Analysis of the nearly complete mitochondrial genome of Paederus fuscipes (Coleoptera: Staphylinidae).

ABSTRACT: The nearly complete mitochondrial genome of Paederus fuscipes (GenBank accession no. MG581161) is 17,644?bp in size, containing 13 protein-coding genes, 22 transfer RNAs, two ribosomal RNAs, and a partial control region. The gene order is similar to the typical insect mitochondrial genome. Maximum likelihood tree recovered the monophyly of Staphylininae, Pselaphinae, Paederinae and Aleocharinae. Additionally, Staphylininae is a sister group to Paederinae.

Phylogeny and Density Dynamics of Wolbachia Infection of the Health Pest Paederus fuscipes Curtis (Coleoptera: Staphylinidae).

Project description:The maternally inherited obligate intracellular bacteria Wolbachia infects the reproductive tissues of a wide range of arthropods and affects host reproduction. Wolbachia is a credible biocontrol agent for reducing the impact of diseases associated with arthropod vectors. Paederus fuscipes is a small staphylinid beetle that causes dermatitis linearis and conjunctivitis in humans when they come into contact with skin. Wolbachia occur in this beetle, but their relatedness to other Wolbachia, their infection dynamics, and their potential host effects remain unknown. In this study, we report the phylogenetic position and density dynamics of Wolbachia in P. fuscipes. The phylogeny of Wolbachia based on an analysis of MLST genotyping showed that the bacteria from P. fuscipes belong to supergroup B. Quantitative PCR indicated that the infection density in adults was higher than in any other life stage (egg, larva or pupa), and that reproductive tissue in adults had the highest infection densities, with similar densities in the sexes. These findings provide a starting point for understanding the Wolbachia infection dynamics in P. fuscipes, and interactions with other components of the microbiota.

The complete mitochondrial genome of Aleochara postica Walker, 1858 (Coleoptera: Staphylinidae).

Project description:The complete mitochondrial genome of Aleochara postica Walker, 1858 was determined in this study. It is 15,473 bps in length, containing 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, and a 778 bp A + T-rich control region. Most PCGs use the conventional ATN start codon, except for cox1 and nad1. Two genes (cox1 and cox3) use single T residue as stop codon rather than the routinely used TAA or TAG. All tRNAs, except for TrnS1, exhibit the cloverleaf secondary structure. ML phylogenetic analysis using 13 PCGs of 52 beetle species indicated that A. postica was clustered with other members of the subfamily Aleocharinae as conventional taxonomy predicted.

Discrimination of Paederus fuscipes and Paederus littoralis by mtDNA-COI PCR-RFLP.

Project description:BACKGROUND:Linear dermatitis is endemic in Iran where most cases occur in the Caspian Sea coast and Fars province. The disease is caused by beetles of the genus Paederus which are active from early spring to beginning of autumn although its incidence rises from May to August. The classic taxonomy of Paederus spp. is based on the male genitalia that is very complex and needs expertise. In this study, we report a DNA-based method to discriminate Paederus fuscipes and Paederus littoralis (=syn: P. lenkoranus, P. ilsae). METHODS:Type specimens were collected from north and south of Iran. Molecular typing of the species was performed using restriction fragment length polymorphism (RFLP) analysis of polymerase chain reaction (PCR)-amplified fragments of mtDNA-COI. RESULTS:Sequence analyses of the data obtained in this study showed significant DNA polymorphisms. There were 89 substitutions between COI sequences of the two species. The mtDNA-COI fragment comprises several useful species-specific restriction sites comprising HaeIII that could result in distinctively different species-specific PCR-RFLP profiles. The HaeIII enzyme cuts the 872 bp PCR amplicon of P. littoralis into 737 and 100 bp and two small nonvisible bands whereas it does not cut P. fuscipes amplicon into fragments. CONCLUSION:This study demonstrates that molecular typing is useful method and allows one to differentiate between two species and is recommended for discrimination of other Paederus species, which morphologically are indistinguishable or very difficult to be distinguished.

The complete mitochondrial genome of a rove beetle, Megalinus hunanensis Bordoni, 2013 (Coleoptera: Staphylinidae).

Project description:The complete mitogenome of rove beetle Megalinus hunanensis Bordoni, 2013 was determined in this study. It was 17,314 bp in length, and comprised of 13 protein-coding genes, 22 tRNA genes, two rRNA genes, and a 2511 bp A + T-rich control region. Most PCGs used the typical ATN start codon, except for nad1. Five genes (cox1, cox2, cox3, nad4, and nad5) used a single T residue as stop codon rather than the commonly used TAA or TAG. All tRNAs could be folded into the cloverleaf secondary structure. Bayesian inference phylogenetic trees built on 13 PCGs and their translated amino acid sequences from 29 rove beetle taxa showed that Staphylininae was paraphyletic with respect to Paederinae; M. hunanensis was a member of the tribe Xantholinini.
