morphologically and genetically distinct from all other bat genera. We accordingly describe it as belonging to a new, monotypic genus. We provide a detailed description of its external and craniodental traits, measurements, and assessment of genetic relationships, including barcode sequences to facilitate its rapid identification in future. The new genus belongs to a group that includes the recently described Cassistrellus, as well as Tylonycteris, and its closest relative, Philetor. We also describe the echolocation calls emitted by members of the taxon in different situations, which may facilitate finding them in previously unsampled locations. Based on the new data, the species occurs from Nepal to North Vietnam and China, which suggests that it could be more widespread than previously thought.

The complete mitochondrial genome of the far Eastern myotis: Myotis bombinus Thomas, 1906 in mainland of Korea (Chiroptera, Vespertilionidae).

Project description:We have determined the second mitochondrial genome of Myotis bombinus Thomas, 1906 in mainland of Korea. The circular mitogenome of M. bombinus is 17,035 bp long which is slightly shorter than that of the previous mitogenome of M. bombinus. It includes 13 protein-coding genes (PCGs), two ribosomal RNA genes, and 22 transfer RNAs. The base composition was AT-biased (66.1%). Fifty single nucleotide polymorphisms and 14 insertions were identified between two mitogenomes of M. bombinus. Phylogenetic trees show that both M. bominus mitogenomes are clustered in one clade.
